Antibiotic Resistance and Virulence Potentials of Shiga Toxin-Producing Escherichia coli Isolates from Raw Meats of Slaughterhouses and Retail Markets in Korea

  • In this study, the prevalence of Shiga toxin-producing Escherichia coli (STEC) was investigated among raw meat or meat products from slaughterhouses and retail markets in South Korea, and their potential for antibiotic resistance and virulence was further analyzed. A total of 912 raw meats, including beef, pork, and chicken, were collected from 2008 to 2009. E. coli strains were frequently isolated in chicken meats (176/233, 75.9%), beef (102/217, 42.3%), and pork (109/235, 39.2%). Putative STEC isolates were further categorized, based on the presence or absence of the Shiga toxin (stx) genes, followed by standard O-serotyping. Polymerase chain reaction assays were used to detect the previously defined virulence genes in STEC, including Shiga toxins 1 and Shiga toxin 2 (stx1 and 2), enterohemolysin (ehxA), intimin (eaeA), STEC autoagglutination adhesion (saa), and subtilase cytotoxin (subAB). All carried both stx1 and eae genes, but none of them had the stx2, saa, or subAB genes. Six (50.0%) STEC isolates possessed the ehxA gene, which is known to be encoded by the 60-megadalton virulence plasmid. Our antibiogram profiling demonstrated that some STEC strains, particularly pork and chicken isolates, displayed a multiple drug-resistance phenotype. RPLA analysis revealed that all the stx1-positive STEC isolates produced Stx1 only at the undetectable level. Altogether, these results imply that the locus of enterocyte and effacement (LEE)-positive strains STEC are predominant among raw meats or meat products from slaughterhouses or retail markets in Korea.

소와 돼지도체에서 Listeria monocytogenes의 분리 및 PCR 검출 방법에 관한 연구 (Isolation and PCR detection of Listeria monocytogenes on raw beef and pork carcass)

  • From February 2000 to December 2001, A total of 1,785 samples was taken from beef and pork carcasses in Seoul. Seven(0.69%) Listeria spp. were isolated from the 1,014 of beef carcasses, and five(0.65%) were isolated from the 771 of pork carcasses. The isolates were identified L monocytogenes by API listeria, and VIDAS LMO kit, serological test and PCR assay were preformed. A total 12 strains of L monocytogenes were isolated form samples tested and all of the strains were classified into serotype 1. PCR primers are selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene(hlyA) of Listeria monocytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested.

소.돼지 도체표면의 미생물학적 고찰 (Survey on the microbiological quality of meat in Seoul)

  • This survey was conducted to evaluate the microbiological quality of raw beef and pork products from January to December in 1999. A total of 107 beef and 157 hog carcasses were collected from two abattoirs located in Seoul. The result showed that beef carcasses had an average bacterial loading around 139,000 bacteria/$\textrm{cm}^2$ of carcass surface, indicating a little bit higher count than the results reported in USA and Australian meat. However, overall hygienic status was found to be acceptable for all examined carcasses because 84.4% of product rated excellent, good or acceptable comparable to USA of 91.6% and Australia of 88%. The analysis of data on overnight-chilled to weekend-chilled carcasses indicated that the microbiological growth occurred in the chiller during the weekend chill with increases in total viable count from 130,000cfu/$\textrm{cm}^2$ to 400,000cfu/$\textrm{cm}^2$. Qualitative testing for escherichia coli, EC + MUG was used as a most probable number (MPN) method along with the petrifilm method. The average of MPN/$\textrm{cm}^2$ of E coli biotype 1 was 29MPN/$\textrm{cm}^2$ for beef carcasses and 1,100 MPN/$\textrm{cm}^2$ for hog carcasses, respectively. However, 41% of beef and 16.3% of hog carcasses were shown to be less than < 3 MPN/$\textrm{cm}^2$ in E coli biotype 1 examination. Although salmonella enteritis, S typhimurium and E coli O157:H7 were all negative, listeria monocytogenes was recovered from only one hog surface samples of the 89 carcasses tested.

Historical Details about the Meat Consumption and Taeniases in Joseon Period of Korea

  • Previous paleoparasitological studies of Joseon specimens established that the prevalence of Taenia infection was not much different from that of the early 20th century Korean population. As many of taeniases originally diagnosed as Taenia saginata in South Korea were revealed to be actually Taenia asiatica, which share a common intermediate host with T. solium (the pig), Joseon people must have ingested raw pork frequently. However, the current examination of extant Joseon documents revealed that the population ate significant amounts of beef even if the beef ban was enforced; and pork was not consumed as much as we thought. Considering the meat consumption pattern at that time, Joseon people should have been infected by T. saginata more frequently than T. asiatica. This may suggest a low prevalence of T. saginata metacestodes in cattle compared to that of T. asiatica metacestodes in pigs, possibly due to the traditional way of rearing pigs (using human feces). This letter gives us a chance to reconsider the existing preconception about parasitic infections in Korean history though we are still hard to accurately estimate the historical patterns of taeniases at this stage.

