  • Choi group reported that Kamijadowhan (KMD). an oriental herbal medicine, has anti-angiogenic effects and it may be a potential agent for clinical chemoprevention since it inhibits angiogenesis. Objectives of this experiment are to investigate acute, genetic and reproductive/developmental toxicities of KMD preparations. Acute toxicity was performed after single administration of KMO (200-500 mg/kg) to mice. Supravital staining micronucleus assay was conducted using peripheral reticulocytes in mice. (omitted)

  • It has been reported that 2-bromopropane might be a causative agent for reproductive toxicity and have immunotoxic effects. 1-Bromopropane known as an alternative to ozone depleting solvents, which has structural similarity to 2-bromopropane, has been reported to be neurotoxic to rats in long-term inhalation exposure.   • The three medicinal herbs-Curcuman longa Linne, Paeonia japonica Miyabe, Scutellaria baikalensis George-irradiated with gamma rays were tested for their possible genotoxicity. The methanol-soluble and water-soluble fractions of the 10kGy gamma-irradiated herbs were examined in cultured Chinese hamster ovary(CHO) cells for their ability to induce micronuclei. No mutagenicity of each test material was detected in the assay with or without metabolic activation. The safety of the herbs irradiated with gamma rays at practical doses needs further investigations using in vivo genotoxicity and chronic and reproductive toxicity tests.

No mutabenicity was detected in this assay with or without metabolic activation. The safety of the herbs irradiated with gamma rays at practical doses needs to be evaluated in further tests of genotoxicity in vivo and chronic and reproductive toxicity.

감마선 조사 홍삼의 안전성에 관한 유전독성학적 연구 (Genotoxicological Safety of the Gamma-Irradiated Korean Red Ginseng In vitro)

  • This experiment was performed to determine the safety of the Korean red ginseng irradiated with gamma rays with respect to genotoxicity. Ethanol extracts of the 5 and 10 kGy gamma-irradiated red ginseng were examined in two short-term in vitro tests : (1) Salmonella typhimurium reversion assay(Ames test) in strain TA 98, TA 100 and TA 102 (2) Micronucleus test in cultured Chinese hamster ovary(CHO) cells. No mutagenicity was detected in the two assays with or without metabolic activation. It was suggested that the Korean red ginseng irradiated with gamma rays did not cause genotoxicity in vitro. Further tests of genotoxicity in vivo, chronic and reproductive toxicity should be carried out to determine whether it is safe to irradiate Korean red ginseng with practical doses of gamma rays.

Toxicity and Biomedical Imaging of Fluorescence-Conjugated Nanoparticles in Hematopoietic Progenitor Cells

  • Cellular uptake of nanoparticles for stem cell labeling and tracking is a critical technique for biomedical therapeutic applications. However, current techniques suffer from low intracellular labeling efficiency and cytotoxic effects, which has led to great interest in the development of a new labeling strategy. Using silica-coated nanoparticles conjugated with rhodamine B isothiocyanate (RITC) (SR), we tested the cellular uptake efficiency, biocompatibility, proliferation or differentiation ability with murine bone marrow derived hematopoietic stem/progenitor cells. The bone marrow hematopoietic cells showed efficient uptake with SR with dose or time dependent manner and also provided a higher uptake on hematopoietic stem/progenitor cells. Biocompatibility tests revealed that the SR had no deleterious effects on cell cytotoxicity, proliferation, or multi-differentiation capacities in vitro and in vivo. SR nanoparticles are advantageous over traditional labeling techniques as they possess a high level of cellular internalization without limiting the biofunctionality of the cells. Therefore, SR provides a useful alternative for gene or drug delivery into hematopoietic stem/progenitor cells for basic research and clinical applications.

  • 3-Monochloro-1, 2-propanediol(3-MCPD) is a toxic compound, often present in different foods containing acid hydrolyzed(AH) protein, like seasonings and savory food products. In Korea, 3-MCPD is currently being a problem because of its toxicity in AH soybean sauce. The purpose of the present studies was to investigate the effects of 3-MCPD on male fertility, sperm and testosterone secretion. In vivo male fertility test was performed for observing the adverse effects of 3-MCPD on the function of male reproductive system and pregnancy outcome. (omitted)

  • 1-and 2-Bromopropane were reported as the causative agents for reproductive toxicity and immunotoxicity. The glutathione (GSH) metabolites resulting from in vitro treatment of 1- and 2-bromopropane were detected, identified and characterized. For the facile identification, expected GSH metabolites rormed by 1- and 2-bromopropane were chemically synthesized as reference materials (positive controls) and characterized by $^1H$-NMR, $^13C$-NMR, HPLC and LC/MS/MS. The treatment of GSH and S-9 fraction with 1- or 2-bromopropane at a physiological condition (pH 7.4, $37^\times$) for 1hr produced GSH metabolites, which were identified by UV, HPLC and ESI LC/MS/MS analyses.
