유아의 채소섭취행동 강화를 위한 채소노출 및 채소놀이 영양교육 프로그램의 효과 (Effects of Eating Behavior of Preschool Children Using a Direct Visual Stimulating Program on Vegetables and Nutrition Education with Vegetable Playing)

  • The purpose of this study was to examine 1) the relationship between the vegetable eating behavior of care givers and that of children, 2) the effectiveness of nutrition education with vegetable playing using direct visual stimulating programs on vegetable eating behavior of preschool children, and 3) the times and period of nutrition education with vegetable playing for significant changes on vegetable eating behavior. A total number of 56 individuals, aged 42 to 66 months old, participated in this study in which three kinds of vegetables (30 g)/meal were served per individual, and vegetable eating behavior was measured by the residue on the dish during 5 weeks (25 days). To the simple visual stimulating group, vegetable dish was served without education, and other groups included education 1 group (nutrition education 1 time/week), education 2 group (nutrition education 2 times/week), and education 3 group (nutrition education 3 times/week) with simple visual stimulation by the vegetable dish. The results showed 1) the significant relationship (P<0.001) between the vegetable eating behavior of the care giver and that of children by analysis of the questionnaire, 2) the effectiveness of nutrition education using vegetable playing on vegetable eating behavior of preschool children (P<0.05), and 3) the significant changes in vegetable eating behavior by the 3rd week in the education 3 group. This study shows that food neophobia caused behavior problems in children regarding vegetable eating and repeated exposure was able to reduce food neophobia.

A Systematic NMR Determination of α-D-Glucooligosaccharides, Effect of Linkage Type, Anomeric Configuration and Combination of Different Linkages Type on 13C Chemical Shifts for the Determination of Unknown Isomaltooligosaccharides

  • Prebiotic isomaltooligosaccharide preparations contain $\alpha$-D-glucooligosaccharides comprising isomaltooligosaccharides (IMOs) and non-prebiotic maltooligosaccharides (MOs). They are both glucose oligosaccharides characterized by their degree of polymerization (DP) value (from 2 to $\sim$10), linkages types and positions (IMOs: $\alpha$-(1$\rightarrow$2, 3, 6 and in a lower proportion internal 1$\rightarrow$4) linkages, MOs: α-(1$\rightarrow$4) linkages). Their structure is the key factor for their prebiotic potential. In order to determine and elucidate the exact structure of unknown IMOs and MOs, unambiguous assignments of $^{13}C$ and $^1H$ chemical shifts of commercial standards, representative of IMOs and MOs diversity, have been determined using optimized standard one and two-dimensional experiments such as $^1H$ NMR, $^{13}C$ NMR, APT and ${^1}H-{^1}H$ COSY, TOCSY, NOESY and <$^1H-{^{13}}C$ heteronuclear HSQC, HSQC-TOCSY, and HMBC. Here we point out the differential effect of substitution by a glucose residue at different positions on chemical shifts of anomeric as well as ring carbons together with the effect of the reducing end configuration for low DP oligosaccharides and diasteroisotopic effect for H-6 protons. From this study, structural $^{13}C$ specific spectral features can be identified as tools for structural analysis of isomaltooligosaccharides.

Bacillus Pumilus TX703 유래 Xylanase의 활성에 관여하는 아미노산 잔기의 확인 (Identification of Amino Acid Residues Involved in Xylanase Activity from Bacillus Pumilus TX703)

  • Bacillus pumilus TX703으로부터 xylanase를 정제한 후 효소의 활성부위를 조사하기 위하여 여러 가지 화학수식제를 사용하여 효소활성의 저해도를 측정하였다. 여러 가지 화학수식제 중에서 carbodiimide와 N-bromosuccinimide가 효소활성을 완전히 저 해시 켜 glutamic acid 또는 aspartic acid 잔기와 tryptophan 잔기가 효소의 활성부위에 관여하리라 추측되었다. 각각의 경우에 효소 실활은 수식제의 첨가농도에 따라 pseudo first-order kinetics 양식을 보여주었으며, car-bodiimide와 N-bromosuccinimide는 각각 비경쟁적 저해와 경쟁적 저해방식을 나타내었다. 기질첨가에 의한 효소활성 보호실험을 통하여 tryptophan 잔기가 기질결합부위라 판단되었다. 효소실활속도의 분석에 의해 효소활성에는 2개의 glutamic acid 또는 aspartic acid 잔기와 1개의 tryptophan 잔기가 관여하는 것으로 나타났다.

