Production and properties of ale beer with Nuruk, a Korean fermentation starter (전통누룩을 이용한 ale맥주 제조 및 품질특성)

  • Nuruk, a traditional Korean alcoholic beverage starter, was evaluated as an additional saccharifying agent comprising up to 1.5% (w/w) of malt weight in ale-type beer processing. Sample characteristics were monitored during fermentation, ripening, and storage. Beer containing nuruk showed higher numbers of total viable bacteria and yeast cell counts. Additionally, ethanol (6.19-6.35%), color (Standard Reference Method), foam stability ($228.49-368.24{Sigma}$), saccharogenic power (307-417), and reducing sugar (3.83-5.25%) increased as the amount of nuruk was increased, while viscosity (3.13-2.07 cP) and bitterness unit (19.68-13.13) were lower than in samples without nuruk. Overall acceptance and aftertaste of the beer were significantly higher in a preference test. These results demonstrate that nuruk can be used to produce a new type of ale.

Quantitative Analysis of Ergosterol as a Biomarker of Mold-contaminated Foods Using the Enzyme Biosensor (효소 바이오센서를 이용한 식품의 곰팡이 오염 지표물질인 Ergosterol 정량분석)

  • Ergosterol is the significant component of the cell wall of fungi. Its presence is regarded as evidence of fungi contamination in grain and other foods. Many studies on ergosterol detection have been carried out using chemical methods, but those methods required complicated pre-treatments and long analysis times. In this study, an amperometric biosensor was developed for fast and precise ergosterol detection. The biosensor system used the electron transfer of hydrogen peroxide produced from the reaction of ergosterol with cholesterol oxidase. The biosensor system consisted of a peristaltic pump, a syringe loading sample injector, an enzyme reactor, a fabricated flow-through cell containing a working electrode, a reference electrode and a counter electrode, and a potentiostat/recorder. The working electrode was prepared by coating modified multi-wall carbon nanotube (MWNT) on glassy carbon electrode. The $MWNT-NH_2$ coated glassy carbon electrode linearly responded to hydrogen peroxide in the range of $1{\times}10^{-5}{\sim}8{\times}10^{-5}$ M with a detection limit of $10^{-7}$ M in the basic performance test. The currents produced from the ergosterol biosensor showed the linearity in a range from $1.0{\times}10^{-6}$ M to $1.0{\times}10^{-5}$ M ergosterol.

An Analysis of Success Factors in Internet Shopping Malls (인터넷 쇼핑몰구축의 성공요인에 대한 분석)

  • The main purpose of this research is to show Internet shopping mall the strategic way with the analysis of success factors in Internet shopping malls. To achieve the above purpose, the success factors and variables were defined by the survey of the reference. Sample malls for the research un selected of shopping malls registered in yahoo, Lycos, Empas and Hanmir regardless of their type and class, and did an online-survey of their operation. From the above method, the following results are deduced. First, there are five factors in the success of Internet shopping malls: the effectiveness of customer management the effectiveness of marketing. the competitiveness of product-sales, the convenience of use, the credibility of product. Second, the effectiveness of marketing is positively related to the number of member, visitors, and sales. Third, the credibility of product is negatively related to the number of member, visitors, and sales. At the end, the number of member and visitor are positively related to sales. This result could provide the managers with highly relevant managerial issues. The implication of the study are discussed and futher research directions are proposed.

Red Blood Cell Surface Antigens Responsible for Neonatal Isoerythrolysis in Thoroughbred Horses of Jeju Island (제주도 더러브렛 종에서 신생마 적혈구용혈증을 유발하는 적혈구 표면 항원 조사)

  • This study was conducted to survey red blood cell (RBC) antigens Aa, Ca and Qa types, which are considered to be the most significantly associated with occurrence of neonatal isoerythrolysis, and the results are expected to provide valuable informations in organization of breeding plan, hence preventing the disease. Blood samples were collected from 262 Thoroughbred horses in Jeju island. Two percent cell suspension has been prepared from each sample and they were tested by indirect antiglobulin test. Of the 226 mare's samples, 9(3.98%) were Aa negative, 8(3.54%) were Ca negative, 17(7.52%) were Qa negative. Of the 36 Stallion's samples, 1(2.78%) was Aa negative, 3(8.33%) were Ca negative, 3(8.33%) were Qa negative. On the basis of these data, a database for breeding compatability could be set, and it would play an important role as a reference for arranging the mating partners.

