Ginsenoside Rg3 increases gemcitabine sensitivity of pancreatic adenocarcinoma via reducing ZFP91 mediated TSPYL2 destabilization

  • Background: Ginsenoside Rg3 and gemcitabine have mutual enhancing antitumor effects. However, the underlying mechanisms are not clear. This study explored the influence of ginsenoside Rg3 on Zinc finger protein 91 homolog (ZFP91) expression in pancreatic adenocarcinoma (PAAD) and their regulatory mechanisms on gemcitabine sensitivity. Methods: RNA-seq and survival data from The Cancer Genome Atlas (TCGA)-PAAD and Genotype-Tissue Expression (GTEx) were used for in-silicon analysis. PANC-1, BxPC-3, and PANC-1 gemcitabine-resistant (PANC-1/GR) cells were used for in vitro analysis. PANC-1 derived tumor xenograft nude mice model was used to assess the influence of ginsenoside Rg3 and ZFP91 on tumor growth in vivo. Results: Ginsenoside Rg3 reduced ZFP91 expression in PAAD cells in a dose-dependent manner. ZFP91 upregulation was associated with significantly shorter survival of patients with PAAD. ZFP91 overexpression induced gemcitabine resistance, which was partly conquered by ginsenoside Rg3 treatment. ZFP91 depletion sensitized PANC-1/GR cells to gemcitabine treatment. ZFP91 interacted with Testis-Specific Y-Encoded-Like Protein 2 (TSPYL2), induced its poly-ubiquitination, and promoted proteasomal degradation. Ginsenoside Rg3 treatment weakened ZFP91-induced TSPYL2 poly-ubiquitination and degradation. Enforced TSPYL2 expression increased gemcitabine sensitivity of PAAD cells and partly reversed induced gemcitabine resistance in PANC-1/GR cells. Conclusion: Ginsenoside Rg3 can increase gemcitabine sensitivity of pancreatic adenocarcinoma at least via reducing ZFP91 mediated TSPYL2 destabilization.

Protective Effect of Fermented Red Ginseng on a Transient Focal Ischemic Rats

  • Red ginseng and fermented red ginseng were prepared, and their composition of ginsenosides and antiischemic effect were investigated. When ginseng was steamed at 98-$100{\circ}C$ for 4h and dried for 5h at $60{\circ}C$, and extracted with alcohol, its main components were ginsenoside $Rg_3$ > ginsenoside $Rg_1$> ginsenoside $Rg_2$. When the ginseng was suspended in water and fermented for 5 days by previously cultured Bifidobacterium H-1 and freeze-dried (fermented red ginseng), its main components were compound K > ginsenoside $Rg_3{\geq}$ ginsenoside $Rg_2$. Orally administered red ginseng extract did not protect ischemia-reperfusion brain injury. However, fermented red ginseng significantly protected ischemica-reperfusion brain injury. These results suggest that ginsenoside Rh2 and compound K, which was found to be at a higher content in fermented red ginseng than red ginseng, may improve ischemic brain injury.

  • 본 논문에서는 KSTAR 토카막 진공용기의 세정을 위한 방전세정 시스템의 설계에 대해 기술하였다. 먼저 방전세정효율에 영향을 주는 매개변수들에 대해 논의하였으며. 실제 설계에서 직류방전 보다 낮은 압력에서 방전이 시작되고 유지되며 따라서 세정효율이 높은 RG 방전(RF-assisted DC glow discharge) 방법을 채택하였다. 그리고 방전세정의 균일성을 위해 두 개의 방전 전극을 진공용기의 진단포트(A,I-port)에 설치하였다. 설계된 방전세정 시스템은 KSTAR 진공용기 내벽 세정뿐만 아니라 제1벽의 연료재순환(fuel recycling)이나 보론화 처리(boronization)등의 연구에도 응용될 수 있다.

