Identification of Novel Universal Housekeeping Genes by Statistical Analysis of Microarray Data

  • Housekeeping genes are widely used as internal controls in a variety of study types, including real time RT-PCR, microarrays, Northern analysis and RNase protection assays. However, even commonly used housekeeping genes may vary in stability depending on the cell type or disease being studied. Thus, it is necessary to identify additional housekeeping-type genes that show sample-independent stability. Here, we used statistical analysis to examine a large human microarray database, seeking genes that were stably expressed in various tissues, disease states and cell lines. We further selected genes that were expressed at different levels, because reference and target genes should be present in similar copy numbers to achieve reliable quantitative results. Real time RT-PCR amplification of three newly identified reference genes, CGI-119, CTBP1 and GOLGAl, alongside three well-known housekeeping genes, B2M, GAPD, and TUBB, confirmed that the newly identified genes were more stably expressed in individual samples with similar ranges. These results collectively suggest that statistical analysis of microarray data can be used to identify new candidate housekeeping genes showing consistent expression across tissues and diseases. Our analysis identified three novel candidate housekeeping genes (CGI-119, GOLGA1, and CTBP1) that could prove useful for normalization across a variety of RNA-based techniques.

Present Situation and Analysis of Cold Storage Facilities for Fisheries Products (수산물 냉동냉장설비의 현황 및 분석)

  • This paper describes the analysis on freeze and refrigeration capacity of cold storage facilities for fisheries products during the last 15 years. And based on the analysis results, we prospect the number and scale of cold storage facilities in the near future. The main analysis results are summarized by the followings; Under the present conditions, the number and scale of our facilities is 879 and 3,586,824 ton, respectively. In comparison of the present and last 15 years ago, the business number increases about 22%, the accommodation capacity increases approximately 80%. In the reported update reference with the related data during the last 15 years, the capacity of cold storage facilities in 2011 expects to reach up to 4,600,000 ton. The newlyestablished facility in 2011 has the function of a multi-purpose distribution center as well as the existing cold storage, but this facilities converges in the capital area. Therefore, in this paper, we provided the basic reference data for the present situation and prediction of our cold storage facilities. But it is necessary to investigate and analyse widely our cold storage facilities for fisheries products in the near future.

Genetic characterization and phylogenetic analysis of Clostridium chauvoei isolated from Hanwoo in Jeonbuk (전북지역 한우에서 분리한 기종저 균의 유전학적 특성 규명)

  • Clostridium chauvoei is the etiologic agent of blackleg, a high mortality rated disease infection mainly cattle. In the present study, the partial sequences of 16S rRNA and flagellin gene of C. chauvoei isolated in Jeonbuk, Korea were determined and compared with those of reference strain. Oligonucleotide primers were designed to amplify a 811 bp fragment of 16S rRNA gene and 1229 bp fragment of flagellin gene. Sequencing analysis of 16S rRNA gene showed high homology to the reference strains ranging 82.3% to 100%, while flagellin gene were different from published foreign clostridia, showing 98.7% to 72.0% nucleotide sequence homology. Phylogenetic analysis based on 16S rRNA gene revealed the close phylogenetic relationship of C. chauvoei and C. septicum in cluster I, which includes C. carnis, C. tertium, C. quinii, C. celatum, C. perfringens, C. absonum, C. botulinum B. Phylogentic analysis also revealed that flagellin gene formed a single cluster with C. chauvoei, C. septicum, C. novyi A, C. novyi B, C. tyrobutylicum, C. acetobutylicum. The genetic informations obtained from this study could be useful for the molecular study of C. chauvoei.

  • The fatigue analysis for cutout panel used for the weight reduction of aircraft has been conventionally performed through the open hole concept using the reference stress and stress concentration factor (Kt). However, in the actual structure cases, the goal of weight reduction might be less meaningful due to the conservative approach induced by the difficulties of extracting the confident reference stress from FE-Analysis in the complicated loading behavior. Therefore a new approach is proposed in order to secure the effectiveness of weight reduction and validate the confidence of the analysis results using the interaction of max-min principal stress at the critical location of open hole edge line.

  • The purpose of this paper is design and implement the invisible depth analysis tools. The developed algorithm was basically used reference plan method and to remove first step errors we mix-used point-to-point method. and we consider error due to curvature and refraction for large scale analysis. The final algorithm was developed as ArcToolBox tools, which can be considered convenient and public use as well; as result it reduced experimental errors as compared with conventional method and makes possible high resolution analysis for large scale site.

OPTIMIZATION OF THE PARAMETERS OF FEEDWATER CONTROL SYSTEM FOR OPR1000 NUCLEAR POWER PLANTS

  • In this study, the parameters of the feedwater control system (FWCS) of the OPR1000 type nuclear power plant (NPP) are optimized by response surface methodology (RSM) in order to acquire better level control performance from the FWCS. The objective of the optimization is to minimize the steam generator (SG) water level deviation from the reference level during transients. The objective functions for this optimization are relationships between the SG level deviation and the parameters of the FWCS. However, in this case of FWCS parameter optimization, the objective functions are not available in the form of analytic equations and the responses (the SG level at plant transients) to inputs (FWCS parameters) can be evaluated by computer simulations only. Classical optimization methods cannot be used because the objective function value cannot be calculated directely. Therefore, the simulation optimization methodology is used and the RSM is adopted as the simulation optimization algorithm. Objective functions are evaluated with several typical transients in NPPs using a system simulation computer code that has been utilized for the system performance analysis of actual NPPs. The results show that the optimized parameters have better SG level control performance. The degree of the SG level deviation from the reference level during transients is minimized and consequently the control performance of the FWCS is remarkably improved.

