The Effects of Alkaloid Fraction of Korean Ginseng on the Radiation-Induced DNA Strand Breaks (방사선 조사에 의한 DNA Double Strand Breaks의 생성 및 회복에 미치는 인삼 알칼로이드 분획의 효과)

  • Purpose : To investigate the effect of alkaloid fraction from Korean ginseng on radiation-induced DNA double strand breaks (dsb) formation and repair in murine lymphocytes Materials and Methods : We used the neutral filter elution technique to assay $^{60}Co\;{\gamma}$ ray-induced DNA double strand breaks formation and repair in C57BL/6 mouse spleen lymphocytes for evaluating the dose-response relationship in the presence of alkaloid fraction as a radioprotective agent. The lymphocytes were stimulated with Phytohemagglutinin (PHA, 2 u g/ml) to label $^3[H]-thymidine.$ Isotope-labelled lymphocytes in suspension were exposed to 100 Gy at $0^{\cdot}C$ in the alkaloid fraction-treated group and elution procedure was performed at PH 9.6. The extents of formation of radiation-induced DNA double strand breaks and repair were compared respectively via strand scission factor (SSF) and relative strand scission factor (RSSF). Results: Alkaloid fraction reduced the formation of double strand breaks with dose modification factor of 2 15, compared to control group Rejoining of DNA dsb appeared to take place via two components. The first fast component was completed within 20.4 minutes, but the second slow component was not completed until 220.2 minutes after irradiation. About $30\%$ of dsb formed by irradiation was ultimately unrejoined despite the administration of alkaloid fraction. The administration of alkaloid fraction had a great effect on the second slow component of repair; the half-time of fast component repair was not changed, but that of slow component was 621.8 minutes. Conclusion: Neutral filter elution assay Proved to be a very effective method to quantitate the extents of DNA dsb formation and its repair. By using this technique, we were able to evaluate the efficiency of alkaloid fraction from Korean ginseng as a valuable radioprotector. Alkaloid fraction can be used prophylactically to prevent or ameliorate the severe radiation damages in workers and neighbors around the atomic power plants. For more refined study, however, more advanced purification of alkaloid fraction wil be needed in the near future.

Inelastic behavior of systems with flexible base

  • This study explores the inelastic behavior of systems with flexible base. The use of a single degree of freedom system (ESDOF) with equivalent ductility to represent the response of flexible base systems is discussed. Two different equations to compute equivalent ductility are proposed, one which includes the contribution of rigid body components, and other based on the overstrength of the structure. In order to asses the accuracy of ESDOF approach with the proposed equations, the behavior of a 10-story regular building with reinforced concrete (RC) moment resisting frames is studied. Local and global ductility capacity and demands are used to study the modifications introduced by base flexibility. Three soil types are considered with shear wave velocities of 70, 100 and 250 m/s. Soil-foundation stiffness is included with a set of springs on the base (impedance functions). Capacity curves of the building are computed with pushover analysis. In addition, non linear time history analysis are used to asses the ductility demands. Results show that ductility capacity of the soil-structure system including rigid body components is reduced. Base flexibility does not modify neither yield and maximum base shear. Equivalent ductility estimated with the proposed equations is fits better the results of the numerical model than the one considering elastoplastic behavior. Modification of beams ductility demand due to base flexibility are not constant within the structure. Some elements experience reduced ductility demands while other elements experience increments when flexible base is considered. Soil structure interaction produces changes in the relation between yield strength reduction factor and structure ductility demand. These changes are dependent on the spectral shape and the period of the system with fixed and flexible base.

Seismic Improvement of Staggered Truss Systems using Buckling Restrained Braces (비좌굴 가새를 이용한 스태거드 트러스 시스템의 내진성능향상)

  • In this paper the seismic performances of 4, 10, and 30-story staggered truss systems (STS) were evaluated by observing the force-displacement relationship up io failure. The results were compared with the seismic performance of conventional moment resisting frames and braced frames. According to the analysis results, the STS showed relatively satisfactory lateral load resisting capability. However, in the mid- to high-rise STS, plastic hinges formed first at the chords were transferred to vertical members of the vierendeel panels, which formed a week link and subsequently leaded to brittle collapse of the structure. Therefore to enhance the ductility of STS it would be necessary to reinforce the vertical bracing members of the virendeel panels so that the plastic hinges, once toned in cord members of a virendeel panel, spread out to virendeel panels of neighboring stories.

Analysis on In-Plane Behavior of Unreinforced Masonry Walls (비보강 조적벽체의 면내거동 해석)

  • A series of unreinforced masonry(URM) walls were analytically investigated by FEM for a limited version of seismic in-plane performance. For this, URM walls were assumed to be continum and modeled as isotropic plane stress elements, within which the nature of cracking was propogated. Accordingly, behavioral mode of cracking in URM was modeled by smeared-crack approach. Total of 70 cases were considered for various parameters such as axial load ratio, aspect ratio and effective section area ratio due to the existence of opening, etc. The analysis results indicate that these parameters significantly and interactively influence over the ultimate strength of URM walls. Finally, it is suggested that the response modification factor for URM adopted in the current Korean Standard should be validated considering various forms of brittleness and probable failure modes in URM.

Correlation of Experimental and Analytical Inelastic Responses of A 1:12 Scale 10-Story Reinforced Concrete Frame with Non-seismic Details (비내진 상세를 가진 1:12축소 10층 R.C.골조의 비선형 거동에 대한 실험과 해석의 상관성)

  • The pushover analysis technique is now attracting the world-wide interest for the prediction of elastic and inelastic behavior of structures in the seismic evaluation of existing buildings. However, the reliability of this analysis technique has not been fully checked by the test results in the case of structures with nonseismic details. The objective of this study is to verify the correlation between the experimental and analytical responses of a 1:12 scale 10-story reinforced concrete frame with non-seismic details by using DRAIN-2DX program and the test results performed previously. It is concluded from this comparison that the overall responses such as the relations between story shear versus interstory drift and the local deformations such as plastic rotations can be predicted with quite high reliability.

