Suppression of colon cancer by administration of Canavalia gladiata D.C. and Arctium lappa L., Redix extracts in tumor-bearing mice model (종양이식 생쥐모델에서 도두(刀豆), 우방근(牛蒡根) 추출물의 대장암 억제 효과)

  • Objective : In the present study, we examined whether Canavalia gladiata D.C. (CG) and Arctium lappa L., Redix (AL) mixture (CGAL), their components, lupeol and chicoric acid, regulate immune system and suppress the tumor in vitro and in vivo. Methods : LPS-induced reactive oxygen species (ROS) and nitric oxide (NO) were measured after treatment with CG extract (CGE), CGAL, lupeol, chicoric acid and lupeol and chicoric acid mixture (lupeol+CA) in Raw264.7 cell. To determine the effect of CGE on immune responses, immune cell population and IgG production were assessed in mice. To investigate the effect of CGAL and their component on anti-tumor activity, tumor volume and weight were measured, cell cycles and immune cell population were analyzed in MC38 injected tumor bearing mice. Also, NK cell activity was determined in splenocyte isolated from tumor bearing mice. Results : CGE, CGAL, lupeol, chicoric acid and lupeol+CA decreased the LPS-induced ROS and NO production without cell toxicity in RAW264.7 cells. CGE increased the immune cell populations of $CD4^+T$, $CD8^+T$ and macrophages in various immune organ of mice. In tumor bearing mice, CGAL, lupeol, chicoric acid and lupeol+CA suppressed tumor volume and weight. In cell cycle analysis, they decreased the percentages of S phase. In addition, CGAL, lupeol, chicoric acid and lupeol+CA immune cell populations of $CD4^+T$, $CD8^+Tcell$, NK cell and macrophage in tumor as well as NK cell activity. Conclusion : CGAL and its compounds may enhance immune responses and suppress tumor growth, and may be capable of developing health functional foods.

Pathological Mechanism of Taeyang Blood Retention Pattern Based on Cases of Thrombosis in Patients with COVID-19 Infection : A Literature Review (코로나19 감염증 환자의 혈전 사례를 바탕으로 본 태양병 축혈증의 병리기전에 관한 문헌고찰)

  • COVID-19 infection heightens the risk of thromboembolism. To see the similarities between the COVID-19 infection and Taeyang blood retention pattern, we conducted a PubMed search using specific terms related to blood circulation issues in the context of COVID-19, summarizing findings from 13 cases and 4 observational studies involving actual patients. Patients with COVID-19 are at risk of blood coagulation due to factors such as viral-induced cytokine storms, vascular endothelial dysfunction, reduced mobility in bedridden or isolated individuals, and resulting constipation. Additionally, cytokine storms and severe inflammation can lead to delirium in COVID-19 patients. The Taeyang blood retention pattern manifests as symptoms arising from delirium and an increased blood coagulation tendency in patients with a robust immune response. According to the Sanghan theory, certain herbal treatments can alleviate symptoms in patients with a tight lower abdomen who do not experience urinary issues. Studies show that components like Persicae Semen and Rhei Redix et Rhizoma in these prescriptions enhance blood circulation and reduce hypercoagulability. Additionally, these treatments aim to promote blood flow by relieving abdominal pressure through facilitating bowel movements. The excessive inflammation and heightened blood coagulation tendency in COVID-19 resemble the Taeyang blood retention pattern, although they are caused by different pathogens. Reinterpreting classical oriental medicine's principles in a modern context may enhance our understanding of traditional East Asian Medicine and foster future developments.

Effect of N, P and K Application on the Contents of the Decursin and Total Sugar in Angelicae gigantis Radix (N,P,K 시용(施用)이 당귀근(當歸根)(Angelicae gigantis Radix)의 Decursin 및 당함량(糖含量)에 미치는 영향(影響))