Prediction of Chemical Organic Composition of Manure by Near Infrared Reflectance Spectroscopy

  • The organic materials included in excreta of livestock are important resources for organic manure and for improving soil quality, although there is still far from effective using. One reason for this is still unclearly standard of quality for evaluation of manure made from excreta of livestock. Therefore, the objective of this study is to develop rapid and accurate analytical method for analyzing organic compositions of manure made from excreta of livestock, and to establish quality evaluation method based on the compositions predicted by near infrared reflectance spectroscopy (NIRS). Sixteen samples of manure, each eight samples prepared from two treatments, were used in this study. The manure samples were prepared by mixing 560 kg feces of swine,60 kg sawdust with moisture content was adjusted to be 65%. The mixture was then keep under two kinds of shelter, black and clear sheets, as a treatment on the effect of sunlight. Samples were taken in every week (form week-0 to 7) during the process of manure making. Samples were analyzed to determine neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) by detergent methods, and organic cell wall (OCW) and fibrous content of low digestibility in OCW (Ob) by enzymatic methods. Biological oxygen demand (BOD) was analyzed by coulometric respirometer method. These compositions were carbohydrateds and lignin that were hardly digested. Spectra of samples were scanned by NIR instrument model 6500 (Pacific Scientific) and read over the range of wavelength between 400 and 2500nm. Calibration equations were developed using eight manure samples collected from black sheet shelter, while prediction was conducted to the other eight samples from clear sheet shelter. Accuracy of NTRS prediction was evaluated by correlation coefficients (r), standard error of prediction (SEP) and ration of standard deviation of reference data in prediction sample set to SEP (RPD). The r, SEP and RPD value of forage were 0.99, 0.69 and 7.6 for ADL, 0.96, 1.03 and 4.1 for NDF, 0.98, 0.60 and 4.9 for ADF, 0.92, 1.24 and 2.6 for Ob, and 0.91, 1.02 and 7.3 for BOD, respectively. The results indicated that NIRS could be used to measure the organic composition of forage used in manure samples.

The importance of NIR spectroscopy in the estimation of nutritional quality of grains for ruminants

  • The production of grain for export and domestic use is one of Australia's most important agricultural industries, and the NIR technique has been used extensively over many years for the routine monitoring of grain quality, particularly moisture and protein content. Because most Australian grain is intended for human food production, the determinants of grain quality for livestock feed, apart from protein, have been largely ignored. However the increasing use of grain for feeding to pigs, poultry, beef cattle and dairy cows has led to an important national research project entitled “Premium Grains for Livestock”. Two of the objectives of this project are to determine the compositional and functional characteristics of grains which influence their nutritional quality for the various classes of livestock, and to adopt rapid and objective analytical tests for these quality criteria. NIR has been used in this project firstly to identify a set of grain samples from a large population of breeders' lines which showed a wide spectral variation, and hence a potentially wide variation in nutritional value. The selected samples were not only subjected to an extensive array of chemical, physical and in vitro analyses, but also were grown out to produce sufficient quantities of grain to feed to animals in vivo studies. Additional grains were also strategically selected from farms in order to include the effect of weather damage, such as rain, drought and frost. In this study to date, NIR calibrations have been derived or attempted, on both ground and whole grains, for in vivo dry matter digestibility (DMD), pepsin-cellulase dry matter disappearance, protein, fat, acid detergent fibre, neutral detergent fibre, starch, in sacco DMD and in vitro assays to simulate starch digestion in the lumen and small intestine. Results so far indicate high calibration accuracy for chemical components (SECV 0.3 to 2.6%) and very promising statistics for in vivo DMD (SECV 1.8, $R^2$ 0.93, SD 7.0, range 61.9 to 92.3, n=60). There appears to be some potential for NIR to estimate some in vitro properties, depending upon the accuracy of reference methods and appropriate sample populations. Current work is in progress to extend the range of grains with in vivo DMD values (a very laborious and expensive process) and to increase the robustness of the various NIR calibrations, with the aim of implementing uniform testing procedures for nutritional value of grains throughout Australia.

Diversity of vir Genes in Plasmodium vivax from Endemic Regions in the Republic of Korea: an Initial Evaluation

  • Variant surface antigens (VSAs) encoded by pir families are considered to be the key proteins used by many Plasmodium spp. to escape the host immune system by antigenic variation. This attribute of VSAs is a critical issue in the development of a novel vaccine. In this regard, a population genetic study of vir genes from Plasmodium vivax was performed in the Republic of Korea (ROK). Eighty-five venous blood samples and 4 of the vir genes, namely vir 27, vir 21, vir 12, and vir 4, were selected for study. The number of segregating sites (S), number of haplotypes (H), haplotype diversity (Hd), DNA diversity (${\pi}$ and ${\Theta}_w$), and Tajima's D test value were conducted. Phylogenetic trees of each gene were constructed. The vir 21 (S=143, H=22, Hd=0.827) was the most genetically diverse gene, and the vir 4 (S=6, H=4, Hd=0.556) was the opposite one. Tajima's D values for vir 27 (1.08530, P>0.1), vir 12 (2.89007, P<0.01), and vir 21 (0.40782, P>0.1) were positive, and that of vir 4 (-1.32162, P>0.1) was negative. All phylogenetic trees showed 2 clades with no particular branching according to the geographical differences and cluster. This study is the first survey on the vir genes in ROK, providing information on the genetic level. The sample sequences from vir 4 showed a clear difference to the Sal-1 reference gene sequence, whereas they were very similar to those from Indian isolates.