Crystal Structure and Comparative Sequence Analysis of GmhA from Colwellia psychrerythraea Strain 34H Provides Insight into Functional Similarity with DiaA

  • The psychrophilic organism Colwellia psychrerythraea strain 34H produces extracellular polysaccharide substances to tolerate cold environments. Sedoheptulose 7-phosphate isomerase (GmhA) is essential for producing $\small{D}$-glycero-$\small{D}$-mannoheptose 7-phosphate, a key mediator in the lipopolysaccharide biosynthetic pathway. We determined the crystal structure of GmhA from C. psychrerythraea strain 34H (CpsGmhA, UniProtKB code: Q47VU0) at a resolution of $2.8{\AA}$. The tetrameric structure is similar to that of homologous GmhA structures. Interestingly, one of the catalytic residues, glutamate, which has been reported to be critical for the activity of other homologous GmhA enzymes, is replaced by a glutamine residue in the CpsGmhA protein. We also found differences in the conformations of several other catalytic residues. Extensive structural and sequence analyses reveal that CpsGmhA shows high similarity to Escherichia coli DnaA initiatorassociating protein A (DiaA). Therefore, the CpsGmhA structure reported here may provide insight into the structural and functional correlations between GmhA and DiaA among specific microorganisms.

토양에서 분리된 Aeromonas hydrophila AM-28이 생산하는 단백질 가수분해효소의 특성 (Properties of Protease from Aeromonas hydrophila AM-28 Isolated from Soil)

  • 토양으로부터 탈지대두박을 잘 분해하는 단백질 가수분해효소를 생산하는 미생물을 분리하여 형태적, 생화학적 특성과 세포벽 지방산조성을 조사한 결과, 분리된 세균은 Aeromonas hydrophila로 동정되었다. 분리된 세균은 glycerol을 탄소원으로, tryptone 및 $(NH_4)_2HPO_4$을 진소원으로, $CaCl_2$를 무기원으로 사용한 배지에서 효소를 가장 많이 생산하였으며, 생산된 효소의 반응최적 pH는 8.0이고, 반응최적온도는 $65^{\circ}C$였다. 이 효소는 $55^{\circ}C$ 이하의 온도에서, pH 7.0에서 13.0의 범위에서 비교적 안정한 것으로 나타났다. 또한 이 효소는 금속이온 $Cu^{2+},\;Hg^{2+}.\;Mn^{2+}$$Zn^{2+}$에 의해서 효소활성이 저해를 받고, 효소저해제 phenylmethylsulfonyl fluoride (PMSF)와 ethylenediaminetetraacetic acid (EDTA)에 의해서 효소활성이 강력히 저해되는 성질로부터 효소활성부위에 serine기와 금속이온을 갖고 있는 것으로 밝혀졌다.

Cl2/CF4 플라즈마에 Ar, O2 첨가에 따른 PZT 박막의 식각 손상 개선 효과 (Reduce of Etching Damage of PZT Thin Firms with Addition of Ar and O2 in Cl2/CF4 Plasma)

  • In this study, the reduce of plasma etching damage in PZT thin film with addictive gas and re-annealing after etching have been investigated. The PZT thin films were etched as a function of $Cl_2/CF_4$ with addition of Ar and $O_2$ with inductively coupled plasma. The etch rates of PZT thin films were 1450 ${\AA}/min$ at 30% additive Ar and 1100 ${\AA}/min$ at 10% auditive $O_2$ into $Cl_2/CF_4$ gas mixing ratio of 8/2. In order to reduce plasma damage of PZT thin films after etching, the etched PZT thin films were re-annealed at various temperatures at $O_2$ atmosphere. From the hysteresis curries, the ferroelectric properties are improved by $O_2$ re-annealing process. The improvement of ferroelectric behavior at annealed PZT films is consistent wish the increase of the (100) and (200) PZT peaks revealed by x-ray diffraction (XRD). From x ray photoelectron spectroscopy (XPS) analysis, the intensity of Pb-O, Zr-O and Ti-O peak are increased and the chemical residue peak is reduced by $O_2$ re-annealing. The ferroelectric behavior consistent with the dielectric nature of $Ti_xO_y$ is recovered by $O_2$ recombination during rapid thermal annealing process.

