코덱스 영양소 기준치 설정시 영양섭취기준의 적용 방안 (Application of Dietary Reference Intakes for Codex Nutrient Reference Values)

  • 현재 코덱스 NRVs 중에는 새로이 각국에서 제정된 영양 섭취기준과 상당히 차이가 있는 영양소들이 있어서 개정을 위한 검토 작업이 시급히 필요하다. DRIs 중 EARs 혹은 RDAs를 코덱스 NRVs 기준으로 설정하는 것에는 각각이 장점과 단점이 있으므로, 각 기준에 대한 장점과 문제점을 충분히 고려하여 선정하여야 한다. 각 나라의 DRIs를 NRVs 기준으로 이용하거나 국제적으로 영양소별로 단일 값을 정한 후 각 나라의 인구 구조를 근거로 NRVs 기준을 설정하는 것은 국가마다 서로 다른 기준을 설정하는 결과를 초래하고 이로 인한 소비자의 혼란 및 국가 간 교역에 혼란을 초래할 가능성이 크므로, 각 나라의 인구 구조를 적용한 값을 NRVs기준으로 설정하는 것은 문제가 있다고 사료된다. 코덱스 NRVs 설정시 인구 구조에 근거한 RDA를 사용할 경우 연령 집단 간 편차가 크면 산출된 값이 lowest UL 보다 높을 수 있으므로, 성인 이외의 연령층 (어린이)에 대해서 바람직한 적용 방안을 후속 연구를 통해 별도로 고려해 볼 필요가 있다. 따라서 기존의 Codex 기준과 같이 국제적으로 새로이 설정된 DRIs를 전면적으로 검토하고 이를 최대한 반영하여 영양소별로 단일 값을 제시하여 각 나라에서 이용하도록 하는 것이 바람직하다고 사료된다.

모집단 분포를 이용한 핵의학 혈액검사의 통계적 품질관리의 유용성 (Availability of Statistical Quality Control of Nuclear Medicine Blood Test Using Population Distribution)

  • 정도관리(Quality Control)는 오차를 최소한으로 하여 검사의 신뢰도를 증진 시키는 것을 목적으로 하고 있으며 이러한 정도관리 활동을 통해 환자의 진단과 치료방침에 영향을 주는 핵의학 혈액 검사의 신뢰도가 확보되고 있다. 이에 연구자들은 신뢰도 향상의 추가적인 기법으로 건강검진 수검자를 대상으로 시행한 핵의학 혈액검사의 모집단 분포를 이용한 통계적 품질관리의 유용성을 평가해 보고자 한다. 2014년 1월부터 2014년 12월까지 서울아산병원 건강증진센터를 이용한 건강검진 수검자 41,341명을 대상으로 핵의학 혈액 검사 12종목에 대해 통계 분석을 시행 하였다. 통계분석은 핵의학 혈액 검사 12종목에 대한 결과 보고 치를 참고치 이내, 참고치 이상, 참고치 이하의 세 가지 그룹의 월간 백분율로 구분하여 모집단 분포의 평균값과 표준편차, 그리고 표준편차 지수(Standard Deviation Index: SDI)를 산출 하였고 그 결과 모집단 분포의 표준편차는 모든 그룹에서 대부분 ${\pm}2SD$ 이내의 결과를 보였다. 건강검진 수검자에 대한 핵의학 혈액검사 12종목(AFP, CEA, CA19-9, CA125, PSA, TSH, FT4, Anti-Tg-Ab, Anti-TPO-Ab, Calcitonin, 25-OH-VitD3, Insulin)의 모집단 분포를 분석한 결과 월간 백분율의 표준편차 지수(SDI)가 ${\pm}2.0$ 이상인 경우는, 1월과 3월 Insulin검사에서 각각 2.3, 2.4, 5월 Anti-Tg-Ab검사에서 2.2, 그리고 9월 CA19-9에서 2.2의 결과를 보였다. 이는 검사 Kit 시약의 이상 (동위원소 시약의 최대 결합률 저하), 검사반응 시간의 저하가 원인 이었다. 연구대상이 되는 전체의 속성을 포함하고 모집단 분포의 특성을 이용한 통계적 품질관리는 검사실에서 시행하고 있는 내부정도관리 프로그램을 보완하는 역할을 할 수 있으리라 생각된다.

