Cytotoxicity of Diesel Exhaust Particles from Various Vehicles toward Macrophage Cells (국내 디젤 차량 배기 입자가 쥐 대식세포에 미치는 세포독성 평가)

  • DEPs (diesel exhaust particles) like any other particles can be also inhaled into lung to participate in a damaging reaction to the organ. Possible damages might be apoptosis and inflammatory responses to the cells in respiratory track. The aim of this study was cytotoxicity evaluation of DEPs from five in-use diesel vehicles using a murine macrophage cell (RAW 254.7). We found that most DEPs have a considerable cytotoxicity compared to the control and SRM 2975. When measured by MTT assay and extents of apoptosis, DEPs of two highmileage vehicles had higher toxicity than those of the other three low-mileage vehicles tested. Although mRNA expression level of TNF-${\alpha$ somewhat explains the trend of cytotoxicity and apoptosis, that of IL-1$\beta$ did not. Correlation studies among the extents of MTT assay, apoptosis, and TNF-$\alpha$ expression showed that the extents between apoptosis and TNF-$\alpha$ expression was most highly correlated (r=0.96). These results suggest that cytotoxicity of various DEPs could be compared easily by measuring the extent of apoptosis or TNF-$\alpha$ expression by DEPs.

Alaternin and Emodin with Hydroxyl Radical inhibitory and/or Scavenging Activities and Hepatoprotective Activity on Tacrine-Induced Cytotoxicity in HepG2 Cells

  • The antioxidative and hepatoprotective potentials of two anthraquinones, alaternin (2-hydroxy-emodin) and emodin, to scavenge and/or inhibit hydroxyl radicals generated by the Fenton reaction and to protect tacrine-induced cytotoxicity in human liver derived HepG2 cells were evaluated, respectively. The inhibitory activity on hydroxyl radical generated in a cell-free chemical system (FeSO$_4$/$H_2O$$_2$) was investigated by a fluorescence spectrophotometer using a highly fluorescent probe, 2$^1$,7$^1$-dichlorofluorescein. The hydroxyl radical scavenging activity was determined by electron spin resonance spectroscopy using 5,5-dimethy-1-pyrroline-N-oxide as hydroxyl radicals trapping agents. Tacrine-induced HepG2 cell toxicity was determined by a 3-[4,5-dimethylthiazole-2yl]-2,5-diphenyltertrazolium bromide assay. Although the scavenging activity of alaternin on hydroxyl radical was similar to that of emodin in dose-dependent pat-terns, the inhibitory activity exhibited by the former on hydroxyl radical generation was stron-ger than that of the latter, with $IC_{50}$/ values of 3.05$\pm$0.26 $\mu$M and 13.29$\pm$3.20 $\mu$M, respectively. In addition, the two anthraquinones, alaternin and emodin showed their hepatoprotective activ-ities on tacrine-induced cytotoxicity, and the EC$_{50}$ values were 4.02 11M and 2.37 $\mu$M, respec-tively. Silymarin, an antihepatotoxic agent used as a positive control exhibited the EC$_{50}$ value of 2.00 $\mu$M. These results demonstrated that both alaternin and emodin had the simultaneous antioxidant and hepatoprotective activities.ies.

Development of SMH Actuator System Using Hydrogen-Absorbing Alloy

  • This paper presents the temperature-pressure characteristics of a new SMH actuator using a Peltier module. The SMH actuator is characterized by its small size, low weight, noiseless operation, and compliance similar to that of the human body. The simple SMH actuator, consisting of the plated hydrogen-absorbing alloys as a power source, Peltier elements as a heat source, and a cylinder with metal bellows as a functioning part has been developed. To improve the thermal conductivity of the hydrogen-absorbing alloy, an assembly of copper pipes has been used. It is well known that hydrogen-absorbing alloys can reversibly absorb and desorb a large amount of hydrogen, more than about 1000 times of their own volume. The hydrogen equilibrium pressure increases when hydrogen is desorbed by heating of the hydrogen-absorbing alloys, whereas by cooling the alloys, the hydrogen equilibrium pressure decreases and hydrogen is absorbed. The new special metal hydride (SMH) actuator uses the reversible reaction between the heat energy and mechanical energy of a hydrogen absorbing alloys. The desirable characteristics of SMH actuator, which makes it suitable for the uses in medical and rehabilitation applications, have been also studied. For this purpose, the characteristics of the new SMH actuator for different temperature, pressure, and external load were explored.

