The Significance of the Analytical Sciences In Environmental Assessment

  • The quality of human life is directly related to the quality of the environment. To assess environmental quality we must first determine the MCLG(Maximum Contaminant Level Goal), MCL(Maximum Contaminant Level), environmental impact and so on. The MCLG is the concentration at which no known adverse health effects occur. The MCLG is determined by risk assessment identifying which process is hazardous assessing, dose-response, human exposure, and characteristics of risk. With consideration of analytical methods, treatment technology, cost and regulatory impact, the MCL is set as close to the MCLG as possible. In this way, determination of the concentration and national distribution of contaminants is important for assessment of environmental quality The analytical sciences pose potential problems in assessing environmental quality. Continuing improvement in the performance of analytical instruments and operating technique has been lowering the limits of detectability. Contaminant concentration below the detection limit has usually been reported as ND(Not-Detected) and this has often been misunderstood as equivalent to zero. Because of this, more the contaminant concentration in the past was below the detection limit, whereas contaminants can be quantified now even though the contaminant concentration might remain the same or may even have decreased. In addition, environmental sampling has various components due to heterogeneous matrices. These samples are used to overestimate the concentration of the contaminant due to large variability, resulting in excess readings for MCL. In this paper, the significance of the analytical sciences is emphasized in both a conceptual and a technical approach to environmental assessment.

Discrimination of three highly toxic Alexandrium tamarense(Dinophyceae) isolates using FITC-conjugated lectin probesLectin probe

  • Lectin binding assay was conducted on 3 A. tamarense isolates (AT-A, AT-2 and AT-6). Fatty acid composition of all 3 isolates was analyzed, and total carotenoid content and $\beta$-carotene were also determined. AT-A and AT-2 treated with different lectins in this study showed the positive response, whereas potentially toxic AT-6 did not bind DBA lectin, regardless of different growth phase, but conjugated ConA, PNA, RCA, SBA, UEA and WGA. It is possible that DBA is a desirable method for rapid and easy discrimination of highly toxic A. tamarense. AT-A, AT-2 and AT-6 comprised saturated fatty acids (49.0-61.9%), monounsaturated fatty acids (8.0-20.5%) and polyunsaturated fatty acids (23.2-30.5%). In particular, 22:6 (n-3) polyunsaturated fatty acid in AT-6 had a high abundance, compared with AT-A and AT-2. However, carotenoid content and $\beta$-carotene were not contributed to discriminate each isolate. Due to variability in biochemical composition at different isolates, possibly DBA and 22:6 (n-3) polyunsaturate fatty acid provide a good information for discrimination of AT-6.

목건초의 정량급여시 소 반추행동의 변이성 (Variability of the Rumination-Behaviour in Strrrs fed a Constant Amount of Hay)

  • The variation of rumination response in steers fed a same amount of orcharugrass hay was investigated. With three steers (6, 12, 24 months) of the Japanese Black Breed, rumination behaviour was measured continuously during a 5 days period by masseter EMG telemetry system. The results are summarized as follows: 1. The average daily rumination time of respective animals were 438,447 and 433 min. with small daily variation both within and among animal. But the rumination time per DM kg of hay consumed varied from 66 to 138 min., and calf spent longer than older calf. 2. Actural chewing time showed small variation, and percentage of actural chewing time spent in rumination time was from 88 to 93% on individual average. 3. The daily number of chewing in rumination was approximately 25,000 to 30,000 with large individual differences. The chews per DM kg intake varied from 3,800 to 9,600, and calf chewed more than older calf. 4. The average rate of chewing per 100 sec. (chewing speed), there was a large difference between animals i.e. 104, 114, 131 respectively, but very little variance between days in individual. 5. No relation between day to day variations in eating time and rumination behavior, but significantly positive correlation was observed in the relation among rumination time, chewing time, No. of boluses and No. of chews. 6. Active time of day in rumination altered considerably day to day, but variation in the amount of rumination per day as expressed by the rumination time was relatively small.

