Comparison of the antioxidant properties and flavonols in various parts of Korean red onions by multivariate data analysis

  • To compare the antioxidant properties and flavonols in various parts; dry skin (DS) and edible portion (EP), of 8 red onions (Allium cepa L, ROs), total content of phenolics (TPC), flavonoids (TFC), and anthocyanins (TAC) and DPPH radical scavenging properties were estimated and the content of six flavonols were quantified by HPLC-PDA analysis. The major component of DS and EP of RO was quercetin and quercetin-4'-glucoside, respectively. Score plots of the PCA and PLS-DA were segregated by flavonols content and antioxidant properties according to the EP and DS of ROs. Loading plot of the PCA showed that the quercetin and sum of flavonol content were highly correlated with antioxidant activity of ROs. Therefore, flavonol content and antioxidant activity can be used as markers for distinct parts of ROs.

Bulb Storability of Red and Yellow Onion (Allium cepa L.) Cultivars Grown in Korea

  • The bulb storability of eight red and ten yellow onion (Allium cepa L.) cultivars grown primarily in Korea was evaluated. During storage, sprouting occurred earlier in red cultivars than in yellow cultivars. In addition, the ratio of sprouted to unsprouted bulbs increased more rapidly in red cultivars than in yellow cultivars. However, not all yellow cultivars had strong storabilities. Bulb storability of three yellow cultivars was as poor as that of red cultivars, suggesting only a slight possibility of a pleiotropic effect of color and bulb storability. Meanwhile, $F_3$ lines of a red cultivar selected based on stronger storabilities showed intermediate storabilities between those of the red and yellow cultivars, implying that strong storability could be obtained by successive selection. In contrast, $F_1$ hybrids from crosses between yellow and red breeding lines showed poor storability compared with the yellow cultivars, indicating that poor storability may be dominant over strong storability. The relationships between color and other traits that potentially affect storability, such as fresh and dry weight, water content, and firmness of bulbs, were evaluated. No significant differences in these traits were observed between the two colors, although the water content of yellow cultivars and the dry weight of red cultivars were slightly higher than their counterparts. Correlation analyses between bulb storability and other traits, including weight, water content, and firmness, also showed no significant correlations. In this study, no correlations were identified between bulb color, storability, and other traits. However, based on these results, red cultivars with stronger storabilities could be developed through successive selection of bulbs with stronger storabilities.

Development of Functional Markers for Detection of Inactive DFR-A Alleles Responsible for Failure of Anthocyanin Production in Onions (Allium cepa L.)

  • Inactivation of the gene coding for dihydroflavonol 4-reductase (DFR) is responsible for the color difference between red and yellow onions (Allium cepa L.). Two inactive DFR-A alleles, DFR-$A^{PS}$ and DFR-$A^{DEL}$, were identified in our previous study. A functional marker was developed on the basis of the premature stop codon that inactivated the DFR-$A^{PS}$ allele. A derived cleaved amplified polymorphic sequences (dCAPS) primer was designed to detect the single nucleotide polymorphism, an A/T transition, which produced the premature stop codon. Digested PCR products clearly distinguished the homozygous and heterozygous red $F_2$ individuals. Meanwhile, to develop a molecular marker for detection of the DFR-$A^{DEL}$ allele in which entire DFR-A gene was deleted, genome walking was performed and approximately 3 kb 5' and 3' flanking sequences of the DFR-$A^R$ coding region were obtained. PCR amplification using multiple primers binding to the extended flanking regions showed that more of the extended region of the DFR-A gene was deleted in the DFR-$A^{DEL}$ allele. A dominant simple PCR marker was developed to identify the DFR-$A^{DEL}$ allele using the dissimilar 3' flanking sequences of the DFR-A gene and homologous DFR-B pseudogene. Distribution of the DFR-$A^{PS}$ and DFR-$A^{DEL}$ alleles in yellow onion cultivars bred in Korea and Japan was surveyed using molecular makers developed in this study. Results showed predominant existence of the DFR-$A^{PS}$ allele in yellow onion cultivars.

국내 육종 양파 품종별 주요성분의 함량 및 항산화활성 비교 (Major constituents and antioxidant activities of domestic onion (Allium cepa L.) cultivars)

  • 본 연구에서는 국내 육종 양파[황색(맵시황, 신선황, 연신황, 영보황, 영풍황), 적색(엄지나라), 연녹색(스위트그린)]와 외국 육종 양파(황색, 선파워; 적색, 천주적) 간의 일반형질, 주요성분 및 항산화능을 비교하였다. 그 결과, 각 페놀성 화합물과 flavonoid의 성분별 총 함량 및 총 페놀성 화합물과 총 flavonoid 함량에 있어 외래품종인 썬파워를 포함한 6종의 황색양파들 중 영보황이 가장 우수한 값을 보였으며, 적색품종에 있어서는 천주적이 엄지나라보다 약간 높은 함량을 보였다. 총 anthocyan의 함량은 엄지나라가 천주적보다 낮았다. 반면 thiosulfate의 함량은 모든 품종들에 있어 유의차를 보이지 않았으며, pyruvic acid 함량은 황색양파들 중 영보황과 썬파워가 동일한 정도로 가장 높았고, 적색품종에 있어서는 엄지나라가 천주적보다 더 높은 함량을 보였다. 그러나 총 당함량에 있어서는 영신황이 썬파워를 제외한 모든 품종들 중 가장 높은 값을 보였다. 그리고 항산화능은 모든 평가항목에 있어 공통적으로 적색품종이 가장 우수하였으며, 환원력에 있어서는 천주적이 엄지나라보다 우수하였으나 DPPH radical scavenging 활성은 천주적보다 엄지나라가 다소 높은 경향을 보였고, ABTS+ radical scavenging 활성은 두 품종들 간에 유의차를 보이지 않았다. 황색품종들에 있어서는 신선황과 영보황이 우수한 DPPH radical scavenging 활성을 보였으며, 환원력은 국내산 품종들이 썬파워보다 약간 높은 경향을 보였고, ABTS+ radical scavenging 활성은 영신황이 썬파워보다 높았다. 이상의 결과로부터 국내 육성 품종인 영보황과 엄지나라의 성분학적 및 기능학적 우수성이 부분적으로 제시되어진 바, 본 연구결과가 국산 양파 품종의 우수성 홍보를 위한 유용 자료로 활용되기를 기대한다.

