Effects of Astragali Radix Herbal-acupuncture at $ST_{36}$ (Jok-samni) on Collagen-induced Arthritis in Mice (황기약침이 Collagen으로 유발된 생쥐의 Arthritis에 미치는 영향)

  • Objectives : The aim of the present study was to observe the effect of Astragali Radix Herbal-Acupuncture Solution(AR-HAS) at $ST_{36}$(jok-samni, $Z\acute{u}s\bar{a}n$ Li) on collagen- II -induced arthritis(CIA) in mice. Methods : DBA/1J mice were immunized with bovine type II collagen(CII) on days 0 and 21 to induce arthritis. The mice were divided into 5 groups : normal group(no CIA), control group(CIA+no treatment), needle prick group(CIA+single prick with an injection needle), saline group(CIA+saline injection) and ARHA group(CIA+ R-HA treatment). The needle prick, saline injection, and AR-HA groups were injected on the right $ST_{36}$(jok-samni) of mice for 9 weeks, 3 times a week, beginning 4 weeks after the booster immunization. Results : 1. The incidence of arthritis, AI(arthritis index), and joint edema decreased in the AR-HA group. 2. Weight gain, hypertrophy of the spleen, adhesion of the tissues, and transformation of the joint were restrained in the AR-HA group. 3. The concentrations of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, $IFN-{\gamma}$ in CIA mouse serum and $IFN-{\gamma}$, IL-10 in the CIA mouse spleen cell culture decreased significantly at $ST_{36}$ in the AR-HA group. 4. Total cell counts increased significantly, and the ratio of $CD3e^+$ to $CD45R^+$, $CD4^+$ to $CD8^+$, and $CD4^+$ to $CD25^+$ cells decreased significantly at $ST_{36}$ in the CIA mouse spleen cell culture of the AR-HA group. 5. Total cell counts decreased significantly, and $CD4^+/CD25^+$ and $CD45R^+/CD69^+$ decreased significantly at $ST_{36}$ in the CIA mouse lymph nodes of the AR-HA group. 6. $CD3e^+/CD69^+$ and $CD11b^+/Gr-1^+$ cells decreased significantly at $ST_{36}$ in the CIA mouse joints of the AR-HA group. 7. The histological examination showed that cartilage destruction and synoviocyte proliferation decreased in the CIA mouse joints of the AR-HA group, and collagen fiber was expressed similar to that seen in the normal group. Conclusions : Our experiments show that at $ST_{36}$, an anti-inflammatory mechanism of AR-HA controls synovial cell proliferation and protects against cartilage destruction in rheumatoid arthritis.

Body Image, Eating Problems and Dietary Intakes among Female College Students in Urban Area of Korea (일부 도시지역 여대생들의 신체상과 섭식 장애 및 영양 섭취 양상)

