Anti-inflammatory Effect of Dianthi Semen Herbal-acupuncture at KI10 on nephritis in rats (음곡에 시술한 구맥 약침이 LPS로 유도된 흰쥐의 신장염에 미치는 영향)

  • Objective : This study aimed to evaluate the effects of Dianthi Semen Herba herbal-acupuncture (DS-HA) at KI10 (Umgok) on nephritis induced by LPS in rat. Methods : Rats were injected with LPS and treated with DS-HA at KI10 3 times for a week(DS-HA), N.P. group was treated 26 gauge needle at KI10, saline group was treated with normal saline at KI10. To estimate the effects of DS-HA at KI10 on nephritis in rats, WBC, neutrophils in blood, BUN, creatinine TNF-${\alpha}$, CINC-1 in serum, urinal volume and creatinean and total protein in urine, reanl TNF-${\alpha}$, renal MPO were measured and histological analysis of renal tissue was performed. Results : DS-HA at KI10 significantly decreased WBC and neutrophil in blood and BUN and creatinin in serum, MPO in kidney, and significantly increased urinary volume in LPS-stimulated rats. DS-HA at KI10 reduced accumulation of neutrophil in renal tissue of LPS-stimulated rats. Conclusions : Taken together, DS-HA at KI10 has a protective or therapeutic effect for nephritis in LPS-stimulated rat. Therefore, it is suggested that DS-HA at KI10 may be an useful therapeutics in clinical field after further researches.

  • This cDNAs were cloned with the aid of the polymerase chain reaction (PCR) by sequences based on cloned rat LH/CG and FSH receptor cDNAs. A cDNAs of LHR and FSHR were transfected into the 293 cells. Several clonal cell lines were obtained expressing different numbers of cell surface receptors. One cell lines for each LHR and FSHR were chosen, and a corresponding cell lines expressing the wild type LHR and FSHR were selected based on the number of cell surface receptor for the particular LHR and FSHR. The abilities of the LHR and FSHR to transduce the hCG and FSH signals were measured by quantitating cAMP accumulation in cells incubated with increasing concentrations of hCG and FSH. The cAMP accumulation effects for these receptors were increased by the increasing concentrations of hCG and FSH. Thus, most of the receptors expressed in cells transfected with LHR and FSHR could be detected by measuring hormone binding and cAMP response, and can utilize to study the structure function and signal transduction of the choriogonadotropins and glycoprotein hormones.

Cholesterol Biosynthesis from Lanosterol: Development of a Novel Assay Method, Characterization, and Solubilization of Rat Hepatic Microsomal Sterol Δ7-Reductase

  • A novel assay method is described for rapid quantitation of reaction rate of sterol ${\Delta}^7$-reductase (${\Delta}^7$-SR) which catalyzes reduction of the ${\Delta}^7$-double bond of sterols. Of six different organ tissues-liver, small intestine, brain, lung, kidney, and testis-. ${\Delta}^7$-SR activity was detected only in liver (2.30 nmol/min/mg protein) and testis (0.11 nmol/min/mg protein). Using a newly developed method which employs diet-induced enzyme proteins and ergosterol as substrate, we assessed both kinetics ($K_m=210\;{\mu}M$, $V_{max}=1.93\;nmol/min/mg$) and inhibition of the rat hepatic ${\Delta}^7$-SR against well-studied cholesterol lowering agents such as triparanol ($IC_{50}=16\;{\mu}M$). 3-$\beta$-[2-(diethylamino)ethoxy]androst-5-en-17-one (U18666A) ($IC_{50}=5.2\;{\mu}M$), and trans-1.4-bis(2-chlorobenzylaminomethyl)cyclohexane dihydrochloride (AY-9944) ($IC_{50}=0.25\;{\mu}M$). Of the three well-known AY-9944-sensitive cholesterogenic enzymes (i.e., ${\Delta}^7$-SR, sterol ${\Delta}^8$-isomerase, and sterol ${\Delta}^14$-reductase). ${\Delta}^7$-SR was found to be the most sensitive enzyme with a noncompetitive inhibition of this compound ($K_i=0.109\;{\mu}M$). Substrate specificity studies of the microsomal ${\Delta}^7$-SR indicate that the relative reaction rate for 7-dehydrocholesterol and ergosterol are 5.6-fold and 1.6-fold higher than that for lathosterol. ${\Delta}^7$-SR activity was also modulated by feeding rats a diet supplemented with 0.5% ergosterol (>2.6-fold) in addition to 5.0% cholestyramine plus 0.1% lovastatin ($\simeq$5.0-fold). Finally, microsomal ${\Delta}^7$-SR was solubilized by 1.5% 3-[3-(cholamidopropyl)-dimethylammonio]-1-propanesulfonate (CHAPS) and enriched on PEG (0~10%) precipitation, which should be suitable for further purification of the enzyme.