Improved antimicrobial effect of ginseng extract by heat transformation

  • Background: The incidence of halitosis has a prevalence of 22-50% throughout the world and is generally caused by anaerobic oral microorganisms, such as Fusobacterium nucleatum, Clostridium perfringens, and Porphyromonas gingivalis. Previous investigations on the structure-activity relationships of ginsenosides have led to contrasting results. Particularly, the antibacterial activity of less polar ginsenosides against halitosis-related bacteria has not been reported. Methods: Crude saponins extracted from the Panax quinquefolius leaf-stem (AGS) were treated at $130^{\circ}C$ for 3 h to obtain heat-transformed saponins (HTS). Five ginsenoside-enriched fractions (HTS-1, HTS-2, HTS-3, HTS-4, and HTS-5) and less polar ginsenosides were separated by HP-20 resin absorption and HPLC, and the antimicrobial activity and mechanism were investigated. Results: HPLC with diode-array detection analysis revealed that heat treatment induced an extensive conversion of polar ginsenosides (-Rg1/Re, -Rc, -Rb2, and -Rd) to less polar compounds (-Rg2, -Rg3, -Rg6, -F4, -Rg5, and -Rk1). The antimicrobial assays showed that HTS, HTS-3, and HTS-4 were effective at inhibiting the growth of F. nucleatum, C. perfringens, and P. gingivalis. Ginsenosides-Rg5 showed the best antimicrobial activity against the three bacteria, with the lowest values of minimum inhibitory concentration and minimum bactericidal concentration. One major reason for this result is that less polar ginsenosides can more easily damage membrane integrity. Conclusion: The results indicated that the less polar ginsenoside-enriched fraction from heat transformation can be used as an antibacterial agent to control halitosis.

Stereoisomer-specific ginsenoside 20(S)-Rg3 reverses replicative senescence of human diploid fibroblasts via Akt-mTOR-Sirtuin signaling

  • Background: The replicative senescence of human dermal fibroblasts (HDFs) is accompanied by growth arrest. In our previous study, the treatment of senescent HDFs with Rg3(S) lowered the intrinsic reactive oxygen species (ROS) levels and reversed cellular senescence by inducing peroxiredoxin-3, an antioxidant enzyme. However, the signaling pathways involved in Rg3(S)-induced senescence reversal in HDFs and the relatedness of the stereoisomer Rg3(R) in corresponding signaling pathways are not known yet. Methods: We performed senescence-associated β-galactosidase and cell cycle assays in Rg3(S)-treated senescent HDFs. The levels of ROS, adenosine triphosphate (ATP), and cyclic adenosine monophosphate (cAMP) as well as the mitochondrial DNA copy number, nicotinamide adenine dinucleotide (NAD)+/1,4-dihydronicotinamide adenine dinucleotide (NADH) ratio, and NAD-dependent sirtuins expression were measured and compared among young, old, and Rg3(S)-pretreated old HDFs. Major signaling pathways of phosphatidylinositol 3-kinase/Akt, 5' adenosine monophosphate-activated protein kinase (AMPK), and sirtuin 1/3, including cell cycle regulatory proteins, were examined by immunoblot analysis. Results: Ginsenoside Rg3(S) reversed the replicative senescence of HDFs by restoring the ATP level and NAD+/NADH ratio in downregulated senescent HDFs. Rg3(S) recovered directly the cellular levels of ROS and the NAD+/NADH ratio in young HDFs inactivated by rotenone. Rg3(S) mainly downregulated phosphatidylinositol 3-kinase/Akt through the inhibition of mTOR by cell cycle regulators like p53/p21 in senescent HDFs, whereas Rg3(R) did not alter the corresponding signaling pathways. Rg3(S)-activated sirtuin 3/PGC1α to stimulate mitochondrial biogenesis. Conclusion: Cellular molecular analysis suggests that Rg3(S) specifically reverses the replicative senescence of HDFs by modulating Akt-mTOR-sirtuin signaling to promote the biogenesis of mitochondria.

아임계수 추출에 의한 홍삼 추출물의 진세노사이드 조성 및 이화학적 특성 (Physicochemical Properties and Composition of Ginsenosides in Red Ginseng Extract as Revealed by Subcritical Water Extraction)

