Objectives : The Lycoris squamigera Maxim has been used traditionally for treatment of various diseases. However, the studies on the effect of Lycoris squamigera Maxim have not been carried out. In the present study, extract of Lycoris squamigera Maxim were tested for their anti-inflammatory and anti-oxidation effect. Methods : The anti-inflammatory effect of the various solvent extract was studied in lipopolysaccharide (lps)-treated mouse macrophage cells. RAW 264.7 cells were pre-incubated with Lycoris squamigera Maxim extracts for 4h and treated with $1\;{\mu}g/m{\ell}$ lps for 18h, and then the anti-inflammatory effects of extracts were determined. The anti-oxidation effect of extracts measured by DPPH method, reductive potential test, total phenolics test. Results : Extracted root's ethyl acetate layer showed a significant decrease in nitric oxide. And that layer (root's ethyl acetate extract) was showed decrease in TNF-${\alpha}$ concentration dependently. Separated from Root's ethyl acetate extract was fraction 1 has $0.1{\sim}5\;{\mu}M$ range, fraction 2 has $0.1{\sim}10\;{\mu}M$ range did not showed cytotoxicity. Anti-oxidation result as DPPH test showed the best was root ethyl acetate extract. Redusing power was made a comparison between fractions and standard. They were showed similar value. Fraction's total phenol containing result was better then standard. Conclusions : These results suggest that these extracts can be used as anti-inflammatory, anti-axidation materials.
A total of 37 bacterial isolates were obtained from dye-contaminated soil samples at a textile processing factory in Nakhon Ratchasima Province, Thailand, and the potential of the isolates to decolorize and biotransform azo dye Reactive Red 141 (RR141) was investigated. The most potent bacterium was identified as Paenibacillus terrigena KKW2-005, which showed the ability to decolorize 96.45% of RR141 (50 mg/l) within 20 h under static conditions at pH 8.0 and a broad temperature range of 30-40℃. The biotransformation products were analyzed by using UV-Vis spectrophotometry and Fourier-transform infrared spectroscopy. Gas chromatography-mass spectroscopy analysis revealed four metabolites generated from the reductive biodegradation, namely sodium 3-diazenylnaphthalene-1,5-disulfonate (I), sodium naphthalene-2-sufonate (II), 4-chloro-1,3,5-triazin-2-amine (III) and N1-(1,3,5-triazin-2-yl) benzene-1,4-diamine (IV). Decolorization intermediates reduced phytotoxicity as compared with the untreated dye. However, they had phytotoxicity when compared with control, probably due to naphthalene and triazine derivatives. Moreover, genotoxicity testing by high annealing temperature-random amplified polymorphic DNA technique exhibited different DNA polymorphism bands in seedlings exposed to the metabolites. They compared to the bands found in seedlings subjected to the untreated dye or distilled water. The data from this study provide evidence that the biodegradation of Reactive Red 141 by P. terrigena KKW2-005 was genotoxic to the DNA seedlings.
보이차를 물로 추출하고, 항산화성 물질을 얻기위해 n-hexane, ethyl acetate(EA), butanol(BuOH)로 분획하였다. 분획은 자유라티칼 소거 활성과 환원력 측정의 두가지 방법으로 항산화활성 정도를 조사하였다. 이들 항산화활성을 butylated hydroxyanisole(BHA), butylated hydroxytoluene(BHT) 그리고 ${\alpha}$-tocopherol 같은 비교표준 항산화제와 비교하였다. 보이차의 EA분획은 BHA와 BHT 보다 더 높은 항산화활성을 보였다.
비정질 루테늄 산화물을 사용한 수퍼캐패시터를 개발하였다. 삼염화루테늄 수화물$(RuCl_3{\cdot}xH_2O)$로부터 제조한 비정질의 이산화루테늄 수화물$(RuO_2{\cdot}nH_2O)$을 사용하여 수퍼캐패시터 전극을 제조하였다. 집전체로는 티타늄 및 STS 304박막에 비해 보다 넓은 전위창을 가지는 탄탈륨 박막을 사용하였다. 제조한 전극과 4.8M 황산 전해액을 사용하여 수퍼캐패시터를 제조하였다. 전극의 비정전용량은 순환전위전류분석에서 미분 최대 값으로 산화 및 환원 과정 각각 710 및 $645\;F/g-RuO_2{\cdot}nH_2O$이었으며, 평균값은 $521\;F/g-RuO_2{\cdot}nH_2O$으로 나타났다. 수퍼캐패시터를 포화카로멜기준전극에 대하여 0.5 V로 protonation level을 조정하고, 충방전 시험한 바, $151\;F/g-RuO_2{\cdot}nH_2O$의 비정전용량을 나타내었다.
