Comparative analysis of nutritional values of riverine and marine hilsa (Tenualosa ilisha; Hamilton, 1882)

  • A study was performed to analyze the biochemical composition (moisture, protein, fat, ash, salt value, iron, calcium and phosphorus) of raw and salted hilsa. Pure (with less than 1% impurities) and clean dry salt was used (fish weight : salt weight = 3 : 1) for salting the hilsa. The nutrients values of the hilsa from two different regions were significantly (p < 0.05) varied. The biochemical compositions were also different before and after the processing of the hilsa. Riverine hilsa contains relatively more moisture ($57.79{\pm}0.51%$) and protein ($15.65{\pm}0.50%$) than marine hilsa. Fat ($16.39{\pm}0.51%$) and salt ($1.80{\pm}0.14%$) contents are higher in marine hilsa; whereas the ash ($7.88{\pm}0.35%$) content was higher in the riverine hilsa. Minerals like iron ($4.92{\pm}0.32mg/100g$) and calcium ($480.02{\pm}6.73mg/100g$) remain in large amounts in the marine hilsa, but the phosphorus ($112.36{\pm}4.40mg/100g$) content remains at a high level in the riverine hilsa. In addition, the protein (raw condition, $18.54{\pm}0.46%$, riverine; $17.12{\pm}0.42%$, marine and salted condition, $32.54{\pm}0.5%$, riverine; $27.31{\pm}0.48%$, marine) and fat (raw condition, $15.41{\pm}0.46%$, riverine; $19.07{\pm}0.51%$, marine and salted condition, $11.58{\pm}0.39%$, riverine; $13.6{\pm}0.55%$, marine) contents were higher in the abdominal region of the riverine and marine hilsa both in the raw and salted conditions than in the head and caudal region.

생선묵의 보장성에 관한 세균학적 연구 (Microbiological studies on the preservation of fish-paste products)

  • This study is concerned to the microbial population and its effects in the fish paste products. Experimental results were summarized as following orders ; 1) The number of bacteria in accordance with raw materials and coocking site waas known to as ; Wheat flor - $7.0{\times}10^8$/gram , Brayed fish meat - $2.0{\times}10^7$/gram, Cooking table - $6.1{\times}10^6$/$1.54cm^2$Chopped fish meat -$4.1{\times}10^6$/gram, Wooden plate - $5.5{\times}10^5$/218.32cm, Sodium chloride - $8.8{\times}10^4$/gram, Wheat starch - $4.5{\times}10^4$/gram, Fish meat - $2.3{\times}10^4$/gram. However, there were a few bacterial numbers less than 300 per gram of raw materials, such as monosodium glutamate, tap water, sugar, saccharine and eggs. 2) Bacterial population in the raw materials might be changed according to the conditions of preservation. 3) When the heat tratment is used for the preservation, the thermal effects were rarely found at the interior part of fish paste products. Conduction of heat into the interior part dose not rise about $84^{\circ}C$ and the survival of bacteria could be possible under the condition. Therefore, cooking being used requires further studies. 4) Number of bacteria in the fish paste products sold in the market was higher on April and May than those on June. Especially on June, the number of bacteriz in the interior part were found higher than those in the exterior part of the products.

모과(木瓜) 물추출물의 항염증 효능에 관한 실험적 연구 (The Antiinflammatory Effects of Chaenomelis Fructus Herba Water Extract on Mouse RAW 264.7 Cell)

  • Objectives: The purpose of this study was to investigate the effects of Chaenomelis Fructus Herba Water Extract(CF) on the production of inflammatory mediators in RAW 264.7 cell mouse macrophages stimulated with LPS. Methods: We have not examined effect of CF on the cell viability of RAW 264.7 cell until we investigated effects of CF on LPS-induced productions of NO, Ca and various cytokines in RAW 264.7 cell. And when p-value is below 0.05, it is judged to have the significant difference statistically(P<0.05). Results: 1. CF increased the cell viability in the RAW 264.7 cell at the density of 25, 50, 100 and 200 ${\mu}g/ml$. 2. CF inhibited significantly increasing the production of NO in LPS-induced RAW 264.7 cell at the density of 25, 50, 100 and 200 ${\mu}g/ml$. 3. CF inhibited significantly increasing the production of Intracellular Ca in LPS-induced RAW 264.7 cell at the density of 25, 50, 100 and 200 ${\mu}g/ml$. 4. CF inhibited significantly the IL-2, IL-10, IL-12p70, TNF-${\alpha}$, GM-CSF, M-CSF, LIF and VEGF of the RAW 264.7 cell induced by LPS at the density of 25, 50, 100 and 200 ${\mu}g/ml$. 5. CF inhibited significantly the IL-4 at the density of 25, 50 ${\mu}g/ml$, the IL-5, IL-15 and MIG at the density of 25, 50 and 200 ${\mu}g/ml$ and IFN-${\gamma}$ at the density of 25, 100 ${\mu}g/ml$ respectively in the RAW 264.7 cell increased by LPS. Conclusions: CF inhibited significantly increasing IL-2, IL-10, IL-12p70, TNF-${\alpha}$, GM-CSF, M-CSF, LIF, VEGF, NO and Ca in LPS-induced RAW 264.7 cell at the density of more than 25 ${\mu}g/ml$ without causing the toxicity. These results signify that CF has antiinflammatory effect on controlling the over inflammatory reaction by the RAW 264.7 cell.

