Screening of Herbal Medicines from China with Inhibitory Activity on Advanced Glycation End Products Formation (XIII) (중국 약용식물의 최종당화산물 생성저해활성 검색(XIII))

  • Advanced glycation end products (AGEs) have been postulated to play a central role in the development of diabetic complications. A variety of different agents that inhibit AGEs have been under investigation. In this study, 111 herbal medicines from China have been investigated with an in vitro evaluation system using AGEs formation inhibitory activity. Of these, 9 herbal medicines (IC50: <5 μg/ml) were found to have significant AGEs formation inhibitory activity. Particularly, herbal medicines Barleria cristata (leaves), Calotropis gigantea (stems), Ardisia virens (leaves), Dalbergia yunnanensis (leaves) Pittosporum runcatum (leaves), Ardisia japonica (leaves), Rhododendron racemosum (twigs), Oxyria sinensiss (aerial parts), Pyrrosia calvata (whole plants), showed more potent inhibitory activity (approximately 15-40 fold) than the positive control aminoguanidine (IC50: 76.47 μg/ml).

  • In this study, 80 herbal medicines from Vietnam have been investigated with an in vitro evaluation system using advanced glycation end products (AGEs) formation inhibitory activity. Of these, 10 herbal medicines (IC50: <5 μg/ml) were found to have significant AGEs formation inhibitory activity. Particularly, herbal medicines Strobilanthes pateriformis (aerial parts), Rhodamnia dumetorum (twigs), Glochidion rubrum (twigs), Dipterocarpus obtusifolius (twigs), Bombax ceiba (twigs), Amesiodendron chinense (twigs), Bauhinia coccinea (twigs), Lithocarpus laouanensis (twigs), Bauhinia bracteata (twigs) and Connarus paniculatus (twigs), showed more potent inhibitory activity (approximately 16-31 fold) than the positive control aminoguanidine (IC50: 76.47 μg/ml).

Establishment of in vitro 3-Dimensional Tumor Model for Evaluation of Anticancer Activity Against Human Solid Tumors (항고형암제의 활성평가를 위한 in vitro 삼차원 암세포 배양계의 확립)

  • For the efficient determination of activity against solid tumors, an in vitro tumor model that resembles the condition of in vivo solid tumors, is required. The purpose of this study was to establish a rapid culture method and viability assay for an in vitro 3-dimensional tumor model, multicellular spheroid (MCS). Among 12 human cancer cell lines, a few cell lines including DLD-1 (human colorectal carcinoma cells) formed fully compact MCS which was adequate for in vitro viability assay. DLD-1 MCS showed steady growth reaching $700\;{\mu}m$ diameter after 11 day culture. DLD-1 cells grown as MCS showed significant increase in $G_0/G_1$ phase compared to the monolayer cells (73.9% vs 45.7%), but necrotic regions or apoptotic cells were not observed. The cells cultured as MCS showed resistance to 5-FU (10.3 fold higher $IC_{50}$) compared to monolayers, however, tirapazamine (a hypotoxin) showed similar activity in both culture systems. In summary, MCS may be a valid in vitro model for activity screening of anticancer agents against human solid tumors and also exploitable for studying molecular markers of drug resistance in human solid tumors.

Of these, 6 herbal medicines ($IC_{50}&lt;5{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Particularly, herbal medicines Punica granatum (peels), Terminalia chebula (fruits), Rheum palmatum (roots), Oxyria digyna (stems and leaves), Anisodus luridus (roots) and Quercus schottkyana(stems and leaves) showed more potent inhibitory activity (approximately 9-43 fold) than the positive control aminoguanidine ($IC_{50}=77.04{\mu}g/ml$).

  • In recent, the abuse of newly-emerging psychoactive drugs, ('designer drugs') is a rapidly increasing problem in Korean society. Quantitative-structure activity relationship (QSAR) is an alternative method to predict bioactivities of new abused compounds. In this study, cathinone-related new designer drugs, 4-methylbuphedrone and 4-methoxy-N,N-dimethylcathinone were tested for prediction of the bioactivity with QSAR model. The bioactivity of 4-methylbuphedrone and 4-methoxy-N,N-dimethylcathinone was similar to those of methylone. These results suggest that the prediction with QSAR model may provide scientific evidences for regulatory decision.

