Growth performances of the selected and the non-selected red sea bream (Pagrus major) lines were evaluated up to marketable size. The offspring were mass-produced either from the selected line (the fourth generation of broods from mass selection) or the non-selected line (the second generation of wild broods). Early (59 to 103 days old, trial I) and late growth performances (4 to 24 months old, trial II) were evaluated based on communal rearing in land-based tanks and sea cages, respectively. In trial I, the selected line exhibited significantly higher values in both weight gain (WG) and specific growth rate (SGR) when compared to those observed in the non-selected line (P<0.05). In trial II comprising of three growth phases (4-9, 9-16 and 16-24 months old), selected line showed only slightly improved in WG and SGR at the beginning two growth phases (4-9 and 9-16 months old) (P>0.05), however, displayed significantly higher values at the last growth phase (16-24 months old) (P<0.05). Overall WG and SGR throughout the experiment (4-24 months) were also significantly higher in the selected line than the non-selected line (P<0.05). Consequently, mean body weight of the selected line at 24 months old was 1.4-fold (40.8{\%}\;increased) as compared to that of the non-selected line. This present result indicates that mass selection of this species against the growth traits might be quite effective, and the present advance has important Implication in the productivity enhancement of red sea bream aquaculture.
Tissue distribution and residue depletion of oxytetracycline (OTC) and tetracycline (TC) following dipping administration were evaluated in olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major) under field conditions. Fishes were held in floating cages placed in sea water and fed a commercial diet for 15 days to acclimate to a new surrounding. Fishes were dipped in OTC 50 g/ton water for 30min and TC 18 g/ton water for 5 hours. Blood and muscle were sampled from fishes on 0th, 1th, 2th, 3th, and 5th day after administration. After solid-phase extraction, OTC and TC analyses were carried out by HPLC. The recovery rate of OTC in serum and muscle samples was 71-77% and 78-84%, respectively. Also, the recovery rate of TC in serum and muscle samples was 70-79% and 73-78%, respectively. The results of recovery rate were similar to previous studies reported. At the termination of dipping administration of OTC and TC, residue concentration in muscle samples of rockfish was significantly higher than those of olive flounder and red sea bream. At day 5, residue concentrations of all samples were believed to decrease to lower than 0.05 mg/kg, the detection limit. The present study showed that residue concentrations of OTC and TC decreased to below 0.05 mg/kg after treatment 5th day, faster than the established withdrawal period. The tissue reside depletion time of dipping administration of OTC and TC seems to be shorter than those of oral or parenteral administration.
Pathological symptoms and hematological parameters of red sea bream, Pagrus major and water temperature in the culture ground was investigated to clarlify the cause of winter moratility. Dead fish showed green liver and accumulation of ascites in the cavity. A few Bivagina tai were also found on the gill but either bacteria or virus were not. When hematological parameters from fish taken before/after the winter mortality were compared, blood glucose, serum AST and ALT, and total cholesterol, trigyceride and total protein were significantly decreased in the fish after the winter mortality. These results may explain that the nutritional level of fish was decreased because fish could not fed during winter season. According to the CORI monitoring system operating by KODC, NFRDI a long term water temperature from Dec. 25, 2005 to Feb. 24, 2006 (60 days) were exposed the low water temperature environments to the red sea bream. First of all, mass mortality began at Sarayngdo where low temperature below 8℃ continued for 42 days. The winter mortality did not occured in the depths of 9 m to 19.2 m where difference of water temperature in the surface and bottom was only within 0.1℃. But in the depth of 7.5 m to 11 m winter mortality occured where water temperature in the surface and bottom showed much variation ranged from 0.2℃ to 1.4℃. From these results, great difference of water temperature in the surface and bottom of the culturing area might results in winter mortality of red sea bream.

Fluoroquinoles have a wide range of antimicrobial properties and are effective in the treatment of bacterial diseases in fish. The use of fluoroquinoles continues to grow steadily. Fluoroquinolone antibiotics are probably the most important class used among synthetic antibiotics in human and veterinary medicines because of their broad activity spectrum and good oral absorption. This study was conducted to estimate the residue of antibiotics in four species of farmed fishes, including olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major), and sea bass (Lateolabrax japonicus), collected from fish farms located in the southern coastal area of Korea. The residues of fluoroquinolones were determined using high performance liquid chromatography (HPLC) with a fluorescence detector. Residuals of five fluoroquinolones in muscle tissue of farmed fish were analyzed. We found that enrofloxacin was the most common agent in fish muscle, and that ciprofloxacin was the next most common. The range of detected concentrations of fluoroquinolones in olive flounder muscle was 0-0.859 mg/kg in 32.6% of all samples. Enrofloxacin was commonly detected in sea bass muscle at a range of 0-0.143 mg/kg in 38.9% of all samples. Fluoroquinolones were detected in 6.9% of black rock fish muscle and in 16.6% of sea bream, although the detected concentration was below 0.01 mg/kg. The maximum detection value of enrofloxacin and ofloxacin in olive flounder at the time of shipping was 0.102 mg/kg and 0.09 mg/kg, respectively; no other antimicrobials were detected. We detected no antimicrobial substances in red sea bream.

