Spatial Genetic Structure and Genetic Diversity of a Rare Endemic Juniperus chinensis var. sargentii in Mt. Halla, Korea (희귀식물인 눈향나무(Juniperus chinensis var. sargentii)의 공간분포에 따른 유전구조 및 유전적 다양성)

  • Juniperus chinensis var. sargentii Henry is a short and creeping evergreen shrub which reaches about 50㎝ in height and occurs in the northeast Asia and in high mountains over the South Korea. Its distribution is restricted, and the number of individuals are gradually decreasing. This study was conducted to estimate spatial pattern, genetic diversity and spatial genetic structure of J. chinensis var. sargentii. A total of 131 clumps were studied in the study area (40m × 60m). The spatial pattern of this population was random (Aggregation index R=1.031). In spite of the small number and the limited distribution, the level of genetic diversity (Shannon's index 1=0.463) was relatively high as compared with those of other plant species with similar ecological characteristics. ISSR genotypes of all individuals were investigated to find the genetic relationship of clumps and genets. Fifteen clumps were composed to be clones, and a total of 116 unique genotypes were composed by separate genets. Spatial autocorrelation analysis using Tanimoto distance showed that the genetic patch was established within 8m. The effect of clonal reproduction on genetic structure was almost nothing.

  • Five cases infected with Rhabditis sp. were detected in a survey to examine the stool specimens from rural primary school children. A large number of the larvae of Rhabditis sp. detected by the direct cellophane thick smear were cultured by the alter paper method. The eBamination was carried out in April through June, 1980 in Tangjeong-Myon, Ahsan.Gun, and in September 1983 in Sandong-Eup, Yeongwol-Gun. The results obtained in this study were as follows: 1. Prevalence: Out of 925 children, 5(0.54%) children were found to be positive. The number of the detected larvae was 110/0.1gm of feces in case 1, 35 in case 2, 130 in case 3, 86 in case 4 and 62 in case 5. 2. Larvae: The larvae were prepared by means of the direct fecal smear and measured by a micrometer equipped in the microscope. Twelve (12) through 15-day old larvae in culture were $197.1{\mu}{\textrm{m}}$ long in average, and the maximum size of the matured stage larvae was $884.0{\times}25.9{\mu}{\textrm{m}}$. However, the length variation was ranged as 173.0 to $884.0{\mu}{\textrm{m}}$. 3. Adults: The size of clubbed adult female was $1, 357{\mu}{\textrm{m}}(1, 176~1, 419)$ in length and $80{\mu}{\textrm{m}}(79~82)$ in width. Length of buccal cavity was $33{\mu}{\textrm{m}}$. A long cylindrical esophagus($273{\mu}{\textrm{m}}$) of the worms with a valved posterior cardiac bulb and with median bulbar swelling was morphologically indicated. Ditance from mouth to vulva was occupied 58% of body length. Male worm was $1, 006{\mu}{\textrm{m}}(890~1, 148)$ in length and $49{\mu}{\textrm{m}}(48~49)$ wide. Caudal alee of bursa and spicules ($75{\mu}{\textrm{m}}$ in length) were well developed. 4. Eggs: The oval shaped eggs in the female uterus, when cultured, were $66{\times}56{\mu}{\textrm{m}}$ in sixte, and the eggs laid by the adult could not be detected. So, reproduction might be thought to be ovoviviparity. 5. The five cases were re-examined during the period from the 1st to the 3rd week after stool examinations, but Rhabditis sp. were detected again in 4 cases in 1st week. When they were examined in 3rd week, larvae could not be detected. So, it was thought that the infection of Rhabditis sp. to humans was facultative.

Spawning Periodicity and Behavior of Amphiprion melanopus and Development of Mass Hatching System for Clownfishes (Cinnamon Clownfish Amphiprion melanopus의 산란 주기, 산란 행동 및 Clownfish류의 부화장치 개발)

  • The marine ornamental industry has become a multi-billion dollar industry these days. As developing, however, this industry has been criticized for the indiscriminate captures and the destruction of the surrounding environment. To circumvent these problems, it is suggested to breed the organisms artificially. While clownfishes Amphiprion sp. and Premnas sp. are the most famous ornamental organisms in the trade, few studies are yet available on the culture and commercial production of these fishes. These studies were performed to investigate the spawning periodicity, behavior and the habits during egg incubation, and to provide the information on the mass hatching system. The spawning periodicity and frequency were different in 4 pairs under the constant condition, temperature, salinity and photoperiod. On the contrary, the male's behaviors for egg incubation are almost same in the all. The egg-fanning activity of the male increased as the developing eggs reaching to the hatching day. Based on the above results, we designed a new artificial hatching system, the rotating type (RT), and compared it with the aeration type (AT) and spray type (ST) that were previously described. RT showed higher hatching rate of 87.3% than AT (74.4%) and ST (60.5%). Also, there were no significant differences in the hatching rate regardless of the number (2, 3, 5) of hatching plates. We suggest RT may accommodate various number of hatching plates and constitute a better hatching system for clownfishes.

