We prepared a number of reaction-bonded silicon carbides (RBSCs) made from various mixing ratios of raw SiC particles, and investigated their microstructure and etch characteristics by Reactive Ion Etch (RIE). Increasing the amount of $9.5{\mu}m$-SiC particles results in a microstructure with relatively coarser Si regions. On the other hand, increasing that of $2.6{\mu}m$-SiC particles produces much finer Si regions. The addition of more than 50 wt% of $2.6{\mu}m$-SiC particles, however, causes the microstructure to become partially coarse. We also evaluated their etching behaviors in terms of surface roughness (Ra), density and weight changes, and microstructure development by employing Confocal Laser Scanning Microscope (CLSM) and Scanning Electron Microscope (SEM) techniques. During the etching process of the prepared samples, we confirmed that the residual Si region was rapidly removed and formed pits isolating SiC particles as islands. This leads to more intensified ion field on the SiC islands, and causes physical corrosion on them. Increased addition of $2.6{\mu}m$-SiC particles produces finer residual Si region, and thus decreases the surface roughness (Ra.) as well as causing weight loss after etching process by following the above etching mechanism.
The aromatic compound p-hydroxybenzoate (PHBA) is an important material with multiple applications, including as a building block of liquid crystal polymers in chemical industries. The cytochrome P450 (CYP) enzymes are beneficial monooxygenases for the synthesis of chemicals, and CYP53A15 from fungus Cochliobolus lunatus is capable of executing the hydroxylation from benzoate to PHBA. Here, we constructed a system for the bioconversion of benzoate to PHBA in Escherichia coli cells coexpressing CYP53A15 and human NADPH-P450 oxidoreductase (CPR) genes as a redox partner. For suitable coexpression of CYP53A15 and CPR, we originally constructed five plasmids in which we replaced the N-terminal transmembrane region of CYP53A15 with a portion of the N-terminus of various mammalian P450s. PHBA productivity was the greatest when CYP53A15 expression was induced at $20^{\circ}C$ in $2{\times}YT$ medium in host E. coli strain ${\Delta}gcvR$ transformed with an N-terminal transmembrane region of rabbit CYP2C3. By optimizing each reaction condition (reaction temperature, substrate concentration, reaction time, and E. coli cell concentration), we achieved 90% whole-cell conversion of benzoate. Our data demonstrate that the described novel E. coli bioconversion system is a more efficient tool for PHBA production from benzoate than the previously described yeast system.

This research applied and evaluated a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) using cytochrome b gene to detect pork contamination in meatballs from local markets in Surabaya and Yogyakarta regions, Indonesia. To confirm the effectiveness and specificity of this fragment, thirty nine DNA samples from different meatball shops were isolated and amplified, and then the PCR amplicon was digested by BseDI restriction enzyme to detect the presence of pork in meatballs. BseDI restriction enzyme was able to cleave porcine cytochrome b gene into two fragments (131 bp and 228 bp). Testing the meatballs from the local market showed that nine of twenty meatball shops in Yogyakarta region were detected to have pork contamination, but there was no pork contamination in meatball shops in Surabaya region. In conclusion, specific PCR amplification of cytochrome b gen and cleaved by BseDI restriction enzymes seems to be a powerful technique for the identification of pork presence in meatball because of its simplicity, specificity and sensitivity. Furthermore, pork contamination intended for commercial products of sausage, nugget, steak and meat burger can be checked. The procedure is also much cheaper than other methods based on PCR, immunodiffusion and other techniques that need expensive equipment.