  • Concern about body weight among young females is too much, so distorted body image and restrained eating are serious problems among Korean young females. A society preoccupied with thinness give a psychological burden to women, this burden may have adverse effects on health physically and psychologically. We tried this study to investigate body image, eating problems, and dietary intakes among female college students in urban areas of Korea. We satisfactorily surveyed using their body weight, desired weight dieting, binge eating, and night eating by self-questionnaires with 500 female college students. We evaluated their dietary intakes by semi-quantitative food frequency questionnaires to assess their usual intakes. After removing incomplete data, we analyzed 469 data by SAS 6.10. The 469 women aged 21.1 years. Their mean height, weight, and BMI were 161.4cm, 52.5kg, and 20.2kg/$m^2$ respectively. Though the frequency of obesity was only 2.6$\%$, the frequency of dissatisfaction with their body weight was 84.7$\%$. Female college students desired their hight, weight, and BMI to be 165.8cm, 48.2kg, and 17.5kg/$m^2$, respectively. The frequency of binge eating and night eating were 20.7$\%$and 6.8$\%$. The odds ratios for binge eating and night eating were 10.9(95$\%$ CI : 2.6-15.1) and 6.0(95% CI : 1.8-14.8) in the group satisfied with their body weight compared with the dissatisfied group. The odds ratios for binge eating and night eating were 2.6(95% CI : 1.6-4.4) and 2.4(95% CI : 1.1-5.4) in the dieting group compared with the non-dieting group. The mean caloric intake was 1, 840kacl and percent intakes of carbohydrate, protein, and fat wee 57.2$\%$, 16.3$\%$, and 24.0$\%$, respectively. The dieting group showed deficient intakes of protein and calcium significantly compared with the non-dieting group showed deficient intakes of protein and calcium significantly compared with the non-dieting group. Almost all women college students in urban area had dissatisfaction with their body shape, and desired severe undrerweight. The frequency of binge eating and night eating increased according to dissatisfaction with body weight, diet, and distortion of body image. Dieting has adverse effect on the intakes of essential nutrients. Educations is needed to correct distorted body image, and bad eating habits in young females, and marked preoccupation with thinness among women should then be reduced. LncRNA CRNDE Promotes the Progression of B-cell Precursor Acute Lymphoblastic Leukemia by Targeting the miR-345-5p/CREB Axis

  • The imbalance between the proliferation and apoptosis of B-cell precursors is an important contributor to the pathogenesis of B-cell precursor acute lymphoblastic leukemia (BCP-ALL), while its specific regulatory mechanism remains perplexing. This study aimed to expound the underlying mechanism of the proliferation and apoptosis of BCP-ALL cells from the perspective of non-coding RNA. In this study, long non-coding RNA colorectal neoplasia differentially expressed (LncRNA CRNDE) was upregulated in the bone marrow of BCP-ALL patients and BCP-ALL cell lines (NALM-6 and RS4;11). Functionally, LncRNA CRNDE knockdown restrained cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. The subsequent investigation confirmed that LncRNA CRNDE bound to miR-345-5p and negatively regulated miR-345-5p expression. The overexpression of miR-345-5p suppressed cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. Further experiments revealed that miR-345-5p downregulated cyclic AMP response element-binding protein (CREB) expression by targeting its mRNA directly. CREB overexpression reversed the effect of miR-345-5p mimic on cell proliferation and apoptosis in NALM-6 and RS4;11 cells. Finally, in vivo experiments showed that LncRNA CRNDE knockdown prolonged the survival of mice xenotransplanted with NALM-6 cells. In conclusion, LncRNA CRNDE upregulated CREB expression by suppressing miR-345-5p, thus promoting cell proliferation and reducing cell apoptosis in BCP-ALL.

Stress-Reducing Effects of Brown Rice Koji

  • The primary objective of this study is to determine whether a diet supplemented with brown rice koji (BRK) results in a reduced stress response in rats and mice. BRK, which has been suggested as a candidate for use as a stress- and fatigue-fighting supplement, was compared with red ginseng extract (RG) for its stress-reducing potential. The animals in this study were divided into no-stress, stress, RG, and BRK groups of 8 to 10 animals each. Stress was induced by means of immobilization (being restrained in plastic tubes for 30 min and electroshock (0.5 mA in mice or 2 mA in rats for 5 min). The no-stress group was not exposed to stress. Rats in the RG group received oral doses of 200 mg RG extract/kg body weight daily. The BRK group was fed a 30% BRK diet and exposed to stress. Animals were given supplements for 7 days before being exposed to stress, and then were given supplements for 5 days with exposure to stress. When the stress exposure ended, the animals were observed for stress-related changes in behavior and their plasma corticosterone levels were measured. BRK supplementation was associated with a partial blockade of the effects of stress on locomotion and elevated plus-maze test results in rats and mice. It was also associated with a partial reduction in stress-induced behaviors such as freezing, burrowing, smelling, face-washing, and rearing. BRK supplementation did not have a significant effect on plasma corticosterone levels, which were increased in the animals exposed to stress (p<0.01). The mice in the RG group received RG in water (2 mg RG/ mL $H_2O$), and the BRK group received a 30% BRK diet (weight) for 7 days. Both groups were evaluated for signs of fatigue. BRK supplementation increased endurance, as indicated by time on the rota-rod, in cold water, and on the horizontal wire. These results suggest that BRK supplementation partially protects the animal from the effects of stress and may also contribute to resistance to fatigue on physical exertion.

