TT Mutant Homozygote of Kruppel-like Factor 5 Is a Key Factor for Increasing Basal Metabolic Rate and Resting Metabolic Rate in Korean Elementary School Children

  • We investigated the contribution of genetic variations of KLF5 to basal metabolic rate (BMR) and resting metabolic rate (RMR) and the inhibition of obesity in Korean children. A variation of KLF5 (rs3782933) was genotyped in 62 Korean children. Using multiple linear regression analysis, we developed a model to predict BMR in children. We divided them into several groups; normal versus overweight by body mass index (BMI) and low BMR versus high BMR by BMR. There were no differences in the distributions of alleles and genotypes between each group. The genetic variation of KLF5 gene showed a significant correlation with several clinical factors, such as BMR, muscle, low-density lipoprotein cholesterol, and insulin. Children with the TT had significantly higher BMR than those with CC (p=0.030). The highest muscle was observed in the children with TT compared with CC (p=0.032). The insulin and C-peptide values were higher in children with TT than those with CC (p=0.029 vs. p=0.004, respectively). In linear regression analysis, BMI and muscle mass were correlated with BMR, whereas insulin and C-peptide were not associated with BMR. In the high-BMR group, we observed that higher muscle, fat mass, and C-peptide affect the increase of BMR in children with TT (p < 0.001, p < 0.001, and p=0.018, respectively), while Rohrer's index could explain the usual decrease in BMR (adjust $r^2$=1.000, p < 0.001, respectively). We identified a novel association between TT of KLF5 rs3782933 and BMR in Korean children. We could make better use of the variation within KLF5 in a future clinical intervention study of obesity.

  • Objective: To evaluate the effect of Ephedra Sinica and Evodia Rutaecarpa on resting metabolic rate(RMR), weight and body composition in obese women during low-calorie diet. Methods: We recruited 90 healthy premenopausal women$(body\;mass\;index{\geq}25kg/m^2)$ and randomly assigned to three groups(ephedra group, evodia group and placebo group). Subjects were administered Ephedra Sinica(6g twice a day) or Evodia Rutaecarpa(6g twice a day) or placebo in a double-blind fashion and participated in low-calorie diet for 8 weeks. Resting metabolic rate and body composition were measured at baseline, 4 and 8 weeks. Results: RMR change for 4 weeks was significantly higher in the ephedra group compared with the placebo group(p<.05). But the change for 8 weeks was not significant and the Evodia group showed no significant RMR change. Weight and percent body fat changes for 4 weeks and 8 weeks were significantly higher than the placebo group in the ephedra group(p<.05), but the Evodia group didn't show significant level. Conclusions: This study supports and emphasizes the benefits of herbal medicine in maintaining or increasing RMR during low-calorie diet. Especially Ephedra treatment was effective on significant maintainment of RMR, loss of weight and percent body fat(P<.05).

운동군과 비운동군 고등학생의 활동량, 활동계수, 예측 휴식대사량, 1일 에너지 및 영양소 섭취량의 비교 (Comparison of Activity Factor, Predicted Resting Metabolic Rate, and Intakes of Energy and Nutrients Between Athletic and Non-Athletic High School Students)

  • This study compared activity factor. predicted resting metabolic rate (RMR), and nutrient intakes between athletic and non-athletic high school students in Gangwon-do. Fifty soccer players (30 males and 20 females; mean ages 16.7${\pm}$1.0 years and 16.4${\pm}$1.1 years. respectively) and 50 non-athletic (30 males and 20 females: mean ages 17.5${\pm}$0.4 years and 16.4${\pm}$1.1 years respectively) high school students were included. Anthropometric measurements included: weight and height. triceps skinfold, mid-ann circumference, and body fat. Prediction equations consisted of those from the Harris-Benedict. FAO/WHO/VNU, IMNA, Cunningham, Mifflin et al., and Owen et al. A one-day activity diary was collected by interview, and the 24-hour recall method was used to analyze nutrient intakes of subjects. The activity factors of the male and female athletic groups (2.23 and 2.16, respectively) were significantly higher than those (1.52 and 1.46, respectively) of the non-athletic group. There was only a significant difference in RMR by use of the Cunningham's equation between two groups. For the males. almost all nutrient intakes of the athletic group (except carbohydrate, iron, vitamin $B_1$, $B_6$, and niacin) of athletic group were significantly higher than those of the non-athletic group. The female athletic group showed significantly higher nutrient intakes with the exception of most vitamins. These results suggest that assessments of energy balance between energy intake and energy expenditure by employing RMR and activity factors would be useful to prevent and treat obesity in high school athletes. In addition, the Cunningham's equation would be appropriate for predicting their energy needs.