Evaluation of clinical usefulness of reference interval of some selected hematological parameters in canine blood

  • To estimate the source of variance components for some hematological parameters and assess the utility of the conventional population-based reference interval, this study computed index of individuality for blood samples, which were from 13 dogs drawn once weekly for 4 consecutive weeks. Results were subjected to nested analysis of variance. For all parameters measured between-dog variations were greater than within-dog variation. Except for the parameters RBC and MCHC the index of individuality was <1.4. The low reliability coefficient and high index of individuality of ${\leq}0.8$ were found for the majority of hematological parameters. In practical term, the present study indicated that use of hemogram profiles alone in the evaluating clinical state of a single patient should be avoided because of their physiological or natural random variations, and that comparing a single measurement on the blood analytes from an individual dog to the conventional population-based reference range may be too insensitive to detect any significant changes in the blood components of that particular dog. A single measurement may not characterize an individual's average concentration of the parameters even shortterm period.

Optimal population size to detect quantitative trait loci in Korean native chicken: a simulation study

  • Objective: A genomic region associated with a particular phenotype is called quantitative trait loci (QTL). To detect the optimal F2 population size associated with QTLs in native chicken, we performed a simulation study on F2 population derived from crosses between two different breeds. Methods: A total of 15 males and 150 females were randomly selected from the last generation of each F1 population which was composed of different breed to create two different F2 populations. The progenies produced from these selected individuals were simulated for six more generations. Their marker genotypes were simulated with a density of 50K at three different heritability levels for the traits such as 0.1, 0.3, and 0.5. Our study compared 100, 500, 1,000 reference population (RP) groups to each other with three different heritability levels. And a total of 35 QTLs were used, and their locations were randomly created. Results: With a RP size of 100, no QTL was detected to satisfy Bonferroni value at three different heritability levels. In a RP size of 500, two QTLs were detected when the heritability was 0.5. With a RP size of 1,000, 0.1 heritability was detected only one QTL, and 0.5 heritability detected five QTLs. To sum up, RP size and heritability play a key role in detecting QTLs in a QTL study. The larger RP size and greater heritability value, the higher the probability of detection of QTLs. Conclusion: Our study suggests that the use of a large RP and heritability can improve QTL detection in an F2 chicken population.

Genetic Similarity and Variation in the Cultivated and Wild Carassius carassius Estimated with Random Amplified Polymorphic DNAs

  • RAPD analysis based on numerous markers have been used to investigate patterns of genetic differentiation ann and within two cultured and wild populations represented by the species crucian carp(Carassius carassius). From RAPD analysis using five primers, a total of 442 polymorphic bands were obtained in two populations and 273 were found to be specific to a wild population. According to RAPD-based estimates, average number of polymorphic bands in wild population was approximately 1.5 times as diverse as that in cultured. The average level of bandsharing values was $0.40 \pm 0.05$ in wild population, but was $0.69 \pm 0.08$ in cultured population, With reference to bandsharing values and banding patterns, wild population was considerably more diverse than cultured population. Knowledge of the genetic diversity of crucian carp should help in formulating more effective strategies for managing this aquacultural fish species.

코덱스 영양소기준치 설정을 위한 인구집단 및 영양소의 종류 선정 (The Scope of Population Groups and Nutrients for Codex Nutrient Reference Values)

  • The purposes of this study were to establish the scope of population groups to be set and nutrients to be included for Codex Nutrient Reference Values (NRVs). Examination of a variety of documents and reports on the principles of scientific evidence was gone through to choose representative groups from a variety of population groups and kinds of nutrients for setting Codex NRVs. Also, hearings from the groups of NRVs expertise has been formed and was gone through by e-working process. The differences of food and foods being consumed in general and by specific population groups and limited actual space on packages for food labeling were considered. This paper suggest the options on the scope of population groups to be set and nutrients to be included for Codex NRVs on the basis of the above procedures. Also, the advantages and disadvantages of these options are commented when the options are applied for establishment and revision of the Codex NRVs.

Genetic evaluation and accuracy analysis of commercial Hanwoo population using genomic data

  • This study has evaluated the genomic estimated breeding value (GEBV) of the commercial Hanwoo population using the genomic best linear unbiased prediction (GBLUP) method and genomic information. Furthermore, it analyzed the accuracy and realized accuracy of the GEBV. 1,740 heads of the Hanwoo population which were analyzed using a single nucleotide polymorphism (SNP) Chip has selected as the test population. For carcass weight (CWT), eye muscle area (EMA), back fat thickness (BFT), and marbling score (MS), the mean GEBVs estimated using the GBLUP method were 3.819, 0.740, -0.248, and 0.041, respectively and the accuracy of each trait was 0.743, 0.728, 0.737, and 0.765, respectively. The accuracy of the breeding value was affected by heritability. The accuracy was estimated to be low in EMA with low heritability and high in MS with high heritability. Realized accuracy values of 0.522, 0.404, 0.444, and 0.539 for CWT, EMA, BFT, and MS, respectively, showing the same pattern as the accuracy value. The results of this study suggest that the breeding value of each individual can be estimated with higher accuracy by estimating the GEBV using the genomic information of 18,499 reference populations. If this method is used and applied to individual selection in a commercial Hanwoo population, more precise and economical individual selection is possible. In addition, continuous verification of the GBLUP model and establishment of a reference population suitable for commercial Hanwoo populations in Korea will enable a more accurate evaluation of individuals.