Elementary Science-gifted Teachers' Views and Attitudes Toward Teaching on Nature of Science (초등 과학영재 지도교사의 과학의 본성에 대한 인식 및 교수태도 분석)

  • This study aimed to investigate a perception of teachers engaged in special education for scientifically gifted regarding nature of science and identify attitudes toward teaching nature of science. The sample of this study consists of 122 science teachers who are teaching or taught students attending gifted classes of any primary school in Korea and any gifted education center of every District Office of Education. We made a partial amendment of a VOSE that was developed by Chen(2006) and then used. We tested their perception and attitudes in regard to nature of science. In terms of nature of science, we devide it into 7 sub-areas to analyse. For attitudes toward teaching nature of science, we investigate and analyse following 5 sub-areas; tentativeness of scientific knowledge, nature of observation, scientific methods, theories and laws and subjectivity and objectivity. The result showed that the generally teachers have a desirable recognitions about a nature of science. For attitudes toward teaching nature of science, the teachers showed that they have positive attitudes. However between degrees of teachers' recognition about a nature of science and attitudes toward teaching nature of science showed a low correlation. To increase their understanding of nature of science and develop attitudes toward teaching nature of science, there should be more training time for the teachers and training contents also should be changed. In addition, we hope that this study contribute to develop contents and direction of training for the teachers as a basic reference.

Personalized Dietary SikdanOme Recommendation based on Obesity Related SNP Genotype and Phenotype (비만 관련 SNP genotype-phenotype 정보기반의 맞춤 식단옴 추천)

  • Obesity extends the global economic burden and it causes that the failure of a reduction of physical activity, and diet management. In this work, nutritional information and personalized diet based on calorie supply system and is discriminatory utilized the obesity-related SNP information in order to recommend a personalized functional foods. This study performed a GWAS analysis for the excavation of a Korean-specific and obesity-related SNP, which utilizes genetic information were recommended by entering a personalized diet in accordance with the SNP genotype-phenotype information. In addition, we integrated Database with relation of nutrient for utilizing the USDA Food information and it was applied to recommend with Sickdanome. As a result, the obesity-related SNP information was confirmed in the sample which has the normal value BMI. In this study, we have recognized that the phenotype information related obesity, BMI is inconsistent with the SNP genotype information. This result is shown that it is necessary to provide the personalized dietary SickdanOme recommendation based on the both pheotype-genotype information.

Efficiency Calibration of HPGe Detector in Normal ana Coincidence Mode for the Determination of Prompt Gamma-ray (즉발감마선 측정을 위한 HPGe 검출기의 전계수 또는 동시계수모드에서의 광대역 계측효율 보정)

  • Neutron induced prompt gamma-ray spectroscopy(NIPS) system measures the prompt gamma-ray emitting by the interaction of a neutron with various materials. This system will be of great benefit to scientists worldwide, since it provides the non-destructive measurement of many element in either solid or liquid wastes. In this study, the full-energy-peak (FEP) efficiency calibration for a HPGe detector was constructed in the ${\gamma}$-ray energy range from 80 keV to 8 MeV, using $^{l33}$Ba and >TEX>$^{152}Eu$ RI sources and $ ^{35}Cl(n, ${\gamma}$)^{36}Cl$ thermal neutron captured reaction. The FEP efficiency curve for the higher energies using the $^{35}Cl(n, ${\gamma}$)^{36}Cl$ reaction was normalized with the curve obtained from the RI sources, since the accurate activity of its prompt ${\gamma}$-ray is unknown. The average thermal neutron flux was theoretically calculated using the FEP efficiency curve for the KCl standard solutions. The NIPS system equipped with a ${\gamma}$-${\gamma}$ coincidence setup with two n-type coaxial HPGe detectors was considered in order to reduce the interfering ${\gamma}$-ray background. The FEP efficiency curve for the ${\gamma}$-${\gamma}$ coincidence system was also obtained for full energy range. The performance of the normal and coincidence NIPS system was tested by comparing signal-to-noise ratio in each mode using the reference sample.e.
