Inhibition of lipid and protein oxidation in raw ground pork by Terminalia arjuna fruit extract during refrigerated storage

  • Objective: Terminalia arjuna plant, specially its leaves, bark, and roots, are widely used in traditional herbal medicine due to presence of bioactive components and being a rich source of natural antioxidants. But its fruit has not been used for any such purposes despite its potential to retard oxidation. Hence, the antioxidant potential of Arjuna fruit extract (AFE) in retarding lipid and protein oxidation of raw ground pork was evaluated during refrigerated storage for 9 days. Methods: The AFEs were prepared using different solvents viz. ethanol (EH), water, ethanol: water (60:40) and methanol:hot water (60:40). The AFEs were analysed for total phenolic content (TPC), 2, 2-diphenyl-1-picrylhydrazyl radical scavenging activity and reducing power. Water extract (WE) and ethanol-water extract (EH-WE) were selected and incorporated at 1.0% into freshly minced pork meat and compared with a synthetic antioxidant, in retarding lipid and protein oxidation during storage. Results: The TPC in AFEs using different solvents ranged from 11.04 to 16.53 mg gallic acid equivalents/g and extracts exhibited appreciable scavenging activity ranging from 50.02% to 58.62%. Arjuna extracts significantly (p<0.05) improved the colour score of meat samples by reducing the formation of metmyoglobin during storage. Both the AFEs (WE and EH-WE) significantly (p<0.05) lowered the thiobarbituric acid reactive substances value, peroxide formation and formation of protein carbonyls in raw pork than control sample during storage. Upon sensory evaluation of all samples, it was found that AFE treatment could prolong the storage period of meat samples, without influencing the colour and odour score, up to 6 days. Conclusion: AFEs used at 1% improved the oxidative stability, colour and odour score and prolonged the refrigerated shelf life of ground pork up 6 days. Therefore, AFE could be explored as an alternative natural antioxidant in retarding lipid and protein oxidation in meat products.

  • Microbiological and sensory evaluations of chicken wings treated with acetic acid and trisodium phosphate(TSP) during retail(1${\pm}$2$^{\circ}C$) and refrigerated storage(4$^{\circ}C$). Chicken wings(average weight of 15${\pm}$2g per leg) were treated with 0.5∼2% aerobic plate counts(APC) on the surface of chicken wings for storage of 16 days at 4$^{\circ}C$. During storage of 12 days at retail store(1${\pm}$2$^{\circ}C$), the treating chicken wings with 7.5∼10% TSP solutions for 10 min showed significantly inhibitory effect in preventing the growth of aerobic microbes. For odor and appearance scores, chicken wings treated with 0.5∼2% acetic solutions resembled untreated wings for storage of 16 days at 4$^{\circ}C$. However, both odor and appearance scores of chicken wings treated with 2.5∼10% TSP solutions were similar to those of untreated wings for storage of 4 days, and a higher in the storage of beyond 8 to 12 days at retail store.

Effect of Marination with Black Currant Juice on the Formation of Biogenic Amines in Pork Belly during Refrigerated Storage

  • The effect of marination with black currant juice (BCJ) was investigated for their effects on meat quality and content of biogenic amines (BAs) [putrescine (PUT), cadaverine (CAD), histamine (HIM), tyramine (TYM), and spermidine (SPD)] in pork belly during storage at 9℃. BCJ was shown to have antibacterial activities against Escherichia coli and Pseudomonas aeruginosa. Additionally, the pH of pork belly marinated with BCJ (PBB) was significantly lower than that of raw pork belly (RPB) during storage. No significant difference in microorganisms between RPB and PBB was observed at day 0 of storage. However, at days 5 and 10 of storage, volatile basic nitrogen (VBN) was significantly decreased in PBB compared to RPB, and PBB also demonstrated significantly lower numbers of bacteria associated with spoilage (Enterobacteriaceae and Pseudomonas spp.) at these time-points. PBB was also associated with significantly reduced formation of BAs (PUT, CAD, TYM, and total BAs) compared to RPB at days 5 and 10 of storage. These results indicated that BCJ can be regarded as a natural additive for improving meat quality by preventing increased pH, VBN, bacterial spoilage, and inhibiting BAs formation during refrigerated storage.

