Synthesis of Stigmastanol as a Serum Cholesterol-lowering substance Using Pd Catalyst (Pd 촉매를 이용한 혈청 콜레스테롤 저하제 Stigmastanol의 합성)

  • Stigmastanol, a functional agent of cholesterol-lowering in humans, was synthesized from stigmasterol. To investigate the usability as a raw material, the contents of sterol in vegetable oils and extract of soybean chaff were analyzed. The total sterol contents showed high values of 213.7 and 209.8 mg/100g in corn and soybean oils respectively. The extract of soybean chaff has played a good role as a raw material with high sterol contents. The kinetics of hydrogenation of stigmasterol was studied using a 5% Pd/AC catalyst in the temperature range of 30~$60^{\circ}$C. Increasing temperature showed a prominent decrease in conversion. The optimum temperature was $40^{\circ}$C for high yield of stigmastanol. The effects of $H_2$ pressure, agitation speed, catalyst loading, and stigmasterol concentration on reaction rate profile were also examined. From the power law model analysis using the initial rates of reaction, the reaction order was calculated as 0.705 for stigmasterol concentration and 0.147 for hydrogen pressure.

  • With the intention of preparing cation exchangers from the domestic coals, and using these for softening hard water and some other purposes, seven kinds of raw coal were tested and the results are as follows. 1) The following conditions of preparation were given the good results. Reaction time 5 hours Reaction temperature $95^{\circ}$ Concentration of sulfuric acid 98% Amount of sulfuric acid 10 times to the sample (as weight) 2) The raw coals which is rich in fixed carbon and have the fuel ratio 0.8 to 1.0 were suitable, and Kampo lignite has shown the best results. 3) The cation exchangers from coals were able to exchange the cation, both hydrogen and sodium type dynamically and statically, like the synthetic ion exchange resin. PTCR Effects in KCI Molten Salt Synthesized $BaTiO_3$ (KCI 용융염 합성법에 의한 $BaTiO_3$ 의 PTCR 효과)

  • The PTCR characteristics of the $BaTiO_3$ as a function of the molten KCl and dopant $Sb_2O_3$ were investitated. When the weight ratio of KCl to raw materials was above 0.1, $BaTiO_3$ was synthesized by calcining at 80$0^{\circ}C$ for 6 hrs. As the amount of the KCl increased the resistivity of the $BaTiO_3$ at room temperature incr-eased. This can be explained by charge compensation effect between electrons and holes and with microstruc-tures change of the specimens. The resistivity of the $BaTiO_3$ decreased with increasing amount of $Sb_2O_3$. In the time-current characteristics initial current decreased with increasing the ratio of KCl to raw materials but initial current increased and then decreased with the increase of the dopant $Sb_2O_3$. These results of the time-current characteristics can be explained by the resistivity-temperature characteristics.

Oxidized Cholesterols in Dried Alaska Pollacks (건조 명태 중 콜레스테롤 산화물의 생성)

  • This study attempted to establish the basic data for ensuring the safety of dried Alaska pollack. The changes of such compounds in the curse of drying as cholesterol, malonaldehyde, 7-ketocho-lesterol, 7$\alpha$-, 7$\beta$-hydroxycholesterol and 25-hydroxycholesterol were analyzed. The contensts of cholesterol decreased rapidly in the samples durint sun, hot-air and frozen drying and the decreased levels of cholesterol in dried products were about 134.0~191.3mg/100gas compared with 307.1mg/100g in raw samples. 4.8~6.1$\mu\textrm{g}$/100g in dried samples. In the raw samples, oxidized cholesterols were not detected hydroxycholesterol(34.1~41.7$\mu\textrm{g}$), 7$\beta$-hydroxycholesterol(26.8~40.2$\mu\textrm{g}$/g) and 25-hydroxycholesterol(0.3~1.3$\mu\textrm{g}$/g) were detected. Frozen drying was formed to be the most effective to minimize the formation of oxidized cholesterol in Alaska pollacks.

