천연 및 가교 키토산에 의한 희토류 원소의 흡착과 회수에 관한 연구 (A Study on the Adsorption of Rare Earth Elememts by Raw and Crosslinked Chitosan)

  • 본 연구에서는 천연 키토산과 glutaraldehyde을 이용하여 가교한 가교 키토산을 사용하여 희토류 금속과의 흡착특성을 조사하였다. 최대 흡착량을 나타내는 최적 pH 영역은 $Nd^{3+}$$Tm^{3+}$ 금속이온의 경우, 천연 및 가교 키토산에 대해 4.5~5.5 영역이었으며, $La^{3+}$$Ce^{3+}$ 금속이온의 경우, 가교 키토산은 pH 4. 0~5.5 영역, 천연 키토산은 pH 2.0에서 최대 흡착량을 보였다. 단일상 (pH 4.0)에서 희토류 금속의 최대 흡착량은 $Er^{3+}$ > $Gd^{3+}$ > $Yb^{3+}$ > $Nd^{3+}$ > $Lu^{3+}$ > $Eu^{3+}$ > $Tm^{3+}$ > $Ho^{3+}$ > $Dy^{3+}$ > $La^{3+}$ > $Ce^{3+}$ > $Y^{3+}$ > $Pr^{3+}$ 같은 순서로, 혼합상에서는 $Lu^{3+}$ > $Yb^{3+}$ > $Tm^{3+}$ > $Dy^{3+}$ > $Ho^{3+}$ > $Er^{3+}$ > $Eu^{3+}$ > $Gd^{3+}$ > $Nd^{3+}$ > $Y^{3+}$ > $La^{3+}=Ce^{3+}=Pr^{3+}$의 순서로 경쟁적 흡착반응을 나타내었다. 혼합상에서 희토류 금속의 경쟁반응은 원자번호가 증가할수록, 그리고 이온반지름이 작을수록 높은 흡착량을 나타내었다. 최대 흡착량에 도달하는데 걸리는 흡착평형시간은 이전 금속이온과의 연구에서와 상이하게 5시간 이상 소요되었다. 키토산에 대한 희토류 금속이온의 회수율은 $Nd^{3+}$ 이온의 경우, 83~95%이고, $Tm^{3+}$ 이온의 경우, 90~106%을 나타내었다.

이용도가 낮은 수산자원의 효소적 가수분해 조건 (Enzymatical Hydrolysis of Low-Usefulness Marine Resources)

  • In present work, the development of processing for various fermented sea foods using low-usefulness marine resources were investigated. The optimum temperatures of autolysis were 35$^{\circ}C$ for hair tail, 45$^{\circ}C$ for gizzard shad, 30$^{\circ}C$ for kangdale, 30$^{\circ}C$ for pen shell and 30$^{\circ}C$ for oyster and when alcalse(Novo Co.) were added, optimum temperatures were 60$^{\circ}C$, 50$^{\circ}C$, 50$^{\circ}C$, 50$^{\circ}C$ and 50$^{\circ}C$, respectively, and protease N, P. (Pacific chem. enzyme mixture 2,000) were 55$^{\circ}C$, 60$^{\circ}C$, 50$^{\circ}C$, 50$^{\circ}C$ and 50$^{\circ}C$, respectively. Especially although exozymes and endozymes reacted at same time, hydrolysis rate of raw materials got to maximum at optimum temperatures of exozymes. The facts showed that exozymes dominated the hydrolysis reached max8imum at pH 9.0, and optimum hydrolysis time of all raw materials were 6 hours. And the optimum concentrations of exozymes were about 3.0% for hair tail, 4.5% for gizzard shad, 3.5% for kangdale, 3.0% for pen shell and 3.0% for oyster, respectively.