  • 본 논문은 전자파 장해 및 복사 내성 측정의 대용 시험 시설로서 활용될 수 있는, 다중 유전체로 구성된 평탄형 QRD(Quadratic Residue Diffuser)를 사용한 전자파 잔향실에 대한 연구이다. 잔향실은 $2.5{\sim}3$ GHz 대역에서 사용 가능하도록 설계되었으며, 기존 QRD의 특성을, 다양한 유전체를 이용하여 낮은 평탄형 구조물로 구현하였고, 이를 적용한 전자파 잔향실 내부에서의 전자기장 특성 변화 및 시험 공간 확장에 관해서 비교 분석하였다. 잔향실 내부의 전자기장 분포 해석을 위해서 FDTD(Finite-Difference Time-Domain) 수치 해석 방식을 이용하였다. 잔향실 내에 새로 제안된 확산기를 사용하면, 기존 확산기에 비해 물리적인 크기가 줄어들 뿐만 아니라 전자기장 균일도, 편파 특성 및 공차 등의 성능이 비슷하거나 우수해짐을 확인하였다. 따라서 다중 유전체로 구성된 평탄형 확산기를 사용한 잔향실이 기존의 QRD를 사용한 잔향실에 비해 전자기적 성능 및 공간 활용면에서 우수한 시험시설임을 확인하였다.

서울특별시 종로구 대중목욕탕의 수질 중 미생물 오염도 조사 연구 (Investigation of Microbial Contamination of Public Bath in Jongno-gu, Seoul)

  • The aim of this study is to investigate microbial sanitary condition of public baths in Seoul, Korea. A total of 28 water samples were collected from 14 different public baths and sudatoriums. The prevalence of fecal indicator microorganisms such as total coliform, fecal coliform, and Escherichia coli was characterized. In addition, bacteria in water was membrane filtered by 0.45um nitrocellulose membrane, and the filter was analyzed by both cultivation and PCR amplification of partial 16S rRNA gene. The levels of chlorine were measured for each of water samples. More than 40% of 14 collected water samples, the concentrations of total coliform bacteria exceeded the water quality for bath water guideline. There was no significant correlation between chlorine residue and the presence of total coliform. Various microorganisms including pathogenic microorganisms were identified from cultivation and subsequent analysis of 16s rRNA gene sequences. Our results suggest that appropriate hygiene practice and continuous monitoring is needed for reducing health risk associated with public bathhouses.

Performance Test of a Real-Time Measurement System for Horizontal Soil Strength in the Field

  • Purpose: Soil strength has been measured using a cone penetrometer, which is making it difficult to obtain the spatial data required for precision agriculture. Our objectives were to evaluate real-time horizontal soil strength (RHSS) to measure soil strength in real time while moving across the field. Using the RHSS data, the tillage depth was determined, and the power consumption of a tractor and rotavators were compared. Methods: The horizontal soil-strength index (HSSI) obtained by the RHSS was compared with the cone index (CI), which was measured using a cone penetrometer. Comparison analysis in accordance with the measurement depth that increased at 5-cm interval was conducted using kriged maps at six sensing depths. For tillage control and evaluation of the power consumption, the system was installed with a potentiometer for tillage depth, a torque sensor from the rear axle, and a power take-off (PTO) shaft. Results: The HSSI was lower than the CI, but they were the same at 54.81% of the total grids for the 5-cm depth and at 3.85% for the 10-cm depth. In accordance with the recommended tillage map, tillage operations between 0 and 15 cm left 2.3% and 7% residue cover on the soil, and that between 20 and 10 cm covered a wider utilization of 3% and 18.4%, respectively. When the tillage depth was 15 cm, the comparison result of the power requirements between the PTO and rear axle in terms of control performance revealed that the maximum power requirements of the axle and PTO were 44.63 and 23.24 kW, respectively. Conclusions: An HSSI measurement system was evaluated by comparison with the conventional soil strength measurement system (CI) and applied to a tractor to compare the tillage power consumption. Further study is needed on its application to various farm works using a tractor for precision agriculture.