Bioreduction Characteristics of Perchlorate in Raw Sewage (생하수의 퍼클로레이트 생분해 특성)

  • This research was done to investigate the bioreduction characteristics of perchlorate in raw sewage because sewage contains biodegradable organics and various microorganisms for biological perchlorate reduction. Two different types of sewage were tested for biological perchlorate reduction in the flasks. Sewage A was collected from the screening equipment and sewage B was collected from the primary settlement in the municipal wastewater treatment facilities. Perchlorate was completely reduced within 72hours from 8.2 and 10.4 mg/L in the sewage A and sewage B flask tests. When perchlorate and nitrate were added in sewage A, both perchlorate and nitrate were reduced. However, perchlorate and nitrate removal rates were 9.3% and 64.0% at 72hours in sewage B. Perchlorate reduction was significantly inhibited by high salinity(0.5% NaCl) in the sewage A and B. These results showed the sewage has potential for the biological perchlorate reduction in the sewage pipe.

Diagnostic test for bovine mastitis by the determination of ATP based on firefly bioluminescence (Bioluminescence 반응에 의한 ATP 측정을 이용한 젖소 유방염 진단에 관한 연구)

  • This study was carried out to diagnostic test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are follow; 1. The infection rate of bovine mastitis investigated with 521 cows in 47 dairy farms were found to be 3.6% of clinical form and 44.1% of subclinical form according to the degree of infection. 2. The light yield produced in firefly bioluminescence system was proportional to the concentration of ATP giving stright line within the range of 100PM~1uM. 3. When the number of somatic cell in milk was determined by the ATP assay and compared with three conventional methods such Fossomatic, California mastatic test (CMT), and rolling ball viscometer (RBV), it was shown that r=0.92 for Fossomatic, 0.63 for CMT and 0.7 for RBV. 4. The microorganisms causing mastitis were isolated Staphylococcus sp. (53.3%), Streptococcus sp. (17.9%), Micrococcus sp. (13.5%), Gram negative bacilli (6.3%), Gram positive bacilli (5.5%) and Yeast-like fungi (5.4%). 5. The endogeneous ATP levels of bacteria in a raw milk determined by the firefly bioluminescence system and compared with the results of the conventional methods. The correlation was 0.88 for raw milk.

The immune enhancing effects and characteristics of Bifidobacterium longum and Bifidobacterium breve for the probiotic use in humans and animals

  • The purpose of this study was to investigate probiotic characteristics and immune enhancing effects of Bifidobacterium (B.) longum KBB1-26 and BIF-4, B. breve KBB5-22 isolated from human intestine for probiotic use in humans and animals. We measured acid, bile and heat tolerance, antimicrobial activity against pathogenic bacteria, Escherichia (E.) coli, Salmonella (S.) Enteritidis, Staphylococcus (S.) aureus, and Listeria (L.) monocytogenes. Immune enhancing effects of B. longum and B. breve were investigated by measuring nitric oxide (NO), nuclear factor ($NF-{\kappa}b$), $interleukin-1{\beta}$ ($IL-1{\beta}$), interleukin-6 (IL-6), interleukin-12 (IL-12) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in RAW 264.7 cells or RAW BLUE cells. B. longum KBB1-26 was survived at pH 2.0. B. longum KBB1-26 and BIF-4, B. breve KBB5-22 also showed tolerance to 0.3% of oxgall bile salt. B. longum KBB1-26 was able to survive at $70^{\circ}C$ and $80^{\circ}C$ for 20 min. KBB1-26 showed the antimicrobial inhibition zone to pathogenic bacteria such as E. coli (12 mm), S. Enteritidis (14 mm), S. aureus (14 mm) and L. monocytogenes (41 mm). The production of NO ($4.5{\pm}0.00{\mu}M/mL$) and $IL-1{\beta}$ ($39.7{\pm}0.55pg/mL$) of KBB1-26 significantly higher than BIF-4 and KBB5-22, respectively. In addition, KBB1-26 and KBB5-22 induce the production of high level of $TNF-{\alpha}$ and IL-6 in macrophages. Collectively, B. longum KBB1-26 have acid, bile, heat tolerance, antimicrobial activity and immune enhancing effects. These results suggest that KBB1-26 can be used as probiotics for humans and animals.

