Cultivation of Chlorella Sp. Using Light Emitting Diode (발광다이오드를 이용한 클로렐라 배양 연구)

  • The purpose of this study was to determine optimum conditions for the cultivation of Chlorella sp. FC-21 using light emitting diodes (LEDs). Specific growth rate and cell concentration were measured for the reactors at the illuminations of different wavelengths of LEDs. Among various types of LEDs, red LEDs were the most effective light source, and also greatest increases of specific growth rate and cell concentrations were obtained when light intensity of red LEDs increased. The specific growth rate decreased when initial cell concentration increased due to the shading effect of each cell in the reactor. To determine beneficial effect of aeration to cell cultivation, micro-air bubbles were aerated at 0.35 vvm in the reactor at the illumination of red LEDs. Two and ten times greater specific growth rate and cell concentration were obtained when aeration was applied. From this study, we found that red LEDs with aeration were the most appropriate light source for the cultivation of Chlorella sp. FC-21.

A Study on the Lighting and the Photosynthetic Photon Flux Density with LED for Light Reinforcement (보광용 LED의 광특성과 광자속밀도 특성)

  • This study determined the characteristics of LED illumination and photosynthetically active radiation flux density (PPFD) for LED lighting design in an indoor plant factory. This was done based on the light wavelength and PPFD intensity required for plant growth. It has been found that the wavelength and intensity are decreased according to the measuring distance, and green light has an important role in illumination characteristics, while blue light has an important role in the PPFD characteristics. Considering only the photosynthetic properties of plants, the effective order of photosynthesis was blue>red>white>green. When the measurement distance was 30 [cm], it was found that reduction levels of 60 [cm], 90 [cm], and 120 [cm] decreased to about 36 [%], 18 [%], and 10 [%], respectively. As a result of the characteristics of mixed light (red:blue=2:1, 3:1, 4:1) and the measurement distance, when the measured value at 30 [cm] is 100%, the measured value at 120 [cm] is 10-11 [%]. From the obtained results, an optimal structure was proposed for maximizing the light efficiency of an indoor greenhouse for future research.

Proteomic Analysis of Protein Expression Patterns Associated with Astaxanthin Accumulation by Green Alga Haematococcus pluvialis (Chlorophyceae) Under High Light Stress

  • Two kinds of Haematococcus pluvialis cells (green vegetative cells cultivated under optimal cell culture conditions and red cyst cells maintained under high light stress conditions to induce astaxanthin production) were used to investigate the protein expression profiles by two-dimensional electrophoresis, image analysis, and peptide mass fingerprinting. The cellular accumulation of astaxanthin was evident after exposure to high light intensity and reached the maximum cellular level after 78 h of high light stress. In a 2-D electrophoresis analysis, 22 proteins were upregulated over 2-fold in the red cyst cells when compared with the green vegetative cells and selected for further analysis by chemically assisted fragmentation (CAF)-MALDI-TOF sequencing to identify the protein functions. Among 22 different spots, several key enzymes specific to the carotenoid pathway, including isopentenyl pyrophosphate isomerase (IPP) and lycopene $\beta$-cyclase, appeared in H. pluvialis after exposure to high light intensity. Therefore, IPP and lycopene $\beta$-cyclase would appear to be involved with carotenoid accumulation in the cytoplasm, as these peptides were preferentially upregulated by high light intensity preceding an increase in carotenoid, and only these forms were detected in the red cyst cells.

  • In order to establish the basic data for the growth of fish in the aquaculture industry, the behavior analysis of rockfish (Sebastes inermis) depending on the temperature and LED lights was conducted. In this study, water temperatures were set from $3^{\circ}C$ to $30^{\circ}C$ were used. One red light (wave length: 622 nm; light power: 811 mW) and one green lights (wave length: 518 nm; light power: 648 mW) were used. Behavior of the rockfish was expressed as average moving distance (AMD) for 1 minutes and a rate of movement. The mean AMD depending on the temperature was 1.0 m and the mean rates of movement was 50%. The mean AMD were 1.5 m, 1.9 m and 0.7 m in the red LED light, green LED light and control condition respectively. The mean rates of movement were 54%, 65% and 45% in the red LED light, green LED light and control condition respectively.