Control of Catabolite Repression by Limit Feed of Cellobiose in Cellulomomas sp. (Cellulomonas sp.에 있어서 셀로비오스의 미량공급에 의한 생성물 저해의 조절)

  • As an investigation on the catabolite repression system in cellulase production by Cellulomons sp. CS1-1, the organism was tested on the avicel overlay plates containing glucose or cellobiose at a range of concentration and was grown in continuous culture vessel, supplied by cellobiose medium, aiming the enhanced production of extracellular CM-cellulase at low dilution rates. Product inhibition of cellulase action by cellobiose was also tested. The results obtained are: i) no inhibition of CM-cellulase was observed up to 10 mM(3.4mg/ml) cellobiose in the reaction mixture, however 30% inhibition was observed at 20mM and 55% at 50mM, ii) the tests of catabolite repression on the solid media were successful, and avicel degradation was markedly repressed by glucose or cellobiose, iii) at low concentrations of cellobiose, dilution rate 0.05 and $1.0hour^{-1}$, no significant increase was observed in the production of either intra or extracellular CM-cellulase.

  • Recent studies dealing with the fundamental understanding and applications of bimetallic catalysts are discussed. Bimetallic catalysts have had a major industrial impact, specifically for the reforming of petroleum naphtha, for the hydrogen reduction of carbon monoxide, and for the three way catalytic converter system. The action of the bimetallic catalysts in these reactions may be interpreted in terms of ensembles, electronic influences and surface structure. Various combinations of metal pairs have been considered in order to evaluate the role played by the added metals. For catalyst selectivity control, the possibility of surface enrichment of one element has been recognised. More generally, the influence of preparative variables on the formation of supported catalysts has been clarified, In particular by temperature programmed reduction (TPR). Information on the structure of bimetallic catalysts has been obtained with chemical probes, such as chemisorption and reaction rate measurement and physical probes, such as extended X-ray absorption fine structure (EXAFS), scanning transmission electron microscopy (STEM) and Xe-NMR.

A Preliminary Study for Continuous Performance Test by Subtypes of Attention-Deficit Hyperactivity Disorder (주의력결핍 과잉행동장애 하위 유형별 연속수행과제 양상의 예비 연구)

  • Objectives : The purpose of this study was to compare two subtypes of attention-deficit hyperactivity disorder (ADHD) and controls in their attention using ADHD diagnostic system (ADS), a kind of continuous performance test. Methods : Subjects of this study were 8 children with ADHD-predominantly inattentive type, 28 children with ADHD-combined type and 20 control children. Ages ranged from 5 to 14 years. The Korean Educational Development Institute version of the Wechsler Intelligence Scale for Children (KEDI-WISC) and the ADS were administered to all subjects to provide measures of intelligence and attention. Results : Significant differences were found in the ADS variables for the three groups. When compared with controls, the ADHD-predominantly inattentive type children scored significantly higher on the omission error at middle stage and later stage and ADHD-combined type children scored significantly higher on the omission error and reaction time at later stage. When compared with the other two groups, the ADHD-combined type children scored significantly higher on the commission error at all three stages. Conclusion : Our results suggest that the two subtypes of ADHD have different features in attention deficit. And there was a possibility that commission error is available variable to show cognitive characteristics of ADHD-combined type than any other ADS variables. Further studies are needed to evaluate this finding.

Transgenic Expression of MsHsp23 Confers Enhanced Tolerance to Abiotic Stresses in Tall Fescue

  • Tall fescue (Festuca arundinacea Schreb.) is an important cool season forage plant that is not well suited to extreme heat, salts, or heavy metals. To develop transgenic tall fescue plants with enhanced tolerance to abiotic stress, we introduced an alfalfa Hsp23 gene expression vector construct through Agrobacterium-mediated transformation. Integration and expression of the transgene were confirmed by polymerase chain reaction, northern blot, and western blot analyses. Under normal growth conditions, there was no significant difference in the growth of the transgenic plants and the non-transgenic controls. However, when exposed to various stresses such as salt or arsenic, transgenic plants showed a significantly lower accumulation of hydrogen peroxide and thiobarbituric acid reactive substances than control plants. The reduced accumulation of thiobarbituric acid reactive substances indicates that the transgenic plants possessed a more efficient reactive oxygen species-scavenging system. We speculate that the high levels of MsHsp23 proteins in the transgenic plants protect leaves from oxidative damage through chaperon and antioxidant activities. These results suggest that MsHsp23 confers abiotic stress tolerance in transgenic tall fescue and may be useful in developing stress tolerance in other crops.