  • 가열 조리된 냉장 돼지고기 패티에 사용된 상업용 유통 ${\gamma}$-oryzanol과 ${\alpha}$-tocopherol의 항산화 효과가 돼지고기 육질 내의 지질 산화로부터 기인되는 warmed-over flavor(WOF) 값과 콜레스테롤 산화로부터 기인되는 이차 산화물인 7-ketocholesterol의 생성 추이 변화로 비교 분석되었다. 지질산화로부터 측정된 파라미터는 이화학 데이터인 TBARs 값과 관능 데이터인 WOF 값에 의해 비교 분석되었으며, 콜레스테롤 산화로부터 유래된 측정 파라미터는 산화 유도기간에 따른 가열 조리된 돼지고기의 콜레스테롤 함량 변화 정도와 주요 콜레스테롤 산화물인 7-ketocholesterol 생성 정도에 의해 비교 분석 되었다. 콜레스테롤과 롤레스테롤 산화물은 cold saponification 추출 기법과 고속액체크로마토그래프 (HPLC) 테크닉을 이용하여 정량적으로 분석되었다. 아울러, 측정된 각 파라미터 변수들 간의 통계적 상관관계가 검토되었으며, 가열 조리된 돼지고기 패티의 산화로부터 기인되는 이차 산화물에 대한 ${\gamma}$-oryzanol과 ${\alpha}$-tocopherol의 항 산화력이 비교 분석되었다. 분석 결과 모든 파라미터 변수들은 서로 간에 유의적인 상관관계를 나타내었으며, 특히, ${\gamma}$-oryzanol처리 군에서 유의적인 지질 및 콜레스테롤 항 산화 효과를 보였다.

Study on Suitable Semen Additives Incorporation into the Extender Stored at Refrigerated Temperature

  • The objective of this study was to compare the effect of Butylated Hydroxy Toluene (BHT), Pentoxifylline (PTX) and ${\alpha}$-tocopherol (Vit E) on semen quality parameters of Karan Fries bulls. The fortification of extender by various semen additives improves motility as well as fertility of spermatozoa. Split samples of 24 ejaculates of four Karan Fries bulls were extended in extender with or without various additives such as BHT, PTX and Vit E, and performance was evaluated at an interval of 0, 24, 48 and 72 h at refrigerated temperature (4-$7^{\circ}C$). Results of the present study revealed that addition of BHT, PTX and Vit E in extender improved sperm cell function, such as motility, viability, HOST, and acrosome integrity, as compared to the control during liquid storage up to 48 h of preservation at refrigerated temperature. There was no significant (p<0.05) difference between any of the additives up to 48 h of preservation. Overall, the results showed a significant (p<0.05) deterioration in motility after each storage interval. The results showed a significant deterioration in the acrosome integrity and plasma membrane integrity up to 48 h; subsequently, there was not much degradation of both the semen quality parameters. There was a significant increase in spermatozoal tail and total abnormality after each storage interval at refrigerator temperature (4 to $7^{\circ}C$); however, the head and mid-piece abnormalities were almost unaffected. Tail and total abnormality were least in extender fortified with BHT, PTX and Vit E at different hours of incubation as compared to the control. The addition of 1.5 mM BHT, 3.6 mM PTX and 1 mg/ml Vit E in the semen extender has more beneficial effect in terms of semen quality and preservability of spermatozoa.

Effects of High Relative Humidity on Weight Loss, Color Change, and Microbial Activity of Tomatoes during Refrigerated Storage

  • The effects of high relative humidity (RH) on the physicochemical properties and microbial activity of mature green tomatoes ('Dombito') during refrigerated storage were determined at three temperatures (5, 10, and $15^{\circ}C$) and four different RH levels (91, 94, 97, and 99%). At each temperature, the weight loss rates of tomatoes at different levels of RH were significantly (p<0.05) different from each other. For the samples stored at $10^{\circ}C$, the weight losses were generally higher than those for the samples at $15^{\circ}C$ within the same RH level (i.e., greater vapor pressure deficit). The color change rates ('a' value) showed positive slopes, indicating that the tomato color was changing from green to red. Neither bacteria nor fungi caused visible damages to the samples, and the microbial counts were below 650 colony forming units/$cm^2$ during the test period.

High pressure processing for dark-firm-dry beef: effect on physical properties and oxidative deterioration during refrigerated storage

  • Objective: Study on the application of high pressure processing (HPP) for dark-firm-dry (DFD) beef was conducted to observe whether HPP has any impact on physical properties and to evaluate oxidative deterioration during refrigerated storage under vacuum. Methods: The longissimus lumborum muscles obtained from Friesian Holstein steers ($33{\pm}0.5$ months old) with 24-h postmortem pH higher than 6.0 were vacuum-packed and subjected to pressurization at 200, 400, and 600 MPa for 180 s at $15^{\circ}C{\pm}2^{\circ}C$; the samples were then stored for 9 days at $4^{\circ}C{\pm}1^{\circ}C$ and compared with control (0.1 MPa). Results: HPP increased meat pH by 0.1 to 0.2 units and the tenderness of cooked DFD beef significantly with no significant effects on meat texture profile. The stability of meat pH was well maintained during refrigerated storage under vacuum. No clear effects were found on the activity of catalase and superoxide dismutase, however, glutathione peroxidase activity was significantly reduced by high pressure. HPP and storage time resulted in aroma changes and the increasing amount of malondialdehyde and metmyoglobin relative composition. Conclusion: Although the increasing amount of malondialdehyde content, metmyoglobin formation and aroma changes in HPP-treated samples could not be avoided, HPP at 200 MPa increased $L^*$ and $a^*$ values with less discoloration and oxidative deterioration during storage.

