Antioxidant Activity of Low Molecular Weight Laminarin Prepared with Gamma Irradiation (감마선 조사된 저분자 laminarin의 항산화 활성 연구)

  • In this study, it was investigated the antioxidant activity of laminarin degraded by gamma irradiation. Because the activities of antioxidants have been attributed to various mechanisms, different assay methods have been conducted and compared. 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power, and lipid peroxidation inhibitory activity of degraded laminarin were measured and compared with non-degraded. All of these results showed that the antioxidant activity of laminarin degraded by irradiation was increased depending on the absorbed dose. Therefore, gamma irradiation could be an alternative method for the preparation of degraded laminarin with higher antioxidant activity.

Antioxidant, Anti-inflammatory and Cytotoxicity on Human Lung Epithelial A549 Cells of Jerusalem artichoke (Helianthus tuberosus L.) Tuber

  • This study investigated in vitro antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells of different solvent extracts from Jerusalem artichoke (Helianthus tuberosus L.) tuber. The EtOH extract contained amounts of phenolics (22.20 tannic acid equivalent ㎎/ɡ) and exhibited the highest antioxidant activity and anti-inflammatory activity. Several methods were employed for measure the antioxidant activity: 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (IC50 = 206.79 ㎍/㎖), reducing power activity (21.26 ascorbic acid equivalent ㎎/ɡ) and total antioxidant activity (19.05 ascorbic acid equivalent ㎎/ɡ). Meantime, the EtOH extract inhibited the NO production completely with a concentration of 800 ㎍/㎖. Besides, the H2O extract exhibited more potent effect on human lung epithelial A549 cells. This study suggested that Jerusalem artichoke tuber had antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells.

Antioxidant and Cell Proliferation Effects of Acanthopanax senticosus for Extract in Human Osteoblast-like MG-63 Cell Line (가시오가피 추출물의 항산화활성 및 MG-63 조골세포 증식과 alkaline phosphatase 활성에 미치는 효과)

  • Acanthopanax senticosus is a common Asian herb also known as "Siberian Ginseng". It is often used as a traditional herbal medicine for reducing damage in the liver, kidney, bone and muscle. In the present study we investigated the ferric reducing/antioxidant power and total phenolic contents of the ethanol-/water-extracts obtained from the stems and leaves of Acanthopanax senticosus. Osteoblast cellular proliferation was evaluated using the MTT and alkaline phosphatase activity assays in the human osteoblast-like MG-63 cell line. Acanthopanax senticosus extracts exerted remarkable ferric reducing/antioxidant power and contained high amount of phenolics. Among the extracts the stem-/ethanol-extract showed the highest antioxidant activity and total phenol content. Interestingly a highly positive correlation was found between antioxidant activity and total phenol content (p < 0.01). Proliferation of MG-63 osteoblast cells was highest in the stem-/ethanol-extract and alkaline phosphatase activity significantly increased in the water-extract of the stems (p < 0.05). These findings suggest that Acanthopanax senticosus extracts have antioxidant activity for preventing oxidative stress-related diseases and may have beneficial effects on bone health through the proliferation of osteoblast cells.

Antioxidative activities of various solvent extracts from haw (Crataegus pinnatifida Bunge) (산사(Crataegus pinnatifida Bunge)의 추출 용매별 항산화 활성)

  • The objective of this study was to investigate the antioxidative acitivities of various solvent extracts from haw (Crataegus pinnatifida Bunge) pulpy. The Freeze-dried pulpy of haw was extracted with five solvents (70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v), n-butanol, and ethyl acetate or EA), and the DPPH, reducing power, ferric-reducing antioxidant power (FRAP), ABTS, NO, hydroxyl radical scavenging activity were then measured. The DPPH radical scavenging activity of the 70% methanol and 70% ethanol extracts were 92.89% and 91.17% in the 0.8 mg/mL concentrations, respectively, which were higher than those of the other extracts. The reducing power and FRAP decreased in the following order: 70% methanol, 70% ethanol, n-butanol, EA, and CM extracts. The ABTS radical scavenging activity was high in the 70% methanol and 70% ethanol extracts and low in the n-butanol extract. The NO radical scavenging activity was high in the 70% methanol and 70% ethanol extracts but low in the CM extracts. On the other hand, the hydroxyl radical scavenging activity was high in the n-butanol and 70% methanol extracts but low in the EA extract. These results suggest that the 70% methanol and 70% ethanol extracts from haw will be useful as natural antioxidant and biohealth foods.

  • Astragalus membranaceus has a long history of medicinal use in Chinese herbal medicine. It has been shown to have immunostimulant, tonic, antioxidant, antiperspirant, diuretic, anti-diabetic, expectorant properties, and a supplementary medicine during cancer therapy. In this study, we investigated the effect of anti-oxidation of Astragalus membranaceus root extract. The anti-oxidative activities of water, 80% methanol, and 100% methanol extracts from Astragalus membranaceus were analyzed by DPPH free radical scavenging activity, Superoxide dismutase-like activity, reducing power, and crude ash. The water extract demonstrated to be more effective than methanol extract for a DPPH radicals scavenging activities and reducing power. Superoxide dismutase-like activity showed higher efficiency in 80% methanol extract. Our results indicate that Astragalus membranaceus extracts could be used as a source of antioxidant ingredients in the food industry.

