Effects of rhubarb extract on osteoclast differentiation in bone marrow-derived macrophages (대황 추출물이 골수유래 대식세포의 파골세포 분화에 미치는 영향)

  • Objectives: This study aimed to investigate the effects of rhubarb extract on osteoclast differentiation in bone marrow-derived macrophages (BMMs). Osteoclasts are vital for bone resorption and remodeling. Osteoclast dysregulation can contribute to various bone-related disorders that directly affect oral health. Rhubarb, a medicinal plant with anti-inflammatory properties, has been shown to modulate bone metabolism. Methods: BMMs were isolated from the femurs and tibias of 5-week-old C57BL/6 mice and cultured in the presence of mouse macrophage colony-stimulating factor (M-CSF) for 3 days. Subsequently, BMMs were treated with M-CSF and receptor activator of nuclear factor-κB ligand (RANKL) to induce osteoclast differentiation. Results: Rhubarb extract effectively suppressed osteoclast differentiation in BMMs. Furthermore, rhubarb extract inhibited the mRNA expression of tartrate-resistant acid phosphatase (TRAP) and cathepsin K (CTSK), which are essential for osteoclastogenesis. Moreover, it inhibited the RANKL-induced expression of nuclear factor of activated T cell c1 (NFATc1), a crucial transcription factor in osteoclast differentiation. Conclusions: These results suggest that rhubarb extract promotes oral health by inhibiting osteoclastogenesis in BMMs. Thus, rhubarb extract shows promise as a therapeutic agent for bone-related disorders that directly affect oral health, particularly those associated with abnormal osteoclast activity. Further research and exploration of the underlying mechanisms are warranted to fully understand their potential clinical applications.

Evaluation of the Confidence and Learning Effects of Dental Hygiene Ethical Decision-Making through Dental Hygiene Ethics Subjects (치위생(학)과 학생들의 치위생윤리 교과목을 통한 치위생 윤리적 의사결정에 대한 자신감과 학습성과 평가)

  • Background: This study evaluated the learning outcomes of dental hygiene students' ethical consciousness and ethical decision-making competence through dental ethics courses conducted in some universities. Methods: The subjects were 35 and 29 fourth-year dental hygiene students at G University in the first semester of 2021 and 2022, respectively, and 53 and 43 third-year dental hygiene students at D University, respectively, for a total of 160 students. After implementing the dental hygiene ethics course, classroom performance was evaluated in terms of moral sensitivity, confidence in making ethical decisions, classroom practicality, learning outcomes, and class satisfaction. Statistical analysis was conducted using independent t-test and paired t-test, and the statistical significance level was 0.05. Results: Both universities reported an increase in moral sensitivity and confidence in ethical decision-making after the course (p<0.001). Classroom practicality and class satisfaction for the dental hygiene ethics course did not differ between disciplines and were rated positively with a score of 4 or higher (p>0.05). Learning outcomes were higher among 4-year students than 3-year students (p<0.001). Conclusions: It was evaluated that the ethics in dental hygiene curriculum can strengthen students' competence in ethical decision-making, including moral sensitivity and confidence in solving ethical problems in dental hygiene.

Validity of midsagittal reference planes constructed in 3D CT images (전산화단층사진을 이용한 3차원 영상에서 정중시상기준평면 설정에 관한 연구)

  • Objective: The purpose of this study was to evaluate the validity of midsagittal reference (MSR) planes constructed in maxillofacial 3D images. Methods: Maxillofacial computed tomography (CT) images were obtained in 36 normal occlusion individuals who did not have apparent facial asymmetry, and 3D images were reconstructed using a computer software. Six MSR planes (Cg-ANS-Ba, Cg-ANS-Op, Cg-PNS-Ba, Cg-PNS-OP, FH${\perp}$(Cg, Ba), FH${\perp}$(Cg, Op)) were constructed using the landmarks located in the midsagittal area of the maxillofacial structure, such as Cg, ANS, PNS, Ba and Op, and FH plane constructed with Po and Or. The six pairs of landmarks (Z, Fr, Fs, Zy, Mx, Ms), which represent right and left symmetry in the maxillofacial structure, were selected. Statistically significant differences of the right and the left measurements were examined through t-test, and the difference of the right and the left measurement was compared among the six MSR planes. Results: The distances from the right and the left landmarks in each pair to each MSR plane did not show a statistically significant difference. The reproducibility of the landmark identification was excellent. Conclusion: All the six planes constructed in this study can be used as a MSR plane in maxillofacial 3D analysis, particularly, the planes including Cg and ANS.

