Age-and Sex-Related Differences in Serum Levels of Lipid Peroxide, Retinol an d$\alpha$-Tocopherol in Korean Adults

  • The present study was aimed to investigated whether there are age-and sex-related differences in serum levels of lipid peroxides, retinol , and $\alpha$-tocopherol in Korean adults. The subjects were 441 persons , including 268 men and 173 women. Those of each sex were divided into four age group : 20-29, 30-39, 40-49 and 50-65 years, and their lifestyles and serum levels were compared . Men smoked and drank less as age increased , but 99.9% of women of did not smoke and only 11% drank more than once a month. Lipid peroxides of males increased gradually with age, while those of females showed greater levels in the 50-65 years group thon younger groups. Lipid peroxides, retinol and $\alpha$-tocopherol concentrations, which were adjusted for age, were significantly higher in males than in females. Lipid peroxide levels adjusted for total lipid were positively correlated with age in males than in females. Serum levels of $\alpha$-tocopherol adjusted for total lipid were positively correlated with age both in males and in females , while retinol was neither. The results indicate that serum levels of lipid peroxides, retinol , and $\alpha$-tocopherol are affected by age and that the response could be different between males and females.

정상과 설사증 송아지에서 혈중 retinol과 $\alpha$-tocopherol의 비교 (The Comparison of Blood Retinol and $\alpha$-Tocopherol Concentrations between Diarrhea and Normal Calves)

  • The purpose of this study is to examine and compare the concentrations of blood retinol and $\alpha$-tocopherol in normal and diarrhea calves. The subjects of the experiments are from three groups. Each group consists of 5 calves(diarrhea, treatment and control group). The CBC, serum chemistry, serum retinol and a-tocopherol concentrations were estimated in each group. Blood retinol concentration was $13.3{\pm}7.0{\mu}g/100ml$ in diarrhea group, $31.5{\pm}6.9{\mu}g/100ml$ in treatment group and $28.1{\pm}11.8{\mu}g/100ml$ in the control group. The blood concentration in diarrhea group was significantly lower than that of the control group in the case of retinol(p<0.05). However, there were no significance between the treatment poop and the control group. The $\alpha$-tocopherol concentration in blood was $266.0{\pm}127.6{\mu}g/100ml$ in diarrhea group, $432.2{\pm}172.7{\mu}g/100ml$ in the treated group and $579.3{\pm}145.8{\mu}g/100ml$ in the control group. In the case of $\alpha$-tocopherol, the laboratory group were significantly lower than the control group(p<0.05), except for the treatment group. As in retinol concentration there were no significance between treatment group and the control group. In the test of CBC, PCV was significantly lower in the group with diarrhea than the control group(p<0.05). Fibrinogen concentrations in diarrhea calves were significantly higher than the treatment and control group. In conclusion, the blood retinol and $\alpha$-tocopherol concentration in diarrhea calves are lower than normal calves. Medication of retinol and $\alpha$-tocopherol on calves with diarrhea is recommended.

돼지 체외 수정란 생산 효율 제고를 위한 레티놀 첨가 효과 (Retinol Supplemented to a Maturation Medium Stimulated In Vitro Development of Porcine Oocytes after In Vitro Fertilization)