Reset-first resistive switching mechanism based on reduction reaction in HfO2-x with oxygen drift-diffusion was studied. we first report that the indirect evidence of local filamentary conductive path formation in bulk HfO2 film with local TiOx region at Ti top electrode formed during forming process and presence of anion-migration at interface between electrode and HfO2 during resistive switching through high resolution transmission electron microscopy (HRTEM), electron disperse x-ray (EDX), and electron energy loss spectroscopy (EELS) mapping. Based on forming process mechanism, we expected that redox reaction from Ti/HfO2 to TiOx/HfO2-x was responsible for an increase of initial current with increasing the post-annealing process. First-reset resistive switching in above $350^{\circ}C$ annealed Ti/HfO2 film was exhibited and the redox phenomenon from Ti/HfO2 to TiOx/HfO2-x was observed with high angle annular dark field (HAADF) - scanning transmission electron microscopy (STEM), EDX and x-ray photoelectron spectroscopy. Therefore, we demonstrated that the migration of oxygen ions at interface region under external electrical bias contributed to bipolar resistive switching behavior.
Objective: Aim of present study was the set up of a fast and reliable protocol using species-specific markers for the quali-quantitative analysis of DNA and the detection of ruminant biological components in dairy products. For this purpose, the promoter of the gene coding for the ${\alpha}$-lactoalbumin (LALBA) was chosen as possible candidate for the presence of short interspersed nuclear elements (SINEs). Methods: DNA was isolated from somatic cells of 120 individual milk samples of cattle (30), Mediterranean river buffalo (30), goat (30), and sheep (30) and the gene promoter region (about 600/700 bp) of LALBA (from about 600 bp upstream of exon 1) has been sequenced. For the development of a single polymerase chain reaction (PCR) protocol that allows the simultaneous identification of DNA from the four species of ruminants, the following internal primers pair were used: 5'-CACTGATCTTAAAGCTCAGGTT-3' (forward) and 5'-TCAGA GTAGGCCACAGAAG-3' (reverse). Results: Sequencing results of LALBA gene promoter region confirmed the presence of SINEs as monomorphic "within" and variable in size "among" the selected species. Amplicon lengths were 582 bp in cattle, 592 bp in buffalo, 655 in goat and 729 bp in sheep. PCR specificity was demonstrated by the detection of trace amounts of species-specific DNA from mixed sources ($0.25ng/{\mu}L$). Conclusion: We developed a rapid PCR protocol for the quali-quantitative analysis of DNA and the traceability of dairy products using a species-specific marker with only one pair of primers. Our results validate the proposed technique as a suitable tool for a simple and inexpensive (economic) detection of animal origin components in foodstuffs.

Osteoporotic fractures (OFs) are critical hard outcomes of osteoporosis and are characterized by decreased bone strength induced by low bone density and microarchitectural deterioration in bone tissue. Most OFs cause acute pain, hospitalization, immobilization, and slow recovery in patients and are associated with increased mortality. A variety of genetic studies have suggested associations of genetic variants with the risk of OF. Genome-wide association studies have reported various single-nucleotide polymorphisms and copy number variations (CNVs) in European and Asian populations. To identify CNV regions associated with OF risk, we conducted a genome-wide CNV study in a Korean population. We performed logistic regression analyses in 1,537 Korean subjects (299 OF cases and 1,238 healthy controls) and identified a total of 8 CNV regions significantly associated with OF (p < 0.05). Then, one CNV region located on chromosome 20q13.12 was selected for experimental validation. The selected CNV region was experimentally validated by quantitative polymerase chain reaction. The CNV region of chromosome 20q13.12 is positioned upstream of a family of long non-coding RNAs, LINC01260. Our findings could provide new information on the genetic factors associated with the risk of OF.
As this paper is observed the phase equilibrium diagram of mono- (methane) and multi-component(natural gas) hydrates, and the hydrate growth behavior is analysed and compared by the experiments during the reaction. The difference of mono and multi-component hydrates is an induction delay time and a plateau region. And the concentration of component of gases is changed during the reaction in multi-component hydrates and the concentration of components is changed during the decomposition of hydrate according to each decomposing rates of gases. At 6 MPa, 276.65 K and 600 rpm, the induction delay time of multi-component hydrate formation is observed shorter than that of mono-component hydrate formation because the hydrate nuclei of gases except methane form faster than those of methane. And the plateau region of mono-component hydrate is observed distinctly at 0.055 mole of $CH_4$/mole of water and that of multi-component hydrate is observed at 0.04 mole of $CH_4$/mole of water.