  • In this paper, a method is proposed for establishing an approximate prediction model of rotor-dynamics through modal testing. In particular, the proposed method is applicable to systems that cannot be established according to conventional methods owing to the absence of information regarding the dimensions and material of the rotor-bearing system. The proposed method is demonstrated by employing a motor dynamometer driven by a 1 MW class induction motor without dimension and material information. The proposed method comprises a total of seven steps, wherein an initial model is established by incorporating approximate dimensions and material information, and the model is improved on the basis of the natural frequency characteristics of the system. During model improvement, the modification factor is introduced for adjusting the elastic modulus and shear modulus of the system. Analysis of critical speed and imbalance response indicates that the separation margin is 67% and the maximum vibration amplitude is less than the amplitude limit of 0.032 mm under the API 611 standard, which means that the motor dynamometer can stably operate at a rated speed of 1800 rpm. Hence, the obtained results validate the feasibility of the proposed method. Furthermore, for broad usage, it is necessary to accordingly apply and validate the proposed method for various rotor-bearing systems.

Seismic Performance of Square RC Column Confined with Spirals (나선철근으로 횡구속된 정사각형 RC 기둥의 내진성능)

  • The objective of this research is to investigate the seismic performance and flexure-shear behavior of square reinforced concrete bridge piers with solid and hollow cross section. Test specimens were nonseismically designed with the aspect ratio 4.5 Two reinforced concrete columns were tested under constant axial load while subjected to lateral load reversals with increasing drift levels. Longitudinal steel ratio was 2.217 percent. The transverse reinforcement ratio As/($s{\cdot}h$), corresponding to 58 percent of the minimum lateral reinforcement required by Korean Bridge Design Specifications for seismic detailing, which represent existing columns not designed by the current seismic design specifications or designed by limited ductility concept. This study are to provide quantitative reference data for the limited ductility design concept and tendency for performance or damage assessment based on the performance levels such as cracking, yielding, collapse, etc. Failure behavior, ultimate displacement/drift ratio, displacement ductility, response modification factor, equivalent viscous damping ratio, residual deformation, effective stiffness, plastic hinge length, strain of reinforcements and nonlinear analysis are investigated and discussed in this paper.

Seismic Performance of Octagonal Flared RC Columns using Oblong Hoops (장방형 띠철근을 이용한 팔각형 플레어 RC 기둥의 내진성능)

  • Transverse steel bars are used in the plastic hinge zone of columns to insure adequate confinement, prevention of longitudinal bar buckling and ductile behavior. Fabrication and placement of rectangular hoops and cross-ties in columns are difficult to construct. Details of reinforcement for rectangular section require a lot of rectangular hoops and cross-ties. In this paper, to solve these problems, the new lateral confinement method using oblong hoop is proposed for the transverse confinement of the flared column. It can be the alternative for oblong cross-section and flared column with improved workability and cost-efficiency. The final objectives of this study are to suggest appropriate oblong hoop details and to provide quantitative reference data and tendency for seismic performance or damage assessment based on the drift levels such as residual deformation, elastic strain energy. This paper describes factors of seismic performance such as ultimate displacement/drift ratio, displacement ductility, response modification factor, equivalent viscous damping ratio and effective stiffness.

Seismic Characteristics of Hollow Rectangular Sectional Piers with Reduced Lateral Reinforcements (횡방향철근이 감소된 중공사각단면 교각의 내진거동 특성)

  • The seismic design concept of RC bridges is to attain the proper ductility of piers, yielding a ductile failure mechanism. Therefore, seismic design force for moment is determined by introducing a response modification factor (R), and lateral reinforcements to confine core concrete are specified in the current design code. However, these design provisions have irrationality, which results in excessive amounts of lateral reinforcements for columns in Korea, which are generally designed with large sections. To improve on these provisions, a new design method based on seismic performance has been proposed. To apply this to hollow sectional columns, however, further investigations and improvements must be performed, due to the different seismic behaviors and confinement effects. In this study, hollow sectional columns with different lap-splice of longitudinal bars and lateral reinforcements have been tested. Seismic characteristics and performance were investigated quantitatively. These research results can be used to derive a performance-based design for hollow sectional columns.

Seismic Performance Evaluation of Steel Moment Frames in Korea Using Nonlinear Dynamic Analysis (비선형동적해석을 통한 국내 철골 모멘트골조의 내진성능 평가)

  • Domestic steel moment resisting frames were designed in accordance with the former KBC2005 and the current KBC2009, and then their seismic performance was evaluated in accordance with FEMA355F by utilizing nonlinear dynamic analysis. The results from the procedure in FEMA355F were different with those from the capacity spectrum method utilizing nonlinear static push-over analysis. In particular, the domestic steel moment resisting frames have a weak panel zone, so their behavior can be estimated more precisely by nonlinear dynamic analysis. The domestic steel moment resisting frames satisfied the performance goal if located at a site class $S_B$ or $S_C$, regardless of the story number and the response modification factor. However, if they are located at a site class $S_D$ or $S_E$, performance goal satisfaction cannot be guaranteed. No matter what standard is used for the design, KBC2005 or KBC2009, the domestic steel moment resisting frames may possess satisfactory seismic performance if the site condition is relatively good.