  • 본 연구는 돼지 수정란의 체외 성숙 및 체외 배양액의 retinol 첨가 효과를 규명하기 위하여 체외 성숙 및 체외 배양액에 retinol을 첨가하여 수정란의 체외 발달에 미치는 영향을 구명하고자 수행되었다. 체외 성숙 배양액에 retinol을 첨가한 결과 성숙율은 $66.7{\pm}6.0{\sim}69.2{\pm}5.3%$으로 각 처리구 간의 유의적인 차이가 없었다(p>0.05). 체외 수정 후 배반포로의 발달율은 $5{\mu}M$ 첨가구에서 $20.4{\pm}2.6%$의 발달율을 나타내어 타 처리구에 비하여 유의적으로(p<0.05) 높게 나타났으며, $10{\mu}M$ 첨가구에 있어서는 분할율과 배반포로의 발달율이 타 처리구에 비하여 유의적으로(p<0.05) 낮게 나타났다. 정상 수정율은 $59.3{\pm}9.6{\sim}73.0{\pm}7.3%$로 처리구간에 차이가 없었다. 다정자 침입율은 처리군 간에 유의차는 인정되지 않았다(p>0.05). 배반포로의 발달 후 세포수를 비교한 결과 대조구($29.8{\pm}1.0$)에 비해 $5{\mu}M(37.0{\pm}1.6)$ 첨가구에서 유의적으로(p<0.05) 많은 세포수를 나타내었다. 체외 배양액에 retinol을 첨가한 후 발달율을 조사한 결과, 난할률, 상실배 및 배반포로의 발달율에서 모두 효과를 나타내지 못하였으며, 특히, $10{\mu}M$의 처리구에서는 대조구를 비롯한 타 처리구들보다 유의적으로(p<0.05) 발달율이 낮게 나타났다. 체외 성숙 배지와 체외 배양액 모두 retinol을 첨가한 결과 배반포로의 발달율에 있어서 각 처리구간의 유의적인 차이는 없었다. 이상의 결과를 종합할 때, 돼지 체외 성숙 배양액에 5 uM의 retinol의 첨가는 발달율과 세포수에 있어서 유의적으로(p<0.05) 효과적이었으며 10uM에서는 유해한 결과를 나타내었다.

Serum Carotenoid, Retinol and Tocopherol Concentrations and Risk of Cervical Cancer among Chinese Women

  • Background: Despite many epidemiological studies on the effects of dietary antioxidant micronutrients on risk of cervical cancer, the findings remain uncertain and little evidence is available for serum nutrient markers. The present study aimed to examine the relationship between serum carotenoid, retinol and tocopherol concentrations and risk of cervical cancer among Chinese women. Materials and Methods: We conducted a hospital-based case-control study in which 358 adults (158 incident cases and 200 controls) were recruited from Xinjiang, China. Serum levels of carotenoids (${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lycopene and lutein/zeaxanthin), retinol, and tocopherols (${\alpha}$-tocopherol and ${\gamma}$-tocopherol) were assessed by reverse-phase high-performance liquid chromatography. Results: We found inverse associations between serum carotenoid (${\alpha}$-carotene, ${\beta}$-carotene, and lutein/zeaxanthin) and tocopherol (${\alpha}$-tocopherol) concentrations and the risk of cervical cancer after adjusting for potential confounders, but a null association for retinol. The ORs for 1-SD increase were 0.71 (95 % CI: 0.56-0.92; p=0.003) for total carotenoids and 0.75 (95 % CI: 0.60-0.94; p=0.008) for total tocopherols. Conclusions: These results show that higher serum concentrations of some carotenoids and tocopherols are associated with a lower risk of cervical cancer among Chinese women.

액중건조법으로 제조한 Retinol 함유 마이크로캡슐의 방출 거동 (Controlled Release of Retinol Containing Microcapsules Prepared by Solvent Evaporation Method)

  • 생분해성이 있는 polycaprolactone을 사용하여 용액 상태에서 건조시키는 방법으로 마이크로 캡슐을 제조하였다. 심물질로는 화장품 분야에서 중요한 성분으로 쓰이는 retinol을 선택하였으며 안정제로는 poly(vinyl alcohol)을 사용하였다. 제조 조건에 따른 마이크로캡슐의 형태 변화와 물성 변화를 scanning electron microscope와 differential scanning calorimeter를 이용하여 측정하였으며 심물질의 방출 속도에 미치는 영향들을 UV를 이용하여 측정하였다. 사용한 벽재 물질 용액의 농도, 교반속도, 안정제의 농도 등을 변화 시켜 가면서 마이크로캡슐을 제조하여 보았으며, 최적 조건에서 지름이 5~6 um인 구형 모양의 균일한 크기를 갖는 마이크로캡슐들이 형성되었음을 확인하였다.

  • A novel retinol derivative, polyethoxylated retinamide (Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of [$^3H$]-proline incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells (effective diffusion area: $1, 766{\;}\textrm{cm}^2$) and the excised skin of female hairless mouse aged 8 weeks were used The stabilities of retlnoids were evaluated at two different temperature ($25{\;}^{\circ}C$ and $40{\;}^{\circ}C$) and under UV in solubilized state and in OW emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.

