GDNF secreted by pre-osteoclasts induces migration of bone marrow mesenchymal stem cells and stimulates osteogenesis

  • Bone resorption is linked to bone formation via temporal and spatial coupling within the remodeling cycle. Several lines of evidence point to the critical role of coupling factors derived from pre-osteoclasts (POCs) during the regulation of bone marrow-derived mesenchymal stem cells (BMMSCs). However, the role of glial cell-derived neurotrophic factor (GDNF) in BMMSCs is not completely understood. Herein, we demonstrate the role of POC-derived GDNF in regulating the migration and osteogenic differentiation of BMMSCs. RNA sequencing revealed GDNF upregulation in POCs compared with monocytes/macrophages. Specifically, BMMSC migration was inhibited by a neutralizing antibody against GDNF in pre-osteoclast-conditioned medium (POC-CM), whereas treatment with a recombinant GDNF enhanced migration and osteogenic differentiation. In addition, POC-CM derived from GDNF knock-downed bone marrow macrophages suppressed BMMSC migration and osteogenic differentiation. SPP86, a small molecule inhibitor, inhibits BMMSC migration and osteogenic differentiation by targeting the receptor tyrosine kinase RET, which is recruited by GDNF into the GFRα1 complex. Overall, this study highlights the role of POC-derived GDNF in BMMSC migration and osteogenic differentiation, suggesting that GDNF regulates bone metabolism.

  • This study attempted to analyze the factors that influence the participation of beneficiary companies in the government's defense industry promotion support project. To this end, experimental data were analyzed by constructing a prediction model consisting of highly important variables in beneficiary company decisions among various company information using the decision tree model, one of the data mining techniques. In addition, various rules were derived to determine the beneficiary companies of the government's support project using the analysis results expressed as decision trees. Three policy measures were presented based on the important rules that repeatedly appear in different predictive models to increase the effect of the government's industrial development. Using the analysis methods presented in this study and the determinants of the beneficiary companies of the government support project will help create a sustainable future defense industry growth environment.

  • As high as 50% of pregnancies are known to fail and the majority of such losses occur during the peri-implantation period. For the establishment of pregnancy in mammalian species, therefore, implantation of the conceptus to the maternal endometrium must be completed successfully. Physiological events associated with implantation differ among mammals. In ruminant ungulates, an elongation of the trophohlast in early conceptus development is required before the attachment of the conceptus to the uterine endometrium. Moreover, implantation sites are restricted to each uterine caruncula where tissue remodeling, feto-maternal cell fusion and placentation take place in a coordinated manner. These unique events occur under strict conditions and are regulated by numerous factors from the uterine endometrium and trophoblast in a spatial manner. Interferon-tau (IFN-${\tau}$), a conceptus-derived anti-Iuteolytic factor, which rescues corpus luteum from its regression in ruminants, is particularly apt to play an important role as a local regulator in coordination with other factors, such as TGF-${\beta}$, Cox-2 and MMPs at the attachment and placentation sites.

Evaluation Indicators for Green Libraries and Library Eco-friendliness

  • This study aimed to determine eco-friendly factors on the aspects of library service, materials provided or handled by the library, and programs including educational programs. Furthermore, it was setup to perform a trial evaluation for the eco-friendliness of the library based on the determined evaluation standards by this study. Results are as follows; First, regarding the question of 'Do employees of the library think that establishment of green libraries is necessary?', the employees of the libraries responded yes by 65.52%, but only 4.6% responded that it would not be necessary. Second, in analyzing the most important evaluation areas of green libraries according to the opinions of the libraries' employees, they were ranked in order: the indoor environment, energy and prevention of environmental pollution, and material and resource areas. Third, 11 areas were determined to evaluate the level of greening in the libraries; the highest averages were scored 510 for the area of energy and prevention of environmental pollution, 415 for the area of library resource, and 320 for the area of indoor environment. Last, the libraries were graded based on actual data according to the results to measure the level of greening, two libraries were graded as Diamond, the highest grade, accounting for 15.39% of all libraries; Gold and Silver grades accounted for another 15.39%, together five Certified graded libraries accounted for 38.46%, which was the highest rate among the libraries. Any library scoring less than 20% when compared to the full score was graded as Non-certified, and one library, which scored 15.06% fell into this category. The results of this study set up to serve as basic data for the direction of development for green libraries in our country, as well as reference for the employees of libraries in constructing or remodeling library buildings, establishing service infrastructure, providing information service, and planning library duties in an eco-friendly way.

