Toxicity of 5 Bacillus cereus Enterotoxins in Human Cell Lines and Mice

  • To determine whether the toxicity of Bacillus cereus would be seen in human cell lines and mice, we screened B. cereus B-38B, B. cereus B-50B, and B. cereus KCCM40935 for genes that coded for 5 enterotoxins using the polymerase chain reaction and cultivated them for 17 hr, by whose time they had grown to $10^7-10^8$ colony-forming units (CFU) per milliliter. Cell-free supernatant was added to make up 1% of the total reaction solution. Human cells from normal lung, lung carcinoma, embryonic kidney, and cervical adenocarcinoma cell lines were grown in culture. The cytotoxicity induced by adding the reaction solution was indicated by cell death rates of 0 to 70%, depending on the bacterial strain involved and the cell line. A lethality of 20% was observed when B. cereus cultures containing $10^7-10^8$ viable cells were administrated orally to mice. Therefore, the culture of B. cereus containing $10^7-10^8$ viable cells seems to have high cytotoxicity on human cell lines and lethality on mice.

3-Phenyloxadiazole 유도체의 1,3-dipolar Cycloaddition 반응에 미치는 온도의 영향 (Effect of Themperature on the 1,3-dipolar Cycloaddition of 3-phenyloxadiazole Derivatives)

  • Ultraviolet spectrophotometric investigation has been carried out on the rate constants for 1,3-dipolar cycloaddition of 4-substituted-3-phenyloxadiazole derivatives with dipolarophiles such as phenyl acetylene, propiolic acid methyl ester and dimethylacetylene dicarboxylate. From there, the rate constants for 1,3-dipolar cycloaddition were determined at 80, 100 and $120^{\circ}C$, and the reaction rates were increased with increasing temperature. From these rate constants, the values of the thermodynamic activation parameters were obtained. Some thermodynamic activation parameters such as $E_{\alpha}$, ${\Delta}H^{\ast}$, ${\Delta}S^{\ast}$ and ${\Delta}G^{\ast}$ from Arrhenius equation were also calculated for the electrophilic 1,3-dipolar cycloaddition of 3-phenyloxadiazole derivatives with dipolarophiles. In order to the proposal the mechanism and reactivity of 1,3-dipolar cycloaddition reaction, the effect of substituents having various kinds of electron withdrawing or releasing groups were examinated. Considering the effect of substituents, an electron withdrawing group attached at the 4-carbon position in 3-phenyloxadiazole derivatives decreases the reaction rate because of the lack of electron density in 3-phenyloxadiazole ring.

Reaction of Lithium Gallium Hydride with Selected Organic Compounds Containing Representative Functional Groups

  • The approximate rates and stoichiometry of the reaction of excess lithium gallium hydride with selected organic compounds containing representative functional groups were examined under the standard conditions (diethyl ether, 0 $^{\circ}C)$ in order to compare its reducing characteristics with lithium aluminum hydride and lithium borohydride previously reported, and enlarge the scope of its applicability as a reducing agent. Alcohols, phenol, and amines evolve hydrogen rapidly and quantitatively. However lithium gallium hydride reacts with only one active hydrogen of primary amine. Aldehydes and ketones of diverse structure are rapidly reduced to the corresponding alcohols. Conjugated aldehyde and ketone such as cinnamaldehyde and methyl vinyl ketone are rapidly reduced to the corresponding saturated alcohols. p-Benzoquinone is mainly reduces to hydroquinone. Caproic acid and benzoic acid liberate hydrogen rapidly and quantitatively, but reduction proceeds slowly. The acid chlorides and esters tested are all rapidly reduced to the corresponding alcohols. Alkyl halides and epoxides are reduced rapidly with an uptake of 1 equiv of hydride. Styrene oxide is reduced to give 1-phenylethanol quantitatively. Primary amides are reduced slowly. Benzonitrile consumes 2.0 equiv of hydride rapidly, whereas capronitrile is reduced slowly. Nitro compounds consumed 2.9 equiv of hydride, of which 1.9 equiv is for reduction, whereas azobenzene, and azoxybenzene are inert toward this reagent. Cyclohexanone oxime is reduced consuming 2.0 equiv of hydride for reduction at a moderate rate. Pyridine is inert toward this reagent. Disulfides and sulfoxides are reduced slowly, whereas sulfide, sulfone, and sulfonate are inert under these reaction conditions. Sulfonic acid evolves 1 equiv of hydrogen instantly, but reduction is not proceeded.

Kinetics and Mechanisms of the Oxidation of Carbon Monoxide on $Eu_{1-x}Sr_xCoO_{3-y}$ Perovskite Catalysts

  • The catalytic oxidation of CO on perovskite $Eu_{1-x}Sr_xCoO_{3-y}$, has been investigated at reaction temperatures from 100 to $250^{\circ}C$ under stoichiometric CO and $O_2$ partial pressures. The microstructure and Sr-substitution site of the catalyst were studied by means of infrared spectroscopy. The reaction rates were found to be correlated with 1.5-and 1.0-order kinetics with and without a $CO_2$ trap, respectively; first-and 0.5-order with respect to CO and 0.5-order to $O_2$ with the activation energy of 0.37 eV $mol^{-1}$. It was found from IR, ${\sigma}$ and kinetic data that $O_2$ adsorbs as an ionic species on the oxygen vacancies, while CO adsorbs on the lattice oxygens. The oxidation reaction mechanism is suggested from the agreement between IR, ${\sigma}$ and kinetic data.