The study on voluntary satisfaction and voluntary activities of dental hygiene students (치위생과 학생의 자원봉사 만족도와 자원봉사활동에 관한 연구)

  • This study aimed to investigate that what factors affect the volunteer activities of dental hygiene students and present a plan and problems with devising a program for activating their volunteer activities. A self-administered questionnaire survey was conducted in 265 dental hygiene students in Gwangju from November to December 2012, obtaining the following results: 51.3% of the respondents were freshmen; 74.3% had experience of volunteer activities; and 31.7% had the activities in the field of disabilities. 56.2% were motivated to participate in the activities by 'credit-earning and volunteer activity achievements'. Most of students participated in volunteer activities through clubs. Satisfaction with major-related volunteer activities was associated with higher support by others, higher job perception, higher job adequacy, and higher support by family. To increase satisfaction with major-related volunteer activities of students research is needed to develop systematic education for volunteers, subjects suitable for colleges, and an effective program in order to alter students' perception.

Morphology of Tooth and Smad4 Expression in NFI-C Deficient Mouse (Nuclear Factor I-C 결손생쥐에서 치아의 형태학적 변화와 Smad4의 발현)

  • Over expression of TGF-${\beta}1$ revealed the same phenotype as NFI-C deficient mouse. It has been reported that NFI-C deficient mice demonstrated abnormal odontoblast differentiation and aberrant dentin formation during root development. In the present study, in order to investigate the histological differences between wild type (WT) mouse and NFI-C deficient mouse, we compared morphological characteristics and smad4 expression between those mice. Hematoxyline-eosin (H-E) staining was used to investigate morphological changes and immunohistochemistry was also performed to observe the Smad4 expression pattern. In H-E staining, incisor of NFI-C deficient mouse showed an open area in the lingual root, irregular odontoblasts and osteodentin. Also, NFI-C deficient mouse showed short root and osteodentin in molar. In addition, Smad4 protein was strongly expressed in NFI-C deficient mouse compared with wild type. These findings suggest that NFI-C deficiency affects odontoblast differentiation and result in the formation of abnormal roots. Therefore, balancing between NFI-C and TGF-${\beta}$ signaling including Smad4 is important for the regulation of normal odontoblast differentiation and dentin formation.

Effect of bFGF and fibroblasts combined with hyaluronic acid-based hydrogels on soft tissue augmentation: an experimental study in rats

  • Background: Hyaluronic acid (HA) has been applied as a primary biomaterial for temporary soft tissue augmentation and as a carrier for cells and the delivery of growth factors to promote tissue regeneration. Although HA derivatives are the most versatile soft tissue fillers on the market, they are resorbed early, within 3 to 12 months. To overcome their short duration, they can be combined with cells or growth factors. The purpose of this study was to investigate the stimulating effects of human fibroblasts and basic fibroblast growth factors (bFGF) on collagen synthesis during soft tissue augmentation by HA hydrogels and to compare these with the effects of a commercial HA derivative (Restylane®). Methods: The hydrogel group included four conditions. The first condition consisted of hydrogel (H) alone as a negative control, and the other three conditions were bFGF-containing hydrogel (HB), human fibroblast-containing hydrogel (HF), and human fibroblast/bFGF-containing hydrogel (HBF). In the Restylane® group (HGF), the hydrogel was replaced with Restylane® (R, RB, RF, RBF). The gels were implanted subdermally into the back of each nude mouse at four separate sites. Twelve nude mice were used for the hydrogel (n = 6) and Restylane® groups (n = 6). The specimens were harvested 8 weeks after implantation and assessed histomorphometrically, and collagen synthesis was evaluated by RT-PCR. Results: The hydrogel group showed good biocompatibility with the surrounding tissues and stimulated the formation of a fibrous matrix. HBF and HF showed significantly higher soft tissue synthesis compared to H (p < 0.05), and human collagen type I was well expressed in HB, HF, and HBF; HBF showed the strongest expression. The Restylane® filler was surrounded by a fibrous capsule without any soft tissue infiltration from the neighboring tissue, and collagen synthesis within the Restylane® filler could not be observed, even though no inflammatory reactions were observed. Conclusion: This study revealed that HA-based hydrogel alone or hydrogel combined with fibroblasts and/or bFGF can be effectively used for soft tissue augmentation.

Regenerative Endodontic Procedure in Korean Children and Adolescents: A Case Report (한국 소아청소년 근관치료 영역에서 재생치료, 근관치료재생술에 대한 증례보고)

  • Mineral trioxide aggregate (MTA) is widely used in endodontic therapy as a pulp-capping material, root or furcal perforation repair material, and for apexification and obturation of the root canal system. The purpose of this study was to formally document cases of MTA application in South Korean children and adolescents. Through this research, the practice of using MTA will be introduced and familiarized to the clinical practitioners. This study involved endodontic treatment using MTA for fractured crowns in 11- and 12-year-old. The children were followed up for 12 months until the pulp vitality was confirmed; in young permanent teeth with immature roots, the pulp is integral to the process of apexogenesis. These observational results regarding the use of MTA as an apexification material in non-vital immature permanent incisors appear to provide promising results in the search for new materials to meet existing endodontic needs.

