암컷 마우스 생식기관의 형태에 미치는 에스트로겐 수용체 촉진제의 영향 (Effects of Estrogen Receptor Agonist on Morphology in the Female Mouse Reproductive Organs)

  • 에스트로겐은 수컷과 암컷의 생식기관 내에서 구조적이고 기능적인 변화를 유발하지만 그 정확한 기작에 대하여는 잘 알려져 있지 않다. 에스트로겐의 기능과 연관지어, 본 연구에서는 암컷 생식기관 내에서 에스트로겐 수용체 촉진제인 4,4',4"-(4-propyl-[1H]-pyrazole-1,3,5-triyl)tris phenol (PPT)의 작용을 확인하고자 하였다. 성체 암컷 마우스에 castor oil 0.06 mL에 PPT 3mg 농도로 희석한 용액을 1주일에 1회씩 3주, 5주, 8주 동안 각각 피하주사 하였으며, 대조군에는 castor oil 0.06 mL만을 투여하였다. 광학현미경을 이용하여 생식기관에 대한 PPT의 영향을 분석하였다. PPT에 의해 실험기간 동안 체중, 난소 및 지방조직의 무게가 감소되었다. PPT 투여군에서는 대조군에 비해 난소의 직경이 감소되었으며, 난소 내 그라프여포와 황체의 수도 감소되었다. 자궁의 경우, PPT에 의해 근육층과 내막층의 높이가 감소함에 따라 내강이 증가되었다. 또한 자궁샘의 수도 감소되었다. 이런 결과들로 볼 때 PPT에 의해 암컷 생식기관의 형태학적 변화가 유발되고 이로 인해 생식력에도 변화가 있을 것으로 보인다.

Consequences of Post-grazing Residues Control and Birth Season on the Body Traits, Reproductive Performance and Offspring's Growth of Suckling Goats and Ewes Reared at Pasture in Guadeloupe (FWI)

  • In Guadeloupe small ruminants (SR) are reared for meat production under pasture conditions. Intensive rotational grazing systems (irrigated, fertilised and high stocked) allow reasonable levels of production but generate high post-grazing residues. Experiments were designed to control them. A system in which residuals were mown (RM) was tested in comparison to the control system (Residuals Remained, RR). The same design was carried out for two years with Creole goat (G) and Martinik sheep (S). An accelerated reproductive rate (3 parturitions over 2 years) was carried out. Systems were compared at three parturition seasons per year(dry, intermediate and rainy seasons). Each group was composed of 20 goats ($36.0{\pm}2.5kg$) or 20 ewes ($46.8{\pm}2.4kg$). The female body traits did not vary according to pasture management and seasons. The stocking rate averaged 1,400 kg LW/ha. The mean fertility rate for does varied significantly (p<0.05) within the kidding season, from $80.4{\pm}0.5%$ to $93.7{\pm}2.9%$ while the mean litter size was $2.30{\pm}0.07$ total kids born. No effect of pasture system was observed. Corresponding values for ewes were $83.2{\pm}12.8%$ vs. $75.6{\pm}12.5%$ (p<0.05) and $2.43{\pm}0.24$ vs. $2.03{\pm}0.29$ (p<0.01) total lambs born for SRM and SRR ewes, respectively. A seasonal effect was observed upon ewe performances. The preweaning mortality of kids and lambs averaged 16.3% and 14.4%, respectively. It was 7 and 9 percentage points more (p<0.01) for RR than for RM kids and lambs, respectively. For both species, weaning took place at an average age of $81.4{\pm}3.6days$. In Creole kids, live weight at birth and at weaning were $1.9{\pm}0.2kg$ and $8.9{\pm}0.8kg$, respectively. In the Martinik sheep, the traits averaged $2.9{\pm}0.2kg$ and $18.9{\pm}0.9kg$. For both traits in both species, significant (p<0.05) group${\times}$season interactions were recorded. The consequences of elimination of post-grazing residues varied according to the SR species, the environmental conditions and the animal physiological status. The forage characteristics were not limiting factors since forage availability in the RM systems (2,300 and 2,600 kg DM/ha, respectively) and chemical composition were at satisfactory levels (CP content averaged 12 and 10%). It is recommended to develop new grazing system which would allow the use of post-grazing residues instead of mowing the refusals.

