낙동강 상수원수의 오존처리 효과 I. 회분식 오존처리에 의한 수처리효과 (Effect of Ozone Treatment for Nakdong River Raw Water I. Efficiency of Water Treatment by Conducting Batch Test of Ozonation Experiments)

  • This study was carried out to evaluate the pollutant removal efficiencies of the advanced drinking water treatment using ozonation. for raw water, Nakdong River was used. By conducting batch ozonation test, the following results were obtained. when ozone dosage of 5mg/$\ell$was used, preozonation of raw water reduced turbidity, KMnO$_4$consumption, DOC(dissolved organic carbon), UV254 absorbance, THMFP(trihalomethane formation potential) as much as 3.9NTU, 5.5mg/$\ell$, 1.15mg/$\ell$, 0.112 and 0.065mg/$\ell$, respectively. In case of postozonation of sand filtered water, water quality was also improved with decrease in turbidity, KMnO$_4$consumption, DOC, UV254 absorbance and THMFP at the amount of 0.08NTU, 2.6mg/$\ell$, 0.88mg/$\ell$, 0.042 and 0.018mg/$\ell$, respectively. On the other hand, contents of dissolved oxygen increased at the level of 1.3mg/$\ell$ after preozonation process and 1.0mg/$\ell$ after postozonation process. The effect of ozone dosage was higher than that of its contact time for the removal of the pollutants.

Baicalein이 Lipoteichoic acid로 자극된 RAW 264.7 mouse macrophages의 hydrogen peroxide 생성에 미치는 영향 (Effects of Baicalein on hydrogen peroxide productions in RAW 264.7 macrophages stimulated by lipoteichoic acid)

  • Objectives : The aim of this study was to investigate the effect of Baicalein (BA) on the production of hydrogen peroxide in lipoteichoic acid-stimulated RAW 264.7 mouse macrophages. Methods : Lipoteichoic acid-stressed RAW 264.7 mouse macrophages were incubated with baicalein at concentrations of 50 and 100 µM. Incubation time is 30 minutes, 2 h, 12 h, and 18 h. After incubation, The production of hydrogen peroxide in RAW 264.7 mouse macrophages was measured with dihydrorhodamine 123 assay. Streptococcus aureus lipoteichoic acid and Streptococcus pyogenes lipoteichoic acid were used as cell-stimulating lipoteichoic acid. Cell viabilities were measured with a modified MTT assay. Berberine, indomethacin, and gallic acid were incubated for the same time as the comparative materials. Results : BA at the concentration of 50 and 100 µM did not show cytotoxicity on RAW 264.7 mouse macrophages for 24 h incubation. For 30 minutes, 2 h, 12 h, and 18 h incubation, BA at the concentration of 50 and 100 µM significantly inhibited the production of hydrogen peroxide in RAW 264.7 mouse macrophages stimulated by Streptococcus aureus lipoteichoic acid (p < 0.05); also, BA at the concentration of 50 and 100 µM also inhibited the productions of hydrogen peroxide in RAW 264.7 mouse macrophages stimulated by Streptococcus pyogenes lipoteichoic acid significantly (p < 0.05). Conclusions : BA might have anti-bacterial activity related to its inhibition of hydrogen peroxide production in lipoteichoic acid-stimulated RAW 264.7 mouse macrophages.

예열처리(豫熱處理)가 원유(原乳)의 단백질(蛋白質) 및 미생물(微生物)에 미치는 영향(影響) (Effects of Pre-heat Treatments on Milk Protein and Microorganism Aspects in Raw Milk)