Effects of Daeboeum-hwan Aqueous Extracts on Streptozotocin-induced Rat Diabetes and Related Complications (대보음환(大補陰丸)이 Streptozotocin으로 유도된 백서(白鼠)의 당뇨병 및 당뇨병 합병증에 미치는 영향)

  • Objectives : The object of this study was to investigate the effects of Daeboeum-hwan (DBEH) aqueous extracts on Streptozotocin (STZ)-induced rat diabetes and related complications. Methods : SD rats were divided into 6 groups (n =8) : 50 mM citrate buffer and vehicle (distilled water 5 ml/kg) administered group (Intact control), STZ treatment (60 mg/kg, single intraperitoneally administered) and vehicle administered group (STZ control), STZ treatment and silymarin 100 mg/kg administered group (Reference), and STZ treatment and DBEH extracts 50, 100 and 200 mg/kg administered groups. DBEH extracts were orally administered once a day for 28 days from 3 weeks after STZ treatment. The results were compared with silymarin 100 mg/kg. Results : Decreases in the body weights, increase of kidney and liver weights, blood glucose, BUN, creatinine, AST, ALT, LDL, triglyceride and total cholesterol levels with decreases of HD L levels, increases of pancreatic MDA contents and decreases of GSH contents were detected in STZ control as compared with intact control. These diabetes and related complications were inhibited by treatment with 100 and 200 mg/kg of DBEH extracts. Conclusion : DBEH extracts showed favorable effects on STZ-induced diabetes and related complications mediated by their antioxidant effects as similar to silymarin. Because the lowest dosage (50 mg/kg) of DBEH treated group did not show any favorable effects as compared with STZ control, the effective dosages of DBEH is considered as about 100 mg/kg, while DBEH extracts at 200 mg/kg showed similar effects as compared with 100 mg/kg of silymarin. It, therefore, is expected that DBEH will show favorable effects on diabetes and various diabetic complications.

Study of single dose test of Sweet Bee Venom in rats (Sweet BV의 rat를 이용한 단회 근육시술 독성시험)

  • Objectives: This study was performed to analyse single dose toxicity of pure melittin(Sweet Bee Venom-Sweet BV) extracted from the bee venom by utilizing protein isolation method of gel filtration. Methods: All experiments were conducted at Biotoxtech, a non-clinical studies authorized institution, under the regulations of Good Laboratory Practice (GLP). Six weeks old female Sprague-Dawley rats were chosen for the pilot study and determined 30㎎/㎏ which is 4285 times higher than the clinical application dosage as the high dosage, followed by 15 and 7.5㎎/㎏ as mid and lose dosage, respectively. Equal amount of excipient to the Sweet BV experiment groups was administered as the control group. Results: 1. No mortality was witnessed in all of the experiment groups. 2. Hyperemia and movement disorder were observed around the area of administration in all groups, and higher occurrence in the higher dosage groups. Hyperemia and movement disorder diminished with elapsed time. 3. For the weight measurement, male groups showed larger reduction in weight in accordance with higher dosage. Female groups didn't s how significant changes. 4. To verify abnormalities of organs and tissues, cerebellum, cerebrum, liver, lung, kidney, and spinal nerves were removed and conducted histological observation with H-E staining. No abnormalities were detected in any of organs and tissues. 5. One female rat in the 30㎎/㎏ group had amputated toe near the administered area and histopathological finding was hemorrhage with inflammation. This is presumed as a secondary infection after the administration of Sweet BV. Conclusion: Above findings suggest Sweet BV is relatively s safe treatment medium. Further studies on the subject should be conducted to yield more concrete evidences.