HAS-Analyzer: Detecting HTTP-based C&C based on the Analysis of HTTP Activity Sets

  • Because HTTP-related ports are allowed through firewalls, they are an obvious point for launching cyber attacks. In particular, malware uses HTTP protocols to communicate with their master servers. We call this an HTTP-based command and control (C&C) server. Most previous studies concentrated on the behavioral pattern of C&Cs. However, these approaches need a well-defined white list to reduce the false positive rate because there are many benign applications, such as automatic update checks and web refreshes, that have a periodic access pattern. In this paper, we focus on finding new discriminative features of HTTP-based C&Cs by analyzing HTTP activity sets. First, a C&C shows a few connections at a time (low density). Second, the content of a request or a response is changed frequently among consecutive C&Cs (high content variability). Based on these two features, we propose a novel C&C analysis mechanism that detects the HTTP-based C&C. The HAS-Analyzer can classify the HTTP-based C&C with an accuracy of more than 96% and a false positive rate of 1.3% without using any white list.

Clq-coated ELISA법을 이용한 정맥용 면역글로불린제제의 항보체성 측정 (Clq-Coated Microtitre Enzyme-linked Immunosorbent Assay for Measuring the Anticomplementary Activity of Intravenous Immunoglobulin Preparations)

  • The quality of an intravenous immunoglobulin preparation (IVIG) is reflected by the degree of nonspecific activation of complements, the so-called anticomplementary activity (ACA). ACA of aggregates in IVIG was investigated using method by the European Pharmacopoeia and Clq-coated microtiter enzyme-linked immunosorbent assay (ELISA). Both the EP method and the ELISA method showed a dose response curve with the amount of complements bound increasing with the percentage content of aggregates in immunoglobulin standard. The correlation between the two tests was good (r=0.96, r=0.99). However, the correlation was not found when the ACA (EP method) of IVIG product was compared with its aggregate percentage. These results emphasize that the method of aggregate formation affects ACA and that estimation of the percentage distribution of aggregates by HPLC may not reflect ACA. In analysing WIG product for Clq binding activity test with the ELISA, the result by using Protein A-HRP correlated with aggregate percentage (r=0.84). But the correlation decreased (r=0.48) when the result used Protein A-AP(having poorer sensitivity than HRP) was compared with aggregate percentage. As a result, some variation between the two methods, due to differences in assay principles, is to be expected. However, ELISA technique has the advantage in that it is easier to perform, more precise and less subject to reagent variability, and is the more suitable screening method than HPLC analysis.

  • We are exposed to the various external stimuli input from the environment, which cause emotional changes based on the characteristics of the stimuli. Unfortunately, there are noquantitative results on relationship between human sensibility and the characteristics of physiological signals. The objective of this study was to quantify EEG signals evoked by visual stimulation based on the assumption that the analysis of the variability on the characteristics of the EEG waveform may provide the significant information regarding changes in psychological states of the subject. Seven university students were participated in this study. The experiment was devised with eleven experimental conditions, which are control and ten different types of visual stimulation based on IAPS(International Affective Picture Systems). Seven subjects were used to obtain EEGs while introducing visual stimulation. Wavelet transformation was employed to analyze the EEG signals. Most Positive and negative emotional response were pairely compared. The results showed that the reconstructed signals at the decomposition level revealed the different energy value on the EEG signals. Also, general patterns of EEG signals in rest state compare with negative and positive stimulus were found. This study could be extended to estabish an algorithm which distinguishes psychophysiological states of the subjects exposed to the visual stimulation.

