Heterologous Microarray Hybridization Used for Differential Gene Expression Profiling in Benzo[a]pyrene-exposed Marine Medaka

  • Differential gene expression profiling was performed in the hepatic tissue of marine medaka fish (Oryzias javanicus) after exposure to benzo[a]pyrene (BaP), a polycyclic aromatic hydrocarbon (PAH), by heterologous hybridization using a medaka cDNA microarray. Thirty-eight differentially expressed candidate genes, of which 23 were induced and 15 repressed (P<0.01), were identified and found to be associated with cell cycle, development, endocrine/reproduction, immune, metabolism, nucleic acid/protein binding, signal transduction, or non-categorized. The presumptive physiological changes induced by BaP exposure were identified after considering the biological function of each gene candidate. The results obtained in this study will allow future studies to assess the molecular mechanisms of BaP toxicity and the development of a systems biology approach to the stress biology of organic chemicals.

  • Penaeidins are members of a special family of antimicrobial peptides existing in several species of shrimp and play a crucial role in the immunological defense of shrimp. In this study, we isolated and characterized one EST clone (penaeidin) from cDNA library of fleshy prawn Fenneropenaeus chinensis hemocytes. Amino acids sequence comparison and phylogenetic analysis with other known penaeidins revealed that our clone was completely identical to F. chinensis Penaeidin 3-2 (Accession no. ABC33920), which composed of 71 amino acids with a putative signal peptide (1-19) and a cysteine-rich domain (C-terminal part). The expression and distribution of Penaeidin 3-2 transcripts in shrimp were detected in hemocytes, hepatopancreas, and muscles, and that Penaeidin 3-2 was constitutively expressed mainly in hemocytes. The artificial infection of white spot syndrome virus to F. chinensis resulted in Penaeidin 3-2 mRNA up-regulation in hemocytes, suggesting that the possible role of Penaeidin 3-2 in host defense system of F. chinensis.

Distribution of Whales and Dolphins in Korean Waters Based on a Sighting Survey from 2000 to 2010 (목시조사(2000-2010)에 의한 한국 연안 고래류의 종류 및 분포)

  • In the late 1970s, the National Fisheries Research & Development Institute (NFRDI) started cetacean research to submit the Korean whale catch record to the International Whaling Commission. This continued until the moratorium on commercial whaling in 1986. The NFRDI resumed cetacean research with a pilot whale sighting survey in 1999. Subsequently, the NFRDI has conducted 53 cetacean sighting surveys within the Korean exclusive economic zone between 2000 and 2010. The surveys took a total of 760 days and cruising for 23,866 nautical miles. The finless porpoise Neophocaena asiaeorientalis was sighted most frequently (735 times), followed by the minke whale Balaenoptera acutorostrata (396 times), the long-beaked common dolphin Delphinus capensis (102 times), and the Pacific white-sided dolphin Lagenorhynchus obliquidens (27 times). Minke whales were distributed in the Yellow Sea and coastal area of the East Sea from spring to fall. Pacific white-sided dolphin sightings were restricted to the middle and upper coastal areas of the East Sea in summer. Common dolphins were sighted from east of the southern coast to the eastern coast of the Korean Peninsula from spring to fall. Finless porpoise occurred in all Korean coastal areas, except the middle and upper eastern coast.

Two Spiny Lobsters of the Genus Panulirus (Crustacea: Decapoda: Palinuridae) from Korean Waters

  • Two spiny lobsters of the genus Panulirus, P. stimpsoni Holthuis, 1963 from Jeju Island and P. versicolor (Latreille, 1804) from Busan, are briefly described with colorations. These lobsters are new members of the Korean decapod crustacean fauna. Morphological and distributional accounts of these species are discussed.

Screening of Selected Korean Sweetpotato (Ipomoea batatas) Varieties for Fusarium Storage Root Rot (Fusarium solani) Resistance

  • A common post-harvest disease of sweetpotato tuber is root rot caused by Fusarium solani in Korea as well as the other countries. Storage root rot disease was monitored earlier on sweetpotato (Ipomoea batatas) in storehouses of different locations in Korea. In the present study, an isolate SPL16124 was choosen and collected from Sweetpotato Research Lab., Bioenergy Crop Research Institute, NICS, Muan, Korea, and confirmed the identification as Fusarium solani by conidial and molecular phylogenetic analysis (internal transcribed spacer ITS and translation elongation factor EF 1-α gene sequences). The isolate was cultured on potato dextrose agar, and conidiation was induced. The fungus was screened for Fusarium root rot on tuber of 14 different varieties. Among the tested variety, Yenjami, Singeonmi, Daeyumi, and Sinjami showed resistant to root rot disease. Additionally, the pathogen was tested for pathogenicity on stalks of these varieties. No symptom was observed on the stalk, and it was confirmed that the disease is tissue specific.

Development of Chemical Vapor Sampler for Man-in-Simulant Test(MIST) (화생방 개인보호체계 시험평가용 화학증기 흡착 샘플러의 개발)

  • We have recently developed a cost-effective and pouch-type chemical vapor sampler which consists of a selectively permeable high density polyethylene(HDPE) membrane, aluminum/nylon barrier film, and adsorbents. Since the sampler mimics the actual adsorption process that occurs when the skin is exposed to chemical vapors, it can be applied to man-in-simulant test(MIST) to determine the protective capability of individual protective ensembles for chemical warfare agents. In this study, we describe the manufacturing process of samplers and results for performance testing on MIST. Methyl salicylate(MeS) is used to simulate chemical agent vapor and the vapor sampler was used to monitor chemical concentration of MeS inside the protective suit system while worn. Values of protection factors(PF) were also analyzed to provide an indication of the protection level of the suit system evaluate by MIST. The results obtained by home-made samplers(ADD samplers) and commercially avaliable ones(Natick samplers) showed no significant differences.