  • This study was performed to examine antimicrobial resistance of Enterococcus spp. strains from retail raw meat and fish products purchased in 2012. 43 Enterococcus spp. strains were isolated from a total of 207 samples (beef, pork, chicken, fish) with contamination rate of 20.8%. The isolated strains were identified as E. faecalis (22 strains), E. gallinarum, E. hirae (5 strains), E. avium (4 strains), E. faecium (3 strains), E. duram, E. casseliflavus (2 strains). Susceptibility to 10 antibiotics was tested, and the highest resistance was observed to tetracycline. And antimicrobial resistance rates were presented below 20% with most of the other antimicrobial agents. The isolated Enterococci from chicken showed higher resistance also to ciprofloxacin and erythromycin, not only to tetracycline, compared to the isolated Enterococci from beef, pork and fish. Sixteen isolates (37.2%) were sensitive to all antibiotics. Four isolates (9.3%) were resistant to 3 or more antibiotics. Vancomycin-resistant enterococci (VRE) was not identified. According to the results of genetic similarity pattern analysis via PFGE and rep-PCR, Enterococci strains showed different patterns from these collected in 2011. This indicates that there is no genetic similarity among all the strains.

Monitoring of Microbial Contaminants of Beef, Pork, and Chicken in HACCP Implemented Meat Processing Plants of Korea

  • This research was to evaluate microbial contamination levels in meat samples at hazard analysis critical control point (HACCP)-implemented processing plants that produce beef, pork, and chicken. During a period of about a year, a total of 178 samples (76 from beef, 89 from pork, and 13 from chicken) were obtained from raw materials (21.3%) and final products (78.7%). All samples were determined for each 25 g homogenized one. Samples were analyzed to determine the total aerobic plate count (APC), coliform count (CC), and E. coli count (ECC). By month, APC levels were the highest in September and the lowest in February (p<0.001). In comparison among season, APC levels in meat samples were the highest in the summer and the lowest in winter (p<0.001). By month, the highest CC prevalence was found in August, followed by October and then July (p<0.001). By season, the highest CC was obtained in summer, followed by autumn and then spring (p<0.001). All samples were negative for ECC. There was a direct correlation between the product form and coliform presence (p<0.001). In addition, there was a positive correlation between the APC and CC (r=0.261). The APCs in analyzed samples ranged from below <$10^1CFU/g$ to <$10^7CFU/g$. In conclusion, the month and season had significant effects on microbial contamination levels at HACCP implemented processing plants. Interrelationships between (i) the product form and coliform, (ii) the APC and CC were revealed.

식육감별을 위한 미토콘드리아 12S rRNA와 16S rRNA 유전자의 종 특이적 multiplex PCR 기법 개발 (Development of species-specific multiplex PCR assays of mitochondrial 12S rRNA and 16S rRNA for the identification of animal species)

  • Species-specific PCR assay was developed for detection of cattle, sheep, goat, horse, dog, pig, chicken, duck, goose, and turkey using mitochondrial 12S rRNA and 16S rRNA as target genes. Also, an internal positive control was used to detect possible false negatives by using 18S rRNA gene. We designed species-specific primers with amplicon length of 190, 219, 350, 467, 241, 119, 171, 229, 111 and 268 bp for cattle, sheep, goat, horse, dog, pig, chicken, duck, goose, and turkey respectively. The specificity of the primers was tested against the other 10 non-target animal species and a cross-reaction was not observed. We developed two multiplex PCR assays for the simultaneous identification of Korea's major livestock species (cattle, pig, chicken and duck) and poultry species (chicken, duck, goose and turkey) from analogous samples, retaining the same specificity. The limit of detection of the multiplex PCR assay (cattle, pig, chicken and duck) ranged between 1 pg and 0.1 pg of template DNA extracts from raw meat. Applying multiplex PCR assays to DNA extracts from experimental pork/beef and pork/chicken tested raw and heat-treated ($120^{\circ}C$ for 30 min) mixtures respectively, detection limit was 0.1% level beef in pork, pork in beef and chicken in pork and 1.0% level pork in chicken. In conclusion, this assay using gel-based capillary electrophoresis would be very useful in highly sensitive and rapid identification of animal species or ingredients in minced meat and other meat products.