Fluctuations in Phenolic Content and Antioxidant Capacity of Green Vegetable Juices during Refrigerated Storage

  • Shinseoncho and kale were made into green vegetable juices by building block [shinsenocho branch (SB), shinsenocho leaf (SL), kale branch (KB), and kale leaf (KL)]. Fluctuations in their phenolic contents and antioxidant capacities were analyzed during refrigerated storage at $4^{\circ}C$ for 28 days. Total polyphenolic contents of leaf parts showed a decreasing tendency after 4 days (SL) or 7 days (KL), whereas branch parts showed fluctuating values during the entire storage period. The 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging capacity was rapidly decreased in SB and in SL at 28 days (P<0.001), whereas KL showed a slightly increasing tendency after 14 days. For the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity, SL showed a sharp fall at 28 days (P<0.001), and KL showed a decreasing tendency after 14 days (P<0.001). SB showed a steady decrease during the entire storage period and KB indicated a nearly zero (0.97%) at 28 days. Pearson's coefficients for the correlation between antioxidant capacities measured by the ABTS and DPPH assays, and the total polyphenolic contents were determined. The results showed that the ABTS assay (r=0.934, P<0.001) was more strongly positively correlated with the total phenolic contents than the DPPH assay (r=0.630, P<0.001). In conclusion, when considering all building blocks, green vegetable juices, including kale and shinseoncho may have kept antioxidant capacities for up to 14 days under refrigeration, and the ABTS assay better reflects a positive correlation with the total phenolic contents when compared to the DPPH assay.

전자 코를 이용한 한우 등심육의 냉장저장 중 향기 패턴 분석 (Aroma Pattern Analysis of Hanwoo Beef (M. longissimus) using Electronic Nose during Refrigerated Storage)

  • 본 연구는 전자 코 시스템을 이용하여 센서의 감응도, 주성분 분석, 판별분석을 통해 한우육의 향기 패턴을 구명하고, 이를 pH 및 TBARS 값과 비교함으로써 쇠고기의 신속한 향기 평가방법의 가능성을 모색하기 위해 실시하였다. 시료는 도축 후 24시간이 경과한 쇠고기의 등심을 5$\pm$1$^{\circ}C$에서 7일간 저장하면서 실험에 사용하였다. 저장기간이 증가함에 따라 SY/G, SY/AA, SY/Gh, SY/gCTl, SY/gCT 센서의 감응도(dR/R0) 값은 감소하였으나, SY/LG, T30/1, P10/1, P10/2, P40/1, T70/2, PA2 센서의 dR/R0 값은 증가하였다. 저장 1일과 3일째에서는 dR/R0 값이 SY/AA#, T70/2 센서에서만 유의적인 차이를 보였으나(p<0.05), 저장 7일째에서 저장1일과 비교하였을 때 12개의 센서가 모두 유의적인 차이를 보였다(p<0.05). 감응도의 결과를 주성분 분석으로 mapping 하였을 때 저장 1일에 오른쪽 중간, 3일에 오른쪽 하단, 7일에 왼쪽에 길게 퍼져 군락이 형성되었다. 다시 판별분석으로 mapping을 하면 동일 저장기간내 군락이 집중되고, 저장기간별 군락군이 분산되어 뚜렷하게 구별되었다. 저장기간동안 육의 pH와 TBARS는 모두 어떤 항목과도 상관성을 보이지 않았으나, 모든 센서의 값이 각각 정 또는 역의 높은상관성을 가지고 있었다(p<0.001).

Effects of Annatto (Bixa orellana L.) Seeds Powder on Physicochemical Properties, Antioxidant and Antimicrobial Activities of Pork Patties during Refrigerated Storage

  • This study investigated the effect of the powder produced by ball-milling the outer layer of annatto (Bixa orellana L.) seeds on the physicochemical properties as well as the antioxidant and antimicrobial activities of pork patties over 14 d of refrigerated storage (4±1℃). Five pork patty treatments were produced containing three different concentrations of annatto seeds, 0.1, 0.25 and 0.5% (ANT0.1, ANT0.25, ANT0.5), 0.1% ascorbic acid (AA0.1), and a control (CTL). Based on the results, annatto seed powder appeared to show antioxidant activity. The Hunter color values of pork patties were affected by the addition of annatto seed powder, which increased the redness and yellowness values, but decreased the lightness of the patties (p<0.05). To evaluate the antioxidative effects of annatto on pork patties, thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) were analyzed over 14 d of refrigerated storage. Treatments containing annatto seed showed lower TBARS and POV than control (CTL) samples (p<0.05). The volatile basic nitrogen (VBN) of the pork patties containing annatto seeds were lower than that of CTL at the end of storage (p<0.05). Although no differences in total bacterial counts were observed between control and treated patties, those containing annatto seeds had lower microbial counts for Enterobacteriacease than CTL or AA 0.1%. Therefore, annatto seed powder might be a good source of natural antioxidants for the production of meat products.
