Trial for Drug Susceptibility Testing of Mycobacterium tuberculosis with Live and Dead Cell Differentiation (세포 염색 방법을 이용한 결핵균 감수성 검사법)

  • Background : The resurgence of tuberculosis and outbreaks of multidrug resistant (MDR) tuberculosis have increased the emphasis for the development of new susceptibility testing of the Mycobacterium tuberculosis for the effective treatment and control of the disease. Conventional drug susceptibility testings, such as those using egg-based or agar-based media have some limits, such as the time required and difficulties in determining critical inhibitory concentrations, but these are still being used in many diagnostic laboratories because of no better lternatives, considering cost and accuracy. To overcome these limits, a rapid and simple method for new susceptibility testing, using live and dead assays, was applied for a bacterial cell viability assay to distinguish dead from live bacterial cells based on two-color fluorescence. Materials and Methods Strains : Forty strains were used in this study, 20 susceptible to all antituberculosis drugs and the other 20 resistant to the four first line antituberculosis drugs isoniazid, rifampicin, streptomycin and ethambutol. Antibiotics : The four antibiotics were dissolved in 7H9 broth to make the following solutions: $0.1{\mu}g\;isoniazid(INH)/m{\ell}$, $0.4{\mu}g\;rifampicin(RMP)/m{\ell}$, $4.0{\mu}g\;streptomycin(SM)/m{\ell}$ and $4.0{\mu}g\;ethambutol(EMB)/m{\ell}$. Results : Live and dead Mycobacterium tuberculosis cells fluoresced green and red with the acridin (Syto 9) and propidium treatments, respectively. These results are very well accorded with conventional drug susceptibility testing by proportional method on Lowensen-Jensen media (L-J) containing 4 drugs (INH, RMP, EMB and SM), showing a 93.7 % accordance rate in susceptible strains and 95% in resistant strains. Conclusion : The results of the drug susceptibility testing using the live and dead bacterial cell assay showed high accordance rates compared with the conventional proportion method on L-J. This finding suggests that the live and dead bacterial cell assay can be used as an alternative to conventional drug susceptibility testing for M. tuberculosis strains.

Morphological Characteristics and Classification Criteria for Azalea Cultivars for Landscaping in Korea (조경용 철쭉재배품종의 형태적 특성 및 분류기준)

  • This study was conducted to examine the morphological characteristics of those Azalea Cultivars(hereinafter, Azalea Cultivars) that are mainly used for landscaping in Korea in order to prepare classification criteria. As testing materials, major Azalea Cultivars cultivated in large quantities by its producing companies were collected. Thereafter, the qualitative and quantitative characteristics of the traits of Azalea Cultivars were investigated in the way of characteristic investigation for new cultivars of azalea used by the Korea Seed and Variety Service in order to classify them and prepare the classification criteria. Since cultivar names have not been established for Azalea Cultivars for landscaping thus far, the data were compiled using the names used by cultivating companies. According to the results, Azalea Cultivars cultivated in Suncheon, Jeonnam mainly for landscaping were 10 in number; Beni, Daewang, Three, Zasanhong, Hancheol, Sancheoljuk, Gyeobsancheoljuk, Baekcheoljuk, Akado, and Seok-am. Among them, the cultivars Beni, Daewang, and Three could not be easily distinguished from each other because they are commonly called Yeongsanhong by cultivating companies and the shapes of their leaves and flowers are similar to each other. In particular, the flower color of Beni was 'bright red', that of Daewang was 'vivid purple', and that of Three was 'bright purple'. In addition, Zasanhong and Hancheol were similar to each other in shape the degree of expression of spots on the flowers and the gloss on the front side of the leaves of Hancheol were higher and stronger compared to that of Zasanhong. Sancheoljuk flowered in early April, earlier compared to other Azalea Cultivars. Gyeobsancheoljuk is an elementary species of Sancheoljuk and it had double flowers although all other traits were similar to those of Sancheoljuk. Although Baekcheoljuk was easily distinguished because it had white flowers, its leaves were similar to those of Akado the reason why these two cultivars could not be easily distinguished from each other. The cultivar Akado flowered early May later compared to other Azalea Cultivars and its flowers were relatively large in diameter as with Baekcheoljuk and Sancheoljuk. Finally, the cultivar Seok-am was easily distinguished because it had smaller leaves compared to other cultivars and it flowered late as with the cultivar Akado.

