Enhanced Transformation Efficiency of an Anticoagulant Hirudin Gene into Saccharomyces cerevisiae by a Double ${\delta}-Sequence$

  • Delta-integration vectors were constructed for the purpose of achieving homologous integration of the hirudin expression cassette into the chromosome of Saccharomyces cerevisiae. A double $\delta$ system truncated with the unnecessary bacterial genes, and consequently having a reduced insert size for integration, showed a four-fold increase in transformation efficiency at given DNA concentrations, and as a result, the constructed recombinant yeast strain had a 1.3-fold enhancement in hirudin expression level compared with a single $\delta$ system.

  • The Steam Assisted Gravity Drainage (SAGD) process is an enhanced method to extract oil from bitumen which involves surface and central process facilities. This paper describes the Central Process Facilities (CPF) of SAGD and proposes several retrofit plans to the Heat Exchanger Network (HEN). In this approach, the process integration scheme is applied to estimate the energy saving in HENs, and various cases are modeled in favor of a commercial simulator. Throughout this work, a minimum approach temperature of 10℃ is assumed. The results reveal that, due to the HEN optimization using process integration, the heating and cooling duties can be reduced to 29.68MW and 1.886MW, respectively. Compared with the Husky case, all cases considered in this study indicate a potential reduction of at least 6% in total cost, including investment and operation costs.

  • In case of traditional Rule-Based Reasoning(RBR) and Case-Based Reasoning(CBR), although knowledge is reasoned either by one of them or by the integration of RBR and CBR, there is a problem that much time should be consumed by numerous rules and cases. In order to improve this time-consuming problem, in this paper, a new type of reasoning technique, which is a kind of integration of reduced RB and CB, is to be introduced. Such a new type of reasoning uses Rough Set, by which we can represent multi-meaning and/or random knowledge easily. In Rough Set, solution is to be obtained by its own complementary rules, using the process of RB and CB into equivalence class by the classification and approximation of Rough Set. and then using reduced RB and CB through the integrated reasoning.

  • In this paper, behavior of rectangular storage structures under earthquake loadings are investigated. Linear sloshing is assumed in this study. The effect of the wall flexibility is considered. Eulerian and lagrangian approaches are presented. The Eulerian approach is carried out by solving the boundary value problem for the fluid motion. In the lagrangian approach, the fluid as well as the storage structure is modelled by the finite element method. The fluid region is discretized by using fluid elements. The (1 $\times$ 1)-reduced integration is carried out for constructing the stiffness matrices of the fluid elements. Seismic analysis of the coupled system is carried out by the response spectra method. The numerical results show that the fluid forces on the wall obtained by two approaches are in good agreements. By including the effect of the wall flexibility, the forces due to fluid motion can be increased very significantly.

  • We discuss finite element approximation and use a Mindlin plate element based upon uniformly reduced numerical integration. The finite element selected for use in this work is a four-node, bilinear displacement element based upon the Mindlin theory of plates. Such elements show good accuracy for laminated composite plates when reduced numerical integration is used to evaluate the element marices. This study presents both the experimental and F.E. results for the natural frequencies of CFRPURETHANE-CFRP Composite plate. Good agreement between experimental and calculated frequencies is achived.

Analyses of Non-linear Behavior of Axisymmetric Structure by Finite Element Method (유한요소법을 이용한 축대칭 구조물의 비선형 거동해석)

  • A finite element method is programmed to analyse the nonlinear behavior of axisymmetric structures. The lst order Mindlin shell theory which takes into account the transversal shear deformation is used to formulate a conical two node element with six degrees of freedom. To evade the shear locking phenomenon which arises in Mindlin type element when the effect of shear deformation tends to zero, the reduced integration of one point Gauss Quadrature at the center of element is employed. This method is the Updated Lagrangian formulation which refers the variables to the state of the most recent iteration. The solution is searched by Newton-Raphson iteration method. The tangent matrix of this method is obtained by a finite difference method by perturbating the degrees of freedom with small values. For the moment this program is limited to the analyses of non-linear elastic problems. For structures which could have elastic stability problem, the calculation is controled by displacement.

  • This paper presents a new scheme for constructing locking-free finite elements in thick and thin plates, called Discrete Shear Gap element (DSG), using multiphase material topology optimization for triangular elements of Reissner-Mindlin plates. Besides, common methods are also presented in this article, such as quadrilateral element (Q4) and reduced integration method. Moreover, when the plate gets too thin, the transverse shear-locking problem arises. To avoid that phenomenon, the stabilized discrete shear gap technique is utilized in the DSG3 system stiffness matrix formulation. The accuracy and efficiency of DSG are demonstrated by the numerical examples, and many superior properties are presented, such as being a strong competitor to the common kind of Q4 elements in the static topology optimization and its computed results are confirmed against those derived from the three-node triangular element, and other existing solutions.

  • By the SOG (System-on-Glass) technology with excimer laser anneal process, the number of IC chips and the area of the mounted IC chips on the printed circuit board are reduced. In new circuit integrations on the glass substrate, we have developed D/A converter including the new capacitor array, amplifier comprising the original comparators and new display device with capturing images by integrated sensor into a pixel. This paper discusses the application of circuit integration of low-temperature poly-Si.

  • Smoke emissions from light duty diesel vehicles were measured using light extinction method with the free acceleration test mode. The smoke emissions for each measurement cycle of the free acceleration method showed large variations according to driver's pedal pushing pattern. The smoke values for each measurement cycle initially increased and reach a peak value. Integration of the smoke emissions with time for each measurement cycle was performed to get a representative smoke value which was obtained by averaging the integrated results. Two kinds of integration time range were used. One is range over the whole measurement cycle of the free acceleration method. The other is only the acceleration range in the measurement cycle. Overall, variation of the representative smoke values obtained by the integration method was reduced comparing to the traditional representative smoke value which was obtained from a peak smoke value over the measurement cycle. Ten vehicles of the same model with 2.5 liter diesel engines, and seven vehicles of the same model with 2.7 liter diesel engines, were tested using the free acceleration test method.

Construction Cost-Schedule Integration Management Methodolgy by using Progress Integration Unit (성과측정유닛을 활용한 건설 비용 - 일정 통합관리 방안)

  • Measuring and evaluating project progress and performance are the key element of the construction project success. Construction progress is typically measured quantitatively by evaluating cost and time allocated to the project deliverable, and thus properly integrating cost and time is essential to the project management. This research was performed to propose an alternative methodology to integrate the cost and time and provide a framework for the progress measurement. The researchers developed a typical work process for the cost and schedule planning and also developed an alternative cost-schedule integration method by using progress integration units (PIU). A discipline of a construction phase served as a common level for WBS and CBS integration, so the PIUs'were defined under discipline. A case study project was selected to validate the developed methodology. The result showed the proposed method improved efficiency of cost and time integration. The result also showed the excluding material for the progress measurement purpose significantly reduced the bias of progress measurement.