Effects of Vitamin E on Liver Cytochrome $P_{450}$ Content and Xanthine Oxidase Activity in Acute Cadmium-Poisoned Rats (식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 Cytochrome $P_{450}$ 함량과 Xanthine Oxidase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of vitamin E on liver microsomal cytochrome P450 contents and xanthine oxidase activity in acute cadmium poisoned rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(0E Cd group), 40mg vitamin E per kg diet(40E Cd group) or 400mg vitamin E per kg diet(400E Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd2+/kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4 weeks. Body weight, food intake and feed efficiency ratio of cadmium injected animals, were decreased compared with those of normal group. The weights of liver and kidney in cadmium injected groups were not different from those of normal group. Cadmium contents of liver in cadmium groups were 160 fold higher those that of normal group. Accumulation of cadmium poisoned rat liver was reduced by vitamin E supplementation. Contents of blood hemoglobin and hematocrit in 0E Cd groups were decreased to 2~ 14% of those of the other groups. Contents of serum triglyceride in all experimental groups were not significantly different each other. Levels of serum total cholesterol, LDL cholesterol and antherogenic index in 0E Cd and 40E Cd groups were higher than those of normal group, while the contents of HDL cholesterol in 0E Cd and 40E Cd groups were lower than those of normal group. Xanthine oxidase(XOD) activity and cytochrome P450 contents in the liver were significantly increased in cadmium injected groups but these were reduced by vitamin E supplementations. The present results indicate that acute cadmium poisoning in rats causes increasing free radical generation systems in the liver and that leads to liver tissue damage. But these abnormalities can be reduced by dietary vitamin E supplementations.

Effect of the Level of Ginseng Powder in Diet on Growth Rate and Blood Glucose Concentration in Rat (식사에 첨가된 인삼분의 수준이 백서의 성장 및 체성분함량에 미치는 영향 -혈당을 중심으로-)

  • This study was carried out to investigate the effect of the diet supplemented with various amounts of ginseng powder on the growth rate and blood glucose level in rat. Male Albino rats, weighing between 90 to 100g, were divided into 5 diet groups of 6 rats each. experimental diets contained 0, 2, 4, 6 and 8% ginseng powder which was substituted for rice powder and were designated C, G-2, G-4, G-6 and G-8, respectively. The diets were fed for 3 weeks. Throughout feeding period, feed consumption, body weight, feed and protein efficiency ratios were measured, organ weights and blood glucose level were determined at the end of the experimental period. the results are summarized as follows. 1. rats in G-2 group consumed the highest amount of diet among all groups. 2. Body weight gain was significantly higher in G-2 and G-4 than in other groups. 3. Feed and protein efficiency ratios were a little higher in G-4 than in control group. And those of group G-8 were the lowest among those of other groups. 4. The weight of liver was significantly lower I G-4 than in control group. The weight of kidney was significantly lower I G-4 and G-8 than in control group. The weight of heart was significantly lower I G-4 and G-8 than in control group. While, the weight of spleen was greater in ginseng powder diet than in control group. 5. Supplementing ginseng powder in diet resulted in deceased nitrogen in liver. 6. Blood glucose was decreased with increased level of ginseng powder diet.

Anti-proliferative and angio-suppressive effect of Stoechospermum marginatum (C. Agardh) Kutzing extract using various experimental models

