Biological function of CpSlt2, an ortholog of the cell wall integrity (CWI) MAPK of Saccharomyces cerevisiae, in the chestnut blight fungus Cryphonectria parasitica

  • Cryphonectria parasitica, chestnut blight fungus, has a characteristic of decreasing pathogenicity when infected with Cryphonectria hypovirus 1. C. parasitica is known to be one of the most representative model systems used to observe the interaction between viruses, plants and fungi. The mitogen-activated protein kinase (MAPK) pathway, which is well conserved in various organisms ranging from yeast to humans, functions in relaying phosphorylation-dependent signals within MAPK cascades to diverse cellular functions involved in the regulation of pheromone, cell wall integrity, and osmotolerance in filamentous fungi. Several genes in the MAPK pathway were revealed to be regulated by hypovirus, or to be involved in pathogenicity in C. parasitica. Among these pathways, the CWI pathway has aroused interest because CpBck1, an ortholog of yeast Bck1 (a CWI MAPKKK), was previously reported to be involved in cell wall integrity and sectorization. Interestingly, sporadic sectorization was observed in the CpBck1 mutant and sectored phenotypes were stably inherited in the progeny that were successively transferred from sectored mycelia. In this study, we analyzed the biological function of CpSlt2, downstream gene of CpBck1, to confirm whether the sectorization phenomenon occurred in the specific single gene or cell wall integrity (CWI) pathway. As results, the CpSlt2-null mutant exhibited marked changes in colonial growth, near absence of conidiation and aerial hyphae, abnormal pigmentation, CWI-related phenotypic defects, and dramatically impaired virulence. As cultivation of the mutant strains progressed, the majority of the colonies showed sporadic sectorization and mycelia from the sectored area stably maintained the sectored phenotype. These results suggest that the unique sectorization is CWI pathway-specific, though the components in the same CWI pathway have common and specific functions.

A Study on Complex Field Network Coding Scheme for Wireless Relay System (무선 릴레이 시스템에서의 Complex Field Network Coding 기법 적용에 관한 연구)

  • In this paper, a CFNC (Complex Field Network Coding) scheme is presented, which focuses on improving transmission efficiency by reducing time slots that are needed to exchange data frames. Conventional CFNC scheme has the advantage of minimizing the number of time slots required for information exchanges. However, there exists serious performance deterioration because the transmit signals are interfered with each other. Moreover, when CFNC scheme is applied, the estimation and compensation performance of fine frequency offset severely deteriorates due to the multiple frequency offsets in received signal of relay node. In order to overcome these critical problems, we propose an improved CFNC scheme with modified maximum likelihood decision method which uses uplink transmit diversity. Also, we propose an enhanced fine frequency offset estimation method and a corresponding compensation method to deal with the multiple frequency offsets. Corresponding simulation results verify that the proposed methods are able to effectively solve the problems of CFNC scheme.

Performance of DOT Relay System with MRC/GSC receiver in Rayleigh Fading Channels (레일레이 페이딩 채널에서 MRC/GSC 수신하는 DOT 릴레이 시스템의 성능)

  • Opportunistic transmit cooperative relaying (OTR) system has been interested for its ability to mitigate the fading in wireless channel without multiple antennas in a small terminal. In OTR system, only the relays that the received Signal-to-noise ratio (SNR) from a source is greater than the threshold transmit to the destination. However, the receiving branches of a destination in a realistic system is fixed, the excess number of signals from the transmit relays does not improve the system performance and consequently increases power consumption. In this paper, we adopt Double Opportunistic Transmit (DOT) cooperative diversity system which controls the average number of transmit relays. Although the average number of the transmit relays can be controlled by adjusting the two thresholds in DOT system, the instantaneous number of transmit relays is varying in fading channel. Thus we propose Maximal Ratio Combining (MRC) or Generalized Selection Combining (GSC) according to the number of the signals from relays at the destination. The outage probability of the proposed system is derived in closed form. The analytical results show that the system performance is improved with the number of the branches. Also it is noticed that when the number of the branches is fixed, the outage probability decreases with the increase of the average SNR of S-R path and R-D path.

Analysis of the Effects of Overexpression of Specific Phospholipid Binding Proteins on Cellular Morphological Changes in HEK293T Cells (특정 인지질 결합 단백질의 과발현이 HEK293 세포모양에 미치는 영향 분석)

  • The plasma membrane plays a crucial role in relaying signals from the outside environment to the inside of the cells. In eukaryotic cells, the inner leaflets of the plasma membrane are composed mostly of phospholipids, including phosphatidylethanolamine (PE), phosphatidylserine (PS), and phosphatidylinositides (PIs). In this study, we tried to analyze the morphological changes induced by EGFP-fused membrane binding proteins, which are targeted to the plasma membrane via specific phospholipids binding. As a result, we found that overexpression of EGFP-P4M-SidM, a specific PI4P binding protein, or EGFP alone, did not induce any morphological changes. On the other hand, overexpression of EGFP-PLCδ1(PH), which is a specific PI(4,5)P2 binding protein, EGFP-AKT1(PH) which binds to PI(3,4,5)P3, or EGFP-OSH2(PH)×2 which binds to PI4P and PI(4,5)P2, could induce the filopodia and lamilapodia formation as well as cell shrinkage. Overexpression of Lact-C2-EGFP which is a specific PS-binding probe, EGFP fused Aplysia phosphodiesterase 4 (ApPDE4) long-form (L(N20)-EGFP) which is localized to the plasma membrane via hydrophobic interaction, or EGFP fused Aplysia PDE4 short-form (S(N-UCR1-2)-EGFP) which is localized to the plasma membrane via electrostatic interaction, could induce cell shrinkage, but not filopodia or lamilapodia formation. Taken together, our data support that the different phospholipid bindings in the plasma membrane could induce different characteristic morphological changes. Thus, we can analyze, characterize, and classify the cellular morphological changes induced by the various phospholipid binding proteins.