  • This study presents a teaching model to increase the participation and interest, and to improve their understanding of physical concepts of first-year engineering students taking physics(2) course at a three-year college. In the class, a team task solution based on teamwork and a peer learning method through questions and answers between participants in each team were applied so that learners could actively participate in the class to discuss and present. We examined how the activities of each team affected students' interest in subjects, motivation to learn, and the degree of understanding of physical concepts. In the team activity, students were able to actively participate through emotional sharing between learners and free questions and explanations, and it was confirmed that academic achievement was improved by comparing the final exam evaluation results with the evaluation results of the previous three years.

Thiobenzamide S-oxidation in Perfused Rat Liver: Ex Vivo Determination of Hepatic Flavin-Containing Monooxygenase Activity

  • An ex vivo assay determining the flavin-containing monooxygenase (FMO) activity in perfused rat liver has been developed by assessing the rate of thiobenzamide S-oxide (TBSO) formation from the infused thiobenzamide (TB). The hepatotoxicity by TB or TBSO was not a critical factor for maintaining the FMO activity for up to 50 min. The FMO activity expressed in nmoles TBSO produced/g liver/min was the same for the recycling and non-recycling perfusion. This implies that reduction of the oxidized TBSO back to the parent compound (TB) is negligible. Hydrolysis of the collected perfusates with either ${\beta}-glucuronidase$ or arylsulfatase did not increase the TBSO level and thus, TBSO does not appear to undergo conjugation either to glucuronide or sulfate esters. Thus, measuring the rate of TB S-oxidation in the isolated perfused liver with 1 mM TB for 50 min provides a useful tool for evaluation of the hepatic FMO activity in the absence of hepatic necrosis and without the interferences caused by further conjugation or back reduction of the TBSO to the parent TB.

Anti-Photoaging Effects of Angelica acutiloba Root Ethanol Extract in Human Dermal Fibroblasts

  • The effects that ultraviolet rays elicit on collagen synthesis and degradation are the most common causes of wrinkle formation and photo-aging in skin. The objectives of this study were to evaluate the effects of Angelica acutiloba root ethanol extract (AAEE) to promote collagen synthesis and inhibit collagen degradation in human dermal fibroblasts. By examining total polyphenol and flavonoid contents, electron donating ability, radical scavenging activity, and superoxide dismutase-like activity, we found that AAEE exhibited fairly good antioxidant activity. Treatment with AAEE significantly increased type I procollagen production by cultured fibroblasts, as well as reduced ultraviolet-induced matrix metalloproteinase-1 (MMP-1) expression and MMP-2 activity in a dose-dependent manner (p < 0.05). In addition, AAEE significantly increased TIMP-1 mRNA expression (p < 0.05), although without an associated dose-dependent increase in TIMP-1 protein expression. In summary, we suggest that AAEE may be a potentially effective agent for the prevention or alleviation of skin-wrinkle formation induced by ultraviolet rays.

Physiological Functionalities of Vitis hybrid (Sheridan)-Rubus coreanus Red Wine Made by Saccharomyces cerevisiae

  • Vitis hybrid (Sheridan)-Robus coreanus red wine was vinified by fermentation of a mixture of Vitis hybrid and Robus coreanus must at $25^{\circ}C$ for 10 days. The Vitis hybrid-Robus coreanus red wine had ethanol contents of 10.9%. It had high antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of 57.8% and antioxidant activity of 64.8%. Changes in the physicochemical properties and functionality of the Vitis hybrid-Robus coreanus red wine was investigated during a post-fermentation period of three months. The ACE inhibitory activity of the red wine increased as the post-fermentation period prolonged, and showed the highest ACE inhibitory activity of 70.4% 60 days post-fermentation. However, the antioxidant activity declined significantly to 47.2% during the post-fermentation period of 60 days. In terms of sensory evaluation, the Vitis hybrid-Robus coreanus red wine had the best acceptability 60 days post-fermentation.