  • BACKGROUND/OBJECTIVES: Abundant consumption of seaweeds in the diet is epidemiologically linked to the reduction in risk of developing cancer. In larger cases, however, identification of particular seaweeds that are accountable for these effects is still lacking, hindering the recognition of competent dietary-based chemo preventive approaches. The aim of this research was to establish the antiproliferative potency and angiosuppressive mode of action of Stoechospermum marginatum seaweed methanolic extract using various experimental models. MATERIALS/METHODS: Among the 15 seaweeds screened for antiproliferative activity against Ehrlich ascites tumor (EAT) cell line, Stoechospermum marginatum extract (SME) was found to be the most promising. Therefore, it was further investigated for its anti-proliferative activity in-vitro against choriocarcinoma (BeWo) and non-transformed Human embryonic kidney (HEK 293) cells, and for its anti-migratory/tube formation activity against HUVEC cells in-vitro. Subsequently, the angiosuppressive activity of S. marginatum was established by inhibition of angiogenesis in in-vivo (peritoneal angiogenesis and chorioallantoic membrane assay) and ex-vivo (rat cornea assay) models. RESULTS: Most brown seaweed extracts inhibited the proliferation of EAT cells, while green and red seaweed extracts were much less effective. According to the results, SME selectively inhibited proliferation of BeWo cells in-vitro in a dose-dependent manner, but had a lesser effect on HEK 293 cells. SME also suppressed the migration and tube formation of HUVEC cells in-vitro. In addition, SME was able to suppress VEGF-induced angiogenesis in the chorio allantoic membrane, rat cornea, and tumor induced angiogenesis in the peritoneum of EAT bearing mice. A decrease in the microvessel density count and CD31 antigen staining of treated mice peritoneum provided further evidence of its angiosuppressive activity. CONCLUSIONS: Altogether, the data underline that VEGF mediated angiogenesis is the target for the angiosuppressive action of SME and could potentially be useful in cancer prevention or treatment involving stimulated angiogenesis.

Comparison of the Effects of Deer Antler, Old Antler, and Antler Glue on Osteoporosis in Ovariectomized Rats

  • Background: Examination of the effects of deer antler, old antler, and antler glue on postmenopausal osteoporosis in an ovariectomized Sprague-Dawley rat model. Methods: The study involved 7 experimental groups; SHAM (sham-operated rats), OVX (ovariectomized rats), E2 (ovariectomized rats with estradiol $10{\mu}g/kg$ daily, orally), DA (ovariectomized rats with deer antler extract 5.83 mg/kg), OA (ovariectomized rats with old antler extract 3.8 mg/kg), low-AG (ovariectomized rats with low dose of antler glue powder 12.5 mg/kg), high-AG (ovariectomized rats with high dose of antler glue powder 37.5 mg/kg). After 6 weeks of treatment, body weight, blood calcium, phosphorus, estradiol, liver [alkaline phosphatase (ALP), aspartate transaminase (AST), alanine transaminase (ALT)] and kidney [blood urea nitrogen (BUN)/creatinine ratio] function, and femoral bone mineral density (BMD) were measured. Results: The body weights of DA, OA, low-AG, and high-AG groups did not significantly differ from OVX group. Blood estradiol levels were significantly increased in the DA, low-AG, and high-AG groups compared to the OVX group. Blood calcium, phosphorus, ALP, AST, and ALT levels and BUN/creatinine ratio did not show significant changes in the DA, OA, low-AG, and high-AG groups. BMDs of the femur, and femoral head and neck were significantly increased in the low-AG group. In the OA group, the BMD of the femoral head and neck was significantly increased. Conclusion: Treatment with deer antler, or antler glue for 6 weeks was effective for increasing estradiol and femoral BMD in ovariectomized rats, suggesting that this may be of therapeutic benefit for osteoporosis.

Effects of Polygoni Multiflori Radix on prevention of hyperlipidemia and liver damage induced by alcohol (하수오(何首烏)가 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향)

  • Objectives : The Korean medical doctors use PMR(Polygoni Multiflori Radix) for nourishing the liver-kidney, loosing the bowel to relieve constipation, recovering from malaria, and clearing away heat and eliminating toxin, etc. But, this study was performed to investigate the effect of water extracts from PMR on prevention of hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And the extract was administrated for the same period. We measured the serum components in rat's blood, body weight and weight of liver. Results : At first, we observed effects of PMR on prevention of hyperlipidemia induced by alcohol. PMR group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. PMR group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. At second, we observed effects of PMR on prevention of liver damage induced by alcohol. PMR group showed significant decrease of GOT, GPT, ALP, and LDH in comparison with those of the control group. PMR group showed significant increase of liver weight in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from PMR have pharmaceutical efficacy on prevention of hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.