Precise dynamic finite element elastic-plastic seismic analysis considering welds for nuclear power plants

  • This study performed a precise dynamic finite element time history elastic-plastic seismic analysis considering the welds, which have been not considered in design stage, on the nuclear components subjected to severe seismic loadings such as beyond-design basis earthquakes for sustainable nuclear power plants. First, the dynamic finite element elastic-plastic seismic analysis was performed for a general design practice that does not take into account the welds of the pressurizer surge line system, one of safety class I components in nuclear power plants, and then the reference values for the accumulated equivalent plastic strain, equivalent plastic strain, and von Mises effective stress were set. Second, the dynamic finite element elastic-plastic seismic analyses were performed for the case of considering only the mechanical strength over-mismatch of the welds as well as for the case of considering both the strength over-mismatch and welding residual strain. Third, the effects of the strength over-mismatch and welding residual strain were analyzed by comparing the finite element analysis results with the reference values. As a result of the comparison, it was found that not considering the strength over-mismatch may lead to conservative assessment results, whereas not considering the welding residual strain may be non-conservative.

Accuracy Analysis of FKP for Public Surveying and Cadastral Resurvey (공공측량 및 지적재조사 사업 적용을 위한 FKP 정밀도 분석)

  • NGII (National Geographic Information Institute) has been providing VRS (Virtual Reference Station) service so that could determine precise positioning in real time since 2007. However, since the VRS service has to maintain the connected status with VRS server, the number of users who can use VRS service are limited by capacity of VRS server. To solve this problem, NGII has been providing FKP (Virtual Reference Station) service using one way telecommunication from November 1, 2012. Therefore, it is predicted that the usage of FKP service will increase in public surveying and cadastral resurveying in the future. However, the studies with respect to analysis of FKP precision for applying to public surveying and cadastral resurveying is not conducted enough. In this study, to analyse the application possibility of FKP on the public surveying and cadastral resurveying, the two kind analysis were performed. First is the analysis of accuracy according to the configuration of reference station of FKP and VRS. One is consisted of same reference stations, another is consisted of different reference stations. Second is the accuracy anlalysis of horizontal and vertical positioning acquiring VRS and FKP data in various measurement environment based on VRS regulation. Result of first study, Positioning accuracy according to the configuration of the reference stations satisfies related regulation. However, accuracy of FKP in case of different reference stations is worse than in case of same reference stations.. The result of second test shows that the horizontal precision of FKP and VRS in good measurement environment satisfy the allowed precision. However, in some case, horizontal precision of FKP and VRS in poor measurement environment exceed the allowed precision. In addition, the number of exceeding the allowed precision in the FKP is more than the VRS. The vertical precision of the VRS satisfy related work provision. In conclusion, the result of this study shows that the FKP only in open area should be used for public survey and cadastral resurvey. Therefore the additional studies with respect to the improvement of FKP precision should be conducted.

Parameter Regionalization of Hargreaves Equation Based on Climatological Characteristics in Korea (우리나라 기후특성을 고려한 Hargreaves 공식의 매개변수 지역화)

  • The quantitative analysis of evapotranspiration (ET) is a key component in hydrological studies and the establishment of water resources planning. Generally, the quantitative analysis of ET is performed by the estimation method of potential or reference ET based on meteorological factors such as air temperature, wind speed, etc. Hargreaves equation is one of empirical methods for reference ET using air temperature data. In this study, in order to estimate more exact reference ET considering climatological characteristics in Korea, parameter regionalization of Hargreaves equation is carried out. Firstly, modified Hargreaves equation is presented after the analysis of the relationship between solar radiation and temperature. Secondly, parameter ($K_{ET}$) optimization of Hargreaves equation is performed using Penman-Monteith method and modified equation at 71 weather stations. Lastly, the equation for calculating $K_{ET}$ using temperature data is proposed and verified. As a result, reference ET from original Hargreaves equation is overestimated or underestimated compared with Penman-Monteith method. But modified equation in this study is more accurate in the climatic conditions of Korea. In addition, the applicability of the equation between $K_{ET}$ and temperature is confirmed.

A Study on the Highest Exposure Temperatures of Exposed Reinforced Concrete Structures at Fire (화재에 노출된 철근콘크리트 구조물의 최고노출온도 추정을 위한 연구)

  • In this paper, Machinery analysis was conducted, in order to predict highest exposure temperatures and the analyze fire damage in the case of fire on reinforced concrete structure. After analyzing differential thermal of reference materials in accordance with temperature of concrete reference core specimen, it turned out that powerful endothermic peak came resulting from evaporation of capillary water and get water untill $200^{\circ}C$, another endothermic peak came resulting from decomposition of calcium hydroxide at $520^{\circ}C$, and then mass of reference materials remarkably decreased due to endothermic reaction. Another powerful endothermic reaction came after decomposition of calcite at $720^{\circ}C$. After analyzing X-ray diffraction of reference materials in accordance with temperature of concrete reference core specimen, it turned out that calcium hydroxide existed until the temperature of $400^{\circ}C$, but CH almost disappeared and CaO appeared from $600^{\circ}C$. The production increased in proportion to the temperature. This is because that calcium hydroxide and calcite are decomposed and CaO is produced when the temperature of concrete increases with fire. It is estimated that calcium hydroxide and calcite are utterly decomposed and peak disappears, and peak of CaO is remarkably formed instead, at the temperature of $700-800^{\circ}C$.