Metabolic Rate and Thermolabile Properties of Ognev's Great Tube-nosed Bat Murina leucogaster in Response to Variable Ambient Temperature

  • The winter-resident Korean bats, Murina leucogaster ognevi, show a circadian cycle of thermoregulation and locomotion in summer, as do other bat species in temperate regions. They are most active between dusk and dawn with body temperature (Tb) of 35-4OC, and are usually torpid in their roost sites for the rest of day with their Tb close to ambient temperature (Ta) of around 15C. The present study was conducted to determine thermogenic and thermolabile properties of the heterothermic bats that would influence their daily feeding activities and ultimately, their energy conservation strategy. Testing on active male Murina, resting metabolic rate (RMR, gauged by oxygen consumption rate) at the lower limit of thermoneutral zone (31C) was 2.0 L kq-1 h-1. The regression slope of RMR below the thermoneutral zone (an index of metabolic thermal sensitivity) was -0.38 L $kg^{-l} h^{-1} C^{-1}$. The metabolic rate at the roost Ta (15C) was 4.5 times the lowest RMR in the active state but becomes nearly zero in the torpid state. This implies that by being torpid during daytime (between dawn and dusk), the individual bats would save about 4.7 kcal each day in mid-summer. Interspecific comparisons of thermal metabolic response over a mass scale suggest that the smaller bats show a relatively higher metabolic rate in thermoneutral zone and a greater thermal sensitivity of metabolism, which follows the general principle seen in homeothermic metabolism. Thermolabile features in metabolic responses seem to be fairly common for these bats in conditions other than a fully active state. Types of thermolabile responses and their energetic significance are discussed.

Effects of endurance training for 4weeks on resting metabolic rate and excess post-exercise oxygen consumption in mouse

  • This study assessed the amount of energy consumed and fat deposition after endurance training in order to review the effect of 4-week endurance exercise on resting metabolic rate of a mouse during and after exercise and the effect of exercise. A total of 19 seven-week-old ICR male mice were used as the study subject. Those mice were divided into sedentary group (Sed) and trained group (Tr) after a week of environment adaption. The Tr group was trained with endurance exercise five times a week for four weeks. Weight and the amount of food intake were daily weighed and resting metabolic rate and metabolic rate after exercise were assessed before starting exercise and on the fourth week after training. Metabolic rate during exercise were measured four weeks after training. At the end of breeding period, statistically significant difference was shown in weights of trained and sedentary groups (p < 0.05). During a resting period, no significant difference was shown in oxygen intake, respiratory exchange ratio, and the amount of carbohydrate and fat oxidized. Moreover, no significant difference was shown in excess post-exercise oxygen consumption (EPOC) of an hour period after training. In contrast, the maximal oxygen uptake (VO2 max) was approximately 11.1% higher in trained group after training compare to before. However, there was no significant difference in respiratory exchange ratio and carbohydrate and fat oxidization. During exercise, oxygen uptake, carbon dioxide production, and respiratory exchange ratio in energy metabolism during exercise showed no significant difference. However, significant difference was exhibited in the amount of fat oxidized in both groups. Summing up those results, endurance exercise could be concluded to be effective in weight control. However, weight loss is thought to be resulted from increase in fat oxidization during exercise unlike the conclusion made from previous studies where weight loss is prominently influenced by energy metabolism during a resting period and increased fat oxidation during post-exercise recovery. All experimental procedures were carried out at the Animal Experiment Research Center of Konkuk University. This study was conducted in accordance with the ethical guidelines of the Konkuk University Institutional Animal Care and Use Committee.