Comparison of Quality of Bologna Sausage Manufactured by Electron Beam or X-Ray Irradiated Ground Pork

  • Ground lean pork was irradiated by an electron beam or X-rays to compare the effects of two types of radiation generated by a linear accelerator on the quality of Bologna sausage as a model meat product. Raw ground lean pork was vacuum packaged at a thickness of 1.5 cm and irradiated at doses of 2, 4, 6, 8, or 10 kGy by an electron beam (2.5 MeV) or X-rays (5 MeV). Solubility of myofibrillar proteins, bacterial counts, and thiobarbituric acid reactive substance (TBARS) values were determined for raw meat samples. Bologna sausage was manufactured using the irradiated lean pork, and total bacterial counts, TBARS values, and quality properties (color differences, cooking yield, texture, and palatability) were determined. Irradiation increased the solubility of myofibrillar proteins in a dose-dependent manner (p<0.05). Bacterial contamination of the raw meat was reduced as the absorbed dose increased, and the reduction was the same for both radiation types. Differences were observed only between irradiated and non-irradiated samples (p<0.05). X-ray irradiation may serve as an alternative to gamma irradiation and electron beam irradiation.

A Study on Haloacetic Acids Formation Potentials by Chlorination in Drinking Water (상수의 염소처리시 생성되는 소독부산물 중 Haloacetic acid류의 생성능에 관한 연구 - 일부 상수원수를 대상으로 -)

  • The main reason of applying chlorination is to sterilize microbes existing in the drinking water treatment. But chlorination could lead to the formation of disinfection by-products (DBPs) by the reaction of free chlorine with humic substance in the water. Especially the DBPs including trihalomethanes (THMs), haloacetic acids (HAAs), haloacetonitriles (HANs), and haloketones (HKs) exist in the tap water. The US environmental protection agency (US EPA) defines that trihalomethanes, dichloroacetic acid, trichloroacetic acid, and dichloroacetonitrile among DBPs are probable/possible human carcinogens. US EPA suggests maximum contaminant levels (MCLs) for THMs (80$\mu$g/L) and HAAs (60$\mu$g/L) in drinking water. In Korea, THMs in drinking water has been surveyed but DBPs in general has not been studied in drinking water practically. Therefore only THMs have been regulating as criteria compounds since 1990 but neither HAAs nor HANs. Researches on HAAs are yet to be found. HAA formation potentials(HAAFPs) have not been practiced. HAAs depends on the characteristics of water sources by chlorination. In this study, HAAFPs from three distinct sources were investigated by laboratory chlorination experiments. This study was performed to measure the level of HAAs in drinking water in Seoul area. At April 1996, after collecting the raw waters from the three sites with the different properties, the water samples were chlorinated at various conditions(pH 5.5, pH 7.0 and without pH adjustment) in the state of raw water to have 0. 5mg/L of residual chlorine concentration. And the raw water, treated water, and tap water of water treatment were collected to measure the HAAs concentration. The quantitative analysis of HAAs was conducted by US EPA methods.

Identification and Antibacterial Activity of Volatile Flavor Components of Cordyceps Militaris

  • Flavor characteristics of raw Cordyceps militaris significatntly different from those of dried one. In the case of raw Cordyceps militaris , major flavor components were composed of 5 alcohols, 3 ketones, 4 phenols, 9 alkanes , and 3 alkenes. The major alcohol was 1-octen-3-ol(22.56%, 1147.3% ng/ml), which contributed to the characteristic green flavor. Ketones (3-ocatone, inparticular )were present in the highest concentration in raw Cordyceps militaris . In contrast, major flavor components of dried Cordyceps militaris were composed of 4 alcohols, 4 ketones, 3 furans, 4 pyrizines, 2 dithiazines, 5 phenols , 8alkenes , 17 alkanes, and 8 fatty acids. Dried Cordyceps militaris had unique sweet aroma of sesame as wella s a milky flavor. Green or fruit flavor were rarely detected . In alkanes , 10 cosanes, component fo wax were present. Typical flavor components of alkanes such as $\beta$-caryophyllen and Δ-cadinene were also detected. Fatty acids of dried Cordyceps militaris ranged from myristic acid (14 :0) to linoleic acid (18 ; 2). The sweet aroma of dried Cordyceps militaris was mostly due to pryazines, dithaiazines, and furans. Two dithaizines were identified and characteristics of these flavor components was a roasted bacon flavor. Strong antibacterial acitivity was observed toward Vibrio spp. such as V. vulnificus, V.cholerae, V. parahaemlyticus. Relatively high antibacterial acitivity was shown toward Bacillus subtilis , B,cereus, Staphyllococcus aureus, and Corynebacterium xerosis.