  • The suppression of residual settlement is required on earthwork sections as concrete track is introduced. Use of pile-slab structure is one of the settlement restraining methods applied on soft ground. The slab distributes the upper embankment load and piles transfer the load from the slab to the stiff ground. While this method is very effective in terms of load transfer, it has not yet been established for dealing with the vibration transfer effects and interaction characteristics between a structure and the ground. It is possible that vibration caused by a moving train load is propagated in the upper embankment, because the slab acts as a reflection layer and waves are multi-reflected. In this present paper, wave propagation generated by a moving train load is evaluated in the time and frequency domains to consider a roadbed structure using an artificial impact load and field measured train load. The results confirmed the wave reflection effect on the pile-slab structure, if the embankment height is sufficient, vibration propagation can be stably restrained, whereas if the height is not sufficient, the vibration amplitude is increased.

Effect of Soil Amendments on Rice Yield and on Occurence of Harmful Substance in Ill Drained Paddy Soil (습답(濕畓)에서 개량제(改良劑) 시용(施用)이 벼 수량(收量) 유해물질(有害物質) 생성(生成)에 미치는 영향(影響))

  • In order to investigate the application effects of soil amendment on wetland rice soil field and laboratory experiment were conducted on ill drained paddy field on which rice straw, Compost, Lime and Silicate materlials such as wallostonite and fused phosphate were applied. 1. As compared with check plot, rice yield was increased by application of soil amendments significantly in all plots but rice straw one. 2. The effects of soil amendment on rice production were in the order of fused phosphate and N.K, addition > silicate material and N.K, addition=Compost > rice straw and lime plot. 3. The significant correlation was showed among the rice yield and the numbers of panicle and grain. 4. The application of the rice straw restrained the rice growth because of nitrogen defficiency in early stage but increased rice yield due to the number of panicle due to supplying nitrogens late stage. 5. The application of rice straw in wetland soil promoted to occur organic acids such as $PCO_2$ and $HCO_3$, and decreased tillering due to these organic acids occuring in early growth stage.

Effect of High-molecular-weight Maleic Anhydride-grafted Polylactic Acid Compatibilizer on the Properties of Polylactic acid-based Wood Polymer Composites (말레산 무수물로 그래프트된 고분자량의 폴리락트산 상용화제가 폴리락트산 기반의 합성목재에 미치는 영향)

  • High-molecular-weight maleic anhydride-grafted polylactic acids (HMMA-g-PLA) compatibilizers were prepared by melt grafting in a twin screw extruder using di(tert-butyl-perxoyisopropyl)benzene (PK-14; as initiator), maleic anhydride (MA), and divinylbenzene (DVB). To determine the properties of the prepared HMMA-g-PLA compatibilizers, Fourier transform infrared (FTIR), Melt index (MI), and back-titration analyses were performed. On increasing DVB concentration, grafting yield of HMMA-g-PLA increased but MI decreased because 𝛽-scission of PLA was restrained by the DVB, and thus, the molecular weight of HMMA-g-PLA increased. PLA-based wood-plastic composites (WPCs) were prepared using HMMA-g-PLA by melt blending through a single screw extruder. The flexural and impact strengths of WPCs compatibilized with HMMA-g-PLA were greater than those of WPCs produced without HMMA-g-PLA. Scanning electron microscope (SEM) studies indicated that increased mechanical properties were caused by excellent interfacial adhesion between PLA and wood fibers due to the addition of HMMA-g-PLA. However, rather high contents of HMMA-g-PLA reduced the mechanical properties of WPCs. We believe that lower molecular-weight of HMMA-g-PLA added as an compatibilizer, compared with PLA polymer, caused the reduction of mechanical properties.