  • This experiment was conducted to increase the available constituents contents of Angelica gigas Nakai and to decide the application amounts of nitrogen, phosphorus and potassium fertilizer. The effects of the amounts of their application and absorption on the contents of decursin and sugar in Angelicae gigantis Redix were studied. The decursin contents of root was decreased with increasing the application amount of nitrogen more than 10kg/10a, and increased along with the increasing application level until 20kg/10a and 15kg/10a of phosphorus and potassium, respectively. The decursin yield per 10a was increased with increasing application level until 10kg/10a, 10kg/10a and 10kg/10a of nitrogen, phosphorus and potassium, respectively. The decursin yield per l0a was decreased with increasing application level more than 20kg/10a of nitrogen. The total sugar yield per 10a was increased with increasing the application level until 15kg/10a, 10kg/10a of nitrogen and potassium respectively. But the effect of phosphorus application on the total sugar yield was not observed. The significant negative correlation was obtained between the decursin contents and the nitrogen contents of root$(r=-0.437^{\ast})$, and the significant positive correlation between the decursin contents and the contents of phosphorus$(r=0.587^{{\ast}{\ast}})$ or potassium$(r=0.467^{\ast})$ in root. But the significant correlations between the decursin contents and the their contents of shoots were not observed. It is recommended for decursin production to apply 10kg/10a, 15kg/10a and 10kg/10a of nitrogen, phosphorus and potassium, respectively.

Antimicrobial Activity of Whangkumtang Extract and Scutellariae Radix Extract on the Food-Borne Pathogens (황금탕 및 황금추출물의 식중독 세균들에 대한 생육억제 효과)

  • The effects of Whangkumtang extract and Scutellariae radix extract on the microbial growth were investigated. The antimicrobial activities and cell growth inhibiting effects were investigated to selected strains with different concentrations of Whangkmtang and Scutellariae radix extracts. Whangkmtang and Scutellariae radix extracts showed the antimicrobial activities on Staphylococcus aureus, Bacillus cereus, Shigella sonnei, Shigella flexneri, Salmonella Typhimurium, Salmonella Enteritidis, Vibrio parahaemolyticus, Escherichia coli O111 and Escherichia coli O126. Whangkumtang and Scutellariae radix extracts did not show the antimicrobial activity on Listeriamonocytogenes. Scutellariae radix extract showed the antimicrobial activity on Escherichia coli O157 but Whangkumtang extract did not, Minimum inhibitory concentrations (MIC) of Whangkumtang extract for each strain appeared to 40 mg/mL on Staphylococcus aureus and Bacillus cerus, 100 mg/mL on Shigella Flexneri and Salmonella enteritidis. The MICs of Sutellariae radix extract appeared to 10 mg/mL on Bacillus cereus, 20 mg/UL on Staphylococn aureus, Shigellanexneri and Salmonella Enteritidis. Scutellariae radix extract showed the higher antimicrobial activity than Whangkumtang extract. The cell growth inhibitions by Whangkumtang and Scutellariae radix extracts were observed from Staphylococcus aureus and Bacillus cereus, repectively, during 48-hr incubation period.

Heavy Metals and Pesticides with Maturing Process of Rehmanniae Radix Preparata (숙지황 제조과정에서 중금속과 잔류농약)

  • To obtain the risk assessment of hazardous materials with maturing process of Rehmanniae Radix Preparata, the residual heavy metals and pesticides in samples are surveyed. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparata of Korea are $0.147{\pm}0.007\;mg/L,\;0.025{\pm}0.001\;mg/L\;and\;0.193{\pm}0.021\;mg/L$, respectively. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparat of China are $0.0145{\pm}0.014\;mg/L,\;0.023{\pm}0.002\;mg/L\;and\;0.299{\pm}0.013\;mg/L$, respectively. Concentration of As, Cd and Hg remain unchanged during the maturing process of Rehmanniae Radix Preparata. Concentration of As, Cd and Pb in Rehmanniae Radix Preparat on the Korea are 3.53%, 6.66% and 5.16% for Korea Food & Drug Administration (KFDA) advisory level, respectively. The residual pesticides, such as aldrin, $BHC-{\alpha},\;BHC-{\beta},\;BHC-{\gamma},\;BHC-{\delta}$, captan, chlorfenapyr, chlorpyrifos, DDD, DDE, DDT-o,p, DDT-p.p, dieldrin, endosulfan-sulfate, $endosulfan-{\alpha}$, $endosulfan-{\beta}$, fenitrothion, penthoate, permethrin, quintozene, tolyfluanid and vinclozolin, are not detected in Rehmanniae Radix Preparat on the Korea and China. The hazardous materials in Rehmanniae Radix Preparata on the Korea and China are verified the safety of the residual heavy metals and pesticides compare with Korea Food & Drug Administration (KFDA) advistory level.