재래흑염소와 교잡종 염소의 Monomorphic SNP 분석을 통한 유전적 다양성과 집단구조의 비교 및 검증 (Comparison and Validation of Genetic Diversity and Population Structure Using Monomorphic SNP Data of the Korean Native Black Goat and Crossbred Goat)

  • 본 연구는 우리나라 고유의 재래흑염소 집단인 당진, 장수, 통영 및 경상대 계통과 교잡종 염소 계통 또는 해외품종의 개체 식별을 위한 유전적 다양성과 관계 조사 및 검증을 위해 수행하였다. 각 염소 집단에 존재하는 Monomorphic SNP를 수집한 이후 공통적으로 존재하는 SNP 133개를 선발하여 분석에 이용하였다. Monomorphic SNP 133개를 통한 재래흑염소와 교잡종 염소의 유전적 구조 차이를 나타냈으며, 주성분 분석 결과 재래흑염소와 교잡종 염소가 명확히 구분되는 것으로 나타났다. 또한, 참조집단 이외의 70두(Native Korean goat = 24, Cross breed = 46)로 구성된 검증집단을 분석한 결과 국내 재래흑염소 계통의 참조집단과 동일한 유전적 구조를 나타냈으며, 교잡종 염소의 경우 참조집단의 일부 유전적 구성을 공유하는 것으로 나타났다. 국내 재래흑염소의 경우는 하나의 군집을 형성한 반면 해외 품종 및 교잡종 계통의 경우 재래흑염소 계통에 비해 넓게 퍼져 군집을 형성하는 것으로 나타났다. 따라서, 본 연구 결과는 국내 재래흑염소 유전자원 집단을 보존을 위한 기초자료로 활용하고 추후 유전적 다양성을 고려한 염소의 개량을 위한 기초자료로 유용하게 활용 할 수 있을 것으로 판단된다.

Constructing Reference Transcriptome Sets of Codonopsis lanceolate(Deodeok) and Ixeridium dentatum

  • As the aging population increases and interest in well-being increases, the importance of developing special crops increases. Natural medicine based on the special crops has been mainly used as an adjunct therapy for many diseases and symptoms based on culture, traditional medicine, and experience. In particular, it is attracting attention as a new resource to develop new drugs such as Artemisinin, a treatment for malaria. In order to efficiently use crops, it is essential to establish omics data such as genomes, transcriptomes, and metabolites of special-purpose crops. However, many special-purpose crops have large, heterogeneous and polyploid genomes that require high cost and long time to reference genome sequencing. Therefore, we built an inexpensive, fast, but very usefill reference transcriptome as the first step. We constructed high-quality reference transcriptom sets of Codonopsis lanceolata and Ixeridium dentatum with PacBio data. Our team will continue to construct reference transcriptoms of more special-purpose crops, and the data will be released by the National Agricultural Biotechnology Information Center (NABIC) in order to be widely used in agricultural as well as medical R&D.

Evaluation of the Serving Sizes of Packaged Processed Food in Korea

  • This study was carried out to evaluate the adequacy of serving sizes of those packaged processed foods in Korea which are designed to provids one meal or snack for one person. There is a lack of data on reference serving sizes which are essential components in the nutrition information provided on food labels. After compiling the average weight of packaged processed foods available in the market in Korea, a questionnaire was formulated to evaluate the adequacy of the serving sizes on 56 packaged processed food items of 188products using 25 female university students as a consumer group. The results showed that 65.5% of the packaged processed foods had adequate serving sizes. In general, current packaged sizes for bread and carbonated drinks were found to be too large. The proposed reference serving sizes in this research appear to be somewhat different from the ones suggested by the Korean Nutrition Society s $7^{th}$ Recommended Dietary Allowances for Koreans and by the Food and Drug Administration (USFDA) in the U.S. The serving sizes suggested by the USFDA appear to be too large for beverages and too small for snack foods when these are applied to the Korean population. It is suggested that the size of beverages in the Korean market should be reduced, and smaller sub-packets of snacks (each for one serving) should be packaged in a larger pack, for ease of use of nutrition information by consumers as well as for the reduction of food waste. In the future, other representative population groups should be included in the determination of reference serving sizes.
