The strobilurin fungicides, azoxystrobin and kresoxim-methyl, were investigated to know the biological half-lives and dissipation patterns in Korean melon under plastic film house condition. Used pesticides for field application were 20% azoxystrobin of suspension concentrate and 47% kresoxim-methyl of water dispersible granule. Two pesticides were sprayed at recommended and double dose rate. Pesticide residues in Korean melon were analyzed until 14 days after application. The azoxystrobin was analyzed by HPLC equipped with UV detector after cleanup with florisil glass column. Initial residue concentrations of azoxystrobin in Korean melon at recommended and double dose rate were 0.09 mg/kg and 0.14 mg/kg, respectively. Those were less than 0.2 mg/kg maximum residue limit of Korean melon established by KFDA. The biological half-lives of azoxystrobin in Korean melon were 4.7 days at recommended dose rate and 7.8 days at double dose rate. Initial concentrations of kresoxim-methyl which was analyzed by GLC-ECD in Korean melon at recommended and double dose rate were 0.10 mg/kg and 0.23 mg/kg, respectively. Those were less than 1.0 mg/kg, MRL. The biological half-lives of kresoxim-methyl in Korean melon were 4.1 days at recommended dose rate and 4.8 days at double dose rate. The residue amounts of both pesticide was lower than MRL and biological half-lives were not so long. Because the weight of Korean melon under plastic film house condition was fast increased during cultivation.
This study was performed to acquire scientific data for establishing the maximum residue limits (MRLs) of pesticides in Korean red ginseng and its extract. Pesticides (azoxystrobin, fenhexamid, cyprodinil) were applied to a cultivated field of ginseng, and the fresh ginseng was harvested and processed to make Korean red ginseng and its extract. The reduction rates of the residue pesticides were calculated by determining the pesticide contents in each stage of ginseng processing. The residue levels in fresh ginseng were 0.12 ppm for azoxystrobin, 0.19 ppm for fenhexamid, and 1.78 ppm for cyprodinil. The residue levels in Korean red ginseng were 0.24, 0.54, and 1.49 ppm, and in the extract 0.81, 1.93, and 3.66 ppm for azoxystrobin, fenhexamid, and cyprodinil, respectively. The steaming and processing of fresh ginseng increased azoxystrobin and fenhexamid residues, but cyprodinil was reduced. The reduction rates (dry basis) of azoxystrobin, fenhexamid, and cyprodinil were 0.66, 0.94, and 0.28 for Korean red ginseng, and 3.25, 4.94, and 1.01 for the extract, respectively.
Assessment of N release pattern from hairy vetch (Vicia villosa Roth) residue is an important step for the efficient use of the residue as a green manure and the specific management strategy development. Objective of this research was to determine the changes of chemical composition, especially in light of the N release, when the residues of hairy vetch were applied in the soil. hairy vetch was harvested at different growth stages (April 23, May 9, and May 28) and the residues were encapsulated in the nylon mesh (1 mm) bag. The bags were placed on the surface of no-tillage plots and buried at 15cm in depth in conventional tillage plots of the corn field in early June. The bags were removed for measurements of dry matter and N content at 1, 2, 3. 5, 8 and 12 weeks after placement. Dry matter and N content of hairy vetch were higher when harvested closer to maturity, as also found in C/N ratio, cellulose and hemicellulose contents. The rate of N releases from hairy vetch residue under conventional tillage condition was greater than that under no-tillage condition. Percentages of N remaining in residue over the initial N contents under conventional tillage condition were in average 40, 23, and 15% for 1, 2, and 3 weeks after placement of mesh bag, respectively, compared with respective values of 71, 31, and 21% under no-tillage condition. The rate of N release was slower as the harvest time was closer to maturity of hairy vetch under both tillage conditions. Most of N in residue was released within 3 and 5 weeks, under conventional and no-tillage conditions, respectively. Results indicated that amounts of N released from hairy vetch residue were more closely related to the variability of N contents in residue due to the harvest time than changes of chemical composition of plant or soil management.
Nek6 belongs to NIMA1 (never in mitosis, gene A) related kinase, which was originally identified in Aspergillus nidulans as a serine/threonine kinase critical for cell cycle progression. We noticed that the putative SUMOylation site is localized on the $K^{252}$ residue in $^{251}FKsD^{254}$ of Nek6, based on the consensus sequence ${\Phi}KxE$; where ${\Phi}$ represents L, I, V or F and x is any amino acid. We observed that the Nek6 SUMO mutant (K252R) has decreased protein kinase activity, nuclear speckle localization and protein stability, compared with that of the Nek6 wild type. However, the Nek6 SUMO mutant increased the cell survival rate of COS-1 cells as determined by FACS analysis. Therefore, our data suggest that SUMOylation on the $K^{252}$ residue of Nek6 is required for its normal functions, such as proper nuclear localization, kinase activity and protein stability, to control cell cycle.
A fast and simple high-performance liquid chromatography (HPLC) method for the simultaneous determination of four pesticides having fungicide properties has been proposed for Panax ginseng, C. A. Meyer grown for 4, 5, or 6 years. Analytical separation was performed on C18 columns using ultraviolet detector under gradient conditions. Spiked blank samples were used as standards to counteract the matrix effect observed in the chromatographic determination. The HPLC response for all pesticides was linear, with determination coefficients > 0.9986. The average rate of recovery for pesticides spiked with 2 fortification levels was > 72% with relative standard deviations < 9%. The limits of quantification (LOQ) ranged from 0.03 to 0.16 ppm. These LOQs were lower than the respective maximum residue limits (MRL) established by the Korean Food and Drug Administration (KFDA), except for cyazofamid. The proposed method was used to determine pesticide residue levels in samples of ginseng obtained from Jeonnam Province (Republic of Korea). None of the pesticides were found in ginseng samples grown for 4, 5, or 6 years.