  • 원료유(原料乳)의 보전성(保存性) 향상을 위하여 원료유와 이를 $65^{\circ}C$에서 30초간 및 $75^{\circ}C$에서 2초간 가열처리(加熱處理)하여 $5^{\circ}C$의 냉장상태에서 4일간 보존(保存)하면서 이화학적(理化學的)인 성장(性狀)과 미생물학적(微生物學的)인 변화(變化)를 시험(試驗)한 결과(結果)는 다음과 같다. 1. 원료유(原料乳) 및 가열처리유(加熱處理乳)의 저장기간중의 우유성분 즉, 유지방(乳脂肪), 유단백질(乳蛋白質), 유당(乳糖) 및 총고형분(總固形分)함량의 변화는 나타나지 않았다. 2. 원료유(原料乳)의 pH와 산도의 범위는 각각 6.73~5.94 및 0.16~0.27%였으며 가열처리유(加熱處理乳)의 pH 및 산도는 각각 6.79~6.62 및 0.16~0.17%이었고 phosphatase시험결과는 모두 음성으로 나타났다. 3. 가열처리온도(加熱處理溫度)가 높아짐에 따라 총질소, NCN, 및 Whey protein의 함량은 감소한 반면 NPN과 Casein의 함량은 증가하였다. 4. 저장기간(貯藏期間)에 따른 총질소 및 Casein의 함량은 감소한 반면에 NCN과 NPN의 함량은 증가 하였으며, Whey protein의 함량에는 변화가 없었다. 5. 가열처리(加熱處理)한 원유는 대장균(大腸菌)이 검출(檢出)되지 않았으나, 가열처리(加熱處理)하지 않은 원유는 저장기간동안 점차적 증가 하였다. 6. 원료유(原料乳)의 총균수는 $5.6{\times}10^6/ml$, 내냉성미생물은 $1.8{\times}10^6/ml$, 내열성미생물은 $1.6{\times}10^5/ml$를 나타내었으나 가열처리(加熱處理) 온도(溫度)의 상승에 따라 감소하여 $75^{\circ}C$에서 2초간 가열처리(加熱處理) 하였을때는 각각 $3.0{\times}10^3/ml$, $1.5{\times}10/ml$$2.7{\times}10^3/ml$으로 감소 하였다. 7. 저장기간(貯藏期間)에 따라 원료유는 내열성미생물, 내냉성미생물 및 총균수가 크게 증가 하였으며 가열처리유(加熱處理乳)는 원료유(原料乳)에 비하여 증가되지 않았다.

  • 유안염석과 column chromatography 및 gel여과 등으로 비활성이 45.2U/mg가 되는 정제된 Rhizopus oryzae의 생전분 분해효소를 16.2%의 수율로 얻었다. 정제효소는 전기영동상에서 그 순도가 인정되었고 분자량은 67000, Km값은 4.082mg/ml이었다. 정제효소는 $50^{\circ}C$, pH $4.0{\sim}5.0$에서 잘 작용하였으며 옥수수amylose가 가장 적합한 전분이었고 옥수수 생전분에 작용한 분해산물은 거의 glucose였다.

메주 제조를 위한 원료 대두의 최적 침지 조건 (Optimum Soaking Condition of Raw Soybean for Meju Preparation)

  • In order to establish scientific foundations for the production of meju, optimum soaking conditions of raw soybean were investigated. 100% hydration of raw soybean by soaking conditions were observed in 17 hrs at 5$^{\circ}C$, 15 hrs at 1$0^{\circ}C$, 11 hrs at 15$^{\circ}C$, 9 hrs at 2$0^{\circ}C$, 7 hrs $25^{\circ}C$, 5 hrs 3$0^{\circ}C$, 4 hrs at 4$0^{\circ}C$ and 3 hrs at 5$0^{\circ}C$. Exudation amounts of total free amino acids and free sugars were the least when soybean was soaked for 2$0^{\circ}C$ for 9 hours.