  • Osteoarthritis is a noninflammatory degenerative disease affecting the articular surfaces and is accompanied by remodeling of the underlying bone. The sympotms of osteoarthritis of the temporomandibular joint are pain in the joint and muscles of mastication, limitation. Osteoarthritis is generally accepted that several factors can contribute to the trauma, aging process, internal derangement and MPDS. Radiographic features of the osteoarthritis are seen flattening of joint, sclerosis on flattened area and osteophyte or anterior lipping. In the past, osteoarthritis was considered to self-limiting disease. Currently, synovial chondromembrane is part of the process of osteoarthritis secondary to trauma. Synovial chondromatosis is an uncommon disease of cartilaginous transformation of synovial membrane with formation of loose bodies within the joint space. The pathogenesis is more an active metaplastic than a neoplastic process. The cause of synovial chondromatosis is unknown. Although trauma and inflammation have all been cited as possible factors in tis pathogenesis. The clinical sign and symptoms are unilateral swelling of the joint region, pain in the joint area and crepitus seem to be the most reliable signs and symptoms. Radiographic evidence of loose bodies may or may not be present. This is a case report of 66 year old female with synovial chondromatosis, that is advanced disease of the osteoarthrits. We treated patient with surgical excision of lose bodies, diskectomy and synovectomy. The defected articular fossa area was reconstructed with temporalis fascia flap. The result was satisfactory.

간세포성장인자가 골결손부의 치유에 미치는 영향 (THE EFFECT OF HEPATOCYTE GROWTH FACTOR FOR REPAIR OF THE BONE DEFECT)

  • Bone healing plays an important role in orthognathic and craniofacial surgery. Bone tissue repair and regeneration are regulated by an array of growth and morphogenetic factors. Bone formation and remodeling require continuous generation of osteoprogenitor cells from bone marrow stromal cells, which generate and respond to a variety of growth factors with putative roles in hematopoiesis and mesenchymal differentiation. In this study, the efficacy of a single application of hepatocyte growth factor to promote bone regeneration in 5-mm experimental calvarial defects of adult male rats was assessed histologically and immunohistochemically. The result of the experimental site were compared with those of the contralateral contral side. None of the control and experimental bone defects demonstrated complete bone closure. Bone regeneration was found close th the margine and central part of the defects. At 1, 2 weeks, there were found much significant cellural mitotic activity and many inflammatory cells and osteoblasts on the experimental site than control site. At 4, 6 weeks, new bone apposition was founded in both site but, more apposition was seen at experimental site. At 8, 12 weeks, also, some differences was found that more apposition of new bone and collagen fiber was seen on experimental site. Our results have some possibility that HGF do a early positive role to repair the bone defect. More study will be needed.

흰쥐의 급성 위점막 손상에서 황연탕(黃蓮湯)이 apoptosis 관련단백질 및 성장인자 발현에 미치는 영향 (The Protective Effects of $Hwangyeon-tang$ on Acute Gastric Ulcer induced by HCl/EtOH solution in Rats)

  • The apoptotic process of gastric mucosa triggered by induction of proapoptotic gene expression, such as Bax. Stress-inducing factors may affect Bcl-2/Bax ratio and thus the rate of apoptosis through modulation of the expression of both proteins depending upon the experimental model. TGF-${\beta}$ is believed to be essential in wound healing for regulation of cell growth and differentiation and is known to be involved in tissue repair and remodeling. The polypeptide growth factors, such as vascular endothelial growth factor(VEGF), regulate essential cell functions involved in tissue healing including cell proliferation, migration, and differentiation. The purpose of this study was to investigate whether the oral administration of $Hwangyeon-tang$ (HYT) would have protect effects on gastric ulcer in rat. Sprague-Dawley rats (n=40) were randomly divided into 4 groups ; Normal, Saline, Cimetidine and HYT group. The saline, cimetidine and HYT extract were orally administrated to each group and gastric ulcer was induced with HCl/EtOH solution. After 1 hour, the stomachs were collected for histological observation and immunohistochemistry. In Results, the wound healing of gastric ulcer was promoted by HYT and the significant alterations of BAX/Bcl-2, TGF-${\beta}1$ and VEGF proteins in gastric mucosa were observed. These results suggest that Fritillaria ussuriensis extract promotes wound healing and has protective effects on gastric ulcer in rats.

