  • Expressions of cellular adhesion molecules (CAMs) are closely related to the formation of early atherosclerosis, an age-dependent vascular disorder. However. previous research provided only limited and conflicted reports on age-related alterations of CAMs' expressions and even much less is known the modulation of CAMs by calorie restriction (CR), In this study, expression of vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule-1 (ICAM-1), E-selectin, P-selectin and platelet/endothelial cell adhesion molecule-1 (PECAM-1) in aorta and kidney were investigated by western blot and immuno-histochemical stain utilizing ad libitum (AL) and CR rat.   • Great efforts have been made to improve both the quality of life and life expectancy of diabetes by treating problems associated with chronic complications such as neuropathy, retinopathy and nephropathy. In particular, diabetes is an increased risk of developing several types of kidney disease, and the predominant cause of end-stage renal disease in patients with this disorder is diabetic nephropathy. Therefore, prevention of the occurrence and progression of diabetes and its complications has become a very important issue. The scientific observations of an animal model of streptozotocin-induced diabetes, spontaneously occurring diabetes and diabetic nephropathy in this study suggest that one of the Kampo prescriptions, Hachimi-jio-gan comprising eight constituents, is a novel therapeutic agent.

광물성 미네랄이 동물세포의 손상에 미치는 효과 (Effects of Ore Minerals on the Damages of Animal Cells)

  • In this study, we investigated the suppressive effects of ore minerals on the allergic cell damages and oxidative cell damages. The ore minerals significantly reduced the productions of tumor necrosis factor-alpha (TNF-${\alpha}$) and interleukin-4 (IL-4) in rat basophilic leukemia cells challenged with 2,4-dinitrophenol-bovine serum albumin (DNP-BSA). Lipoxygenase activity was also reduced by the ore minerals. Moreover, the ore minerals showed weak protective effects on the oxidative damage induced by hydrogen peroxide in pig kidney cells and retinal ganglion cells. Photohemolysis of erythrocytes in the presence of rose-bengal as a sensitizer was also inhibited by ore minerals. These results suggest that the ore minerals may be useful as the protectant for allergic and oxidative cell damages.

Protective Effect of Heat-processed Ginseng (Sun Ginseng) in the Adenine-induced Renal Failure Rats

  • The effect of orally administered sun ginseng (SG), which is a ginseng processed by steaming, was examined in adenine-induced chronic renal failure rat. SG significantly decreased both blood urea nitrogen and serum creatinine levels, indicating an improvement of renal function. Also, SG significantly increased the urinary excretion of both urea and creatinine. Furthermore it lowered the blood pressure, and inhibited adenine-induced kidney hypertrophy and edema. Based on these findings, SG may ameliorate chronic renal failures.

  • Ochratoxin A (OA), a potent nephrotoxin in several species, is knownto be a renal carcinogen in animals and is implicated in the etiology of Balkan endemic nephropathy (BEN). The NTP (National Toxicology Program) classified Ochratoxin A as having clear evidence of carcinogenic activity, based on uncommon tubular adenomas and tubular cell carcinomas of the kidney and multiple fibroadenomas of the mammary gland, seen in the rat.(omitted)

Experimental Toxocara catiInfection in Gerbils and Rats

  • Mongolian gerbils and Wistar rats were inoculated orally with 240 and 2,500 Toxocara cati embryonated eggs, respectively, to evaluate the larval recovery in different tissues and organs, such as the liver, lungs, heart, kidney, and skeletal muscles after 5, 30, 49, 70, and 92 days post-infection (PI). Larval recovery rates were 1.7-30.0% in Mongolian gerbils on days 5-92 PI and 0.2-3.8% in rats on the same days. These results indicate that Mongolian gerbils and Wi star rats are suitable experimental paratenic hosts for the study of neurological toxocariasis as well as visceral toxocariasis.

  • Osteoporosis is usually considered a disease of older women reported the rate, pattern and determinants of bone loss, far less information is available for men although it is also common in men [1,2]. The three major causes of osteoporosis in men are excessive alcohol intake, long-term glucocorticoid therapy, and hypogonadism [3,4]. In process of bone resorption, type I collagen crosslinking molecules, pyridinoline (PYD) are released into the circulation and cleared by the kidney. $^2$H$_2$O as a tracer has been applied to measure synthesis rates of slow-turnover proteins and successfully applied to bone collagen synthesis, skeletal muscle and cardiac muscle in rat. The objective of this study was to examine osteoporosis and alcohol-induced changes of femur and liver in post-menopausal males using the developed method. (omitted)

Beneficial effects of Phellodendri Cortex extract on hyperglycemia and diabetic nephropathy in streptozotocin-induced diabetic rats

  • This study investigated the effect of Phellodendri Cortex extract on hyperglycemia and diabetic nephropathy in streptozotocin-induced diabetic rats. Male Sprague-Dawley rats were divided into normal control (NC), diabetic control (DC), and diabetic treatment with Phellodendri Cortex extract (DP). Over a 4-week experimental period, Phellodendri Cortex extract was administered orally at 379 mg/kg BW/day. The final fasting serum glucose level, urine total protein level, and relative left kidney weight in the DP group were significantly lower than the DC group. Renal XO and SOD activities in the DP group were significantly lower than the DC group and renal CAT activity in the DP group was significantly higher than the DC group. Tubular epithelial change was reduced in the DP group compared to the DC group. These results indicated that Phellodendri Cortex can reduce glucose level and prevent or retard the development of diabetic nephropathy in streptozotocin-induced diabetic rats.

Effect of atorvastatin on dendritic cells of tubulointerstitium in diabetic rats

  • Inflammatory reactology has become increasingly important in diabetic kidney disease. In this study, we estabilished STZ-induced diabetic rat model to investigate whether dendritic cells (DCs) mediated tubulointerstitial damages, and whether the effects by DCs were mediated by P-selectin expression and can be inhibited by atorvastatin. The study demonstrated that there was an accumulation of DCs in diabetic rats mediated by P-selectin. It also showed the accumulation of DCs and expression of P-selectin was closely correlated with the degree of renal tubulointerstitial injury. These effects were markedly attenuated by atorvastatin. Thus, DCs play a role in tubulointerstitial damages, atorvasttin can prevent renal tubulointerstitium from damage by inhibiting the P-selectin expression and DCs migration.

영계출감탕의 신기능에 미치는 영향 (Effects of Youngkaechulgamtang on the Kidney Function)

  • The Youngkaechulgamtang(Y) is composed of four herbal drugs, including Hoelen(H), Cinnamomi Ramulus(C), Atractylodis Rhizoma Alba(A) and Glycyrrhizae Radix(G). In oriental medicine literatures, Youngkaechulgamtang is described to be effective in headache, inflammation, uremia, gastritis, diarrhea, hypertension. To estimate the clinical effectiveness of Youngkaechulgamtang, several pharmacological experiments were carried out. The results are summarized as follows: On $HgCl_2-induced$ renal failure rat, Youngkaechulgamtang showed the significant increase of $Na^+,\;K^+,\;Cl^-$ contents in urine and the significant decrease of creatinine and BUN concentrations in serum. Youngkaechulgamtang was more effective than mixture of 4 ingredients in the urine volume and excretion of electrolytes. In the diuretic effect, Hoelen and Atractylodis Rhizoma Alba were more effective than other drugs.
