Antioxidant effect of garlic and aged black garlic in animal model of type 2 diabetes mellitus

  • Hyperglycemia in the diabetic state increases oxidative stress and antioxidant therapy can be strongly correlated with decreased risks for diabetic complications. The purpose of this study is to determine antioxidant effect of garlic and aged black garlic in animal model of type 2 diabetes. The antioxidant activity of garlic and aged black garlic was measured as the activity in scavenging free radicals by the trolox equivalent antioxidant capacity (TEAC) assay. Three week-old db/db mice were fed AIN-93G diet or diet containing 5% freeze-dried garlic or aged black garlic for 7 weeks after 1 week of adaptation. Hepatic levels of lipid peroxides and activities of antioxidant enzymes were measured. TEAC values of garlic and aged black garlic were $13.3{\pm}0.5$ and $59.2{\pm}0.8{\mu}mol/g$ wet weight, respectively. Consumption of aged black garlic significantly decreased hepatic thiobarbituric acid reactive substances (TBARS) level compared with the garlic group which showed lower TBARS level than control group (p<0.05). Activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) of garlic and aged black garlic group were significantly elevated compared to the control group. Catalase (CAT) activity of aged black garlic group was increased compared with the control group. These results show that aged black garlic exerts stronger antioxidant activity than garlic in vitro and in vivo, suggesting garlic and aged black garlic, to a greater extent, could be useful in preventing diabetic complications.

재래종과 개량종 세절 돼지고기의 저장 중 품질 특성 비교 (Quality Comparison between Korean Native Black Ground Pork and Modern Genotype Ground Pork during Refrigerated Storage)

  • 본 연구는 재래종 세절 돼지고기의 저장 중 품질 특성을 구명하기 위하여 개량종과 비교하여 실시하였다. 시험용 돼지고기로 240일간 사육된 평균체중 75 kg의 재래돼지와 210일간 사육된 평균체중 105 kg의 개량돼지를 이용하였다. 도축 후 세절돈육을 4℃에서 9일간 저장하며 실시하였다. 일반성분의 조성을 보면 재래종 돼지고기가 개량종에 비해 조지방과 조단백질 함량이 높고, 수분과 조회분 함량은 낮았다. 재래종 돼지고기의 pH가 개량종에 비해 저장 5일부터는 낮았고, 보수력은 저장기간 내내 유의적으로 높았다(p<0.05). 재래종 돼지고기는 명도가 낮고 적색도가 높아 개량종에 비해 검붉은 색깔을 지니고 있다. 저장기간이 늘어날수록 CIE L/sup */, a/sup */, b/sup */와 h/sup O/값들은 감소하는 경향을 보였다. TBARS, POV, FOX값 모두 저장기간이 증가할수록 유의적으로 증가하였으며 특히 재래종이 저장기간이 길어질수록 현저한 증가현상을 나타내었다. 재래종 돼지고기의 지방산 조성비율을 보면 총 포화지방산과 stearic acid 함량이 개량종에 비해 유의적으로 높았다(p<0.05).

Korean Red Ginseng and Korean black ginseng extracts, JP5 and BG1, prevent hepatic oxidative stress and inflammation induced by environmental heat stress

  • Background: Continuous exposure to high temperatures can lead to heat stress. This stress response alters the expression of multiple genes and can contribute to the onset of various diseases. In particular, heat stress induces oxidative stress by increasing the production of reactive oxygen species. The liver is an essential organ that plays a variety of roles, such as detoxification and protein synthesis. Therefore, it is important to protect the liver from oxidative stress caused by heat stress. Korean ginseng has a variety of beneficial biological properties, and our previous studies showed that it provides an effective defense against heat stress. Methods: We investigated the ability of Korean Red Ginseng and Korean black ginseng extracts (JP5 and BG1) to protect against heat stress using a rat model. We then confirmed the active ingredients and mechanism of action using a cell-based model. Results: Heat stress significantly increased gene and protein expression of oxidative stress-related factors such as catalase and SOD2, but treatment with JP5 (Korean Red Ginseng extract) and BG1 (Korean black ginseng extract) abolished this response in both liver tissue and HepG2 cells. In addition, JP5 and BG1 inhibited the expression of inflammatory proteins such as p-NF-κB and tumor necrosis factor alpha-α. In particular, JP5 and BG1 decreased the expression of components of the NLRP3 inflammasome, a key inflammatory signaling factor. Thus, JP5 and BG1 inhibited both oxidative stress and inflammation. Conclusions: JP5 and BG1 protect against oxidative stress and inflammation induced by heat stress and help maintain liver function by preventing liver damage.

