조피볼락(Sebastes schlegeli)의 Retinol-Binding Protein의 유전자 발현에 미치는 4-Nonylphenol의 영향 (Effect of 4-Nonylphenol on the Gene Expression of Retinol-Binding Protein in the Rockfish, Sebastes schlegeli)

  • 레티놀 결합 단백질(retinol-binding protein, RBP)은 고등 척추동물에서 혈류를 통해 특이적으로 레티놀을 표적세포에 운반해 주는 중요한 역할을 한다. 우리나라의 연안에 서식하고 있으며 산업적으로 중요한 조피볼락(Sebastes schlegeli)을 대상으로 4-nonylphenol(NP)가 RBP mRNA 발현에 미치는 영향을 구명하기 위해 간으로부터 cDNA library를 제작하고 RBP 단편의 염기서열을 분석하였다. 분석된 RBP mRNA 서열로부터 아미노산 서열을 추정하여 다른 종의 RBP 아미노산 서열과 비교한 결과 Sparus aurata와는 80%, 무지개 송어(Oncorhynchus mykiss)와는 72%, 유럽 뱀장어(Anguilla anguilla)와는 78%의 높은 상동성을 보였다. 조피볼락에서 4-NP에 대한 RBP와 VTG mRNAs 발현에 미치는 영향을 northern blot 분석 방법에 의해 조사하였다. 4-NP를 10 mg/kg BW로 주사한 실험구에서는 암수 모두 VTG mRNA 발현에 영향을 주지 않았으나, RBP mRNA 발현은 수컷에서 48시간 후에 감소하였다. 4-NP를 25 mg/kg BW으로 주사한 실험구에서는 24시간 후에 암수 모두 VTG mRNA 발현이 증가하였으며, RBP mRNA 발현은 48시간 후에 암수 모두 감소하였다. 이들 결과로부터 조피볼락에서는 4-NP 등의 에스트로겐 유사물질이 RBP와 VTG 유전자 발현에 상반되는 효과를 유도함을 알 수 있었다.

중등도 유산소 운동이 비만아동의 adiponectin, RBP-4(retinol binding protein-4) 및 혈관 염증인자에 미치는 영향 (The Effect of Moderate Aerobic Exercise on Adiponectin, Retinol Binding Protein-4, and Vascular Inflammation Factors in Obese Children)

  • 본 연구는 비만아동을 대상으로 12주간 중강도 유산소 운동을 통해 신체조성, 아디포넥틴, RBP-4, 혈관염증인자에 미치는 영향을 규명하는데 목적이 있다. 대상자들은 두 집단으로, 운동군(n=15)은 12주간 중강도 유산소 운동을 실시하였고, 대조군(n=13)은 운동 프로그램에 참여하지 않았다. 운동군은 여유심박수 50%의 운동강도로 300 kcal에 도달할 때까지 트레드밀에서 런닝을 하였다. 연구결과는 다음과 같다 : 운동군이 통제군보다 체중, 체질량지수, 체지방률, 수축기 혈압과 이완기 혈압이 각각 유의하게 감소하는 것으로 나타났고(p<0.05), 최대산소섭취량은 운동군에서 높게 나타났다(p<0.05). 공복시 혈당, 인슐린과 인슐린저항성은 운동군에서 유의하게 감소하였고(p<0.05), 아디포넥틴은 12주 운동 후 운동군에서 유의하게 증가하였다(p<0.05). 또한, 혈중 RBP-4, CRP, IL-6 농도는 대조군보다 운동군에서 유의하게 감소하는 것으로 나타났다(p<0.05). 본 연구에서 12주간 규칙적인 유산소 운동 프로그램은 비만 아동들의 염증과 신체지수 개선에 긍정적인 역할을 하는 것으로 나타났다.

