Ginsenoside $Rg_5$, A Genuine Dammarane Glycoside from Korean Red Ginseng

  • A genuine dammarane glycoside, named ginsenoside $Rg_{5}$, has been isolated by repeated column chromatography and preparative HPLC from the MeOH extract of Korean red ginseng (Panax ginseng C.A. Meyer). The chemical structure of ginsenoside$ Rg_{5}$ was determined as $3-O-[{\beta}-D-glucopyranosyl (1{\rightarrow}2)-{\beta}-D-glucopyranosyl]$ dammar-20(22), $24-diene-3{\beta},12{\beta}-diol$ by spectral and chemical methods. The stereostructure of a double bond at C-20(22) of ginsenoside $Rg_{5}$ was characterized as (E) from the chemical shift of C-21 in the $^{13}C-NMR $and a NOESY experiment in the $^{1}H-NMR$.

In Vitro Sex Steroid Metabolism in Red Spotted Grouper, Epinephelus akaara during Oocyte Maturation

  • We studied steroid metabolites produced from red-spotted grouper ovarian follicles during maturation. Oocytes with 350-500 ㎛ diameter were in vitro incubated in the presence of [3H] 17α-hydroxyprogesterone as a precursor. Steroid metabolites were extracted from incubated media and oocytes. The extracts were separated and identified using thin layer chromatography, high performance liquid chromatography and gas chromatography-mass spectrometry. The identified metabolites were androstenedione (A4), testosterone (T) and estrone (E1). The metabolites of A4 was dominant in all size of oocytes and it was the highest in 480 ㎛ diameter oocytes. The metabolites of two progestins, 17α,20β-dihydroxy-4-pregnen-3-one and 17α,20α-dihydroxy-4-pregnen-3-one were detected in the oocytes less than 480 ㎛ diameter although they were not identified definitely. In the oocytes of 480 ㎛ diameter, metabolite of progestin was the highest, and germinal vesicle (GV) was still in the middle of cytoplasm. In the oocytes of 500 ㎛ diameter, GV was began to migrate and the major metabolites were A4 and E1. The metabolite of E1 was detected in all size of oocytes and it was higher than that of E2. These results suggest that oocytes of 480 ㎛ diameter are the transitional stage involving steroidogenic shift to final oocyte maturation and potential function of E1 during maturation process.

On the Red Cell Fragility in the Normal and Anemic Professional Blood Donors (건강인(健康人) 및 직업적(職業的) 매혈(賣血)로 인(因)한 실혈성(失血性) 빈혈자(貧血者)의 적혈구(赤血球) 취약성(脆弱性)에 관(關)하여)

  • The osmotic and mechanical red cell fragility of the professional blood donors, who were found to be anemic as the result of frequent and repeated blood loss the past 5-6 years, were compared with that of the normal person while incubating the blood at $4-6^{\circ}C$ for 28 days. The fragility was expressed as % hemolysis occured during the incubation, and the following results were obtained: 1. The osmotic fragility in the normal persons (i.e, ; control group) progressively increased as the incubation became longer, and % hemolysis in 0.42% NaCl solution at 0, 10, 15, 21 and 28 incubation day was 31.90, 50.20, 41.68, 43.50 and 55.40 respectively. The mechanical fragility. in the normal red cells ranged between the minimum of 0.00% to the maximum of 5.80% both in 0.90 and 0.66% of NaCl solutions. 2. The hemolysis curve obtained in the red cell osmotic fragility from three cases of the anemic persons (i.e,; experimental group) showed a significant left side shift comparing with the normal in general which indicates that the fragility was more increased in the experimental group. The mechanical fragility in the experimental group ranged between the minimum 0.00% to the maximum 19.00% both in 0.90 and 0.66% of NaCl solutions. 3. The red cells of the chronic anemic person due to the frequent blood loss as the professional blood donor exhibit significantly marked increase both in osmotic and mechanical fragility comparing with the normal, and the tendency was more prominent as the incubation period became longer.