Ameliorating Effect of Selenium against Arsenic Induced Male Reproductive Toxicity in Rats

  • Oral exposure of humans by excess amounts of arsenic may cause disturbances of the reproductive system. In the present study, such exposure was modelled in rats, with the support of sperm principal parameters and histopathological observations. Male Sprague-Dawley rats were randomly divided into three groups where the group I was served as a normal control, group II was received sodium meta-arsenite as arsenic (10 mg/kg b.w/day) and a combination of sodium meta-arsenite and sodium selenite (3 mg/kg b.w/day) in group III. After 6 weeks, there was no significant change in testis weight and in total motility of all the three experimental groups, whereas, rapid moving spermatozoa, moderately moving spermatozoa and slow moving spermatozoa were significantly decreased in arsenic treated rats as compared to control rats. The other sperm principal parameters like progressiveness, average path velocity, straightness linear velocity (VSL), curvilinear velocity (VCL), straightness, linearity sperm head elongation ratio, area, linearity amplitude of lateral head department (ALH) and beat cross frequency (BCF) were found to be reduced in arsenic intoxicated rats. These results are not correlated with the histological studies. On oral administration of selenium ameliorated the adverse effects of arsenic as compared to arsenic alone treated rats. Our findings clearly demonstrate that administration of selenium could prevent some of the deleterious effects of arsenic in the testis.

  • The Dromedary camel (Camelus dromedaries) is an important species because of its ability to produce good quality meat, milk, and fibers under harsh environmental conditions. Camels are also crucial for transportation, racing, and as draft animals in agriculture. Therefore, dromedary camels play a critical role in the economy for millions of people living in the arid part of the world. The inherent capability of camels to produce meat and milk is highly correlated with their reproductive performance. Compared with other domestic species, the reproductive efficiency in camelids is low. Although recent reproductive technologies such as in vitro fertilization (IVF) and somatic cell nuclear transfer (SCNT) have been successfully applied to camelids and the birth of live offspring following these technologies has been reported; in vitro embryo production (IVP) has lagged in this species. The development of the IVP system for dromedary camels may be a useful tool for the genetic improvement of this species. IVP in farm animals includes three main steps; in vitro maturation (IVM) of an oocyte, IVF of a matured oocyte, and in vitro culture (IVC) of fertilized oocyte up to the blastocyst stage. This review aims to summarize various factors that influence oocyte quality, IVM, and in vitro embryo development in dromedary camel.

Excess of leptin inhibits hypothalamic KiSS-1 expression in pubertal mice

  • Purpose: Leptin has been considered a link between metabolic state and reproductive activity. Defective reproductive function can occur in leptin-deficient and leptin-excessive conditions. The aim of this study was to examine the effects of centrally injected leptin on the hypothalamic KiSS-1 system in relation to gonadotropin-releasing hormone (GnRH) action in the initial stage of puberty. Methods: Leptin (1 ${\mu}g$) was injected directly into the ventricle of pubertal female mice. The resultant gene expressions of hypothalamic GnRH and KiSS-1 and pituitary LH, 2 and 4 hours after injection, were compared with those of saline-injected control mice. The changes in the gene expressions after blocking the GnRH action were also analyzed. Results: The basal expression levels of KiSS-1, GnRH, and LH were significantly higher in the pubertal mice than in the prepubertal mice. The 1-${\mu}g$ leptin dose significantly decreased the mRNA expression levels of KiSS-1, GnRH, and LH in the pubertal mice. A GnRH antagonist significantly increased the KiSS-1 and GnRH mRNA expression levels, and the additional leptin injection decreased the gene expression levels compared with those in the control group. Conclusion: The excess leptin might have suppressed the central reproductive axis in the pubertal mice by inhibiting the KiSS-1 expression, and this mechanism is independent of the GnRH-LH-estradiol feedback loop.

Effects of Gamisoyosan on In Vitro Fertilization and Ovulation of Stressed Mice by Electric Shock

  • Exposure to stress is known to precipitate or exacerbate many reproductive dysfunctions such as dysmenorrhea and infertility. Abnormalities of the reproductive system, as shown by reduced ovulation, fertilization and early embryonic development, are frequently seen in dysmenorrhea and infertility. It has been generally accepted that Gamisoyosan (GSS) is a useful prescription for treating insomnia, dysmenorrhea and infertility induced by a stress. Also GSS has been used traditionally to improve systemic circulation and biological energy production. Based on these, this study investigates whether GSS improved ovarian dysfunction caused by stress in mice. Mice were subjected to stress by electric shock on the foot for 30 min daily for a week and treated with GSS at 500 / body weight per day for one week. Thereafter, changes body weight, adrenal weight, ovulation rate, in vitro and in vivo fertilization, embryonic development and estradiol concentrations were measured. GSS markedly increased the body weight of mice with stress, but not normal mice. The administration of GSS caused a reduction in adrenal weight in stressed mice. GSS also had significant positive effects on ovulation rate, estradiol production, in vivo and in vitro fertilization rates and embryonic development. These results indicate that GSS can improve the reproductive dysfunctions caused by stress, and these may production biological energy.