Physicochemical and Sensory Textural Properties of Rice Extrudate Depending on Extrusion Conditions

  • Extrusion conditions for production of rice extrudate were studied. The optimal production conditions of rice extrudate were determined by the relationship between dependent variables such as expansion ratio, shear strength and color change and independent variables such as moisture content of raw material, screw speed, and die tem-perature of extruder. The textural quality of rice exturdate was significantly affected by the moisture content of raw material (x1), screw speed (x2), and die temperature (x3) of extruder. The expansion ratio of rice extrudate showed the highest value at the moisture content of 18% of raw material, and the lowest at 24%, and whose regression equation was Y=34.8967 - 3.219X1 - (0.623$\times$10-2)X2 + 0.136X3 + (0.648$\times$10-1)X12 + (0.138$\times$10-3)X1X2 + (0.456$\times$10-4)X22 + (0.719$\times$10-3)X1X3 -(0.515$\times$10-3)X2X3- (0.552$\times$10-3)X32. The most desirable texture of rice extrudate determined by shear test and sensory evaluation was obtained at the following conditions : mois-ture content of 18% of raw material, screw speed of 210 rpm and die temperature of 11$0^{\circ}C$. The rice extrudate prepared under the above conditions showed the lowest shear force of 954g at which the highest sensory score was obtained.

저염 오징어젓갈의 제조를 위한 원료어육의 수분활성도와 papain과 glucose의 첨가조건 (Conditions of Water Activity of Raw Material and Adding Levels of Papain and Glucose for Processing Fermented Squid with Low Salt Concentrations)

  • 발효에 의한 향미특성과 저장성이 양호한 새로운 저염 오징어젓갈의 제조기술을 개발하고자 원료어육의 수분활성도와 염농도, papain과 glucose의 첨가농도 등 적정 제조조건에 대하여 조사하였다. 오징어육의 수분활성도를 0.94, 0.90과 0.88로 조절하고 3, 5와 7%의 식염을 첨가하여 제조한 오징어젓갈을 $10^{\circ}C$에서 6주간 숙성하였을 때 아미노태질소는 수분활성도가 높고 염도가 낮을수록 함량이 높았으며 수분활성도 0.90이고 식염농도가 5%인 경우 관능적 품질이 양호하였고 30일 이상 유통가능하였다. 수분활성도를 0.90으로 조절하고 5% 식염으로 염장한 오징어육을 papain의 첨가농도를 달리하여 $10^{\circ}C$에서 숙성하였을 때 papain의 첨가농도가 높을수록 단백질의 분해가 빨랐으며 관능적 품질은 papain 0.1%를 첨가한 처리구가 가장 양호하였다. 또한 수분활성도를 조절하여 5% 식염으로 염장한 오징어육은 1% glucose의 첨가로 유기산 발효를 촉진시켜 5주경에 pH 5.8로 저하하므로서 대조구의 최저 pH 6.13에 비하여 비교적 낮았으므로 저장성 연장효과가 기대되었다.

삼내자 열수추출물의 항산화 및 항염 효과 (Anti-inflammatory and Antioxidant Effects of Hot Water Extracts from Kaempferia Galanga L)

  • 본 연구의 목적은 삼내자(Kaempferia Galanga) 열수추출물의 항산화, 세포독성, 항염 효능을 조사하여 화장품 성분으로서 활용 가능성을 알아보고자 하였다. 항산화 활성 평가를 위해 DPPH, ABTS 라디컬 소거능 활성, FRAP 환원력, 총폴리페놀 함량을 측정하였다. 쥐의 대식세포인 RAW264.7 세포를 대상으로 MTT assay를 수행하여 세포독성을 확인하였다. 항염 효능을 알아보기 위해 LPS로 유도된 RAW264.7 세포에서 nitric oxide(NO), $TNF-{\alpha}$ 분비 및 iNOS, $TNF-{\alpha}$ mRNA 발현 수준을 조사하였다. 그 결과 DPPH와 ABTS 라디칼 소거능은 농도 의존적으로 라디칼 소거 활성이 증가하였다. FRAP 값은 5 mg/mL에서 24.5 uM로 가장 높게 나타났으며, 총폴리페놀 함량은 $1.28{\pm}0.064mg\;GAE/g$로 나타났다. KG 추출물 농도 0.625~2.5 mg/mL에서 세포독성은 보이지 않았다. 또한 RAW264.7 세포에서 LPS 유도에 의한 NO, $TNF-{\alpha}$ 분비 그리고 iNOS, $TNF-{\alpha}$의 mRNA 발현이 KG 추출물에 의해 유의하게 감소하였다. 이러한 결과는 삼내자 열수추출물이 안전하고 효과적인 화장품 원료로서의 활용 가능성이 있음을 보여준다.