상수도계통에서의 세균 분포 및 변화 (Bacterial Distribution and Variation in Water Supply Systems)

  • 1991년 9월부터 1992년 9월까지 23회에 걸쳐 서울시 구의정수장의 상수도계통 즉 원수와 정수 및 이곳에서 공급되는 단일 배급수계통의 3개 수돗물에서 종속영양세균(heterotrophic plate count : HPC) 과 장내세균(Enterobacteriaceae : total coliforms) 의 분포를 조사하였다. $R_{2}A$ 한천배지에서의 종속영양세균수는 원수 $1.22{\times}10^{3}~3.05{\times}10^{5}$ 정수 $1.50{\times}10^{1}~4.29{\times}10^{3}$, 수돗물 $2~5.41{\times}10^{3}$ cfu/100ml, 수돗물 0-47.5 cfu/100 ml 의 범위였으며, m-T7 한천배지에서의 손상된장내세균을 포함하는 장내세균수는 전수 0-27, 수돗물 0-35 cfu/100 ml/의 범위였다. 이러한 결과로부터 정수장에서 공급된 물이 배급수관을 통하여 공급되면서 종속영양세균 및 장내세균의 개체수가 증가하는 세균재생장 현상이 발생한다는 것을 확인하였다. 종속영양세균의 동정결과 원수, 정수, 수돗물을 포함하는 상수도계통의 물에서는 Peudomonas 와 Acinetobacter 가 우점하였으나, 원수에서는 Pseudomonas 의 비율이, 그리고 정수와 수돗물에서는 Acinetobacter 의 비율이 상대적으로 높은 것으로 나타났다. 장내세균은 Enterobacter agglomerans 가 우점종이었으며, 수돗물에서는 정수에서 검출되지 않았던 Citrobacter freundii, Escherichia coli, Klebsiella pneumoniae, Shigella dysenteriae 등의 기회성 및 병원성 장내세균도 검출되었다.

Protein Quality Evaluation of Cooked Hagfish (Eptatretus burgeri) Meats

  • The effect of cooking methods on in vivo and in vivo indices of the protein quality of hagfish meat were investigated. In vivo protein digestibilities of cooked meats (81.3~83.5 %) were not significant different (p<0.05) from those of van meat (82.9%), with the exception of steamed (11$0^{\circ}C$, 15 min) meat (86.3 %). Convection oven cooking (22$0^{\circ}C$, IS min) resulted in a higher trypsin indigestible substrate (TIS, 49.2 mg/g solid) compared with that of raw meat (38.9 mg/g solid). free amino acid content of raw meat was decreased after boiling (10$0^{\circ}C$, 10min). Both convection oven and microwave cooking (2,450 MHz, 3 min) decreased available lysine from 4.9g/16g N to 3.8~4.1g/16g N. In vivo apparent protein digestibilites (AD) of hagfish meat were similar fur raw (92.4%) and cooked meats, but were somewhat lower than ANRC (Animal Nutrition Research Council) casein (945%). The PERs (3.7~4.1) and NPRs (3.7~4.9) of cooked meats were significantly higher (p<0.05) than those of raw meat (PER 3.3, NPR 3.6 and ANRC casein (PER 2.5, NPR 2.6), despite their lower in vivo protein digestibilities. These results demonstrate that cooking at optimal conditions resulted in remarkably positive effects on in vivo and in vivo protein qualities of hagfish meats. Therefore, steamed hagfish meat is an excellent source of high quality protein from seafood products.

