Protective effect of Kundur (Boswellia serrata) against mercuric chloride induced nephrotoxicity in rats

  • The effect of Kundur (oleo-gum resin of Boswellia serrata) and its fractions viz: methanol soluble (MS) and methanol insoluble (MI) were investigated on mercuric chloride induced nephrotoxicity in rats. The animals of group I and II were administered with 1% carboxymethyl cellulose (CMC) (1,000 mg/kg, p.o.) and the animals of groups III, IV and V were administered with Kundur (1,000 mg/kg, p.o.), MS (650 mg/kg, p.o.) and MI (350 mg/kg, p.o.) respectively for ten days. On 10th day a single dose of the mercuric chloride (3 mg/kg, 5.c.) was also administered to all groups except the group I which received only 1% CMC (10 ml/kg, p.o.). After two days of mercuric chloride administration the blood samples of each animal were collected and analyzed for blood urea nitrogen and creatinine concentration. Rats fed with Kundur and MI fraction showed a significant prevention in the rise of serum markers while MS failed to prevent the rise of these serum makers. These results suggest that Kundur and MI fraction may have potential to reduce the nephrotoxicity in rats.

Engineering Properties of Permeable Polymer Concrete for Pavement Using Polypropylene Fiber (폴리프로필렌섬유를 혼입한 포장용 투수성 폴리머 콘크리트의 공학적 성질)

  • Research on permeable pavement like asphalt and concrete pavement with porous structure has been increasing due to environmental and functional need such as reduction of run off and flood. This study was performed to evaluate void ratio, permeability coefficient, and compressive strength of permeable polymer concrete (PPC) using crushed and recycled coarse aggregate that is obtained from waste concrete. Also, 6 mm length of polypropylene fiber was used to increase toughness and interlocking between aggregate and aggregate surrounded by binder. Binder and filler used were unsaturated polyester resin and CaCO3, respectively. The mix proportions were determined to satisfy the requirement for the workability and slump according to aggregate sizes 5~10 mm. In the test results, regardless of kinds of aggregates and fiber contents, the void ratio, permeability coefficient and compressive strength of all types of PPC showed the higher than the criterion of porous concrete that is used in permeable pavement in Korea. Also, strengths of PPC with increase polypropylene fiber volume fraction showed slightly increased tendency due to increase binder with increase of fiber volume fraction. Accordingly, polypropylene fiber and recycled coarse aggregate can be used for permeable pavement.

Identification of Benzidine Metabolites in Rats by Gas Chromatography/Mass Selective Detector and its Toxicity in vitro (Gas-Chromatography/Mass Selective Detector를 사용하여 쥐의 뇨시료 중 benzidine 대사체의 확인 및 in vitro 독성)

  • Metabolism study of the dye, benzidine, was performed by gas chromatography-mass selective detector (GC/MSD) in the urine of rats orally administered 100 mg/kg benzidine. Urine samples were collected in metabolic cages for 0-24, 24-48, and 48-72 hrs. Ten ml of the urine was extracted with XAD-2 resin and the XAD-2 column was eluted with methanol. After evaporation, benzidine and its metabolites were extracted with diethyl ether (for non-conjugated fraction). For conjugated metabolites, $\beta$-glucu-ronidase was added to the aqueous layer that was incubated for 1 hr at 5$0^{\circ}C$ and the aqueous layer was extracted as in non-conjugated fraction. Aliquot of trimethylsilylated derivatives was applied to the GC/MSD. The mutagenicity of benzidine and its acetylated metabolites was tested by histidine/reversion assay. Five metabolites observed and confirmed either by electron impact and chemical ionization modes of the GC/MSD, or authentic compounds were monoacetyl-, diacetyl-, hydroxyacetyl-, hydroxydiacetyl-, and hydroxy-benzidine. Monoacetyl-benzidine was more potent than benzidine in histidine/reversion assay. This data indicates that monoacetylation of benzidine may be one of the metabolites produced in metabolic activation process.