Quality Characteristics and Antioxidant Activity of Mulberry Leaf Tea Fermented by Monascus pilosus (Monascus pilosus로 발효시킨 뽕잎차의 품질특성과 항산화능)

  • This study was carried out to investigate the contents of monacolin K and citrinin, along with the sensory quality and antioxidant activity of mulberry leaf tea fermented by $Monascus$ $pilsous$ (FMM). Total monacolin K content of FMM was 0.058%, but citrinin was not detected. Redness of brewed FMM was remarkably higher than that of unfermented mulberry leaf tea (UFM). In sensory evaluation of brewed FMM, while astringent taste and savory taste were lower, flavor, color, and overall acceptability were significantly higher than those of UFM. Total polyphenol contents of UFM and FMM were 83.1 and 23.61 mg/g (dry basis), total flavonoid contents of UFM and FMM were 17.96 and 3.99 mg/g (dry basis), respectively. Xanthine oxidase (XO) inhibitory activity and superoxide dismutase (SOD)-like activity of FMM were lower than those of UFM. Electron-donating ability and ferric-reducing antioxidant power of FMM were slightly lower than those of UFM. However, the antioxidant activities of FMM per polyphenol content were markedly higher than those of UFM. These results suggest that FMM may scavenge excessive reactive oxygen spices (ROS) via inhibition of XO and SOD-like activity. Furthermore, FMM demonstrated relatively higher acceptability and antioxidant ability along with functionality of $Hongguk$ (red yeast rice), and therefore could be utilized to prevent various ROS-induced diseases.

Physico-chemical Changes of Pork Prepared by Korean Traditional Sauces During Chilled Aging (한국 전통 양념으로 제조한 돼지고기의 숙성중 물리화학적 변화)

  • This study was conducted to investigate physico-chemical changes of pork prepared by Korean traditional 3 types seasoning such as soybean sauce(Tl); garlic sauce(T2); red pepper sauce(T3). The seasoned samples were aged at 0 ${\pm}$ $1^{\circ}C$ for 30 days. The results obtained were as follows; Salinity(%) and saccharinity(%) of sauce were significantly decreased(P<0.05) among the all treatments during aging periods, whereas fermented meats were significantly increased(P<0.05). pH of sauce and fermented meat were tended to increased as aging period. Water-Holding Capacity(WHC) and shear force of fermented meat increased(P<0.05) among the all treatments at 20 days of storage, and decreased after that period. In color, $L^* and b^* value of T3 were lower, while a^*$ value was higher than other treatments during aging period. Volatile Basic Nitrogen(YBN) value of all treatments were increased(P<0.05) after 20 days of storage. YBN value of T3 was significantly higher(P<0.05) compared with other treatments. Thiobarbituric Acid Reactive Substances(TBARS) was rapidly increased until 10 days of storage and somewhat decreased up to 20 days after 10 days in all treatments, it was re-increased after 20th days. T3 showed the highest TBARS among the all treatment groups. Total bacterial and Lactobacilli spp. were significantly increased(P<0.05) in all treatments at 10 days of storage and decreased after that period, however it tended to increased at 30 days of storage. In sensory evaluation, overall acceptability in all treatments were increased after 20 days of storage compared to initial storage and it score was similar thereafter.

Effects of Bacillus subtilis Supplementation on Egg Quality, Blood Characteristics and Fecal $NH_3-N$ in Laying Hens (산란계에 Bacillus subtilis의 급여가 계란 품질, 혈액 성상 및 분내 암모니아태 질소 함량에 미치는 영향)

  • This study was conducted to investigate the effects of Bacillus subtilis on the egg qualify, blood characteristics and fecal $NH_3-N$ in laying hens. A total of two hundred fifty two laying hens were randomly allocated into three treatments with seven replications for six weeks. Dietary treatments included 1) CON (control; basal diet), 2) BS0.2(control + $0.2\%$ Bacillus subtilis) and 3) BS0.4 (control +$0.4\%$ Bacillus subtilis). For overall Period, hen-day egg production tended to increase by the Bacillus subtilis $0.4\%$ in the diets, but was no significant difference. Egg weight, egg shell breaking strength, egg shell thickness, Haugh Unit, yolk color unit and egg yolk index were not affected by treatments. Difference of egg weight and egg shell breaking strength in the BS0.2 treatment tended to increase without significant difference. Difference of egg yolk index in laying hens fed Bacillus subtilis was increased (P<0.05). The concentrations of red blood cell (RBC) and white blood cell (WBC) were not significant difference. $NH_3-N$ concentration in feces with BS0.4 treatment was significantly (P<0.05) lower than control. In conclusion, dietary Bacillus subtilis could decrease fecal $NH_3-N$.