양파의 발아 및 입묘 향상을 위한 종자처리의 효과 (Effects of Presowing Seed Treatments on Improvement of Seed Germination and Seedling Emergence of Onion)

  • 양파의 생력재배를 위한 플러그 육묘시 성묘율 향상에 필요한 정보를 제공하고자 농우대고와 창녕 대고를 공시 품종으로 종자의 선종, priming, GA$_3$, 저온, 건조, 건조 중 광질의 개 별 처리가 발아율에 미치는 영향과 최적 개별처리를 조합한 종자처리가 유묘출현율에 미치는 영향을 추적하고자 본 연구를 실시하였던 결과 양파 종자에 처리를 가하기 전 선종이 발아율을 향상시키는 것으로 조사되었으며, 양파 종자의 발아율에 대한 priming과 GA$_3$처리 효과는 아주 미미하였던 반면, 저온처리 효과는 아주 큰 것으로 나타났다. 이러한 저온처리는 품종간 약간의 차이를 보일지라도 3일간 처리할 경우 발아율이 가장 양호하였다. 처리 종자의 건조방법으로는 건조하지 않는 것보다는 건조하는 것이, 건조할 경우 적색광을 조사하는 것이 발아율이 높았다. 유묘출현율은 선종된 종자를 저온처리를 가하지 않고 건조하는 것보다는 3-5일간 저온처리를 가한 후 적색광을 조사하면서 건조할 경우 높은 것으로 조사되었다.

양파식초가 뇌혈류 및 안전성에 미치는 영향 (Effects of Onion Vinegar on the Cerebral Blood Flow and the Safety Examination)

  • The aim of this study is to investigate the effects of onion vinegar on the cerebral blood flow by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) and by observing the recovery of focal ischemic brain injury in rats. Rats are divided into thee groups depending on the medication; control group (no medication), 8.8-OV group (vinegar using 8.8 brix onion medication), 14.6-OV group (vinegar using 14.6 brix onion medication). The medication of onion vinegar significantly increased rCBF but decreased MABP. This result suggests that onion vinegar significantly increased rCBF by dilating arterial diameter. In addition, focal ischemic brain injury is induced in rats by middle cerebral arterial occlusion. The recovery from focal ischemic brain injury is more significantly improved in the groups using onion vinegar compared to the control group. The amount of recovery is measured by the GAP-43 and the medication of onion vinegar significantly increased GAP-43. This result suggests that onion vinegar is effective on the nerve regeneration. After the medication, the change of body weight, outcomes of renal and liver function test, and outcomes of CBC are analysed for safety examination. There are no statistical differences among control group and all experimental groups in the body weight, renal and liver function test, and CBC. In conclusion, these results suggest that onion vinegar can increase rCBF in normal state, and improve the stability of rCBF in ischemic state.

무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구 (Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. 품종, 지역에 따른 적색양파 화학성분 (Chemical Component of Red Onion (Allium cepa L.) according to Cultivars and Growing Areas)

  • 육종효율 증진 및 소비확대를 위해 적색양파의 화학성분을 분석한 결과 S&P5294가 가장 높은 당 함량 및 감미도를 나타냈고 선발조합 모두 레드프라임, 썬파워에 비해 당 함량 및 감미도가 높았으며, pyruvic acid는 레드프라임=S&P5295> S&P5294=S&P7273>S&P5296=S&P7276>레드썬=썬파워 순으로 함량이 낮았다. 총 페놀과 총 플라보노이드 함량 또한 선발조합 대부분이 레드프라임, 썬파워에 비해서는 함량이 높고 레드썬에 비해서는 낮은 함량을 보였다. Rutin은 모든 품종에서 가장 높은 함량(136.9-$169.9{\mu}g{\cdot}g^{-1}$ F.W.)을 보였으며, quecetin과 myricetin의 함량은 각각 62.5-135.7, 11.2-$16.1{\mu}g{\cdot}g^{-1}$ 보였고 kaempferol은 검출되지 않았다. Quercetin과 rutin은 레드썬에서, myricetin은 S&P5296에서 유의적으로 가장 높은 함량을 보였고 선발조합 모두 레드프라임, 썬파워에 비해 rutin, quercetin 및 myricetin의 함량이 높았다. 지역별로는 총페놀, 총플라보노이드 함량은 함평지역이 가장 높은 반면 ruin, myricetin 및 quercetin의 함량은 창녕지역이 가장 높은 함량을 보였다. 안토시아닌의 경우는 레드썬에 비해서는 다소 함량이 낮으나 레드프라임에 비해서는 선발조합 모두 높은 함량을 나타내었다.