Separation and Purification of Antioxidant Peptide from Fermented Whey Protein by Lactobacillus rhamnosus B2-1

  • In this study, a antioxidant activity peptide fraction was separated and purified from metabolites of whey protein fermented by Lactobacillus rhamnosus B2-1. The fermentation sample was separated by macroporous resin D101 and Sephadex G-15. The collected fractions were tested for antioxidant and antitumor activities. In order to test the antioxidant activity of fractions, Hydroxyl (·OH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS), and Oxygen Radical Absorbance Capacity (ORAC) were used. The final purified peptide B11 showed highest ABTS and ·OH radical scavenging rate by 84.36±1.89% and 62.43±2.64%, respectively, and had an ORAC activity of 1,726.44±2.76 μM Trolox equivalent/g. Further, the inhibitory effect of B11 on the proliferation of LoVo human colon cancer cells, KB and Cal-27 human oral cancer cells were enhanced with increasing concentrations of B11. B11 contains 51.421% amino acids, with Glu and Asp being the major constituents. In this study, we obtained peptide fraction B11 with antioxidant activity, which is promising for development.

  • Demand of glass fiber reinforced composites (GFRC) increased with developing aircraft and defense industries using resin transfer molding (RTM) process to produce complex product. In this research, wetting, interfacial, and mechanical properties were evaluated with different Cross-linking Density by Molecular Weight of Hardener. Epoxy resin as matrices was used bisphenol-A type and amine-type hardeners with different molecular weight. Specimens were manufactured via RTM and wetting property of resin and glass fiber (GF) mat was evaluated to viscosity of epoxy and injection time of epoxy matrix. Mechanical property of GFRC was determined via flexural strength whereas interfacial properties were determined by interlaminar shear strength (ILSS) and interfacial shear strength (IFSS). The difference in mechanical property depends upon the fiber weight fraction (wt %) of GFRC by RTM as well as the different Molecular Weight of Hardener.

Comparison of Selective Removal of Nitrate Ion in Constant Voltage and Constant Current Operation in Capacitive Deionization (축전식 탈염에서 정전압과 정전류 운전에 따른 질산 이온의 선택적 제거율 비교)

  • The adsorption characteristics of ions were evaluated for the nitrate-selective carbon electrode (NSCE) in accordance with power supply methods. The NSCE was fabricated by coating the surface of a carbon electrode with anion-exchange resin powders with high selectivity for the nitrate ion. Capacitive deionization (CDI) experiments were performed on a mixed solution of nitrate and chloride ion in constant voltage (CV) and constant current (CC) modes. The number of total adsorbed ions in CV mode was 15% greater than that in CC mode. The mole fraction of adsorbed nitrate ion showed the maximum 58%, though the mole fraction was 26% in the mixed solution. This indicates that the fabricated NSCE is highly effective for the selective adsorption of nitrate ions. The mole fraction of adsorbed nitrate was nearly constant value of 55-58% during the adsorption period in CC mode. In the case of CV mode, however, the values increased from the initial 30% to 58% at the end of adsorption. We confirmed that the current supplied to cell is important factor to determine the selective removal of nitrate.

Antiplatelet fraction from Ulmi cortex and its active components (유백피의 항혈전 활성 분획 및 유효성분에 관한 연구)

  • Objectives : The purpose of this study was to identify active fraction and components from antiplatelet Ulmi cortex extract. Methods : The 70% ethanol extract of Ulmi cortex was subjected to column chromatography over D101 resin and eluted with an 20% (W1), 30% (W2), 40% (W3), 50%(W4), 70% (W5), and 100% ethanol (W6) to yield 6 fractions. W6 was further fractioned and its active components were purified using semi-preparative HPLC. The isolated compounds were identified by MS and NMR, and their contents were simultaneously analyzed using HPLC/UV. Antiplatelet aggregation activities of the fractions and the compounds were evaluated using rat platelet-rich plasma in presence of collagen ($5{\mu}g/ml$), arachidonic acid (0.05 U/ml), or thrombin ($100{\mu}M$). Results : Among six fractions, W3 prominently inhibited platelet aggregation. At the concentration of $200{\mu}g/ml$, W3 strongly inhibited arachidonic acid- and collagen-induced platelet aggregations by 78.2% and 65.9%, respectivley, and weakly inhibited thrombin-inducded platelet aggregation by 32.6%. Catechin, epicatehin, and catechin-7-O-${\beta}$-D-glucopyranoside were isolated from W3 and their contents were revealed to be 15.1%, 0.87%, and 0.32%. Catechin and epicatechin at the concentrations of $100{\mu}M$ strongly inhibited collagen-induced platelet aggregation by 79.9% and 86.6%, respectively, but weakly inhibited arachidonic acid- and thrombin-induced platelet aggregations. Conclusions : A main active principle of anitplatelet Ulmi Cortex extract is W3 fraction, of which main active component is catechin considering its antiplatelet activity and content.