본 연구에서는 Slow Release Substrate(SRS)로 사용되는 TBOS의 분해특성과 TBOS 분해생성물인 acetate와 butyrate를 적용한 탈염소화 효율을 파악하고자 하였다. 회분식 실험은 GC/FID를 이용하여 TCE 및 cis-dichloroethene(cis-DCE), 1-butanol, TBOS를 분석하였으며, acetate와 butylate는 HPLC를 이용하여 분석하였다. 혐기성 가수분해 반응을 통해 1M의 TBOS는 4M의 1-butanol로 전환 및 축적되었으며, 가수분해율은 $0.186{\mu}M/day$로 나타났다. 또한, 1-butanol은 퇴적토 내 토착균주에 의해 acetate와 butyrate로 분해되었다. 이 결과 TBOS는 자연에서의 탈염소화 공정에서 SRS로 사용하기에 적합한 것으로 판단된다. Acetate와 butyrate를 전자공여체로 적용한 TCE 탈염소화 반응은 초기 TCE 농도가 낮아짐에 따라 탈염소화 효율은 높아지는 것으로 나타났다. 또한, acetate를 적용한 탈염소화 반응의 1차 반응 상수가 butyrate를 적용한 경우보다 높게 나타났다. 이는 탈염소화 반응에서 Geobacter lovleyi의 기질친화도 및 생분해성, 그리고 다양한 기질에 대한 적응도의 영향으로 판단된다. 그러나 Geobacter lovleyi의 TCE 탈염 소화 반응에 따른 cis-DCE의 축적이 발생할 경우 Geobacter lovleyi의 탈염소화 능력이 감소하는 것으로 나타났다. 결론적으로 SRS는 Geobacter lovleyi를 이용한 TCE 탈염소화 공정 향상에 도움이 될 것이며, 이에 따라 발생되는 cis-DCE는 영가철 같은 환원성 금속이나 공존 가능한 탈염소화 미생물을 이용한 처리가 함께 필요할 것으로 판단된다.
퇴적물 공극수의 화학적 특성과 영양분의 농도변화 및 이동특성 파악은 지표수 수질관리에 중요한 요소가 된다. 밭토양 30cm와 상등수 15cm로 구성된 microcosm을 이용하여 담수에 의한 토양 공극수 및 상등수의 화학적 특성과 영앙분의 농도변화를 6개월 동안 모니터링하였다. 담수 5주가 경과한 후 토양 색은 yellowish red에서 grey로 변하였으며 토양표면에 붉은 색의 산화층이 관찰되었다. 토양 공극수의 산화환원전위와 pH는 감소하였다. 담수에 의하여 상등수의 $NO_3^-$ 농도는 증가하고 PO_4^{3-}$ 농도는 감소하였으나 토양 공극수의 $NH_4^+$, $PO_4^{3-}$, FE, Mn 농도는 증가하였다. 상등수의 $NO_3^-$ 농도 증가는 토양에서 생성된 $NH_4^+$가 상등수로 이동 및 산화에 기인하며, 토양 공극수의 PO_4^{3-}$ 농도증가는 산화철과 산화망간의 용해에 의하여 이에 흡착되어 있던 PO_4^{3-}$가 용출됨에 기인한 것으로 판단된다. PO_4^{3-}$에 대한 흡착력이 강한 산화철과 산화망간을 많이 함유하고 있는 토양표면의 산화층은 PO_4^{3-}$의 토양으로부터 상등수로 확산을 방해하는 것으로 판단된다.
환원상태가 발달된 담수상태의 토양이나 습지생태계에서 $NO_3^-$는 환원상태의 진전을 지연시키는 완충역할을 할 수 있다. 논토양에서 $NO_3^-$가 Fe의 환원과 그에 따른 P의 가용화에 미치는 영향을 조사하였다. 담수 후 10 cm 깊이 토양의 산화환원전위 변화는 $NO_3^-$를 처리한 토양과 처리하지 않은 토양에서 현저하게 달리 나타났으며, $NO_3^-$의 환원이 일어나는 동안에는 산화환원전위가 330${\sim}$360 mV 범위에서 유지되었다. 그리고 이 기간 동안 Fe의 환원은 현저히 억제되었다. $NO_3^-$를 처리한 토양 용액의 $PO_4^{3-}$ 함량은 담수 이후 0.2${\sim}$0.3 mg/L 수준 또는 그 이하로 유지되었으며, 반면 $NO_3^-$를 처리하지 않은 경우에는 담수 후 9일째부터 Fe의 환원과 함께 토양 용액의 $PO_4^{3-}$ 함량이 급격히 증가하였다. 일반적인 토양에서 무기태 P의 상당부분이 Fe 산화물에 고정된 형태 및 Fe와 결합된 형태로 존재하므로 Fe의 환원에 따라서 $PO_4^{3-}$가 함께 용출되는 것이다. 이상의 결과를 보면 토양중의 $NO_3^-$가 $Fe^{2+}$와 $PO_4^{3-}$의 용출을 조절하는 요인인 것으로 확인할 수 있다. 토양 용액중의 $NO_3^-$ 농도가 1 mg/L 이상으로 유지되는 상태에서는 토양의 산화환원전위가 330 mV 이하로 낮아지지 못하며, 따라서 Fe의 환원과 그에 따른 P의 용출 또한 현저히 억제되는 것으로 판단된다.