  • 오래 전부터 생물학적 폐수처리 방법에 대한 다양한 연구들이 꾸준히 진행되고 있으나, 전통적인 염료 염색 폐수처리 방법들은 다소 비효율적이다. 최근 들어 이를 극복하기 위하여 특정 미생물이나 효소를 이용한 연구들이 활발히 진행되고 있다. 본 연구에서는 백색부후균이 생산하는 리그닌 효소에 의한 생물처리 방법으로, 아조계(azo) 염료인 Reactive black 5를 적용하였으며, 다양한 농도에 따른 분해경향을 확인하였다. 미생물 재이용과 장기간 운전, 효율적인 유기물 제거의 가능성을 확인하기 위해 i)생물학적 처리와 ii)분리막 처리공정과 iii)이 두 가지를 결합한 공정의 처리 효율을 비교하였다. 결합공정을 사용하였을 때 색도제거율이 100%였으며, 총유기물(TOC) 제거율은 70% 이상으로 각 단위공정만으로 처리하였을 때보다 더 효율적인 결과를 보였다. 또한, 생물반응에서 배양말기까지 유지된 효소활성(40 U/ml laccase and 300-600 U/ml MnP)을 통해 미생물 재이용의 가능성을 확인하였다.

Primary Restorative Transmission Line Selection for Myanmar's Electric Power System

  • Power system restoration following a massive or complete blackout starts with energizing the primary restorative transmission system. During this primary restoration process, unexpected overvoltage may happen due to nonlinear interaction between the unloaded transformer and the transmission system. In the case of the Myanmar electric power system, there are so many wide outage experiences, including complete blackout cases, caused by 230kV line faults and so on. Consequently, Myanmar's system operators have been well trained to deal with wide blackouts. Howver, system blackout restoration has been conducted by relying on the experience of only a few specialists. So, more scientific analysis is required to meet the requirements necessary to ensure fast and reliable system restoration. This paper presents analytical results on the primary restorative transmission system of Myanmar, focusing on the problems during the early restoration process. Methodologies are presented that handle load pick-up, terminal voltage and the reactive capability limitation of black-start generators to compensate the Ferranti effect. Static and dynamic simulation with the PSSolution and EMTDC programs respectively for the six cases are performed in order to select the primary restorative transmission lines.

인진쑥 첨가사료를 급여한 흑우육의 이화학적 품질특성 (Physicochemical Characteristics of Korean Black Cattle-Fed Mugwort)

  • 본 연구는 인진쑥을 첨가한 TMR 사료가 흑우육의 이화학적 품질 및 관능특성에 미치는 영향을 규명하여 고품질의 기능성 흑우육 생산 가능성을 검토하였다. 대조군(control)은 인진쑥을 첨가하지 않은 비육우 사료를 급여하였고, 급여군(treatment)은 비육우 사료에 인진쑥을 첨가하였다. 총 catechin 함량은 대조군 및 급여군이 각각 0.262 및 0.379 mg/kg, epicatechin은 각각 0.042 및 0.059 mg/kg으로 급여군이 유의하게 높았다($p$ <0.05). 적색도($a^*$)는 급여군이 대조군보다 높았으나($p$ <0.05) 명도($L^*$) 및 황색도($b^*$)는 급여군과 대조군 사이에 유의한 차이가 없었다. 그리고 pH, VBN 함량 및 총균수는 대조군과 급여군 사이에 유의한 차이가 없었으나 TBARS는 대조군이 급여군보다 높았고, EDA는 급여군이 대조군보다 높았다. 보수력, 동결감량, 해동감량 및 가열감량은 대조군과 급여군 사이에 유의한 차이가 없었다. 경도 및 검성은 급여군이 대조군보다 높았고, 탄성은 대조군이 급여군보다 높았다($p$ <0.05). 그러나 응집성, 저작성 및 전단력은 대조군과 급여군 사이에 유의한 차이가 없었다. 생육의 색깔 및 향, 가열육의 맛, 풍미, 연도, 다즙성 및 전체적인 기호성은 대조군 및 급여군 사이에 유의한 차이가 없었다. 이상의 결과에서 인진쑥의 급여로 항산화 물질이 축적되어 흑우육의 색깔이 개선되고, 항산화력이 증가하는 효과가 있기 때문에 기능성 고품질 흑우육 생산을 위한 기능성 사료의 소재로서의 이용이 기대된다.