Evaluation of normal morphology, DNA fragmentation, and hyaluronic acid binding ability of human spermatozoa after using four different commercial media for density gradient centrifugation

  • Objective: Density gradient centrifugation (DGC) is frequently used to isolate high-motility fractions of spermatozoa. We compared the efficacy of four DGC media in terms of the percentage of morphologically normal spermatozoa, DNA fragmentation level, and hyaluronic acid (HA) binding ability. Methods: Thirty men with a total motile spermatozoa count > 80 million participated. Semen samples were divided into four aliquots, which were processed using PureSperm, PureCeption, Sidney, and SpermGrad media, respectively. The DNA fragmentation level was measured using the Halosperm assay kit and HA binding ability was measured using the HBA assay kit. Results: The mean percentage of morphologically normal spermatozoa was significantly enhanced after DGC using all four media (10.3%, 9.9%, 9.8%, and 10.7%, respectively; p< 0.05 for each when compared with 6.9% in raw semen). The DNA fragmentation level was significantly reduced after DGC using PureSperm, PureCeption, and SpermGrad media (6.0%, 6.5%, and 4.9%, respectively; p< 0.05 for each when compared with 11.2% in raw semen), but not after DGC using Sidney media (8.5%, p> 0.05). HA binding ability did not change after DGC using any of the four media. Conclusion: The four media were equally effective for obtaining a sperm fraction with highly motile, morphologically normal sperm. PureSperm, PureCeption, and SpermGrad media were equally effective for acquiring a sperm fraction with less DNA fragmentation.

참나리(Lilium lancifolium) 구근을 이용한 민속발효주의 제조 및 생리활성 (Manufacture and Physiological Functionality of Korean Traditional Alcoholic Beverage by Using Lily (Lilium lancifolium) Scales)

  • 백합구근을 식품으로 활용하기 위한 일환으로 구근을 첨가한 전통민속발효주를 개발하기 위하여 알코올 발효 조건을 검토하고 그의 생리기능성을 조사하였다. 백합구근을 건조분말과 생구근 상태로 첨가량을 달리하여 발효한 결과, 구근건조분말 첨가량이 증가할수록 발효 초기에는 에탄올생성량이 무첨가구보다 낮았으나 발효 후기로 갈수록 에탄올 생성량이 높았으며, $25^{\circ}C$에서 10일간 발효시켰을 때 건조분말 15% 첨가 발효주와 생구근 20% 첨가 발효주의 에탄올함량이 각각 19.4%, 19.0%이었다. 첨가량에 따른 발효주의관능검사 결과 백합구근 건조분말을 5% 또는 생구근 20%정도 첨가하였을 때 색, 향, 맛 및 전체적인 기호도면에서 가장 기호도가 좋았으며, 특히 건조분말을 첨가하였을 때가 생구근 첨가 발효주보다 기호도가 더 높았다. 기호도가 가장 좋았던 건조분말 5% 첨가 발효주와 생구근 20% 첨가 발효주의 생리기능성을 측정한 결과 전자공여능, SOD 유사활성, ACE 저해활성 및 tyrosinase 저해활성이 무첨가구인 대조구보다 우수하였으며, 생구근 첨가 발효주보다 건조분말 첨가 발효주가 생리기능성이 약간 더 높았다.

Effect of Ground Chopi (Zanthoxylum piperitum) on Physicochemical Traits and Microbial Community of Chicken Summer Sausage during Manufacture

  • Changes in microbial community and physicochemical traits of chicken summer sausage made from spent layer thigh added with different level (0%, 0.1%, 0.3%, and 0.5% w/w) of ground chopi (Zanthoxylum piperitum) during manufacture were analyzed. The microbial community was profiled and analyzed by sequencing 16S rRNA gene using Illumina MiSeq. Samples were taken from raw sausage batter, after 15 h of fermentation, 8 h of cooking including cooling down, and 7 d of drying. The final pH of the sausage was reduced by the addition of ground chopi. However, no clear effect on water activity was observed. Ground chopi inhibited the development of red curing color after fermentation as it exhibited antimicrobial effect. However, the effect on species richness and microbial composition after cooking was unclear. Ground chopi delayed lipid oxidation during manufacture and the effect was dependent on the addition level. Fermentation reduced the species richness with a dominancy of lactic acid bacteria. The profile of microbiota in the raw batter was different from other stages, while the closest relationship was observed after cooking and drying. Proteobacteria was predominant, followed by Firmicutes and Bacteroidetes in raw samples. Firmicutes became dominating after fermentation and so forth, whereas other predominant phylum decreased. At genus level, unclassified Lactobacillales was the most abundant group found after fermentation and so forth. Therefore, the overall microbial composition aspects were mainly controlled during fermentation by the abundance of lactic acid bacteria, while bacterial counts and lipid oxidation were controlled by cooking and the addition of ground chopi.