Electrochemical Control of Metabolic Flux of Weissella kimchii sk10: Neutral Red Immobilized in Cytoplasmic Membrane as Electron Channel

  • Electrochemical control of the metabolic flux of W. kimchii sk10 on glucose and pyruvate was studied. The growing cell of W. kimchii sk10 produced 87.4 mM lactate, 69.3 mM ethanol, and 4.9mM lactate from 83.1mM glucose under oxidation condition of the anode compartment, but 98.9 mM lactate, 84.3mM ethanol, and 0.2 mM acetate were produced from 90.8 mM glucose under reduction condition of the cathode compartment for 24 h, respectively. The resting cell of W. kimchii sk10 produced 15.9 mM lactate and 15.2 mM acetate from 32.1 mM pyruvate under oxidation condition of the anode compartment, and 71.3 mM lactate and 3.8 mM acetate from 79.8mM pyruvate under reduction condition of the cathode compartment. The redox balance (NADH/$NAD^+$) of metabolites electrochemically produced from pyruvate was 1.05 and 18.76 under oxidation and reduction conditions, respectively. On the basis of these results, we suggest that the neutral red (NR) immobilized in bacterial membrane can function as an electron channel for the electron transfer between electrode and cytoplasm without dissipation of membrane potential, and that the bacterial fermentation of W. kimchii sk10 can be shifted to oxidized or reduced pathways by the electrochemical oxidation or reduction, respectively.

Effects of Microbial Iron Reduction and Oxidation on the Immobilization and Mobilization of Copper in Synthesized Fe(III) Minerals and Fe-Rich Soils

  • The effects of microbial iron reduction and oxidation on the immobilization and mobilization of copper were investigated in a high concentration of sulfate with synthesized Fe(III) minerals and red earth soils rich in amorphous Fe (hydr)oxides. Batch microcosm experiments showed that red earth soil inoculated with subsurface sediments had a faster Fe(III) bioreduction rate than pure amorphous Fe(III) minerals and resulted in quicker immobilization of Cu in the aqueous fraction. Coinciding with the decrease of aqueous Cu, $SO_4{^{2-}}$ in the inoculated red earth soil decreased acutely after incubation. The shift in the microbial community composite in the inoculated soil was analyzed through denaturing gradient gel electrophoresis. Results revealed the potential cooperative effect of microbial Fe(III) reduction and sulfate reduction on copper immobilization. After exposure to air for 144 h, more than 50% of the immobilized Cu was remobilized from the anaerobic matrices; aqueous sulfate increased significantly. Sequential extraction analysis demonstrated that the organic matter/sulfide-bound Cu increased by 52% after anaerobic incubation relative to the abiotic treatment but decreased by 32% after oxidation, indicating the generation and oxidation of Cu-sulfide coprecipitates in the inoculated red earth soil. These findings suggest that the immobilization of copper could be enhanced by mediating microbial Fe(III) reduction with sulfate reduction under anaerobic conditions. The findings have an important implication for bioremediation in Cu-contaminated and Fe-rich soils, especially in acid-mine-drainage-affected sites.

  • The use of the Internet of things plays a major role in the Fourth Industrial Revolution, and a series of tasks of accumulating, converging, analyzing and reusing various data and services becomes very important. Because the pace and scope if the paradigm shift in Fourth Industrial Revolution is so rapid and unpredictable, the development and utilization of a system to fulfill this role for IOT are urgently required. In this paper, we introduce the Web-based IOT management system, which connects the IOT with OKMindmap, which is a domestic open source software and service, and the Node-RED service. This system combines the advantages of OKMindmap with the advantages of Node-RED, which is capable of visual component based programming, so that it can easily and flexibly connect the IOT based on Web browsers, and various data and services can be integrated and linked. We developed a camera module, a temperature and humidity sensor module, and the motor control module in Raspberry PI basically, and tested the operation successfully. We plan to extend the IOT component gradually by using Arduino and System On Chip.

The Effect of Aroma Inhalation Therapy on Fatigue and Sleep in Nurse Shift Workers (아로마 흡입법이 교대 근무 간호사의 피로와 수면에 미치는 효과)

  • Purpose: This study examines the effect of aroma inhalation therapy with lavender and rosewood essential oils for alleviating fatigue and enhancing quality of sleep in nurse shift workers. Methods: The data were collected from November 9th to 27th in 2009 at four hospitals located in Seoul. Total 60 nurses who started their night-shift works were chosen for the study and allocated to either experimental group or control group. The experimental group had had aroma inhalation therapy with lavender and rosewood for 2 weeks and was compared the level of fatigue and quality of sleep with those of the control group. Results: Significant differences were shown between the experimental group and the control group. After 2 weeks, the nurses in experimental group reported significantly lower level of fatigue and higher quality of sleep compared to the nurses in control group. Conclusion: Therefore, aroma inhalation therapy by using lavender and rosewood essential oils can be an effective intervention for relieving the level of fatigue and enhancing quality of sleepf for nurse shift workers.