Response of Growth and Development of Young Tomato Plants to End-of-day Monochromatic Light from Various LEDs

  • Plant growth and development strongly influenced by light quantity and its spectral composition. Young tomato plants were cultivated in growth cabinets under artificial light provided by red and blue light emitting diodes(LEDs) during 12 hours, then plants were exposed to monochromatic ultraviolet, blue, green and red lights as an end-of-day(EOD) treatment during 4 hours to study their effect on plant growth parameters. EOD lighting from various LEDs increased total fresh and dry weights as well as assimilation area compared to those in control. Blue light increased stem height, internode length and stem diameter. Monochromatic UV-A light reduced stem elongation, highly increased stomatal conductance. Compactness and health index of young tomato plants were increased in UV-A and red light treatments.

The Effect of Light on the Production of Reserpine in Cultured Rauwolfia serpentina Cells

  • When reserpine-producing cell strains of Rauwolfia serpentina were transferred from the dark to the light irradiation, the production of reserpine was extremely enhanced whereas the cell growth was suppressed. In an incubation period of 20 days, the most effective culture condition for reserpine production was the combination of 8 days of dark culture and following 12 days of light culture. The time courses of both cell growth and reserpine production were measured in vitro in order to clarify the effect of wave length range of light on the biosynthesis of reserpine. Although the growth of cultured cells which had been incubated under continuous red, yellow, and green lights, respectively, was similar to that of the cultured cells subcultured in the dark. The cells cultured under red light irradiation produced less reserpine than dark-grown cultures. Both blue and near-ultraviolet light inhibited the growth of cultured cells. The production of reserpine was strikingly enhanced by blue light, but was strongly inhibited by near-ultraviolet light.

A Study on The Photosynthesis Accelerate by Light Color Composition in Plant Factory (식물공장 광원의 색조합에 따른 광합성활성화에 관한 연구)

  • This study examined the criteria for efficient LEDs used throughout the experiment of an LED with another light color growth to be used in a plant factory. The experiment was confirmed by measuring the Red-LED, Blue-LED, plant growth, and amount of carbon reduction in a White-LED environment. The white-LED showed a similar growth trend to the Red-LED. Blue-LED showed the lowest growth. Measurements of the carbon dioxide levels, showed that the Red-LED and blue LED produced the lowest levels. The combination of the ratio of the LED showed four Red-LEDs and one blue LED to be the higher of the two. In addition, three Red-LED and one Blue-LED produced equal growth to that of the white-LED. In addition, as much as possible, red is the light color that obtains the result suitable for plant factories.

Effect of LED Light on Primordium Formation, Morphological Properties, Ergosterol Content and Antioxidant Activity of Fruit Body in Pleurotus eryngii (LED광원이 큰느타리버섯 자실체의 발생, 생육, 에르고스테롤 함량 및 항산화활성에 미치는 영향)

  • Light wavelength is the major factor of fruit body development associated with mushroom cultivation, but its wavelength range in Pleurotus eryngii is poorly understood. Using four kinds of light emitting diode (LED) including blue (475 nm), green (525 nm), yellowed (590 nm) and red (660 nm), we investigated to elucidate suitable light wavelength during primordium formation and fruit body development of P. eryngii on bottle cultivation. Primordia formation did not occur in blue light and red light. The morphological properties of fruit body in fluorescent lamp and blue light irradiation were showed thicker and larger pileus than those in other LEDs. However, length of stipe in fluorescent lamp and blue light was shorter than that of other LEDs. The DPPH radical was high in blue light, green light, and yellow light except for red light, and the polyphenol was high in four kinds of LED sources. And ergosterol was the highest in the green light. Thus, the high-quality mushroom production of P. eryngii is possible to green light condition considering productivity and functional materials.

Measurement of red blood cell aggregation by analysis of light transmission in a pressure-driven slit flow system

  • The aggregation characteristics of red blood cells (RBCs) were measured using a newly developed light-transmission slit rheometer. Conventional methods of RBC disaggregation such as the rotational Couette system were replaced with a pressure-driven slit flow system with a vibrational mechanism. Using a vibration generator, one can disaggregate the RBC aggregates stored in the slit. While shear stress decreases exponentially, instantaneous pressure and the transmitted light intensity were measured over time. Applying an abrupt shearing flow after disaggregation caused a rapid elongation of the RBCs followed by loss of elongation with the decreasing shear stress. While the shear stress is further decreasing, the RBCs start to re-aggregate and the corresponding transmitted intensity increases with time, from which the aggregation indices can be obtained using a curve-fitting program.