Role of exon 7 PTEN Gene in Endometrial Carcinoma

  • Background: Endometrial carcinoma is the most common malignant tumor of the female genital tract and the fourth most common cancer in Iranian women after breast, colorectal and lung cancers. Various genetic alterations appear to be early events in the pathogenesis of endometrial carcinoma and it seems that PTEN is the most commonly mutated gene in the endometrioid subtype. The aim of the present study was to investigate the correlation between mutations in exon 7 of PTEN gene and endometrial carcinoma. Materials and Methods: Seventy-five patients with endometrial carcinoma and 75 females whose underwent hysterectomy for non tumoral indication were selected for evaluation of PTEN mutations in exon 7 by PCR-SSCP and sequencing. Correlations between the frequency and type of mutation and the pathologic findings of the cancer (tumor subtype, stage and grade) were assessed. Results: All of the samples were obtained from Iranian patients. 60 % (45 cases) of the tumors were endometriod and 40% (30 cases) were of serous type. The grade distributions of the 75 cases according to the FIGO staging system were as follows: low grade, 20 cases; high grade 55 cases, low stage, 41 cases; high stage 34 cases. For exon 7 of the PTEN gene, the analysis showed that there were no mutations in our cases. Conclusions: Our findings in the present study suggest that exon 7 of PTEN does not play any significant role in the development of endometrial carcinoma in Iranian cases.

백자인(柏子仁)이 수컷 생쥐의 생식세포에 미치는 항산화 효과 연구 (Study of Antioxidant Effects of Platycladi Semen on Male Reproductive cells)

  • Objectives : The purpose of this study is to examine the antioxidant effects on male mouse reproductive cells of the extract of Platycladi Semen. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay, lipid peroxidation by malondialdehyde (MDA) formation and super oxide dismutase (SOD), respectively. Results : The results showed that the extract scavenged DPPH radical in a dose-dependent manner by up to 74.87%. The cell viability of the extract was within 72~96% on Leydig cells and GC-2 cells at concentrations of 1, 5, 10, 50 and 100 ug/ml. The hydrogen peroxide-induced cytotoxicity of Leydig cells was protected to 72.09% by the extract at concentration of 100 ${\mu}g/ml$. The hydrogen peroxide-induced lipid peroxidation of MDA formation was decreased to 1.80 and 1.65 nmoles/mg protein by the extract at concentrations of 50 and 100 ${\mu}g/ml$. The extract at all concentrations, SOD activity was not significantly changed. Conclusions : In conclusion, the extract of Platycladi Semen has antioxidant effects on Leydig cells and protect male reproductive system against oxidative stress.

A Review of Recent Developments in Buffalo Reproduction - A Review

  • The buffalo is an important livestock resource in several countries of South Asia and the Mediterranean regions. However, reproductive efficiency is compromised due to known problems of biological and management origins, such as lack of animal selection and poor nutrition. Under optimal conditions puberty is attained at 15 to 18 months in river buffalo, 21 to 24 months in swamp buffalo and is influenced by genotype, nutrition, management and climate. However, under field conditions these values deteriorate up to a significant extant. To improve reproductive efficiency, several protocols of oestrus and ovulation synchronization have been adopted from their use in commercial cattle production. These protocols yield encouraging pregnancy rates of (30% to 50%), which are comparable to those achieved in buffaloes bred at natural oestrus. The use of sexed semen in buffalo heifers also showed promising pregnancy rates (50%) when compared with conventional non-sexed semen. Assisted reproductive technologies have been transferred and adapted to buffalo but the efficiency of these technologies are low. However, these latest technologies offer the opportunity to accelerate the genetic gain in the buffalo industry after improving the technology and reducing its cost. Most buffaloes are kept under the small holder farming system in developing countries. Hence, future research should focus on simple, adoptable and impact-oriented approaches which identify the factors determining low fertility and oestrus behaviour in this species. Furthermore, role of kisspeptin needs to be explored in buffalo.

Glioma Epidemiology in the Central Tunisian Population: 1993-2012

  • Background: Glioma is a heterogeneous central nervous system (CNS) tumor group that encompasses different histological subtypes with high variability in prognosis. The lesions account for almost 80% of primary malignant brain tumors. The aim of this study is to extend our understanding of the glioma epidemiology in the central Tunisian region. Materials and Methods: We analyzed 393 gliomas recorded in cancer registry of central Tunisia from 1993 to 2012. Crude incidence rates (CR) and world age-standardized rates (ASR) were estimated using annual population data size and age structure. Statistic correlations were established using Chi-square and Kaplan-Meier test. Results: Tunisian glioma patients were identified with a mean age at diagnosis of 48 years and 1.5 sex ratio (male/female). During the 19 years period of study the highest incidence value was observed in male group between 1998 and 2002 (CR: 0.28, ASR: 0.3). Incidence results underline increasing high grade glioma occurring in the adulthood in the last period (2007-2012). Median survival was 27 months, with 1-, 2- and 5-year survival rates of 42%, 30% and 26%, respectively. Survival was greater in patients with younger age, lower tumor grade, infratentrial tumor location and undergoing a palliative treatment. Conclusions: This central Tunisia gliomas registry study provides important information that could improve glioma management and healthcare practice.