pH, 수온, 염소주입량이 정수장 소독부산물 생성에 미치는 영향 (Effects of pH, Water Temperature and Chlorine Dosage on the Formation of Disinfection Byproducts at Water Treatment Plant)

  • 본 연구는 경북 소재 A정수장의 원수를 대상으로 pH, 수온, 염소주입량에 대한 소독부산물(trihalomethanes, haloacetic acids, haloacetonitriles, chloral hydrate 등 16종) 생성능을 조사하고 여과수의 소독부산물 생성능과 비교하였다. 조사한 소독부산물의 생성능은 대부분 중성 pH에서 높았다. 다만, dichloroacetic acid, chloroform, bromodichloromethane의 생성능은 염기성 pH에서 증가하였다. Chloral hydrate, haloacetic acids, haloacetonitriles의 생성능은 수온의 증가와 함께 직선형으로 증가하였고, trihalomethanes의 생성능은 지수함수의 증가추세를 보였다. Chloral hydrate, trihalomethanes, trihaloacetonitriles의 생성능은 염소주입농도 2.0 mg/L $Cl_2$까지 급격하게 상승하였고, 2.0~5.6 mg/L $Cl_2$에서는 완만하게 증가하였다. 여과수의 소독부산물 생성능은 원수에 비해 대부분의 물질에서 낮게 나타났다. 다만, trihalomethanes은 여과수가 원수보다 약 1.4배 높은 생성률을 나타냈다.

단백질 및 지질함량이 다른 부상배합사료와 생사료의 넙치 사육효과 비교 (Evaluation of Extruded Pellets Containing Different Protein and Lipid Levels, and Raw Fish-Based Moist Pellet for Growth of Flounder (Paralichthys olivaceus))

  • This study was conducted to evaluate the effect of extruded pellets (EP) containing different levels of protein (51%-55%) and lipid (9%-15%) for growth of flounder (Paralichthys olivaceus) comparing with raw fish-based moist pellet (MP). Two replicate groups of 40 fish per each tank (initial mean weight 106 g) were fed one of three experimental EP (EP1, EP2 and EP3) containing different protein and lipid levels, a commercial EP (EP4) and MP for 16 weeks. Survival was not significantly different among all groups. Final mean weight of fish fed MP was significantly lower than that of fish fed EP1, EP2 and EP4 (P<0.05), but not significantly different from fish fed EP3. Feed efficiency of MP-fed fish was significantly lower than fish fed all EP formulations (P<0.05), but no significant difference was observed among the EP groups. Daily feed intake of MP-fed fish was significantly higher than fish fed all EP formulations (P<0.05). Condition factor was not significantly different among all groups. Whole body moisture and crude lipid contents were significantly affected by diet (P<0.05). Growth and feed efficiency of flounder was not affected by EP protein and lipid levels. Dietary formulation used in EP1, EP2 and EP3 can be applied to the practical feeding of flounder.