알레르기질환 아동의 가정 내 환경요인과 문제행동의 관계: 2015년 한국아동패널 자료를 바탕으로 (Association between Environmental Factors in Home and Behavioral Problems in Children with Allergic Diseases: Based on 2015 Panel Study of Korean Children Survey)

  • Purpose: This study aimed to investigate the association between indoor environmental exposures and behavioral problems in children with allergic diseases. Methods: We used data from 2015 Panel Study of Korean Children (PSKC). The subjects of this study included 825 children aged 7 years with asthma, allergic rhinitis, or atopic dermatitis. The data was analyzed using hierarchical multiple regression. Results: Factors influencing behavioral problems in children with allergic diseases were passive smoking (β=.15, p<.001), painting from 1year after birth until 1 year ago (β=.13, p<.001), using of linoleum as floor materials (β=.09, p<.001), change of wallpaper From 2 years before pregnancy until 1year after birth (β=.08, p<.001), change of wallpaper from 1year after birth until 1 year ago (β=.07, p<.001), keeping hot food or water in plastic container (β=.06, p<.001), remodeling from 1 year after birth until 1year ago (β=.04, p<.001), using of plastic container (β=.03, p<.001), and change of floor materials from 1 year after birth until 1year ago (β=.01, p=.006) which explained about 10% of behavioral problems. Conclusion: This study showed that indoor environmental exposures were associated with behavioral problems in children with allergic diseases. Based on the findings of this study, programs focusing on controlling of indoor environmental exposures may help to prevent and reduce behavioral problems in children with allergic diseases.

치유환경을 위한 광선반 부착방법에 따른 노인요양시설 침실 내 자연채광 유입 환경 연구 - 기상데이터 기반 동적 자연채광 시뮬레이션을 기반으로 - (A study on Daylighting inducement within bedroom of Elderly care facility by light shelf attaching method for Therapeutic environment - By Dynamic Daylight Simulation Using Weather Data -)

  • There are high recognitions on the importance of comforts in Elderly living environment, but the circumstance is that studies on seniors facility space itself are approached only in planning level, and studies on lighting environment which is significantly associated with the comfort in the indoor environment of seniors where they actually spend the majority of their time are not that active. This study was intended to deduce cozy bedroom environment to which existing elderly care facility can be improved by using light shelf the lighting system with the advantage of being able to serve both as building sun visor and lighting window simultaneously in order to analyze the interior environment of bedroom space of elderly care facility the indoor space where the aged spend the majority of their life and examine the directions for the improvement of existing building lighting system through remodeling and renovation. In this study, lighting performance analysis was done in a way that the windows of the bedroom unit in existing facility were set in southbound direction based on two standard types and were put under initial simulation with the use of Autodesk Revit 2011, and after the simulation results were converted to Green Building Studio gbXML file to be used in ECOTECT, Daylight Autonomy a dynamic simulation and static natural lighting simulation the existing method of calculating daylight factors were deduced through Ecotect Analysis 2011. In conclusion, exiting standard model was found in such a condition that the daylight factors for both type A and type B were above 5% the proper standard value, and required improvement. In case light shelf the natural lighting system was attached, the daylight factor was improved to proper standard value for type A, and also was improved above existing facility for type B.

신축 아파트의 TVOC 농도 및 거주자의 새집증후군 반응 (TVOC Concentrations and Residents' Responses on Sick House Syndrome of Newly-Built Apartments)

  • The purpose of this study was to identify the influence of living factors on TVOC(Total Volatile Organic Compounds) concentrations and personal factors to reduce the Sick House Syndrome for newly-built apartments. The field measurements of TVOC concentrations were made totally 30 times(5 times per one subject house) in six apartment units in which residents recently moved. Those apartments authorized its business approval before May 2004, the Act of Indoor Air Quality Management for multi-use facilities. The Questionnaire surveys of residents' subjective responses on the Sick House Syndrome were carried out in 2nd measurement of each house. Respondents consisted of 20 residents living in the measured houses. The findings were as follows: The TVOC concentrations of the measured subject apartments ranged from about 1/10 of the recommended standard for multi-use facilities($400{\mu}g/m^3$) to up to 90 times as high as the standard. Since then, the Recommended Standard of Indoor Air Quality Management for newly-built apartment house was announced in December 2005. In accordance with this standard($2390{\mu}g/m^3$) it ranged from about 1/100 to up to 15 times as high as the standard. The subject house whose TVOC concentrations reduced below the recommendation standard in the shortest period had the largest amount of ventilation (all the windows were open for ventilation in the past three months) among all measured houses. The reason of another house whose TVOC concentrations were much higher than the rest was fronted with new furniture in the room. There turned out to be no apparent relations between the TVOC concentrations and the residents' individual responses of Sick House Syndrome. The responses were serious in those who stayed in their new houses for a long period or had disease like allergy. It's recommended that they should open all the windows for at least three months for ventilation in newly-built houses, and it would be better to avoid remodeling than needs be.