To clarify the ichthyotoxic mechanisms of a harmful dinoflagellate Cochlodinium polykrikoides, hematological responses of the flounder Paralichthys olivaceus and red sea bream Pagrus major exposed to this algal bloom were investigated. The mortality of red sea bream was considerably larger than that of flounder, and the threshold lethal density of C. polykrikoides to the test fish was approximately 3,000 cells/ml. Blood $PO_2$declined in proportion to the increasing density of algal cells. The blood $PO_2$ of moribund fish was about $40-60\% of control test fish. Particularly, the fishes began to be killed when the blood $PO_2$ fell below 30-40 mmHg. However, the blood pH dropped almost 1.0 unit just before fish kill. Hemoglobin and hematocrit levels of fish exposed to C. polykrikoides of 5,000 cells/ml for 24 h and of moribund fish did not show great difference. The concentrations of plasma $Na^+$, $K^+$ and $Cl^-$ were slightly elevated to different magnitudes except $Ca^{2+}$ and plasma osmolality was also increased in Cochlodinium-exposed fish. In the plasma cortisol level, these values of moribund flounder and red sea bream were 4- 5 times higher than those of control fish. These results suggest that the drop of blood $PO_2$ was may be one of the principal causes of fish kill by C. polykrikoides, and the changes of other hematological parameters were secondary responses elicited by the decrease in blood $PO_2$.

Parallux (IDEXX Laboratories, USA), a solid-phase fluorescence immunoassay (SPFIA) developed for antibiotics residue detection in milk, was applied for analysis of antibiotics in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major). Fishes were dipped in streptomycin 20 g/ton water, the recommended therapeutic dose, for 3 consecutive days. Muscle was sampled after 1st, 2nd, 3rd, 4th and 5th day drug treatment. The concentration of streptomycin in muscle, determined by SPFIA, was compared with that of internal standard (200 ppb as streptomycin). The absorbance ratio of sample to internal standard (S/C) was employed as an index to determine the muscle residues in fishes. The standard solutions were added to muscle samples to give final concentrations in muscle of 0.2 and 0.5 mg/ml to investigate the recovery rate. The recovery rates of all spiked samples were > 84% of the spiked value. Streptomycin was detected in muscles of fishes treated after the 1st day of withdrawal period. The present study showed that the SPFIA can be easily adopted in predicting muscle tissue residues for streptomycin in farmed fishes.

The author carried out experiments to find out the response of rock bream, Oplegnathus fasciatus (TEMMINCK et SCHLEGEL) and sea bass, Epinephelus septemfasciatus (THUNBERG) to the color nettings. The experimental water tank(180L$\times$50W$\times$55Hcm) was set up n a dark room and water level was maintained 50cm high from the bottom. The tank was devided three longitudinal sections marking 60 cm interval. The illumination systems, consisted of 20 watt fluorescent lamps and filter, were suspended adove the tank. Two different color nettings selected from five colors (red, yellow, green, blue, black) were placed in each end section of the tank. Ten fish were used in each experiment and the fish were acclimatized in the dark for 60 minutes before experiment. After the light on, the number of fish in each section of the tank was counted in every 30 seconds interval for 30 minutes. The results obtained are as follows: 1. The rock bream selected the color nettings in the order of yellow, black, blue, green and red. 2. The sea bass selected the color nettings in the order of green, black, red, blue and yellow.

This study evaluates the pathogenicity in striped beakperch, Oplegnathus fasciatus infected with red sea bream iridovirus (RSIV) at different water temperature (17°C, 20°C, 25°C and 27°C). When the fish group was infected with RSIV at 17°C and 20°C, cumulative mortality did not show any significant difference with control group. In contrast, the case at 25°C and 27°C, cumulative mortality reached more than 80%. However, RSIV was detected from all of the fish in each temperature. To confirm a relationship between temperature change and heat shock protein (HSP), partial HSP70 cDNA was isolated from striped beakperch.

In 2009, a systemic megalocytivirus infection associated with high mortality was detected for the first time in cultured starry flounder Platichthys stellatus in Korea. Diseased starry flounder had pale bodies and gill coloring and enlarged spleens. Histopathological examinations revealed basophilic enlarged cells in various organs of diseased starry flounder. Polymerase chain reaction (PCR) was performed on tissue samples using three published primer sets developed for the red sea bream iridovirus. PCR products were detected for all primer sets, except 1-F/1-R, which are registered by the World Organization for Animal Health (OIE). The part of the gene corresponding to the full open reading frame encoding the viral major capsid protein (MCP) was amplified by PCR. PCR products of approximately 1,581 bp were cloned, and the nucleotide sequences were analyzed phylogenetically. The MCP gene of the starry flounder iridovirus, designated SFIV0909, was identical to that of the turbot reddish body iridovirus (AB166788).