Characterization of the Prolactin Receptor 3 (PRLR3) and Retinol-Binding Protein 4 (RBP4) Genes in the Birth Weight and Early Growth of Berkshire Pigs

  • To investigate the influences of candidate genes on the birth weight and the early stages of life, genotyping of the prolactin receptor 3 (PRLR3) and retinol-binding protein 4 (RBP4) genes was performed in 156 and 141 Berkshire pigs, respectively. The frequency of both PRLR3 alleles A and a was 0.50. The frequencies of the RBP4 alleles B and b were 0.42 and 0.58, respectively. Neither locus was in Hardy-Weinberg equilibrium. No significant associations of the PRLR3 alleles with birth or weaning weights and of the RBP4 alleles with birth weight were observed. The proportions of the phenotype variances due to the genotypes of PRLR3 in the feeder weights was 4.0% and those of RBP4 in the weaning and feeder weights were 11.9 and 3.3%, respectively (P < 0.05). The dominance effect of PRLR3 and RBP4 on feeder weights was 2.40 and -1.86 kg, respectively (P < 0.01). The additive and dominance effects of RBP4 on weaning weights were 0.332 and -0.682 kg, respectively (P < 0.01). Even if no significant epistasis of PRLR3 and RBP4 was detected, a considerable trend of consistent positive epistasis estimates of AA/BB and Aa/Bb was observed for all traits. The results of this study may have a considerable impact on early-stage growth by both loci, and a selection strategy should be designed separately for each marker in Berkshire pigs.

Medullary Thyroid Carcinoma - Circulating Status of Vaspin and Retinol Binding Protein-4 in Iranian Patients

  • Background: Vaspin and Retinol binding protein-4 (RBP4) are new adipokines mainly produced by adipose tissue. Considering that medullary thyroid carcinoma (MTC) is a malignant neuroendocrine tumor, and to date the relationship between serum levels of vaspin and RBP4 with MTC has not been studied, in this matched case-control study we evaluated their possible significance to this tumor type. Materials and Methods: A total of 45 patients with MTC (21 males and 24 females) and 45 healthy persons as a control group (24 males and 21 females) were selected. The two groups were matched for age, sex and body mass index. Serum Vaspin and RBP4 levels were measured by enzyme-linked immunosorbent assay (ELISA) methods in both groups. Also, weight and height were measured and body mass index was calculated too. Results: In total, patients with MTC had significantly higher serum vaspin levels compared to the controls (0.52ng/ml vs. 0.45ng/ml, P=0.0241). However, no significant difference was found in serum RBP4 concentrations between the patients with MTC and the controls ($15.2{\pm}2.55{\mu}g/ml$ versus $15.1{\pm}3.34{\mu}g/ml$, p>0.05). Conclusions: The results of this study demonstrated that serum RBP4 levels in MTC patients are not significantly different from those found in healthy individuals and did not correlate with MTC. On the other hand, higher levels of serum vaspin are associated with an increased risk of MTC. Thus Vaspin may be a novel and promising biomarker for diagnosis or confirmation of MTC in conjunction other specific tumor markers.

Prealbumin and Retinol Binding Proteins Are Not Usable for Nutrition Follow-Up in Pediatric Intensive Care Units

  • Purpose: Feeding children is a problem in pediatric intensive care units (PICU) and it is difficult to know the correct amount. The purpose of this study is to evaluate if prealbumin or retinol binding proteins (RBP) are effective relative to daily enteral nutrition, without being affected by severity of diseases or infections and can be used to follow up nutritional amount. Methods: This is a prospective observational study that includes 81 patients admitted to PICU in Akdeniz University with estimated duration >72 hours, age between 1 month and 8 years. Daily calorie and protein intake were calculated and prealbumin, RBP and C-reactive protein (CRP) levels were measured on the first, third, fifth and seventh mornings. Results: We find moderate correlation between daily calorie intake and prealbumin levels (r=0.432, p<0.001), RBP levels and daily protein intake (r=0.330, p<0.001). When we investigated the relationship between changes of prealbumin, RBP, CRP, calorie and protein intake during intensive care stay, we found that increase of Prealbumin and RBP levels are explained by decrease of CRP levels (r=-0.546 and -0.645, p<0.001) and not with increase of nourishment. Conclusion: Even adjusted for PRISM3, age and CRP, prealbumin and RBP are correlated with last 24 hours' diet. However, it is not convenient to use as a follow up biomarker because increase of their levels is related with decrease of CRP levels.