  • Retinol as well as RA is well known to have many beneficial effects on aged skin. But the skin irritation potential and unstable condition of the products containing them have been some problems in their cosmetic uses. So, retinol containing gel product was developed for less skin irritancy and more stability in cosmetic products. To examine the clinical effects of retinlo containing product, we used cilnicla non-invasive assessment techniques on 40 volunteers for 6 months maintaining double-blind test conditions. According to our results, the use of retinol containing product improved skin color and hydration level slightly. But there was no statistical differences. There was no erythema reaction compared to the use of RA. Especially, the skin elasticity increased above 20% and skin wrinkles of crows' feet region decreased more than 10%. Besides the instrumental analysis, a large majority of volnteers felt that their skin were improved in the case of wrunkles, elasticity, hydration and color.

중년기 성인의 혈중 항산화 물질 분석 및 혈중 지질과 신체계측치와의 상관성 (Analysis of Serum Antioxidant Materials Concentration and Their Relation with Blood Lipids and Anthropometric Indices in Middle-Aged Adults in Korea)

  • 본 연구에서는 우리나라 건강한 중년성인의 혈청 carotenoids류 (${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lutein, lycopene)와 ${\alpha}$-tocopherol, ${\gamma}$-tocopherol, retinol의 수준을 파악하고 혈중 지질상태 및 신체계측치와의 상관성을 평가해보고자 하였으며 조사 대상자는 40대 이후 건강한 성인 373명 (남자 231명, 여자 142명)이었다. 우리나라 건강한 중년기 성인의 혈청 중에 존재하는 주요 carotenoids는 ${\beta}$-cryptoxanthin, ${\beta}$-carotene, lutein이었고 lycopene, ${\alpha}$-carotene은 더 낮은 농도로 존재하였다. 성별에 따른 비교에서는 ${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lycopene 등의 carotenoids 물질들의 농도는 여자가 남자에 비해 유의하게 높았고, retinol은 남자가 여자에 비해 유의하게 높았으며 lutein과 tocopherol은 성별에 따른 유의차를 보이지 않았다. 혈중 항산화 물질의 농도는 연령이 높아짐에 따라 증가하는 경향을 보였으며 retinol과 ${\alpha}$-tocopherol 영양상태는 남자가 여자에 비해 우수하였다. 혈청 항산화 물질들의 농도와 혈청 및 소변 중의 지질과산화물 농도 사이에는 뚜렷한 상관성을 발견할 수가 없었다. 혈청 carotenoids들은 상호간에 또 ${\alpha}$-tocopherol 사이에 유의적인 양의 상관관계를 보였으며, 반면 lutein을 제외하고는 retinol과는 음의 상관성을 나타내었다. 혈 중 ${\alpha}$-tocopherol과 retinol 사이에는 유의적인 양의 상관성을 보였다. 혈 중 지질과의 상관성 분석에서 측정된 모든 항산화 물질들은 총콜레스테롤과 양의 상관관계를 나타내었다. 반면 중성지질은 carotenoids 물질과는 음의 상관성을 보였고 tocopherol, retinol과는 양의 상관성을 보였다. 혈청 항산화 물질들의 농도와 신체계측치 사이에는 성별에 관계없이 전반적으로 혈청 carotenoids류는 신체계측치들 및 혈압과 음의 상관성을 보였으며, tocopherol들과 retinol 수준은 양의 상관성을 보였다. 이 중 특히 유의적인 상관성을 보인 것은 남성에서 ${\beta}$-carotene과 lutein의 혈압과의 음의 상관성, 여성에서의 ${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin과 체지방 관련 지표들 (BMI, WHR, 체지방율)과의 음의 관련성이었다. 본 연구 결과 중년 성인의 혈중 항산화 물질들의 농도는 종류에 따라 성별, 연령별로 유의적인 차이를 나타내었으며, 혈중 지질 수준 및 신체 계측치와도 유의적인 관련성을 가지는 것으로 나타났다. 따라서 이러한 상관성의 원인과 의미에 대해서는 향 후 식생활 및 생활습관 등과 관련된 연구를 통해 밝혀져야 할 것이다.