LPS로 유도된 RAW 264.7 세포에 대한 흑색 방울토마토 주스의 항염증 효과 (Anti-inflammatory Effects of Black Cherry Tomato (Lycopersicon esculentum M.) Juice on LPS-induced RAW 264.7 Cells)

  • 본 연구에서는 흑색 방울토마토 주스의 항산화 활성, 아질산염 소거능, 알코올 분해능, 및 RAW 264.7 세포에서의 항염증 활성에 미치는 영향을 알아보았다. 흑색 방울토마토 주스의 총 페놀 함량은 $156.83{\mu}g\;TAE/ml$로 나타났다. 토마토주스의 항산화활성은 DPPH 라디칼 소거능과 SOD 유사활성으로 측정하였다. 토마토주스의 DPPH radical 소거능은 농도의존적으로 현저히 증가하였으며 40%의 농도에서의 83.39%로 나타났다. SOD 유사활성은 100% 농도에서의 SOD 유사활성은 22.01%로 나타났다. 흑색 방울토마토 주스의 숙취 해소능을 알아보기 위하여 체내 알코올 대사의 일차 효소인 ADH 활성 및 아세트알데히드를 분해하는 ALDH 활성을 알코올 분해와 숙취해소에 효과가 있는 것으로 알려진 hepos를 대조구로 하여 분석한 결과, ADH 활성 및 ALDH 활성 모두 농도 의존적으로 증가하였으며(p<0.05), 40% 농도에서의 ADH 활성과 ALDH 활성은 각각 198.87%와 181.89%로 높은 활성을 보였다. 아질산염 소거능 분석에서는 흑색 방울토마토 주스 50% 농도, pH 1.2, 3.0, 6.0에서 각각 85.06, 58.25, 43.68%로 pH가 낮을수록 아질산염 소거능이 증가하는 것으로 나타났다. 흑색 방울토마토 주스의 항염증 활성을 측정하기 위하여 LPS에 의해 유도된 RAW 264.7 대식세포의 NO 합성을 측정한 결과 농도 10%에서의 NO 합성은 1.28%로 LPS 처리군(7.79%)보다 83.57% 현저하게 감소하였다(p<0.05). 이상의 결과에서와 같이 흑색 방울토마토 주스는 항산화 활성과 알코올 분해능 및 항염증 효과가 높은 것으로 나타났기에 기능성 식품으로서의 가치가 높을 것으로 판단된다.

저열량 생즙 식이가 성인의 혈청 페리틴 농도에 미치는 영향 (Effects of a Low-Calorie Raw Juice Diet on the Level of Serum Ferritin in Korean Adults)

  • This study examines the effects of a low-calorie raw juice diet on the level of serum ferritin in adults and analyzes nutrient intake from the diet. There were significant differences between juices; the highest calorie was provided by pear juice, highest crude protein, vitamin A, and vitamin B2 levels were from green Juice 1; and highest vitamin C and vitamin B1 levels were from fruit juices. The ratio of estimated energy requirements (EER) for the participants was 56.2% from the raw juice diet. The percentages of recommended intake (RI) from the raw juice diet of protein (57.9%), dietary fiber (19.1%), niacin (6.2%), calcium (0.1%), and magnesium (0.2%) were lower than 75%. However, those of RI of vitamin A, vitamin B1, vitamin B2, vitamin B6, and vitamin C were 1796.5%, 7481.7%, 1915.5%, 30858.7%, and 7500%, respectively, exceeding the tolerable upper intake level (UL) for vitamin A, vitamin B6, and vitamin C. There were significant decreases in weight, the body mass index (BMI), body fat mass, and skeletal muscle mass in males and females. After the diet program, serum iron and SOD (superoxide dismutase) showed significant decreases, whereas RBC, hemoglobin, hematocrit, and serum ferritin showed significant increases. There were negative correlations between serum ferritin and weight and between serum ferritin and skeletal muscle mass for all participants. There were negative correlations between serum ferritin and skeletal muscle mass for males and between serum ferritin and body fat mass for females. These results suggest that a raw juice diet can supplement a regular diet to prevent excess or deficient nutrient intake.

LPS로 유도된 RAW264.7 세포주 염증모델에서 소리쟁이의 법제처리에 따른 항염증 효과 (Anti-Inflammatory Effects of Beopje Processed Curly Dock (Rumex crispus L.) in LPS-Induced Murine RAW 264.7 Cell Lines)