Physicochemical Properties of Dioscorea opposita Thunb. Solution Fermented by Lactobacillus bulgaricus

  • The present study was designed to determine the physicochemical properties of yam (Dioscorea) solution fermented by L. bulgaricus, the protective effect of fermented Dioscorea opposita Thunb. on stomach lesions, and the possibility of its application to fermented dairy products. When the concentration of yam increased, the pH increased in raw powder samples but not in extracted powder samples. In 6% raw powder containing the sample, the viable cell counts showed a dramatic increase, whereas those in extracted yam powder significantly decreased in all concentrations during 32 h of fermentation. The viscosity of extracted yam powder showed a lower value than that of raw powder, which was maintained throughout the fermentation. The solution of Dioscorea opposita Thunb. raw powder showed a significantly higher allantoin content compared with the extracted powder (p<0.05). At 2% of added Dioscorea opposita Thunb., 0.65 mg/mL was contained in both samples; however, when 6% of Dioscorea opposita Thunb. was added, the amount of diosgenin increased up to 1.9 mg/mL. When 200 mg of powder/kg body wt. was injected, gastric lesions were apparently reduced compared with those of control rats. With 100 mg/kg body wt. injected into rats, the protective effect was a little lower than that in 200 mg/kg body wt. Based on these data, the present study indicated that yam powder fermented by L. bulgaricus showed a protective effect on gastric lesions in rats; therefore, it may be included as an added ingredient in yogurt manufacture.

미정제 석탄회를 혼합골재의 미립분 보충재로 활용하는 시멘트 모르타르의 공학적 특성 분석 (Analyzing the Engineering Properties of Cement Mortar using Raw Coal Ash as a Microfines for the Mixed Aggregate)

  • 본 연구는 국내의 부족한 골재자원확보, 폐기물 재활용 및 콘크리트의 품질향상을 목적으로 시도되었다. 즉, 잔골재의 혼합은 굵은 입자를 많이 포함하여 표준입도범위를 벗어나는 석산B(CSb) 및 건설현장의 터파기한 발파석을 분쇄한 발파석(BS)과 미세한 입자를 많이 포함하여 표준입도범위를 벗어나는 해사(SS)를 혼합하는 것으로 한다. 또한, CSb+SS, BS+SS를 혼합골재로 제조할 경우, 전반적으로는 표준입도범위에 포함될지라도 0.15mm이상의 미립자가 부족하므로서 모르타르 및 콘크리트 품질에 결함으로 나타날 수 있다. 따라서 여기에 겨울철 초기강도에 기여하지 못하므로서 버려지는 화력발전소의 미정제 석탄회(Ra)를 0~10% 범위에서 치환하는 것을 검토하였다. 연구결과, CSb 혹은 BS와 SS를 6:4의 비율로 혼합하여 혼합골재로 제조하고, 여기에 Ra를 5% 치환할 경우 골재의 입도측면 및 시멘트 모르타르의 품질 측면에서 가장 양호한 결과가 얻어짐을 확인 할 수 있었다.

생태교란종 영국갯끈풀의 기능성화장품 원료로서 효능 연구 (A Study on functional cosmetic ingredients of the invasive plant Spartina anglica)

  • 본 연구에서는 생태교란종으로 지정되어 대량으로 방제되는 영국갯끈풀의 에탄올 추출물을 활용하여 여러가지 생리활성을 평가하고 화장품 원료로서의 활용 가능성을 확인하고자 하였다. 항산화 활성은 라디컬 소거법인 DPPH와 ABTS assay 및 환원력을 통한 FRAP과 같은 화학적 방법으로 조사한 결과, 영국갯끈풀 지상부 에탄올 추출물의 항산화 활성이 우수하였다. 각 추출물 농도 0.5 mg/mL 이하 조건에서 RAW264.7 cell과 NHDF cell에 대한 세포독성은 관찰되지 않았다. LPS로 활성화 된 RAW264.7 세포에서 증가된 염증매개물질 NO와 사이토카인 생성량은 지하부 에탄올 추출물에 의해 뚜렷하게 감소되었다. 또한 NHDF cell을 지상부와 지하부 두 추출물로 처리하여 collagen 합성과 피부 수분 유전자 발현을 조사한 결과 높은 항주름 및 보습 효능을 나타냈다. 이와 같은 결과를 통하여 영국갯끈풀 에탄올추출물은 기능성화장품의 생리활성 물질에 대한 유용한 바이오 자원으로서의 활용 가능성이 우수함을 확인하였다.