Vibration behavior of partially bio-sourced sandwich panel with orthogonally stiffened core: Analytical and experiment study

  • By the present paper, both experimental and analytical models have been proposed to study the vibration behavior of partially bio-sourced sandwich panel with orthogonally stiffened core. For a variable mass fraction of Alfa fibers from 5% to 15%, impregnated in a Medapoxy STR resin, this panel were manufactured by molding the orthogonally stiffened core then attached it with both skins. Using simply supported boundary conditions, a free vibration test was carried out using an impact hammer for predicting the natural frequencies, the mode shapes and the damping coefficient versus the fibers content. In addition, an analytical model based on the Higher order Shear Deformation Theory (HSDT) was developed to predict natural frequencies and the mode shapes according to Navier's solution. From the experimental test, we have found that the frequency increases with the increase in the mass fraction of the fibers until 10%. Beyond this fraction, the frequencies give relatively lower values. For the analytical model, variation of the natural frequencies increased considerably with side-to-thickness ratio (a/H) and equivalent thickness of the core to thickness of the face (hs/h). We concluded that, the vibration behavior was significantly influenced by geometrical and mechanical properties of the partially bio-sourced sandwich panel.

Phytochemical Analysis and Anti-cancer Investigation of Boswellia Serrata Bioactive Constituents In Vitro

  • Cancer is a major health obstacle around the world, with hepatocellular carcinoma (HCC) and colorectal cancer (CRC) as major causes of morbidity and mortality. Nowadays, there isgrowing interest in the therapeutic use of natural products for HCC and CRC, owing to the anticancer activity of their bioactive constituents. Boswellia serrata oleo gum resin has long been used in Ayurvedic and traditional Chinese medicine to alleviate a variety of health problems such as inflammatory and arthritic diseases. The current study aimed to identify and explore the in vitro anticancer effect of B. Serrata bioactive constituents on HepG2 and HCT 116 cell lines. Phytochemical analysis of volatile oils of B. Serrata oleo gum resin was carried out using gas chromatography-mass spectrometry (GC/MS). Oleo-gum-resin of B. Serrata was then successively extracted with petroleum ether (extract 1) and methanol (extract 2). Gas-liquid chromatography (GLC) analysis of the lipoidal matter was also performed. In addition, a methanol extract of B. Serrata oleo gum resin was phytochemically studied using column chromatography (CC) and thin layer chromatography (TLC) to obtain four fractions (I, II, III and IV). Sephadex columns were used to isolate ${\beta}$-boswellic acid and identification of the pure compound was done using UV, mass spectra, $^1H$ NMR and $^{13}C$ NMR analysis. Total extracts, fractions and volatile oils of B. Serrata oleo-gum resin were subsequently applied to HCC cells (HepG2 cell line) and CRC cells (HCT 116 cell line) to assess their cytotoxic effects. GLC analysis of the lipoidal matter resulted in identification of tricosane (75.32%) as a major compound with the presence of cholesterol, stigmasterol and ${\beta}$-sitosterol. Twenty two fatty acids were identified of which saturated fatty acids represented 25.6% and unsaturated fatty acids 74.4% of the total saponifiable fraction. GC/MS analysis of three chromatographic fractions (I,II and III) of B. Serrata oleo gum resin revealed the presence of pent-2-ene-1,4-dione, 2-methyl- levulinic acid methyl ester, 3,5- dimethyl- 1-hexane, methyl-1-methylpentadecanoate, 1,1- dimethoxy cyclohexane, 1-methoxy-4-(1-propenyl)benzene and 17a-hydroxy-17a-cyano, preg-4-en-3-one. GC/MS analysis of volatile oils of B. Serrata oleo gum resin revealed the presence of sabinene (19.11%), terpinen-4-ol (14.64%) and terpinyl acetate (13.01%) as major constituents. The anti-cancer effect of two extracts (1 and 2) and four fractions (I, II, III and IV) as well as volatile oils of B. Serrata oleo gum resin on HepG2 and HCT 116 cell lines was investigated using SRB assay. Regarding HepG2 cell line, extracts 1 and 2 elicited the most pronounced cytotoxic activity with $IC_{50}$ values equal 1.58 and $5.82{\mu}g/mL$ at 48 h, respectively which were comparable to doxorubicin with an $IC_{50}$ equal $4.68{\mu}g/mL$ at 48 h. With respect to HCT 116 cells, extracts 1 and 2 exhibited the most obvious cytotoxic effect; with $IC_{50}$ values equal 0.12 and $6.59{\mu}g/mL$ at 48 h, respectively which were comparable to 5-fluorouracil with an $IC_{50}$ equal $3.43{\mu}g/mL$ at 48 h. In conclusion, total extracts, fractions and volatile oils of B. Serrata oleo gum resin proved their usefulness as cytotoxic mediators against HepG2 and HCT 116 cell lines with different potentiality (extracts > fractions > volatile oil). In the two studied cell lines the cytotoxic acivity of each of extract 1 and 2 was comparable to doxorubicin and 5-fluorouracil, respectively. Extensive in vivo research is warranted to explore the precise molecular mechanisms of these bioactive natural products in cytotoxicity against HCC and CRC cells.