Effect of Far - Infrared Radiation on the Antioxidant Activity of Extracts from Citrus unshiu Peels (감귤 과피 추출물의 항산화능에 대한 원적외선 처리의 효과)

  • The effect of far-infrared (FIR) radiation on the antioxidant activity of extracts from Citrus unshiu peels (CPs) was evaluated. CPs (5 g) were placed in Pyrex petri dishes (8.0 cm diameter) and irradiated at 150$^{\circ}C$ for 10, 20, 30, 40 or 50 min with a FIR heater. After FIR irradiation, 70% ethanol extract of CPs were prepared and total phenol contents (TPC), radical scavenging activity (RSA) and reducing power of the extracts were determined. FIR radiation of CPs at 150$^{\circ}C$ for 30 min increased the TPC and RSA of ethanol extract from 71.9${\mu}$M to 133.9 ${\mu}$M and from 29.64% to 51.27%, respectively, and reducing power was increased from 0.451 to 0.675, compared to those of non-irradiated control. These results indicated that FIR radiation onto CPs could liberate and activate covalently bound phenolic compounds that have antioxidant activities.

Making of Dongchimi Naengmyeun Broth Which Has Enhanced Antioxidant Activity Using Purple Sweet Potato (자색고구마를 사용한 항산화 활성이 강화된 동치미 냉면육수 제조)

  • Lactic fermented dongchimi naengmyeun broth which has enhanced antioxidative activity was developed. During lactic fermentation of dongchimi naengmyeun broth at $25^{\circ}C$, changes of total lactic acid bacteria, pH, acidity, soluble phenolics, and anthocyanins were investigated. After 72 h of fermentation, the stronger antioxidant activities were observed in dongchimi naengmyeun broth supplemented with purple sweet potato than those of control dongchimi naengmyeun broth which showing 96.80% in DPPH radical scavenging activity, 100.82% in $ABTs^{+{\cdot}}$ scavenging activity, 7.77 in reducing power, and 6.89 in ferric reducing/antioxidant power, respectively. These high antioxidant activities related with higher contents of soluble phenolics and anthocyanins in dongchimi naengmyeun broth supplemented with purple sweet potato. The results suggest that the making of functional dongchimi naengmyeun broth by using high soluble phenolics and anthocyanins supplements such as purple sweet potato powder was possible.

Freshness and Antioxidant Activities in Pacific Oyster Crassostrea gigas Using Rack-and-Bag Culture or Suspended Culture Methods (부유망식과 수하식 양성방법에 따른 참굴(Crassostrea gigas)의 선도와 항산화활성)

  • The nucleotides and their related compounds, including ATP (adenosine triphosphate), ADP (adenosine diphosphate), AMP (adenosine monophosphate), IMP (inosine monophosphate), HxR (inosine) and Hx (hypoxanthine), were nearly identical in oysters Crassostrea gigas from the two culture methods. The K-value was lower than the threshold value such as 11.2-12.1. Although oysters have low amount of IMP, it was detected in this experiment. DPPH radical scavenging activity did not vary significantly with sample amounts (100, 300, and $500{\mu}g/mL$). DPPH (1,1-diphenyl-2-picryl-hydrazyl) radical scavenging activity was 76.0-80.7% compare with the ascorbic acid standard. Superoxide anion scavenging activity reached 49.3% in the rack-and-bag culture sample at $500{\mu}g/mL$. However, the reducing power and $Fe^{2+}$ chelating activity were very low compared with their respective standard. The oyster culture methods did not affect oyster quality in terms of antioxidant activities.

Antioxidant, Anticancer and Anticholinesterase Activities of Flower, Fruit and Seed Extracts of Hypericum amblysepalum HOCHST

  • Background: Cancer is an unnatural type of tissue growth in which the cells exhibit unrestrained division, leading to a progressive increase in the number of dividing cells. It is now the second largest cause of death in the world. The present study concerned antioxidant, anticancer and anticholinesterase activities and protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in methanol extracts of flowers, fruits and seeds of Hypericum amblysepalum. Materials and Methods: Antioxidant properties including free radical scavenging activity and reducing power, and amounts of total phenolic compounds were evaluated using different tests. Protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in extracts were determined by HPLC. Cytotoxic effects were determined using the MTT test with human cervix cancer (HeLa) and rat kidney epithelium cell (NRK-52E) lines. Acetyl and butyrylcholinesterase inhibitory activities were measured by by Ellman method. Results: Total phenolic content of H. amblysepalum seeds was found to be higher than in fruit and flower extracts. DPPH free radical scavenging activity of the obtained extracts gave satisfactory results versus butylated hydroxyanisole and butylated hydroxytoluene as controls. Reducing power activity was linearly proportional to the studied concentration range: $10-500{\mu}g/mL\;LC_{50}$ values for H. amblysepalum seeds were 11.7 and 2.86 respectively for HeLa and NRK-52E cell lines. Butyryl-cholinesterase inhibitory activity was $76.9{\pm}0.41$ for seed extract and higher than with other extracts. Conclusions: The present results suggested that H. amblysepalum could be a potential candidate anti-cancer drug for the treatment of human cervical cancer, and good source of natural antioxidants.

Antioxidant Activities of Functional Beverage Concentrates Containing Herbal Medicine Extracts

  • This study investigated the antioxidant activity of functional beverage concentrates containing herbal medicine extracts (FBCH) using various antioxidant assays, such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, and reducing power assay. The total polyphenolic content of FBCH (81.45 mg/100 g) was higher than Ssanghwa tea (SHT, 37.56 mg/100 g). The antioxidant activities of FBCH showed 52.92% DPPH and 55.18% ABTS radical scavenging activities at 100 mg/mL, respectively. FBCH showed significantly higher antioxidant activities compared to the SHT (DPPH, 23.43%; ABTS, 22.21%; reducing power optical density; 0.23, P<0.05). In addition, intracellular reactive oxygen species generation significantly decreased in a concentration-dependent manner following FBCH treatment. These results suggest that the addition of herbal medicine extract contributes to the improved functionality of beverage concentrates.