Maturation and Spawning of the Korean Anchovy Coilia nasus on the West Coast of Korea (한국 서해산 웅어, Coilia nasus 암컷의 성숙과 산란)

  • The gonadosomatic index (GSI), fatness, ovarian development, first sexual maturity, and fecundity of the Korean anchovy Coilia nasus were investigated by histological observations and morphometric analysis from January to December, 2007. The GSI and fatness began to increase in February, and reached the maximum in June when the ovary was getting mature and spawning occurred. Thereafter these parametes rapidly decreased in July when spawning occurred. Therefore, monthly changes in the GSI and fatness were closely related to ovarian maturation and spawning. The duration of ovarian development in females can be classified into five successive stages: early growing stage (February to March), late growing stage (March to April), mature stage (May to June), ripe and spent stage (June to July), and recovery and resting stage (December to January). Maturation and spawning of this species occurred between June and July during the period of high seawater temperature-long day length. Percentages of first sexual maturity in female individuals were over 50% for fish ranging 24.1 to 27.0 cm in total length, and 100% for fish over 30.1 cm in total length. The number of total eggs and mature eggs in the absolute fecundity were increased with the increase of total length and body weight, respectively. The number of total eggs and mature eggs in relative fecundity were also proportional to total length, but rather these numbers decreased in the maximum body weight (126.0${\sim}$150.0 g).

Various Expression Pattern of Beta-catenin in the Preimplantation Stage of Porcine Embryos

  • Beta-catenin is very important in early development including involvement in cell adhesion, cell signaling, and developmental fate specification. Cell-cell interaction is an important process during mammalian embryonic development. In preimplantation embryos, embryonic compaction is the process of increased cellular flattening and adhesion of junctional complexes and results in a polarized distribution. Beta-catenin is associated with embryonic compaction in mammals. Here, we examined the relationship between beta-catenin expression and compaction in porcine embryos derived from in vitro fertilization. First of all, we investigated beta-catenin expression in each embryonic developmental stage and also focused on expression pattern according to full, partial and non-compaction at morula stage. We used the immunocyto-chemical method in this research. To confirm compaction affects on the embryonic development, we compared between compaction and developmental rates to the blastocyst. The result showed that compaction and non-compaction rates were 14.6% and 63.8% at 4 days after IVF, respectively The developmental rates to the blastocyst and their total cell number were 50.9% vs 36.4% and 41.4$\pm$11.5 vs 26.8$\pm$12.7 in compaction and non-compaction groups. Although no difference was detected in the ratio of ICM to total cells between two groups, total cell number of the blastocysts in compaction group was superior to that of the blastocysts in non-compaction group (P<0.05). Expression of beta-catenin appeared in the boundary of membrane surface between blastomeres in 2- and 4-cell stage, and observed irregular pattern from 8-cell to blastocyst stage. We also investigated beta-catenin expression pattern according to the degree of compaction in the 3 groups; full, partial (>50%) and non-compaction. The expression signal in fully compacted embryos was stronger than those of partial and non-compacted embryos. Especially, beta-catenin expression appeared various patterns in morula stage suggesting the aberrant distribution of beta-catenin is affected by compaction patterns. Our results suggest that abnormal beta-catenin expression was affected by embryo quality and further development in porcine embryos in vitro.

Sphingoshine-1-Phosphate Enhances Meiotic Maturation and Further Embryonic Development in Pigs