  • This study investigated the anti-inflammatory effects of processed (Beopje) curly dock (Rumex crispus L.) in LPS (lipopolysaccharide)-stimulated murine RAW 264.7 cells. The experimental group was classified into five groups : LPS no treatment, CD (curly dock), CD-B (CD processed through Beopje), LPS, LPS+CD-B (LPS+CD processed through Beopje) and LPS+CD (LPS+CD). Treatment of the Raw 264.7 cell lines using LPS led to a significant increase in NO production, pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6 and $IL-1{\beta}$), and inflammation related genes (COX-2 and iNOS). Investigation of the inhibitory effects of CD and processed CD on NO production and expression of iNOS and COX-2 was done in LPS-induced RAW 264.7 cells. There was significant inhibition of NO production by LPS+CD and LPS+CD-B in a dose-dependent manner (p<0.05). Particularly, LPS+CD-B exhibited reduced mRNA expression of iNOS and COX-2 and NO production as compared to LPS+CD in Raw 264.7 cell lines (p<0.05). These results may explain some known biological activities of curly dock including the anti-inflammatory effects. CD-B in particular exhibited the highest anti-inflammatory effects of inhibiting production of NO, through the regulation of inflammatory related genes and pro-inflammatory cytokines. These results of Beopje processing might help decrease the anti-biological effects and increase several active substances of curly dock.

pH조정후 트랜스글루타미나제로 처리한 탈지 원유의 전자현미경적 특성 (Electron Microscopical Characteristics of Transglutaminase-treated Raw Skim Milk After pH Adjustment)

  • 원유에서 지방을 제거한 탈지유의 pH를 5.5, 7.0, 8.5로 조정한 다음 TGase를 첨가하여 0, 1, 2, 4, 8시간 반응시킨 후 동결 건조하여 조직의 성상을 주사 전자 현미경을 이용해 관찰, 비교하였다. pH와 TGase를 처리하지 않은 탈지유는 단백질 입자들이 규칙적으로 회합해 있었다. pH 조정 후 TGase를 처리한 다음 반응시간을 달리한 시료에서는 pH를 5.5로 조정한 경우 현저한 변화가 있었는데, 그 변화 양상은 단백질 입자들이 0시간에서 조각을 이루어 회합되어 있다가 1시간 반응시킨 경우 단백질 입자들이 서로 결합하여 넓게 회합하였다. 2시간 반응시킨 경우 단백질 입자들이 다시 뭉쳐서 회합하였으며 4시간 반응시킨 경우 뭉쳐져 있던 단백질 입자들이 조그만 한 구형 성상으로 넓게 회합하였다. 8시간 반응시킨 시료는 구형 성상으로 회합되어 있던 단백질 입자들이 사라지면서 다시 넓게 회합하는 것을 관찰할 수있었다. pH 7.0과 8.5 조건하에서는 단백질 입자들이 조각 형태를 이루고 있었으며 반응시간이 증가할수록 입자들이 넓게 확대되는 현상을 나타냈다. 전반적으로 현미경적 입자의 형태는 pH 5.5로 조정된 시료가 pH 7.0또는 pH 8.5로 처리된 시료들보다 더작게 관찰되었다. 이와 같은 단백질들의 현미경적 변화 양상은 pH TGase처러 그리고 반응시간에 따라 영향을 받고 있는 것으로 사료된다.

봉약침액(蜂藥鍼液)이 RAW 264.7 세포의 iNOS, TNF-α 및 NF-kB에 미치는 영향(影響 ) (The Effects of Bee Venom on iNOS, TNF-α and NF-kB in RAW 264.7 Cells)

  • Objective : The purpose of this study was to investigate the effects of Bee Venom on the lipopolysaccharide(LPS), sodium nitroprusside(SNP), hydrogen peroxide$(H_2O_2)$-induced expression inducilble nitric oxide synthetase(iNOS), tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) and nuclear factor kappa B(NF-kB) in RAW 264.7 cells, a murine macrophage cell line. Method : The expressions of expression iNOS and TNF-${\alpha}$ were determined by western blotting with corresponding antibodies. The expressions of expression NF-kB was assayed by EMSA method. Results : 1. The 0.5, 1 and $5{\mu}g/mg$ of bee venom on LPS-induced expression of iNOS, the $5{\mu}g/mg$ of bee venom on SNP-induced expression of iNOS and the $1{\mu}g/mg$ of bee venom on $H_2O_2$-induced expression of iNOS compared with control were inhibited significantly. 2. The 0.5, 1 and $5{\mu}g/mg$ of bee venom inhibited significantly LPS and $H_2O_2$-induced expression of TNF-${\alpha}$ compared with control, respectively. The $0.5{\mu}g/mg$ of bee venom increased significantly SNP-induced expression of TNF-${\alpha}$ compared with control. 3. The $5{\mu}g/mg$ of bee venom on LPS-induced expression of NF-kB, the $0.5{\mu}g/mg$ of bee venom on SNP-induced expression of NF-kB and the 0.5, $5{\mu}g/mg$ of bee venom on $H_2O_2$-induced expression of NF-kB were inhibited significantly compared with control, respectively.