Effects of Pre-Sintering Surface Treatment and Liner Application on the Shear Bond Strength of Zirconia and Pressable Ceramic (소결 전 지르코니아 표면처리와 라이너 사용에 따른 지르코니아와 열가압성형도재의 전단결합강도)

  • Purpose: This study was intended to investigate the effect of applying liner for chemical bonding and physical surface roughness created on zirconia by using a sandpaper before sintering on the bond strength between the two materials. Methods: Zirconia blocks were cut using a low-speed cutter. Plate-shaped specimen($6mm{\times}6mm{\times}3mm$) was fabricated by sintering after giving surface roughness according to four kinds of sandpapers. Depending on whether or not to use liner, 60 specimens were divided into two groups ZN(non-liner), ZL(liner), and the two groups were subdivided into four groups respectively in accordance with sandpaper used, totaling eight groups (n=10). The surface roughness (Ra) values and shapes before sintering were observed, and shear bond strength after pressing ceramic plasticity was measured with a universal testing machine. For a test of the significance, a one-way ANOVA was performed, and Tukey's multiple comparison test was conducted. Results: The observation of the surface roughness was SB04($2.22{\pm}1.16{\mu}m$), SB08($2.98{\pm}0.33{\mu}m$), SB12($2.44{\pm}1.32{\mu}m$), SB20($2.34{\pm}0.59{\mu}m$) and SA04($2.34{\pm}0.67{\mu}m$), SA08($1.28{\pm}0.90{\mu}m$), SA12($2.03{\pm}1.60{\mu}m$), SA20($2.19{\pm}1.73{\mu}m$). In the case of ZN Group, the shear bond strength was ZN04($23.26{\pm}3.83MPa$), ZN08($21.76{\pm}2.33MPa$), ZN12($20.49{\pm}3.01MPa$), ZN20($24.98{\pm}4.22MPa$)(p<0.05). As for ZL Group, the shear bond strength was ZL04($25.09{\pm}5.67MPa$), ZL08($22.98{\pm}2.26MPa$), ZL12($21.54{\pm}5.70MPa$), ZL20($23.98{\pm}3.23MPa$)(p<0.05). Conclusion: The research results showed that the bond strength of Zirconia core and Pressing ceramic was further improved by physical surface treatment before sintering, rather than by chemical bonding through liner surface treatment.

Big Data based Tourist Attractions Recommendation - Focus on Korean Tourism Organization Linked Open Data - (빅데이터 기반 관광지 추천 시스템 구현 - 한국관광공사 LOD를 중심으로 -)

  • Conventional exhibition management information systems recommend tourist attractions that are close to the place in which an exhibition is held. Some recommended attractions by the location-based recommendation could be meaningless when nothing is related to the exhibition's topic. Our goal is to recommend attractions that are related to the content presented in the exhibition, which can be coined as content-based recommendation. Even though human exhibition curators can do this, the quality is limited to their manual task and knowledge. We propose an automatic way of discovering attractions relevant to an exhibition of interests. Language resources are incorporated to discover attractions that are more meaningful. Because a typical single machine is unable to deal with such large-scale language resources efficiently, we implemented the algorithm on top of Apache Spark, which is a well-known distributed computing framework. As a user interface prototype, a web-based system is implemented that provides users with a list of relevant attractions when users are browsing exhibition information, available at http://bike.snu.ac.kr/WARP. We carried out a case study based on Korean Tourism Organization Linked Open Data with Korean Wikipedia as a language resource. Experimental results are demonstrated to show the efficiency and effectiveness of the proposed system. The effectiveness was evaluated against well-known exhibitions. It is expected that the proposed approach will contribute to the development of both exhibition and tourist industries by motivating exhibition visitors to become active tourists.

Cleaning Methods to Effectively Remove Peanut Allergens from Food Facilities or Utensil Surfaces (식품 시설 또는 조리도구 표면에서 땅콩 알레르겐을 효과적으로 제거하는 세척 방법)

  • Peanut is a well-known food allergen that causes adverse reactions ranging from mild urticaria to life-threatening anaphylaxis. Consumers suffering from peanut allergies should thus avoid consuming undeclared peanuts in processed foods. Therefore, effective cleaning methods are needed to remove food allergens from manufacturing facilities. To address this, wet cleaning methods with washing water at different temperatures, abstergents (peracetic acid, sodium bicarbonate, dilute sodium hypochlorite, detergent), and cleaning tools (brush, sponge, paper towel, and cotton) were investigated to remove peanuts from materials used in food manufacture, including plastics, wood, glass, and stainless steel. Peanut butter was coated on the surface of the glass, wood, stainless steel, and plastic for 30 min and cleaned using wet cleaning. The peanut residue on the cleaned surfaces was swabbed and determined using an optimized enzyme-linked immunosorbent assay (ELISA). Cleaning using a brush and hot water above 50℃ showed an effective reduction of peanut residue from the surface. However, removing peanuts from wooden surfaces was complicated. These results provide information for selecting appropriate materials in food manufacturing facilities and cleaning methods to remove food allergens. Additionally, the cleaning methods developed in this study can be applied to further research on removing other food allergens.