  • 홍삼의 신규 추출 방법으로 아임계수 추출(subcritical water extraction, SWE)을 적용하여 홍삼 추출물의 이화화적 특성 및 진세노사이드 추출 특성을 조사함으로써 최적 아임계수 추출 조건을 결정하고 기존 추출 방법과 비교함으로써 산업적 이용 효율을 평가하였다. 아임계수 추출에 의한 홍삼 추출물의 당도, 고형분 함량, 색차 및 탁도는 추출 온도가 높아지고 추출 시간이 길어질수록 증가하였고 pH는 낮아졌으며, 조 사포닌은 $120^{\circ}C$, 20분에서 최대였다. TLC 및 HPLC 분석 결과, 총 및 극성 진세노사이드 농도는 $120^{\circ}C$, 20분에서 최대였고 홍삼 특유의 저극성 진세노사이드 Rg3, Rh1 등은 $150^{\circ}C$, 15분에서 최대 농도였다. 또한 최적 아임계수 추출과 열수, 에탄올 및 메탄올을 용매로 환류 냉각 추출하여 비교한 결과, 저극성 진세노사이드의 추출 이행률은 $150^{\circ}C$, 15분의 아임계수 추출에서 가장 높았고 특히 Rg3는 3.5-5배, Rh1은 2-2.5배의 높은 농도로 나타났다. 홍삼의 특이 사포닌으로 극미량 존재하는 Rg3 및 Rh1은 강력한 항암 효과 등이 보고되면서 최근 화학적, 물리적, 생물학적 방법에 의한 전환연구가 활발히 진행되고 있으나 낮은 선택성과 생산성의 저하, 부 반응으로 인한 환경 공해 및 대량 생산의 한계 등으로 산업적 적용에 제약이 존재하여 왔다. 본 연구에서는 아임계 상태에서의 물의 특성 변화를 이용한 아임계수 추출을 적용하여 홍삼으로부터 특히 저극성 진세노사이드의 선택적 추출 및 전환에 효과적인 대체 기술로서 가능성을 확인하였다.

벼 흰잎마름병 저항성 유전자(Xa-1, Xa-3)연관 RFLP 마커 탐색 (Mapping of RFLP Markers Linked to Bacterial Blight Resistant Genes (Xa-1, Xa-3) in Rice)

  • 우리나라 자포니카 품종의 흰잎마름병 저항성 유전자와 연관된 마커를 탐색하기 위하여, 밀양121호, 밀양123호 및 HB10624-AC5 등을 교배친으로 한 두 조합의 약배양 계통을 재료로 흰잎마름병 저항성 유전자(Xa-1 and Xa-3)와 DNA 마커간의 연관분석을 통하여 유전자 지도를 작성하고자 수행하였던 결과를 요약하면 다음과 같다. 1. $\textrm{K}_1$ 균주에 대한 흰잎마름병 저항성 검정결과, 밀양121호/HRl1650-1-4-2에서는 저항성과 감수성이 1:1로 분리하였으며, 밀양123호/HR10624-AC5 조합의 $\textrm{K}_1$$\textrm{K}_3$ 균주에 대한 검정결과는 각각 3:1과 1:1로 분리하여 이론치에 합당하였다. 2. 교배친에 대하여 DraI. HindIII, EcoRI, EcoRV, PstI등 5가지 제한효소에 대한 다형현상을 검정한 결과, RZ590, RG303, RZ536 등 3개의 마커가 다형현상을 나타내었다. 3. 흰잎마름병 포장저항성 검정결과와 RFLP 마커와의 연관분석 결과 Xa-1 유전자는 RZ590과 4번 염색체 상에서 3.1$\times$1.5 cM으로 연관되어 있었으며, Xa-3 유전자는 Rz536 및 RG303과 11번 염색체 상에서 각각 7.6$\times$2.3 및 16.0$\times$3.2 cM으로 연관되어 있었다. 4. 11번 염색체 상에서 Xa-3와 Rz536 및 RG303은 "Xa-3-RZ536-RG303" 순으로 위치하였다.순으로 위치하였다.

Ginsenosides Rk1 and Rg5 inhibit transforming growth factor-β1-induced epithelial-mesenchymal transition and suppress migration, invasion, anoikis resistance, and development of stem-like features in lung cancer