유산소운동과 diacylglycerol 섭취가 비만인의 혈중 지질성분 및 안정 시 대사량의 변화에 미치는 영향 (Combined Effects of Aerobic Exercise Training and Diacylglycerol Ingestion on Blood Lipid Profiles and Resting Metabolic Rate Changes in Obese Man)

  • 본 연구는 체지방을 감소시키는데 효과적이라는 diacylglycerol (DG) 섭취와 유산소운동이 체구성비의 변화 및 혈중지질 개선에 미치는 영향을 규명하고 안정 시 대사량 변화를 관찰함으로써 임상적 및 대사적 측면에서 비만 개선 효과를 규명하는데 그 목적이 있었다. 본 연구의 대상은 의학적으로 다른 질병을 동반하지 않은 비만 남자 대학생으로서 연구의 의의를 충분히 이해하고 자발적으로 참여 의사를 밝힌 사람으로 선정하였으며, 체지방 측정 후 25%미만인 자와 흡연자는 대상에서 제외하였다. 모든 피험자들은 기본검사와 사전검사가 끝난 후 세가지(D, E, DE) 집단에 6명씩 무선 배정하였다. 첫 번째 실험군(D)은 diacylglycerol 섭취 조건으로 12주 동안 diacylglycerol을 섭취하였고, 두 번째 실험군(E)은 12주 동안 러닝 트레이닝을 실시하였다. 마지막 세번째 실험군(DE)은 12주 동안 diacylglycerol 섭취와 유산소 트레이닝을 실시하였으며, 본 연구결과 모든 그룹에서 12주 동안의 그룹별 처치가 진행됨에 따라 체지방이 감소하는 경향을 보였다. 안정 시 대사량은 D그룹을 제외하고 모든 그룹에서 증가하는 경향을 보였으며 운동과 다이아실글리세롤 섭취에 따라 콜레스테롤의 감소가 일어났다. 추후 카테콜라민, CAMP, HSL, Insulin 등과 같은 지방대사와 관련된 항목들에 대한 추가적인 분석이 이루어짐으로써, DG섭취와 트레이닝의 비만개선효과를 보다 신뢰성 높게 예측할 수 있으리라 본다.

규칙적 운동 및 식이요법이 비만여성의 안정시대사량과 호르몬 변화에 미치는 영향 (Effects of Regular Exercise and Diet on RMR and Hormonal Changes in Obese Women.)

  • 운동과 식이요법을 병행한 체중감량 프로그램에 참여한 여성 비만인의 체지방량, 안정시대사량, 그리고 갑상선 호르몬 차이를 비교할 목적으로 실험한 결과, 체지방량, 체지방율, 체표면적이 유의하게 감소되었고, 안정시대사량 예측치와 실측치 모두 유의하게 감소되었지만, 갑상선 호르몬의 변화는 나타나지 않았다. 안정시대사량과 신체요인들과의 상관분석 결과, 제지방 체충이 가장 밀접한 관계를 보였으며, 안정시대사량과 갑상선 호르몬들과의 관계에서는 TSH와 가장 밀접한 관계를 나타내었다. 결론적으로 8주간 체중감량 프로그램 참여로 체지방은 효과적으로 감소되었으나, 갑상선 호르몬 차이는 없었고 체중감소에 비해 제지방이 유지됨으로써 안정시대사량의 감소는 최소화되어, 에너지 대사작용에 긍정적인 영향을 준 것으로 사료된다.

  • The purpose of this study was to evaluate the effect of various exercise intensity on Excess post exercise energy expenditure (EPEE), Resting Metabolic Rate (RMR),thyroid hormonal changes and biochemical variables in obese and NIDDM patients. The subject of the present study were divided into four groups and four periods: trained (T; n=10), control (C; n=10), obese (O; n=10) and NIDDM (N; n=10) group. And the periods were divided as follows; Resting (RE), Maximal (MA), High intensity (HI), and Low intensity (LI). There was significant difference in RMR among different intensity of exercise. in the T (p<0.05) not in the C, O, and N groups. however, there was no significant different percent body fat among all groups. In the energy expenditure, there was significant different among C, O, N groups compare to T in HIEE (high intensity exercise energy expenditure), LIEE (low intensity exercise energy expenditure), HIEEPE (high intensity exercise energy expenditure post exercise) and LIEEPE (low intensity exercise expenditure post exercise). In the hormonal level, there was significant different in T4 level in the T group at LI period and there was also significant difference in T4, Free T3, & Free T4 levels in T group at LI period, however there was no significant different in the O and N groups except LI period. In the fatigue variables, there was significant different in lactate and ammonia levels in the N group in the period of HI compare to C. The present cross-sectional study was design to investigate the relationship between exercise intensity and RMR in four groups. The focus of this investigation was to compare RMR in aerobically trained (T), control (C), obese (O) and NIDDM (N) group. The relationship among RMR, exercise intensity and percent body fat would best be investigated using Meta Lyzer 3B, MMX3B and body composition analyzer. Each subject completed measurement of percent body fat, RMR, hormone in the period of maximal oxygen uptake exercise (MA), high intensity exercise (HI), and low intensity exercise (LI). From the results, High and Low intensity of exercise, there was a trend for an increased RMR (kcal/day) in the trained groups and control group (in case of LI) not for the obese and N groups. This is best explained not by the reduced percent body fat but by the highly induced energy expenditure (during exercise and post exercise energy expenditure) and increased T4, Free T3, and Free T4 hormonal levels in the low intensity exercise for the T group and sometimes C group.