반응매질로서의 영가철 및 블랙셰일에 용존무기 인산염 흡착 (Sorption of Dissolved Inorganic Phosphorus to Zero Valent Iron and Black Shale as Reactive Materials)

  • 본 연구는 지표수나 호소수 등에서 유기 및 무기오염물질의 저감을 위해 반응매질로서 사용되는 black shale 및 영가철의 수중의 무기인산염을 흡착제거하는 데 있어서의 정량적 특성을 알아보았다. 무기인산염의 회분식 흡착실험에는 갯벌에서 마련한 퇴적토, black shale, 영가철이 흡착제로 사용되었으며 인산염의 흡착에 미치는 영향을 공존 칼슘이온의 농도를 0, 1, 5 mM로 변화시켜 실험하였다. 칼슘이온의 공존과 함께 세 흡착제에 대한 인산염 농도감소를 빠른 흡착계수(k$_f$), 느린 흡착계수(k$_s$)과 침전계수(k$_p$)로 구성된 kinetic model을 이용하여 해석하였다. 등온흡착의 결과는 Langmuir와 Freundlich 모델을 적용하였으며 퇴적토, black shale, 영가철에 대한 Freundlich 모델의 K$_f$값은 각각 1.141, 7.721, 그리고 4.873였다. 공존하는 칼슘이온과의 침전가능성으로 인하여 kinetic model에서는 칼슘농도와 침전제거간의 상관관계가 있었으나 등온흡착식에서 인산염의 최대흡착량에는 칼슘농도가 영향을 미치지 않았다.

Anthocyanin Extracts from Black Soybean (Glycine max L.) Protect Human Glial Cells Against Oxygen-Glucose Deprivation by Promoting Autophagy

  • Anthocyanins have received growing attention as dietary antioxidants for the prevention of oxidative damage. Astrocytes, which are specialized glial cells, exert numerous essential, complex functions in both healthy and diseased central nervous system (CNS) through a process known as reactive astrogilosis. Therefore, the maintenance of glial cell viability may be important because of its role as a key modulator of neuropathological events. The aim of this study was to investigate the effect of anthocyanin on the survival of glial cells exposed to oxidative stress. Our results demonstrated that anthocyanin extracts from black soybean increased survival of U87 glioma cells in a dose dependent manner upon oxygen-glucose deprivation (OGD), accompanied by decrease levels of reactive oxygen species (ROS). While treatment cells with anthocyanin extracts or OGD stress individually activated autophagy induction, the effect was signifi cantly augmented by pretreatment cells with anthocyanin extracts prior to OGD. The contribution of autophagy induction to the protective effects of anthocyanin was verifi ed by the observation that silencing the Atg5 expression, an essential regulator of autophagy induction, reversed the cytoprotective effect of anthocyanin extracts against OGD stress. Treatment of U87 cells with rapamycin, an autophagy inducer, increased cell survival upon OGD stress comparable to anthocyanin, indicating that autophagy functions as a survival mechanism against oxidative stress-induced cytotoxicity in glial cells. Our results, therefore, provide a rationale for the use of anthocyanin as a preventive agent for brain dysfunction caused by oxidative damage, such as a stroke.

중성염이 견의 반응염색에 미치는 영향 (Ⅱ) - 음이온의 영향 - (Effect of Neutral Salts on the Reactive Dyeing of Silk (Ⅱ) - Effect of Anions -)

  • Four kinds of neutral sodium salts with different anions, NaF, NaCl, NaBr, and NaI, were added to the dye bath to accurately understand the effect of anions on the reactive dyeing of silk with C. I. Reactive Black 5. The sodium cation lowered the negative surface potential of the silk and increased the dye-uptake on fille fabric as reported previously. However, because of the discrepancy in the anions′inhibition power from cation′s lowering the surface negative potential the amount of the dye on the silk fiber was different from each other in the order of $F^-\;>\;Cl^-\;>\; Br^-\;>\;I^-$. The activation energy($E_a$) for the dyeing was in the order of $F^-\;>\;Cl^-\;>\; Br^-\;>\;I^-$ but the dye-uptake on the fabric and the activation free energy(Δ$G^*$), the real energy barrier for the reaction, were in the order of $F^-\;>\;Cl^-\;>\; Br^-\;>\;I^-$ because the strength of the interaction of the anions with sodium cations was the same as the order of the latter. In other words $F^-$ exerted the weakest electrostatic force on $Na^+$and competed with the dyestuff anions least of all. The decrease in Δ$S^*$may be due to the looesly bonded activated complex of dyestuff anions, sodium cations and fiber molecules at transition state. It was clarified from the Bronsted equation that sodium salts with different anions also had fille ionic strength effect and the specific salt effect on the reactive dyeing.

청혈산(淸血散)이 ob/ob mouse의 혈당, 고지혈증, Polyol Pathway 및 Reactive Oxygen Species에 미치는 영향 (Effects of Cheonghyul-san on Blood Glucose, Hyperlipidemia, Polyol Pathway and Reactive Oxygen Species in ob/ob Mice)

  • Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Cheonghyul-san has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs. In male ob/ob mouse with severe obesity, hyperinsulinemia, hypergiycemia, hyperlipidemia, the acting mechanisms of Cheonghyul-san were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. The effects of Cheonghyul-san extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Cheonghyul-san lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Cheonghyul-san treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the generation of ROS in the kidney. Finally, MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Cheonghyul-san-administered groups. The results suggested that Cheonghyul-san exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging ROS, regulating the MDA+HAE and GSH levels in the ob/ob mice.
