Plasmid-Mediated Arsenical and Antimonial Resistance Determinants (ars) of Pseudomonas sp. KM20

  • Bacteria have evolved various types of resistance mechanism to toxic heavy metals, such as arsenic and antimony. An arsenical and antimonial resistant bacterium was isolated from a shallow creek draining a coal-mining area near Taebaek City, in Kangwon-Do, Korea. The isolated bacterium was identified and named as Pseudomonas sp. KM20 after biochemical and physiological studies were conducted. A plasmid was identified and its function was studied. Original cells harboring the plasmid were able to grow in the presence of 15 mM sodium arsenite, while the plasmid-cured (plasmidless) strain was sensitive to as little as 0.5 mM sodium arsenate. These results indicated that the plasmid of Pseudomonas sp. KM20 does indeed encode the arsenic resistance determinant. In growth experiments, prior exposure to 0.1 mM arsenate allowed immediate growth when they were challenged with 5 mM arsenate, 5 mM arsenite, or 0.1 mM antimonite. These results suggested that the arsenate, arsenite, and antimonite resistance determinants of Pseudomonas sp. KM20 plasmid were indeed inducible. When induced, plasmid-bearing resistance cells showed a decreased accumulation $of\;73^As$ and showed an enhanced efflux $of\;^73As$. These results suggested that plasmid encoded a transport system that extruded the toxic metalloids, resulting in the lowering of the intracellular concentration of toxic oxyanion. In a Southern blot study, hybridization with an E. coli R773 arsA-specific probe strongly suggested the absence of an arsA cistron in the plasmid-associated arsenical and antimonial resistance determinant of Pseudomonas sp. KM20.

Inheritance of Resistance to Nuclear Polyhedrosis Virus in Silkworm, Bombyx mori

  • Inheritance pattern of resistance to Bombyx mori nuclear polyhedrosis virus (BmNPV) was studied in an Indian silkworm stock TX by single back-cross test method. The resistant parent [TX], susceptible parent [HM], their Fl, F2, and Fl progeny back-crossed to TX [BC(R)] and HM [BC(S)] were inoculated per os with a fixed concentration of BmNPV($0.5{\times}10^{th} PIB/ml$) on the first day of second stadium. The cumulative mortality was recorded until day $10^{\times}$ post-inoculation. The results show that the resistance to BmNPV in TX fellow mono Mendelian inheritance pattern. The resistance dominated over the susceptibility at Fl. At F2, the resistant and susceptible offspring segregated in 3:1 ratio whereas at BC(S), the resistant and susceptible offspring segregated in 1:1 ratio. The response of BC(R) was more or less like the resistant parent TX which confirms the involvement of a major dominant gene conferring resistance to BmNPV in TX. The possible mechanism of inheritance of resistance in TX is discussed.

Investigation of Spudcan Penetration Resistance in Layered Soil Deposits

  • A suite of 3D large deformation finite element (FE) analyses was performed to investigate the load transfer mechanism and penetration resistance of spudcan foundations in heterogeneous soil profile consisting of sand and clay. The Elasto-Plastic models following Mohr-Coulomb and Tresca failure criteria were adopted for sand and clay, respectively. The accuracy of the numerical model was validated against centrifuge test measurements. The dense sand behavior with dilation is modeled using the non-associated flow rule. An investigation study consisting of key parameters, which includes variation in soil stratigraphy (sand-clay, sand-clay-sand), strength parameters of sand and clay (��' and su) and normalized height ratio of the sand layer (Hs/D) was conducted to assess the penetration behavior of spudcan. Based on calculated outputs, it was demonstrated that these parameters have a significant influence on the penetration resistance of spudcan. The calculated penetration resistance profiles are compared with the published (sand overlying clay) analytical model. It is confirmed that for the case of two-layer soil, the available theoretical model provides an accurate estimate of peak penetration resistance (qpeak). In the case of three-layer soil, the presence of a third stiff layer affects the penetration resistance profile due to the squeezing of the soil.

  • Technology trend and application of laser fuse, anti-fuse, and eFUSE as well as the structure, programming mechanism, and reliability of eFUSE have been reviewed. In order to ensure eFUSE reliability in the field, a sensing circuit trip point consistent with the fuse resistance distribution, process variation, and device degradation in the circuit such as hot carrier or NBTI, as well as fuse resistance reliability must be considered to optimize and define a reliable fuse programming window.

  • A new kind of bulb called Stern-End-Bulb(SEB) for the improvement of the after part of fine hull forms was developed. The reduction of wave resistance and the improvement of the powering performance for the ship with SEB were shown by the ship model tests, At the same time, the characteristics of wave in the vicinity of the stern and the mechanism of the resistance reduction by SEB were investigated. By the systematical variation of the SEB size, the optimum size of SEB was obtained.

  • Visceral fat accumulation is known to be an evident clinical index for the insulin resistance related with obesity. Patients with excessive accumulation of visceral fat frequently suffered from metabolic disorder, such as hyperlipidemia, hypertension, and glucose intolerance. However, molecular mechanism for the pathogenesis of obesity-accompanied metabolic disorders has not been fully elucidated. It has been clarified that adipocytes in visceral fat area have different functions from subcutaneous fat area, and these differences might contribute the pathological significance of excessive accumulation of visceral fat for the accompanied insulin resistance and hyperinsulinemia.

  • For the evaluation of corrosion resistance, Al 6061-T6 alloy was welded by Friction Sti. Welding(FSW) and Gas Metal Arc Welding(GMAW) evaluated by Tafel method and immersion test. The Tafel and immersion test results indicated that GMA weld was severely attacked compared with those of friction stir weld. It may be mainly due to the galvanic corrosion mechanism act on the CMA weld.

  • Six of scaled concrete circular columns were experimentally investigated for the contribution of arch action to the column lateral resistance. For this the specimens with the variation of tranverse hoop steel spacing were tested in absence of axial loading All specimens showed the flexure governing behavior pattern irrelevant to transverse hoop spacing. This indicates that the role of arch action should be understood as the intermediate mechanism causing the interaction between shear and flexural mechanisms A simple truss model was proposed to qualitatively explain this notation but further study is needed to advance its application to general columns.

  • High value sheet resistance (Rs' 1K-33K\ulcorner/) polysilicon resistors were fabricated using double ion implantation with boron as the major dopant and phosphorus compensation. It is observed that Rs sensitivity to the net doping concentration is decreased by one order of magnitude compared to the conventional (boron implanted)polysilicon resistors. The temperature co-efficient of resistance (TCR) measured between 25\ulcorner and 125\ulcorner shows equivalent values to those of non-compensated resistors for the same Rs. A qualitative electrical conductiion mechanism for compensated polysilicon resistor is proposed, based on the existing grain boundary charge trapping theory.