중년여성에 대한 영양교육 및 운동 프로그램이 신체조성, 혈청지질 농도 및 휴식대사량에 미치는 영향 (Effect of Body Composition, Serum Lipid Level and Resting Metabolic Rate by Nutritional Education and Exercise Program in Middle Aged Women)

  • The purpose of this study was to investigate the change of body composition, biochemical index level, resting metabolic rate and daily activities by the public health center's health-promoted program in overweight middle aged women. The workout program of 12 subjects constituted by exercise(aerobic exercise & resistance training) and nutritional education for 10 weeks. As the results, there was a significant decrease in all the weight(59.0$\pm$8.9 vs 57.2$\pm$8.3kg, p<0.05), body fat percentage(33.4$\pm$4.3 vs 30.5$\pm$4.5%, p<0.05) and body fat mass(19.8$\pm$5.4 vs 17.7$\pm$4.9kg, p<0.05) after program. Fat free mass and RMR which tend to be higher after than before program were found not to be significantly. In addition, the serum HDL-cholesterol(54.3$\pm$8.1 vs 59.7$\pm$13.9mg/dl, p<0.05) and TG level(65.2$\pm$418 vs 89.4$\pm$65.6mg/dl, p<0.05) of the subjects increased at the end of the study significantly and significant correlation found between total-cholesterol and TG before program. When the other laboratory parameter were compared before and after program, hematocrit(35.1$\pm$3.3 vs 39.7$\pm$3.3%, p<0.001), platelet(225.9$\pm$49.6 vs 244.3$\pm$49.7cell/ul, p<0.05) and fasting glucose(88.0$\pm$6.5 vs 76.6$\pm$9.7mg/dl, p<0.001) increased significantly but hemoglobin(13.3$\pm$1.3 vs 12.9$\pm$1.3mg/dl, p<0.01) decreased. And in activities analysis, 'moderate activity(18.8 vs 5.8minutes, p<0.05)' clearly showed a significant decrease but 'severe activity(17.7 vs 11.4minutes, p<0.05)' showed a increase. These results suggest that continuous health-promoted program involved exercise and nutritional education at the public health center are effective to reduce body fat and to improve resting metabolic rate(RMR). Further research is required to develop more effective public health program for the middle aged women in community.

Using physical activity levels to estimate energy requirements of female athletes

  • [Purpose] The goal of this study was to review data on physical activity level (PAL), a crucial index for determining estimated energy requirement (EER), calculated as total energy expenditure (TEE, assessed with doubly labeled water [DLW]) divided by resting metabolic rate (RMR, PAL = TEE/RMR) in female athletes and to understand the methods of assessing athletes' EERs in the field. [Methods] For the PAL data review among female athletes, we conducted a PubMed search of the available literature related to the DLW method. DLW studies measuring TEE and RMR were included for the present review. [Results] Briefly, the mean PAL was 1.71 for collegiate swimmers with moderate training, which was relatively low, but the mean PAL was 3.0 for elite swimmers during summer training camp. This shows that PAL can largely vary even within the same sport depending on the amount of training, and the differences in PAL were remarkable depending on the sport. Aside from the DLW method, there is currently no research tool related to athletes' EERs that can be used in the field.