Comparison of the seismic performance of existing RC buildings designed to different codes

  • Static pushover analyses of typical existing reinforced concrete frames, designed according to the previous generations of design codes in Greece, have established these structures' inelastic characteristics, namely overstrength, global ductility capacity and available behaviour factor q, under planar response. These were compared with the corresponding demands at the collapse limit state target performance point. The building stock considered accounted for the typical variability, among different generations of constructed buildings in Greece, in the form, the seismic design code in effect and the material characteristics. These static pushover analyses are extended, in the present study, in the time history domain. Consequently, the static analysis predictions are compared with Incremental Dynamic Analysis results herein, using a large number of spectrum compatible recorded base excitations of recent destructive earthquakes in Greece and abroad, following, for comparison, similar conventional limiting failure criteria as before. It is shown that the buildings constructed in the 70s exhibit the least desirable behaviour, followed by the buildings constructed in the 60s. As the seismic codes evolved, there is a notable improvement for buildings of the 80s, when the seismic code introduced end member confinement and the requirement for a joint capacity criterion. Finally, buildings of the 90s, designed to modern codes exhibit an exceptionally good performance, as expected by the compliance of this code to currently enforced seismic provisions worldwide.

생체신호 측정을 통한 운전자의 감정적 안정상태 평가 (Assessment of Driver's Emotional Stability by Using Bio-signals)

  • Objective: The aim of this study is to introduce a methodology to assess driver's emotion stability by using bio-signals. Background: Psychophysiological analysis of driver's behavior has been conducted to improve the driving safety and comfort. However, the variability of bio-signal and individual difference made it difficult to assess the psychophysiological status of drivers that can be expressed as emotional stability of drivers. Method: Two experimental studies were reviewed and summarized. New techniques assessing emotional stability of drivers were explained. Statistical concept and multidimensional space were used to identify the emotionally stable conditions. Conclusion: Psychophysiological approach can provide information of driver's emotional status. The experimental methodology and algorithm used in this study showed the possibility of parameterization of psychophysiological response. Application: Currently measured statistical and geometrical data can be further applied to develop an interactive device monitoring and reacting driver's emotion when driver experiences emotionally unstable or uncomfortable situation.

수간호사의 관리역량과 전문직 자율성 인식이 간호업무성과에 미치는 영향 (Effects of Head Nurses' Managerial Competencies and Professional Nurse Autonomy in Nursing Performance)

  • Purpose: The aim in this study was to identify head nurses' nursing performance according to their managerial competencies and professional nurse autonomy. Methods: From January 15 to January 31, 2013 a survey was distributed to 200 head nurses working in 5 tertiary hospitals in B city and J city. The response rate was 80% (160 responses). The survey content included the Managerial Competencies Scale (MCS), Schutzenhofer professional nurse autonomy Scale (SPNA), and Nursing Performance Scale (SPNA). Results: The score for MCS was 4.53, for SPNA, 177.05, and for SPNA, 4.62. Nursing performance was related to managerial competencies (${\beta}=0.741$; p<.001; SE=0.059) and professional autonomy (${\beta}=0.135$; p=.010; SE=0.001). These factors accounted for 64.8~67.3% of the variability in nursing performance. Conclusion: Results indicate that head nurses' performance would be improved through increased nursing managerial competencies and expansion of professional autonomy suggesting a need to develop work systems designed to achieve high managerial competency and professional autonomy in head nurses.

Pounding analysis of RC bridge considering spatial variability of ground motion

  • To investigate the seismic pounding response of long-span bridges with high-piers under strong ground motions, shaking table tests were performed on a 1/10-scaled bridge model consisting of three continuous spans with rigid frames and one simply-supported span. The seismic pounding responses of this bridge model under different earthquake excitations including the uniform excitation and the traveling wave excitations were experimentally studied. The influence of dampers to the seismic pounding effects at the expansion joints was analyzed through nonlinear dynamic analyses in this research. The seismic pounding effects obtained from numerical analyses of the bridge model are in favorable agreement with the experimental results. Seismic pounding effect of bridge superstructures is dependent on the structural dynamic properties of the adjacent spans and characteristics of ground motions. Moreover, supplemental damping can effectively mitigate pounding effects of the bridge superstructures, and reduce the base shear forces of the bridge piers.