Metabolic engineering is now a well established discipline, used extensively to determine and execute rational strategies of strain development to improve the performance of micro-organisms employed in industrial fermentations. The basic principle of this approach is that performance of the microbial catalyst should be adequately characterised metabolically so as to clearlyidentify the metabolic network constraints, thereby identifying the most probable targets for genetic engineering and the extent to which improvements can be realistically achieved. In order to harness correctly this potential, it is clear that the physiological analysis of each strain studied needs to be undertaken under conditions as close as possible to the physico-chemical environment in which the strain evolves within the full-scale process. Furthermore, this analysis needs to be undertaken throughoutthe entire fermentation so as to take into account the changing environment in an essentially dynamic situation in which metabolic stress is accentuated by the microbial activity itself, leading to increasingly important stress response at a metabolic level. All too often these industrial fermentation constraints are overlooked, leading to identification of targets whose validity within the industrial context is at best limited. Thus the conceptual error is linked to experimental design rather than inadequate methodology. New tools are becoming available which open up new possibilities in metabolic engineering and the characterisation of complex metabolic networks. Traditionally metabolic analysis was targeted towards pre-identified genes and their corresponding enzymatic activities within pre-selected metabolic pathways. Those pathways not included at the onset were intrinsically removed from the network giving a fundamentally localised vision of pathway functionality. New tools from genome research extend this reductive approach so as to include the global characteristics of a given biological model which can now be seen as an integrated functional unit rather than a specific sub-group of biochemical reactions, thereby facilitating the resolution of complexnetworks whose exact composition cannot be estimated at the onset. This global overview of whole cell physiology enables new targets to be identified which would classically not have been suspected previously. Of course, as with all powerful analytical tools, post-genomic technology must be used carefully so as to avoid expensive errors. This is not always the case and the data obtained need to be examined carefully to avoid embarking on the study of artefacts due to poor understanding of cell biology. These basic developments and the underlying concepts will be illustrated with examples from the author's laboratory concerning the industrial production of commodity chemicals using a number of industrially important bacteria. The different levels of possibleinvestigation and the extent to which the data can be extrapolated will be highlighted together with the extent to which realistic yield targets can be attained. Genetic engineering strategies and the performance of the resulting strains will be examined within the context of the prevailing experimental conditions encountered in the industrial fermentor. Examples used will include the production of amino acids, vitamins and polysaccharides. In each case metabolic constraints can be identified and the extent to which performance can be enhanced predicted
나노 영가철은 산화환원기작을 통하여 염소계 유기화합물과 같은 물질을 효과적으로 처리할 수 있다고 알려져 있지만, 작은 사이즈로 인하여 회수가 어려운 단점으로 인하여 실제 수처리 공정에서는 유출 등의 우려로 널리 적용되지 못하였다. 이와 같은 한계를 극복하기 위하여 활성탄과 같은 담체에 고정화 하여 사용하는 연구가 활발히 진행되었다. 본 연구에서는 활성탄에 영가철의 고정화 시 대표적으로 사용되는 고온 및 상온의 두 가지 경로에 대해 평가하였으며, 결과를 바탕으로 최적의 합성 조건을 도출하였다. 효과적인 나노영가철/입상활성탄 복합체를 합성하기 위해서는 높은 철 함량과 더불어 영가철의 분율을 높이는 것이 중요하며, 이를 위해서는 합성 과정에서 형성되는 철 산화물 및 수산화물의 형성을 억제하는 것이 중요한 것으로 나타났다. 또한 영가철의 분율을 높이기 위한 환원 시간 및 중간 건조 과정의 유무 등 합성 조건의 영향을 살펴보았으며, 그 결과 중간 건조 과정 없이 바로 $NaBH_4$를 이용한 환원 조건을 약 2시간 이상 유지하는 것이 최적 조건임을 확인하였다. 합성된 나노영가철/입상활성탄 복합체는 활성탄의 흡착 능력과 영가철의 환원 능력을 동시에 보유함으로써 나이트로벤젠과 같은 환원이 가능한 오염물질의 제거에 효과적으로 나타났다.