Effects of Corydalis Tuber on Synthesis of NO and $PGE_2$ in Murine Macrophage RAW 264.7 Cells Stimulated by LPS

  • Corydalis Tuber has traditionally been used for the treatment of water retention in the body. Administration of the aqueous extract of Corydalis Tuber has been known to be effective for the control of pain and treatment of arthritis. It was reported that Corydalis Tuber possesses anti-inflammatory activity and modulates the intestinal immune system. The effect of Corydalis Tuber against LPS-stimulated expressions of COX-2, iNOS, and $IL-1{\beta}$ in cells of the murine RAW 264.7 macrophages was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcription-polymerase chain reaction (RT-PCR), $PGE_2$ immunoassay, and NO detection. The aqueous extract of Corydalis Tuber was shown to suppress $PGE_2$ production by inhibition on the LPS-stimulated enhancement of COX-2 enzyme activity, $IL-1{\beta}$, and iNOS expression in the RAW 264.7 macrophages. Present results suggest that Corydalis Tuber exerts anti-inflammatory and analgesic effects probably by suppressing of COX-2, iNOS, and $IL-1{\beta}$ expressions, resulting in inhibition of $PGE_2$ synthesis. Corydalis Tuber has anti-inflammatory and analgesic effects probably by suppressing of COX-2, iNOS, and $IL-1{\beta}$ mRNA expressions, resulting in inhibition of $PGE_2$ and NO synthesis.

Effect of Kumquat on N-nitrosodimethylamine Formation (금귤이 N-nitrosodimethylamine의 생성에 미치는 영향)

  • When it added to each designed diet groups such as control, nitrate rich, amine rich and nitrate-amine rich diet under simulated gastric conditions, effect of kumquat (Fortunella mararita) juice on formation of N-nitrosodimethylamine (NDMA) was studied. Inhibition on NDMA formation was the highest in the amine rich diet, as 69.4 $\pm$2.4%, when the juice of 10m1 added to its digestate. Phenolic portion of $C_{18}$ sep-pak cartridge in kumquat juice were separated into 20 kinds of a phenolic compounds using HPLC. These phenolic fractions such as faction no. 1, 2, 3, 4 and 5 inhibited from 3.0 $\pm$ 1.2% to 66.5 $\pm$2.0% NDMA formation in the reaction system which was mixture of nitrite and dimethylamine.

Studies on Reforming Gas Assisted Regeneration of Multi-channel Catalyzed DPF (합성가스(Reforming gas)를 이용한 멀티채널 CDPF의 재생 특성 연구)

  • Diesel particulate filter (DPF) systems are being used to reduce the particulate matter emission of diesel vehicles. The DPF should be regenerated after certain driving hours or distance to eliminate soot in the filter. The most widely used method is active regeneration with oxygen at $550{\sim}650^{\circ}C$. Syngas (synthetic gas) can be used to lower the regeneration temperature of Catalyzed DPF (CDPF). The syngas is formed by fuel reforming process of CPOx (Catalytic Partial Oxidation) at specific engine condition (1500rpm, 2bar) using 1wt.% $Rh/CeO_2-ZrO_2$ catalyst. The oxidation characteristics of PM with syngas supplied to filter were studied using partial flow system that can control temperature and flow rate independently. The filter is coated with washcoat loading of $25g/ft^3$ $Pt/Al_2O_3-CeO_2$, and multi-channel CDPF (MC-CDPF) was used. The filter regeneration experiments were performed to investigate the effect of syngas exothermic reaction on soot oxidation in the filter. For this purpose, before oxidation experiment, PM was collected about 8g/L to the filter at engine condition of 1500rpm, bmep 8bar and flow temperature of $200^{\circ}C$ Various conditions of temperature and concentration of syngas were used for the tests. Regeneration of filter started at 2% $H_2$ and CO concentration respectively and inlet temperature of $260^{\circ}C$. Filter Regeneration occurs more actively as the syngas concentration becomes higher.