Quality Characteristics of Bread Added with Prickly Pear(Opuntia ficus-indica) Powder (손바닥선인장 열매 분말 첨가 식빵의 품질 특성)

  • The quality characteristics of bread added with the levels of $0\%,\;1.0\%,\;2.0\%\;3.0\%\;and\;4.0\%$ prickly pear powder were investigated. Mixing water absorption capacity was increased by increasing amounts of prickly pear powder. Mixing time of dough was increased compared to that of wheat flour by added prickly pear powder, while it decreased as prickly pear powder content increased. Dough stability of wheat flour added with $1.0\%$ prickly pear powder was similar to wheat flour dough, while decreased by increasing amount of prickly pear powder at the range of $2.0\~4.0\%$. Pasting temperature($66.1\~66.9^{\circ}C$) of wheat flour added with $1.0\%,\;2.0\%\;3.0\%\;and\;4.0\%$ prickly pear powder was lower($67.8^{\circ}C$) than that of wheat flour. Peak viscosity($2,226\~2,375$ cp) of wheat flour added with $1.0\%,\;2.0\%\;3.0\%\;and\;4.0\%$ prickly pear powder were higher(2,288 cp) than control, and increased with increasing amount of prickly pear powder Setback($797\~750$ cp) of wheat flour added with $1.0\%,\;2.0\%\;3.0\%\;and\;4.0\%$ prickly pear powder were lower(803 cp) than that of wheat flour, and decreased with increasing amount of prickly pear powder. The volume of dough added with prickly pear powder decreased compared to that of the wheat dough. The specific loaf volume of bread added with prickly pear powder decreased($8.0\~18.5\%$) compared to that of the wheat bread. Color of crumb was deep red gradually with the increasing amount of substituted prickly pear powder. Hardness, adhesiveness, gumminess and chewiness of bread were increased gradually with the increasing amount of substituted prickly pear powder but the springiness was decreased. The sensory evaluation showed that bread of wheat flour added with $2.0\%$ prickly pew powder was superiority than the wheat flour bread.

Breeding of Oriental Lily 'Pacific Wave' with Upward-facing and White Petals (상향 개화형 백색 오리엔탈나리 'Pacific Wave' 육성)

  • An Oriental lily cultivar 'Pacific Wave' was released in 2007 at National Institute of Horticultural and Herbal Science, Rural Development Administration (RDA), Suwon, Korea. The crossing was made in 1999 between Oriental lily 'Simplon', an outward-facing and white colored cultivar, and 'Spinoza', pink colored cultivar. The first selection was done in 2003 with a line of 'O-03-16'. Multiplication and bulb growth, and performance test was conducted from 2004 to 2006. This selection was later on given as 'Pacific Wave' in 2007 at NHRI. Flowering time of 'Pacific Wave' in plastics house culture is mid June and grows average 115 cm. Flowers are upward-facing with 20.1 cm in diameter and white with yellow centered (RHS W155C + Y9A). Mean petal length and width is 12.2 cm and 4.2 cm, respectively. Leaves are 12.3 cm long, 2.9 cm wide. The throat color is green. It shows gray and purple stigma, and red brown pollen. The weight and size of bulb is 82.5 g and 19.6 cm, respectively. Year-round flowering can be by storing the bulb under -1 to $-2^{\circ}C$ conditions. It is necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. It is needed to control Botrytis disease in summer wet season.