Formation and Removal of Trihalomethanes based on Characterization of Hydrophobic and Hydrophilic Precursors (전구물질의 소수성 및 친수성 특성에 따른 트리할로메탄의 생성과 제거에 관한 연구)

  • The Dissolved Organic Carbon (DOC) existing in a water includes both hydrophobic and hydrophilic substances however, most of the discussion focuses on hydrophobic substances. The hydrophobic fraction was easily removed by absorption or coagulation more than hydrophilic fraction. Therefore, control of the hydrophilic fraction is very important in water treatment process. This study is to determine the variation of DOC, the removal efficiency of DOC, and Trihalomethane formation potential (THMFP) after each stage of water treatment process by fractionating Natural Organic Matters (NOM) into hydrophobic and hydrophilic substance. DOC from raw water was fractionated at acidic pH (pH<2) using XAD 8 resin column, into two fraction : hydrophobic substance (i.e. humic substance) adsorbed on XAD 8 and hydrophilic substance which represent the organics contained in the final effluent. THMFP was carried out according to the following set condition: Cl2/DOC=4 mg/mg, incubation at $25^{\circ}C$ in darkness, pH 7 adjust with HCl or NaOH as necessary, and 72hour-contact time. THMs analyzed in this study were chloroform, bromodichloromethane, dibromochloromethan, and bromoform. Sewage was almost evenly split between the hydrophobic (56%) and hydrophilic fraction (44%). But, Aldrich humic substance (AHS) was found to contain less hydrophilics (14%) than hydrophobics (86%). The formation of THMs may depend on the source which is characterized by the composition of organic matters such as AHS and sewage. The THMFP yield of sewage and AHS were assessed as follows. The value of the THMFP reaction yield, AHS $172.65{\mu}g/mg$, is much higher than that of sewage $41.68{\mu}g/mg$. This illustrates possible significant difference in THMFP according to the component type and the proportion of organic matter existing in water source. Apparently AHS react with chlorine to produce more THMFP than do the smaller molecules found in sewage. Water treatment process may reduce THMFP, nevertheless residual DOC (the more hydrophilic substance) has significant THMFP. Further reduction in organic halide precursors requires application of alternative treatment techniques.