Different Types of Active Region EUV Bright Points by Hinode/EIS

  • We have investigated seven Extreme-Ultraviolet (EUV) bright points in the active region (AR 10926) on 2006 December 2 by the EUV imaging spectrometer (EIS) onboard Hinode spacecraft. We determined their Doppler velocities and non-thermal velocities from 15 EUV spectral lines (log T=4.7-7.2) by fitting each line profile to a Gaussian function. We present the Doppler velocity map as a function of temperature which corresponds to a different height. As a result, these active region bright points show two different types of characteristics. Type 1 bright point shows a systematic increase of Doppler velocity from -68 km/s (blue shift) at log T=4.7 to 27 km/s (red shift) at log T=6.7, while type 2 bright points have Doppler velocities in the range of -20 km/s and 20 km/s. Using MDI magnetograms, we found that only type 1 bright point was associated with the canceling magnetic feature at the rate of $2.4{\times}10^{18}$ Mx/hour. When assuming that these bright points are caused by magnetic reconnection and the Doppler shift indicates reconnection out flow, the pattern of the Doppler shift implies that type 1 bright point should be related to low atmosphere magnetic reconnection. We also determined electron densities from line ratio as well as temperatures from emission measure loci using CHIANTI atomic database. The electron densities of all bright points are comparable to typical values of active regions (log Ne=9.9-10.4). For the temperature analysis, the emission loci plots indicate that these bright points should not be isothermal though background is isothermal. The DEM analysis also show that while the background has a single peak distribution (isothermal), the EUV bright points, double peak distributions.

Stability and Sensory Evaluation of Naphthoquinone Pigments from the Roots of Lithospermum erythrorhizon (자근(紫根)으로부터 분리한 Naphthoquinone류 색소의 pH 안정성 및 관능검사)

  • The purplish red pigment from the roots of Lithospermum erythrorhizon, a Korean edible wild plant, has been investigated concerning it's value as a natural colorant for Korean traditional foods. An attempt was made to isolate pigments and define their characteristics. Two compounds of isobutylshikonin and acetylshikonin were identified by melting point determination and spectra of UV, IR, and $^{1}H-NMR$. To examine the utility of these naphthoquinone pigments for foods, the effect of various pH values on stability were determined over a period of storage. Buffered solutions of acetylshikonin and isobutylshikonin at pH 3 and 5 showed stable purplish red. The absorption maxima if acetylshikonin and isobutylshikonin over the range of pH 3 to 7 were 518 nm and 520 nm, respectively. A bathochromic shift to 588 nm at pH 10 was observed on these two naphthoquinone pigments. Sensory evaluation was performed with acetylshikonin and isobutylshikonin of identical absorbance. These two pigments revealed purplish red color in Munsell system.

Effects of pH and Temperature on the Adsorption of Cationic Dyes from Aqueous Suspension by Maghnia Montmorillonite (수용액으로부터 양이온 염료 흡수에 대한 pH 및 온도 효과)

  • The effects of pH and temperature on the removal of two dyes (neutral red; NR and malachite green oxalates; MG) from aqueous effluents using Maghnia montmorillonite clay in a batch adsorption process were investigated. The results showed the stability of the optical properties of MG in aqueous solution and adsorbed onto clay under wide range of pH 3-9. However, the interaction of NR dye with clay is accompanied by a red shift of the main absorption bands of monomer cations under pH range of 3-5, whereas, those of neutral form remains nearly constant over the pH range of 8-12. The optimal pH for favorable adsorption of the dyes, i.e. ${\geq}$90% has been achieved in aqueous solutions at 6 and 7 for NR and VM respectively. The most suitable adsorption temperatures were 298 and 318 K with maximum adsorption capacities of 465.13mg/g for NR and 459.89 mg/g for MG. The adsorption equilibrium results for both dyes follow Langmuir, Freundlich isotherms. The numerical values of the mean free energy $E_a$ of 4.472-5.559 kj/mol and 2.000-2.886 kj/mol for NR and MG respectively indicated physical adsorption. Various thermodynamic parameters, such as ${\Delta}H^{\circ}$, ${\Delta}S^{\circ}$, ${\Delta}G^{\circ}$ and Ea have been calculated. The data showed that the adsorption process is spontaneous and endothermic. The sticking probability model was further used to assess the potential feasibility of the clay mineral as an alternative adsorbent for organic ion pollutants in aqueous solution.