Study on the Prolactin Receptor 3 (PRLR3) Gene and the Retinol-binding Protein 4 (RBP4) Gene as Candidate Genes for Production Traits in Berkshire Pigs

  • To investigate the influence of the prolactin receptor 3 (PRLR3) gene and the retinol-binding protein 4 (RBP4) gene on the production traits of swine, genotyping was performed on 156 and 141 Berkshire pigs, respectively, that were carefully selected for economic traits. The frequencies of allele A in the PRLR3 locus and allele B in the RBP4 locus were 0.50 and 0.42, respectively. Neither locus was in the Hardy-Weinberg equilibrium. After a genotype was assigned to the individuals whose parents had the homozygous genotype, a statistical analysis was conducted for 291 pigs. The animals with the PRLR3 and RBP4 genotypes included 182 and 227 head, respectively. Even though the genotypic effects of PRLR3 (p<0.05) and RBP4 (p<0.01) had a significant influence on the pigs' back fat thickness, the interaction of both genes was not highly significant in terms of the back fat thickness (p = 0.1235). While the estimated epistasis effects of aaBB and aaBb decreased the back fat thickness and reduced the growth rate, the effects of AAbb and aabb increased the growth rate. Despite the insignificant difference in the PRLR genotypes in terms of the days to 90 kg and the average daily gain, the back fat thickness showed a significant difference (p<0.05), and the additive effect of allele A and the dominant effect of the hetero-genotype were -0.377 and 1.206 mm, respectively. The RBP4 genotypes had a very significant effect (p<0.01) on the back fat thickness, the days to 90 kg, and the average daily gain. The additive effects of allele B of the RBP4 locus on the back fat thickness, the days to 90 kg, and the average daily gain were 0.70 mm, -1.3 days and 6.2 g, respectively. Moreover, the dominant effects of the heterozygote for those traits were 0.63 mm, 9.9 days and -45.0 g, respectively. Allele A of the PRLR3 locus favorably influenced the back fat thickness, the days to 90 kg of the body weight, and the average daily gain and its dominant effect unfavorably influenced those traits. Allele B of RBP4 showed an incremental growth rate and back fat thickness, which could lower the lean meat percentage in the carcass. The RBP4 hetero-genotype negatively affected the pork production. These results strongly imply that the selection of allele A of PRLR3 and allele B of RBP4 would produce highly productive pigs in the Berkshire breed. Careful selection of allele B of RBP4 is required because of the increase in the back fat thickness.

The Dffects of Retinoids on CRABPII cRNA Induction amd Collagen Synthesis on Human Dermal Fibroblast

  • Retinoids are essential regulators of spithelial cell growth and celluar differentiation. They are also known to be effective in photoaging. It was reported that topical application of retinoic acid improves facial wrinkle carsed by collagen synthesis reduction in photodamaged skin. Collagen synthesis by retinoic acid may contribute to the wrinkle effacement. Since celluar retinoic acid binding protein II is slsctively induced in human skin and dermal fibroblasts in vitro by retinoic acid, this response can be used to mesure retinoids potency and activity. In order to know the activity of retinoids and their relations with collagen synthesis, we treated dermal fibroblasts with retinoids for 48 hours at 10-6-10-7M and measured CRABPII mRNA level by quantitative Nortern blotting. We also measured the rate of collagen systhesis by retinoids using 3-dimensional dermal equivalent. CRABPII mRNA level was increased 3-fold by retinoic acid, 2.1-fold by retinol and 1.4-fold by retinaldehyde. Collagen systhesis was increased 34% by all-trans retinioc acid, 26% by retinol, 17% by retinaldehyde and 7% by retinyl palmitate. From the above results, retinoids were found to be a potent indecers of CRABPII mRNA and collagen synthesis. Though retinoic acid was the most effective, its use has been restricted because of the side effects. Instead, retinol can be a best candidate in cosmetics for the treatment of photodamaged skin in terms of efficacy and safety.