  • This study was investigated traditional food utilization actual conditions of Gwangju and Chollanamdo. It is Ssukgatmuchim, squid debt saliva, Jabanmuchim's order that season, hard-boiled food is beef boiled in soy sauce, mackerel radish hard-boiled food, order of bean curd hard-boiled food, panbroiling ate often by order of Kimchi panbroiling, red pepper anchovy panbroiling, pork panbroiling. Steamed dish is egg steamed dish, fish steamed dish, steamed short-ribs order, fried fish egg speech, by Gimchijeon, Pajeon order, meat roasted with seasoning ate often by laver meat roasted with seasoning, hair-tail meat roasted with seasoning, mackerel meat roasted with seasoning order. 느타리버섯이 육류의 연육에 미치는 효과 (Effect of Oyster Mushrooms on Meat Tenderization)

  • 느타리버섯을 생고기와 익힌 고기에 30분간 도포하여 연육효과를 조사한 결과, 상당한 연육효과가 있는 것으로 나타났다. 생고기의 경우, 버섯 무첨가 시 소고기와 돼지고기의 경도가 각각 981, 640 g-force인데 비해, 전체 5% 첨가군 소고기와 돼지고기의 경도는 각각 648, 487 g-force이었고, 갓 5% 첨가군 소고기와 돼지고기는 각각 642, 478 g-force, 줄기 5% 첨가군 소고기와 돼지고기는 각각 657, 488 g-force로 전반적인 버섯 첨가에 의한 연육효과는 약 20~35%이었다. 연육효과는 첨가량이 많을수록 커, 느타리버섯분말 5% 첨가군과 버섯추출분말 2% 첨가군의 경도가 가장 낮았다. 소고기와 돼지고기의 아미노태질소량은 버섯첨가량이 많을수록 증가하였다. 특히 버섯분말 5% 첨가한 생소고기의 아미노태질소량은 무첨가군에 비하여 전체가 34.64%(748 mg%), 갓이 34.64%(748 mg%), 줄기가 32.92%(739 mg%) 증가하였고, 버섯추출분말 2% 첨가한 생소고기는 전체가 65.14%(787 mg%), 갓이 75.21%(834 mg%), 줄기가 71.18%(815 mg%) 증가하였다. 이는 느타리버섯분말과 추출분말이소고기의 단백질을 가수분해하여 수용성 성분이 증가된 것으로 사료된다. 버섯전체, 갓, 줄기의 부위별 연육효과는 별 차이가 없어 사용부위는 굳이 구별할 필요가 없는 것으로 나타났다.

국내에서 유통되는 8종의 식육감별을 위한 multiplex PCR법 개발 (Development of Multiplex PCR Assay for Identification of Eight Species from Meats in Korea)

  • 본 연구에서는 국내 대표 식육인 소, 돼지, 닭, 오리의 4종 식육과 염소, 양, 말, 칠면조의 4종 식육을 동시에 신속하게 감별할 수 있는 2 set의 multiplex PCR법을 개발하고자 미토콘드리아 16S RNA에서 종 특이부위를 선발하고 각 종에 대한 특이도를 높이기 위하여 인위적인 미스매치를 주어 프라이머를 제작한 후 8종 식육의 274개 시료를 대상으로 특이도와 민감도를 조사하였다. 그 결과 소, 돼지, 닭, 오리 모든 시료에서 각각 279, 94, 192, 477 bp의 증폭산물이, 말, 양, 염소, 칠면조의 모든 시료에서 각각 152 bp, 271 bp, 670 bp, 469 bp에서 뚜렷한 PCR 유전자 산물이 확인되어 모든 축종에서 100%의 특이도를 나타내어 축종별 감별력이 우수한 것으로 나타났다. 8종의 축종별로 DNA를 $10ng/{\mu}l$으로 정량한 후 혼합물을 10배씩 단계 희석하여 반응여부를 조사한 결과, 소, 돼지, 오리에서는 100 fg까지, 닭에서는 1 pg까지 검출됨을 확인할 수 있었다. 소, 돼지, 닭, 오리고기를 99.9%, 99%, 90%, 70%, 50%, 30%, 10%, 1%, 0.1%의 비율로 혼합한 식육과 $83^{\circ}C$ 20분, $100^{\circ}C$ 30분, $121^{\circ}C$ 10분에서 각각 열처리한 가열 혼합육에 대하여 검출한계를 조사한 결과 마지막 단계의 희석 비율인 모든 혼합육의 0.1%에서 검출이 가능하였으며, 열처리 혼합육에서는 닭에서는 1% 농도에서 소와 돼지의 혼합육에서 0.1% 농도에서 검출되어 민감도가 높음을 확인할 수 있었다. 본 연구에서 개발된 multiplex PCR법은 특이도 및 민감도에 있어서 국내 대표 식육을 감별하는데 있어서 유용한 것으로 평가된다.