Characteristics of the Strains Selected from Crosses between Introduced Interspecific Hybrids and Cultivars in Hibiscus Species (종간교잡 유래 도입 무궁화와 국내 선발 품종과의 교잡에 의해 육성된 계통들의 특성)

  • This study was carried out to develop new cultivars of Hibiscus species from crosses between introduced interspecific hybrids and cultivars in Hibiscus species. Fruit setting of interspecific crosses of Hibiscus strains was less than 10% and the number of seeds in the fruit was also in low level. Three individuals of specific flower and leaf characteristics were selected from crosses between introduced interspecific hybrid, 'Fujimusme'(♀), and H. syriacus 'Namwon'(♂) in 2004. A new strain, Hibiscus ${\times}$ 'W-26', was selected from the crossing of interspecific hybrid, 'Fujimusme'(♀), and H. syriacus 'Namwon'(♂), which had white flower and narrow separated petal. Hibiscus ${\times}$ 'WRB-2' was selected from the crossing of interspecific hybrid, 'Fujimusme'(♀), and H. syriacus 'Namwon'(♂), which had white flower and blue eye spot. Hibiscus ${\times}$ 'R-141' was selected from crosses between introduced interspecific hybrid, 'Shichisai'(♀) and H. syriacus 'Namwon'(♂), which had large flowers over 13 cm diameter and revealed tall tree type. Hibiscus ${\times}$ 'R-142' was selected from the crossing of interspecific hybrid, 'Shichisai'(♀), and H. syriacus 'Namwon'(♂), which had large flowers over 13 cm diameter and revealed tall tree type. The characteristics were succeded after grafting. Flower of 'R-142' had reddish violet color with red eye spot, whereas its parent had blue and purple flowers.

Effect of Halophilic Bacterium, Haloarcula vallismortis, Extract on UV-induced Skin Change (호염 미생물(Haloarcula vallismortis) 용해물의 자외선유발 피부변화에 대한 효과)

  • Skin carrys out protective role against harmful outer environment assaults including ultraviolet radiation, heavy metals and oxides. Especially, ultraviolet-B (UVB) light causes inflammatory reactions in skin such as sun burn and erythma and stimulates melanin pigmentation. Furthermore, the influx of UVB into skin cells causes DNA damage in keratinocytes and dermal fibroblasts, inhibition of extracellular matrix (ECM) synthesis which leads to a decrease in elasticity of skin and wrinkle formation. It also damages dermal connective tissue and disrupts the skin barrier function. Prolonged exposure of human skin to UVB light is well known to trigger severe skin lesions such as cell death and carcinogenesis. Haloarcula vallismortis is a halophilic microorganism isolated from the Dead Sea, Its growth characteristics have not been studied in detail yet. It generally grows at salinity more than 10%, but the actual growth salinity usually ranges between 20 to 25%. Because H. vallismortis is found mainly in saltern or salt lakes, there could exist defense mechanisms against strong sunlight. One of them is generation of additional ATP using halorhodopsin which absorbs photons and produces energy by potential difference formed by opening the chloride ion channel. It often shows a color of pink or red because of their high content of carotenoid pigments and it is considered to act as a defense mechanism against intense UV irradiation. In this study, the anti-inflammatory effect of the halophilic microorganism, H. vallismortis, extract was investigated. It was found that H. vallismortis extract had protective effect on DNA damage induced by UV irradiation. These results suggest that the extract of halophilic bacterium, H. vallismortis could be used as a bio-sunscreen or natural sunscreen which ameliorate the harmful effects of UV light with its anti-inflammatory and DNA protective properties.

A New Sweetpotato Cultivar for Use of Bioethanol 'Daeyumi' (바이오에탄올용 고구마 신품종 '대유미')

  • A new sweetpotato variety, 'Daeyumi', was developed by Bioenergy Crop Research Center, National Institute of Crop Science (NICS), RDA in 2008. This variety was obtained from the cross between 'Jinhongmi' and 'Xusju 18' in 2000. The seedling and line selections were performed from 2001 to 2003, preliminary and advanced yield trials were carried out from 2004 to 2005, and the regional yield trials were conducted at six locations from 2006 to 2008. 'Daeyumi' has cordate leaf, green vine and petiole, elliptic storage root, red skin and yellow flesh color of storage root. This variety is also resistant to Fusarium wilt and nematode. The starch value was 25.9%, ethanol yield was 418 L/Ton, which was 7% higher than that of 'Yulmi' variety, and the total sugar content was 2.47 g/100g, dry weight. 'Daeyumi's initial temperature of starch gelatinization was lower, 76.2$^{\circ}C$, and the retrogradation process was earlier than 'Yulmi'. The average yield of storage root was 27.8 ton/ha in the regional yield trials, which was 36% higher than that of 'Yulmi' variety. Number of storage roots over 50 gram per plant was 3.0, and the average weight of one storage root was 152 gram. This variety can be used for the production of bioethanol and starch processing.