  • Sphingosine-1-phosphate (S1P) has a many function involved proliferation, differentiation and survival of many cells. In this study, to investigate whether S1P improve the developmental competence of porcine embryos, 50 nM of S1P were supplemented during in vitro maturation (with EGF or without EGF) medium and/or in vitro culture (IVC) medium. Addition of S1P was significantly increased the rate of oocytes reaching metaphase II (MII) compared to the control (83.5 vs. 64.1%) in without EGF medium, but not with EGF medium (89.5 vs. 84.6%). When treated with $1{\mu}M$ of N1N-dimethylsphingosine (DMS), a sphingosine kinase inhibitor which is blocked endogenous generation of S1P, the meiotic progression rates to MII stage (without EGF: 45.2 and with EGF: 66.7%) were significantly decreased and degeneration rates (without EGF: 51.2 and with EGF: 30.1%) were increased in both medium compared to control group during IVM periods. Also, the rates of blastocyst formation was significantly increased in the S1P treated group compared to control group (29.0 vs. 19.2%) of EGF supplemented medium, whereas there were no effect in the EGF free medium (9.0 vs. 10.5%). After 12 h IVM, the phosphorylation of ERK1 and ERK2, which is major signaling pathway of MAP kinase, were increased in the S1P group than that of control or DMS group. When supplemented of S1P during IVC, the rates of blastocyst formation and total cell number (30.2% and 40.6) were significantly increased in S1P-treated group compared with control (20.1% and 32.5), DMS (12.3% and 25.1), and S1P plus DMS group (24.7% and 33.6). The percentage of apoptosis nuclei in the S1P group was significantly decreased than other groups. Also, the rates of blastocyst formation (26.7 vs. 14%) and total cell number (42.8 vs. 32.5) were significantly increased in the S1P group than those of control group when S1P added during the entire IVM and IVC periods. Taken together, our results indicate that S1P supplementation in IVM and/or IVC medium affects beneficial effect of meiotic maturation and subsequent developmental competence of porcine embryos.

Relationship between Estrous Expression, Body Condition Score (BCS) and Transferable Embryos Hanwoo Donors (한우 공란우의 발정발현율, 신체 충실 지수(BCS)와 이식가능 수정란과의 관계)

  • The objective of this study was to investigate the relationship between estrous expression, body condition score (BCS), blood urea nitrogen (BUN) and number of transferable embryos for the purpose of improving reproductive performance in blood of Hanwoo donors. Sixty, at random stages of the estrous cycle, received a CIDR. Four days later, the animals were superovulated with a total of 28AU FSH (Antorin, 2AU=1 ml) administered twice daily in constant doses over 4 days. On the 3th administration of FSH, CIDR was withdrawn and 25 mg PGF2 ${\alpha}$ was administered. Cows were artificially inseminated twice after estrous detection at 12 hr intervals. The cows received 100 ${\mu}g$ GnRH at the time of 1nd insemination. Embryos were recovered 7 or 8 days after the 1st insemination. The estrous inducement rate and estrous expression rate were significantly lower for cows with BCS below 2.25 than for cows with BCS above 2.25. There was 50.0% of rate of mounting in cows with BCS below 2.25 whereas the rate of mounting was markedly increased in cows with BCS above 2.25 (94.1% and 89.5% for BCS 2.25~2.75 and BCS above 2.75 cows, respectively). Cows with BCS <2.25, 2.25~2.75 and ${\geq}$2.75 had number of transferable embryos of $4.5{\pm}0.7$, $5.9{\pm}1.8$ and $5.6{\pm}2.3$ respectively.

Superovulatory Response to 200 mg FSH Level and Production In Vivo Embryos in Korean Native Cattle (Hanwoo) (200 mg FSH 투여에 의한 한우의 과배란 유도 및 체내 수정란 생산)

  • This study was performed to investigate the FSH levels for superovulation procedure in Korean Native Cattle (Hanwoo). The effectiveness of 200 mg and 400 mg of FSH to initiate superovulation was examined in Hanwoo. Donors, at random stages of the estrous cycle, received a CIDR 7 days later, 200 mg FSH group was treated with 40, 30, 20, 10 mg FSH levels in declining doses twice daily by intramuscular injection fur 4 days. Also, 400 mg FSH group was treated with 80, 60, 40, 20 mg FSH levels. On the 3rd day administration of FSH, 25 mg $PGF_2$ ${\alpha}$ was administered and CIDR was withdrawn. Donors were artificially inseminated twice at 12 hr intervals. The donor cattle received 250 ${\mu}g$ GnRH at time of 1st insemination and embryos were recovered 8 days after the 1st insemination. As a results, average number of CL treated with FSH 200 mg was higher as $20.9{\pm}1.20$ than $15.8{\pm}0.63$ for donors treated with FSH 400 mg, respectively(p<0.05). Treated group of 200 mg FSH level increased (p<0.05) the number of embryos recovered per procedure compared to 400 mg FSH level ($18.2{\pm}1.18$ vs. $12.38{\pm}0.52$, respectively). When treatment of 200 mg FSH was performed, average transferable embryos/ova increased (p<0.05) to $14.1{\pm}1.12$ from $6.8{\pm}0.33$ of treated of 400 mg FSH. Group of 200 mg FSH increased (p<0.05) to $8.3{\pm}0.76$ from $2.0{\pm}0.26$ in morula stage compare to 400 mg FSH group. Mean of total early blastocyst and expanded blastocyst stage embryos was similar (p<0.05) between the 200 mg and 400 mg FSH levels group ($4.7{\pm}1.19$ vs. $2.9{\pm}0.18$ and $1.2{\pm}0.40$ vs. $1.9{\pm}0.17$). These results suggest that 200 mg FSH level-based superovulation protocol with CIDR may be effectively used fur production of superior embryos in Hanwoo. In other words, the less level of FSH may be effectively applied for Hanwoo (Korean Native Cattle), because Hanwoo was smaller body size than beef or daily cow.