  • The present study attempts to develop the theoretical model for the damage estimation of offshore tubular members which are subjected to the accidental impact loads due to collision, falling objects and so on. For the reasons of the simplicity of the problem being considered, however, this paper postulates that the accidental load can be approximated to be the quasi-static one, in which dynamic effects are negelcted. Based upon the theoretical and experimental results which are obtained from the present study as well as the existing literature, the load-displacement relations taking the interaction effect between the local denting and the global bending deformation into account are presented in the explicit form when the concentrated lateral load acts on the tubular member whose end condition is supposed to be rotation ally free and axially restrained, in which membrane forces develop. Thus, the practical estimation of damage deformation for the local denting and the global bending damage of tubular members against the accidental loads is possible and also the collision absorption capability of the member can be calculated by performing the integration of the area below the given load-displacement curves, provided that all the energy is dissipated to the deforming the member itself.

Effect of hemp seed oil on lipid metabolism in rats fed a high-cholesterol diet (햄프씨드 오일이 고콜레스테롤식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • Purpose: This study evaluates the potential protective effects of hemp (Cannabis sativa L.) seed oil supplementation in rats fed a high-cholesterol diet. Methods: Rats were fed a 1.25% cholesterol diet for 8 weeks, followed by oral administration of either of the two doses of hemp seed oil (HO) (0.5 mL/kg (HOL group) or 1 mL/kg (HOH group) body weight/day) or simvastatin at 10 mg/kg body weight/day. Oxidative stress, lipids, liver enzymes, and renal markers were measured in the serum. Western blot analysis was applied for evaluating the expressions of inflammatory makers. Results: Except for HDL-cholesterol, the altered levels of lipoproteins, aminotransferases, urea, and creatine kinases in hypercholesterolemic rats were significantly corrected by HO administration. Especially, compared to the HOH group, HOL treatment further reduced AST, ALT, creatinine, TC, and LDL-cholesterol levels. Moreover, both the atherogenic index and cardiac risk factor (CRF) in the HOL group were more restrained compared to the HOH group. Increased levels of p-AMPK coincided with the inhibition of SREBP-2 activation which subsequently suppressed the expression of HMGCR. Nuclear factor (NF)-κB activation coincided with the PI3K/Akt pathway activation and the increased phosphorylation of p38; these levels were significantly suppressed by HO treatment. In addition, HO treatment markedly reversed the changes in chemokines such as ICAM-1, VCAM-1, and MCP-1. Histological alterations induced by cholesterol overload in cardiac and hepatic tissues were ameliorated by HO supplementation. Conclusion: Taken together, our results indicate a low concentration of HO demonstrates improved dysfunctions caused by a high-cholesterol diet via inhibition of the PI3K/Akt/NF-κB signaling pathway.

The Effect of Exhibit Commentary Media on the Communication within Family Visitors: with Docent and Mobile Device (PMP) (박물관에서의 전시설명 매체가 가족단위 관람객들의 소통에 미치는 영향: 도슨트와 모바일 기기(PMP)를 중심으로)

  • There are exhibit commentary media (docent, mobile device) for assisting family visitors in the museum, and the communication within family members could be affected by which media they use. When using a docent, family visitors lowered their voice or restrained talks for paying attention to a docent' explanation. When using a mobile device, family visitors mainly relied on a mobile device, so they did not pay attention to the objects. The results of the survey showed that parents reported no difference between the docent and the mobile device on some measurement items and preferred the docent to the mobile device on other measurement items. On the other hand, children showed strong preference toward the mobile deice over the docent because of their enjoyment for operating the mobile device. Regardless of which media they used, the communication among family members could be enhanced by being together. Therefore, the museum should design and operate a docent program for family visitors to enhance the communication among family members. In addition, the museum should consider how family visitors using a mobile device would pay attention to objects and enhance the communication.