강원도 영동권 지역 생선회 전문점의 유형적 근거가 고객의 감정반응과 재구매 행동에 미치는 영향 (The Influence of Tangible Clues of Raw-fish Restaurants on Consumers' Emotional Responses & Repurchasing Behavior in East-northern Area of Kangwon-Province)

  • The purpose of this article was to study the influence of tangible clues of raw-fish restaurants on consumers' emotional responses and repurchasing behavior in eastern area of Kangwon-province. In this study frequency analysis, reliability analysis, factor analysis and path analysis(SEM) were applied to analyze the data. Through path analysis tangible clues' performances had generally significant influences upon consumers' emotional responses to buy products of raw-fish restaurants. According to the results, exterior facility had positive influence on pleasure(p<0.001) and arousal(p<0.01). Interior facility had positive influence on pleasure(p<0.001) and arousal(p<0.001), but negative on dominance(p<0.01). And the others had positive influence on pleasure(p<0.05). Press copy had positive influence on pleasure(p<0.001) and arousal(p<0.001). And Pleasure and arousal had positive influence on repurchasing behavior(p<0.001). But Dominance didn't have significant influence on repurchasing behavior. As a result, we studied that the tangible clues was an effective marketing tactic to maintain and increase consumers' positive emotional reponses to repurchase at restaurants. Therefore it is finally needed that dining-out corporations manage tangible clues in a customer-maintaining without additional costs, and especially to use mixing tangible clues each other in some suitable condition.

한국 연안산 방사무늬김(Porphyra yezoensis)의 일반성분 및 미네랄 함량 (Proximate Composition and Mineral Content of Laver Porphyra yezoensis from the Korean Coast)

  • To measure the proximate composition and mineral content of laver Porphyra yezoensis, we collected 30 raw and 30 dried laver from the major production area of the south coast of Korea (Busan, Goheung, Haenam, Wando). The approximate composition of a 100 g edible portion of raw laver was $89.9{\pm}1.4$ g moisture, $3.7{\pm}1.0$ g protein, $0.5{\pm}0.2$ g lipids, $2.1{\pm}1.8$ g carbohydrate and $3.9{\pm}0.6$ g ash. The approximate composition of a 100 g portion of dried laver was $8.9{\pm}1.6$ g moisture, $31.5{\pm}6.5$ g protein, $1.9{\pm}0.3$ g lipids, $48.4{\pm}6.5$ g carbohydrate and $9.3{\pm}1.1$ g ash. No clear regional variation in laver composition was observed. The mineral content of laver was expressed as dry weight. The mean macro mineral content per 100 g portion of raw laver was (in descending order): K ($1,979{\pm}863.0$ mg), Na ($1,063.2{\pm}498.8$ mg), P ($658.7{\pm}101.8$ mg), Mg ($432.3{\pm}83.5$ mg) and Ca ($394.2{\pm}136.5$ mg). In comparison, the mean micro mineral content of raw laver was (in descending order): Fe ($243.72{\pm}154.75\;{\mu}g/g$), Zn ($72.76{\pm}30.61\;{\mu}g/g$), Mn ($41.53{\pm}15.33\;{\mu}g/g$), Cu ($4.16{\pm}1.66\;{\mu}g/g$) and Ni ($0.43{\pm}0.70\;{\mu}g/g$) No clear regional variation in the mineral content of laver was observed; however, raw laver contained a higher mineral content than dried laver.