기계적 합금화 투입에너지 계산에 의한 이원합금계의 상변태 시간 예측 (Prediction the Phase Transformation Time of Binary Alloy System by calculating the Input Energy of Mechanical Alloying)

  • The activation energy to create a phase transformation or for the reaction to move to the next stage in the milling process can be calculated from the slop of the DSC plot, obtained at the various heating rates for mechanically activated Al-Ni alloy systems by using Kissinger's equation. The mechanically activated material has been called "the driven material" as it creates new phases or intermetallic compounds of AlNi in Al-Ni alloy systems. The reaction time for phase transformation by milling can be calculated using the activation energy obtained from the above mentioned method and from the real required energy. The real required energy (activation energy) could be calculated by subtracting the loss energy from the total input energy (calculated input energy from electric motor). The loss energy and real required energy divided by the reaction time are considered the "metabolic energy" and "the effective input energy", respectively. The milling time for phase transformation at other Al-Co alloy systems from the calculated data of Al-Ni systems can be predicted accordingly.

The Onset and Growth of the Buoyancy-driven Fingering Driven by the Irreversible A+B→C Reaction in a Porous Medium: Reactant Ratio Effect

  • The effect of a reactant ratio on the growth of a buoyancy-driven instability in an irreversible A+B→C reaction system is analyzed theoretically and numerically. Taking a non-stoichiometric reactant ratio into account, new linear stability equations are derived without the quasi-steady state assumption (QSSA) and solved analytically. It is found that the main parameters to explain the present system are the Damköhler number, the dimensionless density difference of chemical species and the ratio of reactants. The present initial grow rate analysis without QSSA shows that the system is initially unconditionally stable regardless of the parameter values; however, the previous initial growth rate analysis based on the QSSA predicted the system is unstable if the system is physically unstable. For time evolving cases, the present growth rates obtained from the spectral analysis and pseudo-spectral method support each other, but quite differently from that obtained under the conventional QSSA. Adopting the result of the linear stability analysis as an initial condition, fully nonlinear direct numerical simulations are conducted. Both the linear analysis and the nonlinear simulation show that the reactant ratio plays an important role in the onset and the growth of the instability motion.

Endoglucanase와 β-Glucosidase 효소에 의한 셀룰로오스 생분해 모델링 (Cellulose Biodegradation Modeling Using Endoglucanase and β-Glucosidase Enzymes)

  • In this study, a biodegradation model of based on molecular cellulose was established. It is a mathematical, kinetic model, assuming that two major enzymes randomly break glycosidic bonds of cellulose molecules, and calculates the number of molecules by applying the corresponding probability and degradation reaction coefficients. Model calculations considered enzyme dose, cellulose chain length, and reaction rate constant ratio. Degradation increased almost by two folds with increase of temperature (5℃→25℃). The change of degradation was not significant over the higher temperatures. As temperature increased, the degradation rate of the molecules increased along with higher production of shorter chain molecules. As the reaction rates of the two enzymes were comparative the degree of degradation for any combinations of enzyme application was not affected much. Enzyme dose was also tested through experiment. While enzyme dose ranged from 1 mg/L to 10 mg/L, the gap between real data and model calculations was trivial. However, at higher dose of those enzymes (>15 mg/L), the experimental result showed the lower concentrations of reductive sugar than the corresponding model calculation did. We determined that the optimal enzyme dose for maximum generation of reductive sugar was 10 mg/L.

  • The effects of nuclear data uncertainties are studied on a typical PWR fuel assembly model in the framework of the OECD Nuclear Energy Agency UAM (Uncertainty Analysis in Modeling) expert working group. The "Fast Total Monte Carlo" method is applied on a model for the Monte Carlo transport and burnup code SERPENT. Uncertainties on $k_{\infty}$, reaction rates, two-group cross sections, inventory and local pin power density during burnup are obtained, due to transport cross sections for the actinides and fission products, fission yields and thermal scattering data.

  • An electroenzymatic system to oxidize veratryl alcohol of on electrodes with in-situ generated hydrogen peroxide was studied. We investigated hydrogen peroxide generation, current efficiency, and veratryl alcohol oxidation in the electrode system at various conditions. The reaction rates of veratryl alcohol oxidation were compared in an electrochemical, an electroenzymatic, and an usual biochemical systems to prove the concept of electroenzymatic oxidation.

  • Nonlinear feedback control scheme for reactive distillation column has been proposed. The proposed control scheme is derived in the framework of Nonlinear Internal Model Control. The product compositions and liquid and vapor flow rates in sections of the reactive distillation column are estimated from selected tray temperature measurements by an observer. The control scheme is applied to example reactive distillation column in which two products are produced in a single column and the reversible reaction A + B = C + D occurs. The relative volatilities are favorable for reactive distillation so that the reactants are intermediated boilers between the light product C and the heavy product D.