In this study, it is investigated that the possibility of the numerical simulation for the incineration of the hazardous material, crbon tetrachloride($CCl_4$). A 3-dimensional numerical technology is applied for turbulent reacting flows of the full-scale Dow Chemical incinerator. The calculations are made by a CRAY-2S, super computer. The major parameters considered in this study are kiln revolution rate (rpm), filling ratio of the solid waste(f), burner Injection velocity and angle, and turbulent air jets for swirl. And the employed turbulent reaction model is the eddy break-up model which is a kind of fast chemistry model assuming general equilibrium and used for a premixed flame. The calculated flow fields are presented and discussed. 1) The presence of turbulent air nozzles for swirl gives rise to visible increase of the convective motion over the region of the solid waste. This implies the possibility to enhance the mixing of the waste with the surrounding all and thereby to reduce thermal and species stratification, which were reported in a large rotary kiln operation. 2) Considering that the location of the recirculation region has a strong relation with the heating rate of the solid waste, the control of the recirculation region by the burner injection angle Is quite desirable in the sense of the flexible design of the rotary kiln incinerator for a carbon tetrachloride. 3) Finally, it is found that the eddy break-up model Is not suitable for carbon tetrachloride($CCl_4$) because this model is not incorporated the flame inhibition trend due to the presence $CCl_4$compound.

Using DNA sequencing method, we analyzed mutations of rpoB (RNA polymerase beta subunit) rifampin-resistant Mycobaterium tuberculosis strains which were identified by conventional test at Masan National Hospital and The Korean Institute of Tuberculosis. Though it has been reported different mutations of rpoB region of rifampin-resistant M. tuberculosis strains in the south of Korea, it is not confirmed whether these mutations of rpoB region actually express rifampin resistance through experiment. We confirmed experimentally these mutations of rpoB region of M. tuberculosis strains induced rifampin-resistance through ampified rpoB by polymerase chain reaction (PCR) and cloning of mutant rpoB into rifampin sensitive-M. tuberculosis strain.
Background: Activation of air and water in the electron linear accelerator for medical use has not been considered severely. By the new Japanese regulation for protection of radiation hazard, it became indispensable to evaluate of activation of air and water in the accelerator room. The measurement of induced activity in air and water components in the electron energy region of 10 to 20 MeV is very difficult, because this energy region is close to the threshold energy region of photonuclear reactions. Then, we measured the photonuclear reaction yields of $^{13}N$, $^{15}O$, and $^{11}C$ by using the electron linear accelerator. Obtained data were compared with the data calculated by the Monte Carlo method. Materials and Methods: An activation experiment was performed at the Research Center for Electron Photon Science, Tohoku University. Highly purified $SiO_2$, $Si_3N_4$, and carbon disks were irradiated for 10 minutes by bremsstrahlung converted by a tungsten plate. Induced activity from C, N, and O was obtained. Monte Carlo calculation was performed using MCNP5 and AERY (DCHAIN-SP) to simulate the experimental condition. Cross section data were adopted the KAERI dataset. Results and Discussion: In our experiment in hospital, calculated values were not agreed with experimental values. It might be three possible reasons as the cause of this deference, such as irradiation energy, calculation procedure and cross section data. Obtained data of this work, calculated and experimental values were good agreement with each other within one order. In this work, we used KAERI dataset of photonuclear reaction instead of JENDL. Therefore, it was found that the photonuclear cross section data of light elements are most important for yield calculation in these reactions. Conclusion: Further improvement for calculation using a new dataset JENDL/PD-2015 and considering electron energy spreading will be needed.