Essential amino acids involving in the catalytic mechanism of the $\beta$-D-xylosidase of Bacillus stearothermophilus were determined by chemical modification studies. Among various che- mical modifiers tested N-bromosuccinimide (NBS), $\rho$-hydroxymercurybenzoate (PHMB), N-ethylma- leimide, 1-[3-(di-ethylamino)-propyl]$-3-ethylcarbodi-imide (EDC), and Woodward's Reagent K(WRK)inactivated the enzyme, resulting in the residual activity of less than 20%. WRK reduced the enzyme activity by modifying carboxylic amino acids, and the inactivation reacion proceeded in the form of pseudo-first-order kinetics. The double-lagarithmic plot of the observed pseudo-first- order rate constant against the modifier concentration yielded a reaction order of 2, indicating that two carboxylic amino acids were essential for the enzyme activity. The $\beta$-D-xylosidase was also inactivated by N-ethylmaleimide which specifically modified a cysteine residue with a reaction order of 1, implying that one cysteine residue was important for the enzyme activity. Xylobiose protected the enzyme against inactivation by WRK and N-ethylmaleimide, revealing that carboxylic amino acids and a cysteine residue were present at the substrate-binding site of the enzyme molecule.
Incubation of an NADPH-dependent succinic semialdehyde reductase from bovine brain with N-bromosuccinimide (NBS) resulted in a time-dependent loss of enzyme activity. The inactivation followed pseudo-first-order kinetics with the second-order rate constant of $6.8\times{10}^3$$M^-1$$min^{-1}$. The inactivation was prevented by preincubation of the enzyme with substrate succinic semialdehyde, but not with coenzyme NADPH. There was a linear relation-ship between oxindole formation and the loss of enzyme activity. Spectro-photometric studies indicated that about one oxindole group per molecule of the enzyme was formed following complete loss of enzymatic activity. It is suggested that the catalytic function of succinic semialdehyde reductase is modulated by binding of NBS to a specific tryptophan residue at or near the substrate binding site of the enzyme.
Using a flue - cured tobacco variety, KF lH9, recently bred and released to farming, effect of growth regulators(fatty alcohol and C - MH) on the change of alkaloid contents and dynamics of MH uptake and distribution were investigated. An efficient sucker control method was also pursued. Treatment of fatty alcohol at button stage remarkably increased the rate of C - MH uptake. However, except in the plants treated with 2- fold volume of C - MH, residue of C -MH was kept under maximum residue level(80 ppd). C - MH residue in stalk was higher than in foliage in general. Compared to untreated control, leaf weight per unit area was increased by the treatment even in lower leaves where vigorous transfer of C - MH from treated tips was indicated and sucker control higher than 94.9% was achieved with all treatments except for sole half volume C - MH treatment at topping stage. Alkaloid content in foliage was reduced by dual treatment of fatty alcohol and C - MH but increased in stalk and root.

γ -Glutamyl transpeptidase (EC 2.3.2.2) plays a key role in glutathione metabolism by catalyzing the transfer of the γ -glutamyl residue and hydrolysis of glutathione. The functional residues at the active site of the rat kidney γ -glutamyl transpeptidase were investigated by kinetic studies at various pH, the treatment of diethylpyrocarbonate (DEPC), and photooxidation in presence of methylene blue. An ionizable group affecting the enzymatic activity with an apparent pKa value of 7.1, which is in the range of pKa values for a histidine residue in protein, was obtained by examining the pH-dependence of kinetic parameters. The pH effect on the photoinduced inactivation rate of the enzyme corresponds to that expected for the photooxidation of the free histidine. The involvement of a histidine in the catalytic site of the enzyme was further supported by DEPC modification accompanied by an increase in absorbance at 240 nm, indicating the formation of Ncarbethoxyhistidine. The histidine located at the position of 382 in the precursor of the enzyme is primarily suspected based on the amino acid sequence alignment of the transpeptidases from various organisms.
Usage and residue of pesticide were sureyed for five years from three golf courses which have morethan eighteenholes inKangwon-do from 1992 to 1996. A total of 186 samples(soils, lawns, effluents) were collected from these courses and residual levels of four kinds of organochlorines and twelve kinds of organophosphorus pesticides in each samples were determined by GC-ECD and GC-NPD. The amount of pesticide used in each golf course was 1, 635.6kg a year on the average, and that of pesticide to the green and the fairway were 33g/$\textrm{m}^2$ and 3g/$\textrm{m}^2$, respectively. The pesticide which was not permitted for the management of golf course was detected in 24 of 186 samples and its detection rate was 12.9%. The average pesticide residue in soils was 0.208 ppm for captan, 0.031 ppm for daconil, respectively. Detection rate of the prohibited pesticides has decreased every year since 1992, and there were no residues all the tested samples in 1996.Based on these results, it might be judged investigation methods of pesticide residue should be changed with current using pesticide from the prohibited pesticides and the criteria of residual pesticides in effluents of golf course also should be established.