Backbone NMR chemical shift assignment of transthyretin

  • Transthyretin (TTR) is an important transporter protein for thyroxine (T4) and a holo-retinol protein in human. In its native state, TTR forms a tetrameric complex to construct the hydrophobic binding pocket for T4. On the other hand, this protein is also infamous for its amyloidogenic propensity, which causes various human diseases, such as senile systemic amyloidosis and familial amyloid polyneuropathy/cardiomyopathy. In this work, to investigate various structural features of TTR with solution-state nuclear magnetic resonance (NMR) spectroscopy, we conducted backbone NMR signal assignments. Except the N-terminal two residues and prolines, backbone 1H-15N signals of all residues were successfully assigned with additional chemical shift information of 13CO, 13Cα, and 13Cβ for most residues. The chemical shift information reported here will become an important basis for subsequent structural and functional studies of TTR.

규칙적인 운동이 제1형 당뇨병 및 비만 환아의 아디포카인과 염증반응에 미치는 영향 (The Effects of Regulatory Exercise on Adipokines and Inflammatory Reaction in Type 1 Diabetic and Obese Children)

  • 본 연구의 목적은 유산소 운동이 비만과 제1형 당뇨병 어린이들에게 adipokines과 염증반응에 미치는 영향을 알아보고자 실시되었다. 피험자들은 비만군(OG, n=9), 제1형 당뇨병군(DG, n=9), 그리고 정상군(NG, n=9) 어린이로 연구하였다. 모든 인자는 체중, 체지방률, 체질량지수(BMI), 최대산소섭취량(ml/kg/min), 지질 인자와 adipokines를 분석하였다. 그 결과 체중, 체지방률, BMI, 최대산소섭취량에서 OG에 비해 DG와 NG는 유의한 차이가 나타났다(p<0.05). LDL-C와 HDL-C 역시 OG에 비해 DG와 NG에서 유의한 차이가 나타났다(p<0.05). 게다가 adiponectin과 retinol binding protein (RBP)-4는 12주 운동 후에 DG와 NG에서 유의한 변화가 나타났으며 (p<0.05), OG에 비해 DG와 NG에서 유의한 차이가 나타났다(p<0.05). Monocyte chemoattractant protein (MCP)-1에서 모든 그룹은 12주 운동후 유의하게 증가하였다(p<0.05). 결론적으로 규칙적인 운동은 비만과 제1형 당뇨병 어린이에게 신체조성의 변화는 가져오지 못했으나 제1형 당뇨병 어린이의 혈관 염증을 줄여줄 수 있으며, 비만어린이에서는 대사 증후군을 보호할 수 있을 것으로 사료된다.

국내 버크셔 돼지에서 성장 및 산자수의 후보유전자로서 PRLR3와 RBP4에 관한 연구 (A Study on the Prolactin Receptor 3 (PRLR3) Gene and the Retinol-binding Protein 4 (RBP4) Gene as Candidate Genes for Growth and Litter Size Traits of Berkshire in Korea)

  • 본 연구는 버크셔 품종에서 PRLR3와 RBP4 후보유전자의 두 개의 대립유전자가 산육형질과 번식형질에 미치는 영향을 조사하였다. 5,919두의 혈통자료, 3,480두의 산육기록과, 244두의 모돈의 775마리의 산자기록을 이용하여 유전능력 평가를 수행하였다. 유전자형 분석은 144두와 156두에서 PRLR3와 RBP4 유전자의 유전자형을 각각 분석하였다. 평가된 개체들의 육종가를 이용 두 마커의 유전자형 효과와 유의 확률을 추정한 결과 PRLR3 유전자는 번식형질의 총산자수(TBN)와 생존산자수(NBA)에서 -0.28과 -0.13두의 상가적 효과를 각각 나타내었다. RBP4 유전자는 일당증체량에서 10.58 g의 우성적 효과를 나타내었다. 그러나 RBP4 유전자의 다형성은 번식형질의 총 산자수(TBN)와 생존산자수(NBA)에서 -0.34와 -0.33두의 상가적 유전적 효과를 각각 나타났다. 따라서 PRLR3와 RBP의 B 대립유전자를 선호하는 MAS (Marker Assist Selection) Scheme은 버크셔 품종의 산자수의 개량에 이용 할 수 있을 것이다.