Effect of Humulus japonicus Extract on Sperm Motility, Fertilization Status and Subsequent Preimplantation Embryo Development in Cattle (소에서 정자활성, 수정 양상 및 착상전 지속적 수정란 발달에 있어서 환삼덩굴 추출액의 효과)

  • Humulus japonicus is an ornamental plant in the Cannabaceae family. Although the mode of action of Humulus japonicus is not fully understood, a strong relationship was observed between anti-inflammatory and anticancer in some types of cells. Recent studies also have shown that Humulus japonicus possesses anti-inflammatory activities and may significantly improve antioxidant potential in Raw 264.7 macrophage cells. Thus, the aim of this study was evaluated the effect of Humulus japonicus extract on sperm motility and subsequent preimplantation developmental competence of the bovine embryos. After in vitro maturation, the oocytes with sperms were exposed in in vitro fertilization (IVF) medium supplemented with Humulus japonicus extract (0.01, 0.05, $0.1{\mu}g/mL$, respectively) for 1 day. In our results, exposure of IVF medium to Humulus japonicus extract did not affect sperm motility and percentage of penetrated oocytes but ROS intensity was significantly decreased by $0.01{\mu}g/mL$ compared with other groups (p< 0.05). Moreover, treatment with $0.01{\mu}g/mL$ of Humulus japonicus extract was higher the frequency of blastocyst formation than the any other groups (p<0.05). Otherwise, treatment with $0.01{\mu}g/mL$ of Humulus japonicus extract not increased the total cell number but reduced apoptotic-positive nuclei number. In conclusion, our results indicate that supplementation of Humulus japonicus extract in IVF medium may have important implications for improving early embryonic development in bovine embryos.

Effect of EGF on In Vitro Oocyte Maturation and Embryo Development and Expression of EGF mRNA in Bovine Oocytes and Embryo I. Influence of Cumulus Expression and Maturation and Embryo Development during Bovine Oocyte Maturation In vitro by Addition EGF

  • The objective of this study was to the effect on subsequent development of EGF present in defined medium during bovine 1)oocyte maturation or 2)embryo culture. The presence of EGF during IVM, irrespective of concentration(1, 10, 100ng/$m\ell$), stimulated cumulus expansion and significantly increased the proportion of oocytes attaining metaphaseII, the rate of cleavage, and develop to blastocyst. 1. In the group of EGF-added medium(1, 10, 100ng/$m\ell$), nuclear maturation rate for in vitro maturation was 91% to 92% but was not significantly higher than control group(87%). 2. For in vitro maturation, in the group of EGF-added medium(1, 10, 100ng/$m\ell$)the rate of cumulus cell expansion degree 2 ranged from 81% to 87%, which was significantly higher than the control group(medium with EGF not added). The rate of in vitro fertilization, developing to 2-to 4- cell stage, was 76% to 80%, which was also significantly higher(p<0.05)than control group(62%). 3. For in vitro maturation, in the group of EGF added in medium(1, 10, 100ng/$m\ell$)the development rate to blastocyst was 24.3% to 27%, which was significantly higher than control group(13.7%). The total cleavage rate in the group of EGF-added medium was 77% to 82%, which was higher than control group. 4. The development rate to blastocyst for 6 days of cultivation and the hatching blastocyst were 30.6% and 59.1%, respectively, in the group of 100ng/$m\ell$ of EGF, which were significantly higher(p<0.05)than control group(14.0% and 24%, respectively), The numbers of cells in blastocyst were 140.2 and 148, respectively, in 10ng/$m\ell$ and 100ng/$m\ell$ of EGF-added medium, which were higher than 108.5 in control group. 5. The development rate of in vitro fertilized embryos to blastocyst in 10ng/$m\ell$ of EGF-added medium co-cultured with somatic cell was 28%, which was significantly higher(p<0.05)than control group(11.8%). The numbers of cells in blastocyst were 141.6 for EGF-added medium and 145 for EGF+co-culture group, which were higher than control(101.6)and medium co-cultured with somatic cells(110.6). These results showed that in vitro maturation and fertilization, EGF was found a significant effect of increase of development rate to blastocyst and cell number.