  • Background: Lung cancer has a high incidence worldwide, and most lung cancer-associated deaths are attributable to cancer metastasis. Although several medicinal properties of Panax ginseng Meyer have been reported, the effect of ginsenosides Rk1 and Rg5 on epithelial-mesenchymal transition (EMT) stimulated by transforming growth factor beta 1 (TGF-β1) and self-renewal in A549 cells is relatively unknown. Methods: We treated TGF-β1 or alternatively Rk1 and Rg5 in A549 cells. We used western blot analysis, real-time polymerase chain reaction (qPCR), wound healing assay, Matrigel invasion assay, and anoikis assays to determine the effect of Rk1 and Rg5 on TGF-mediated EMT in lung cancer cell. In addition, we performed tumorsphere formation assays and real-time PCR to evaluate the stem-like properties. Results: EMT is induced by TGF-β1 in A549 cells causing the development of cancer stem-like features. Expression of E-cadherin, an epithelial marker, decreased and an increase in vimentin expression was noted. Cell mobility, invasiveness, and anoikis resistance were enhanced with TGF-β1 treatment. In addition, the expression of stem cell markers, CD44, and CD133, was also increased. Treatment with Rk1 and Rg5 suppressed EMT by TGF-β1 and the development of stemness in a dose-dependent manner. Additionally, Rk1 and Rg5 markedly suppressed TGF-β1-induced metalloproteinase-2/9 (MMP2/9) activity, and activation of Smad2/3 and nuclear factor kappa B/extra-cellular signal regulated kinases (NF-kB/ERK) pathways in lung cancer cells. Conclusions: Rk1 and Rg5 regulate the EMT inducing TGF-β1 by suppressing the Smad and NF-κB/ERK pathways (non-Smad pathway).

OMC-2010 구성약재가 마우스의 비장세포 cytokine 생성에 미치는 영향 (Effects of OMC-2010 constituents on cytokine productions in mouse spleen cells)

  • Objective : We recently reported that OMC-2010 has an immuno-modulatory effects via inhibiting tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-5. However, we did not find out which constituents play an important role in immuno-modulatory effect of OMC-2010. Thus, this study was performed to estimate the effects of constituents of OMC-2010 on cytokine production in mouse spleen cells, then ultimately reach to find out effective constituents regulating splenic cytokine production. Methods : Mouse spleen cells were pre-treated with water and ethanol extract of constituents of OMC-2010 such as Rehmannia glutinosa (RG), Pinellia ternata (PT), Citrus unshiu Markovich (CUM), Glycyrrhiza uralensis (GU), Platycodon grandiflorum (PG), Schisandra chinensis (SC). After 1 h, the cells were stimulated with lipopolysaccharide (LPS, 1 ${\mu}g/ml$) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokine productions. Results : The water extract of RG extract significantly inhibited the LPS-induced inTNF-${\alpha}$ and IL-5 mRNA expressions, but the water extract of PT, CUM, GU, PG, and SC did not. The ethanol extract of RG, PT, and SC significantly inhibited the LPS-induced TNF-${\alpha}$, and IL-5 mRNA expressions, but the ethanol extract of CUM, GU, and PG did not. Conclusions : Theses results could suggest that the water extract of RG and the ethanol extract of RG, PT, and SC inhibited the expression of TNF-${\alpha}$ and IL-5, which means that the possible candidate of OMC-2010 water extract's action might be RG, and ethanol extract's action might be RG, PR, and SC.

인체 혈장 중 Ginsenoside Rg1의 정량을 위한 LC-MS/MS 분석법 검증 (Validation of LC-MS/MS method for determination of ginsenoside Rg1 in human plasma)

  • LC-MS/MS를 이용하여 인체 혈장 중 ginsenoside Rg1의 신속하고 정확한 분석법을 개발하고 이 분석법에 대한 검증을 수행하였다. 혈장 분석을 위하여 internal standard인 digoxin을 첨가한 후 acetone과 methanol용액 (80:20)으로 전처리하고, 그 상층액을 진공농축 한 후, LC-MS/MS로 분석하였다. 최적 크로마토그래피 분석은 0.1% formic acid 첨가된 water와 methanol을 이동상으로 하여 Agilent Eclipse XDB-C18 column ($4.6{\times}150mm$, $5{\mu}m$)을 이용하여 0.9 mL/min의 유속으로 gradient mode로 수행하였다. 혈장 중 ginsenoside Rg1의 표준 검량선은 1~500 ng/mL의 농도 범위에서 우수한 직선성($r^2=0.9995$)을 보였으며, 일내, 일간 정밀성은 변동계수 7.53% 이하, 정확성은 98.28% 이상이었다. 결과적으로 본 분석법은 ginsenoside Rg1의 약동학 연구에 적용되기에 충분한 감도와 특이성, 직선성, 정밀성 및 정확성을 가지고 있음을 확인하였다.
