The Effect of Shinchulsan on the Rat Model with Allergic Rhinitis (神朮散이 알레르기 비염 유발 흰쥐 모델에 미치는 영향)

  • Objectives : Allergic rhinitis is highly increased in these days. We aimed to investigate therapeutic effect ofShinchulsanby observing changes in blood cells and the protein expression level of IL-4 in spleen of Sprague-Dawley(SD) rats with allergic rhinitis.Methods : Fifteen SD rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitional and intranasal sensitization with 0.1% and 0.4% Ovalbumin solution. ThenShinchulsanwas orally administered only to the sample group for 28 days, while the rats in the control group was given normal saline.Results : 1.Shinchulsanshowed significantly decreased eosinophil level on the serum of the rat model.2.Shinchulsanshowed significantly decreased IgE level on the serum of the rat model.3.Shinchulsanshowed significantly decreased the protein expression levels of IL-4 in spleen of the rat model.4.Shinchulsanhad no hepatoxicity, according to AST and ALT on the serum.Conclusion : According to the above results, it is considered thatShinchulsanis helpful in treatment of allergic rhinitis.

Effects of Antioxidant and Blood Flow Improvement of Grape Leaf Extract and Resveratrol from Vitis romaneti (남항(Vitis romaneti) 유래 포도잎 추출물과 Resveratrol의 항산화 및 혈행개선 효과)

  • The purpose of this study is to investigate the effects of 80% ethanol grape leaf extract (VGLE) and resveratrol (VGLR) from Vitis romaneti on antioxidant of red blood cells (RBC) of rat and efficacy of blood flow improvement in a rat model of topical ferric chloride ($FeCl_3$)-induced carotid artery damage. RBC oxidative hemolysis and plasma lipid peroxidation induced by the aqueous peroxyl radical generator 2,2'-azobis(2-amidinopropane) dihydrochloride were significantly suppressed by VGLE or VGLR in a dose-dependent manner. The $FeCl_3$ treatment seriously damaged the carotid artery: the walls of the artery and blood flow. However, VGLE or VGLR administration has ameliorated the blood flow and suppressed thrombus in blood vessels. These results suggest that VGLE or VGLR ameliorates the oxidative stress of RBC and thrombosis against blood vessel damage.

Effects of Garlic Juice on Toxicity of Mercury in Rat (마늘즙 투여가 흰쥐의 수은 독성에 미치는 영향)

  • It was attempted to observe the antidotic effects of garlic on mercury intoxicated rat model in vivo. The experimental model group consists of garlic juice treated group (G), garlic juice-mercury treated group(MG), mercury treated group (M), and control group (basal diet), The garlic juice of 2% to the daily diet by weight was administered orally to G and MG groups, The single dose of 2.5mgHG/Kg per week was given orally to M and MG groups. the study was carried out for 4 weeks. The results of experiment were as follows. For the group of Mg, and the weight increasing rate was improved to about 30% compared to that of group M. Furthermore a general tonic efficay of garlic was observed in G group in term of increased weight gain rate (bout 15%) than control. In the biochemical studies of rat blood garlic showed effects on lowering the abnormally elevated GPT, GOT, uric acid creatinine value, and especially in lowering the BUN value of Hg treated rat that was selectively elevated in the case of impared renal function such as acute gromerulonephritis caused by Hg intoxication. In the analytical studies blood and renal Hg contents. HG group showed lower value (0.3, 0.33ppm) than that of M group (0.46, 0.51 ppm) Significant difference in reducing Hg level due to the antidotic effect of garlic was observed. In conclusion, it was revealed out from this research, the main principles of garlic, nonprotein sulfur amino acid (alliin) and sulfur compounds (allicin and diallyl disulfides) seem to almost certainly have an antidotic effect on mercury intoxication of rat in vivo.

Heterogeneity of Renin Released from Renal Cortical Slices (고혈압백서의 신장 Renin Heterogeneity에 관하여)

  • It has been well known that the renal cortical blood flow rate was much higher than that of the medulla and the renal blood flow distribution was affected by hemorrhage, volume expansion or salt-loading. The existance of the heterogeneities of glomerular filtration rate and nephron has also been reported. In order to understand the regulations and physiological roles of the heterogeneities, studies on the intrarenal renin-angiotensin system have been focused. Although it is well known that the granularity of iuxtaglomerular cells and renal renin content are more marked in superficial than in the deep glomeruli, their physiological significance is not quite clear. This study was therefore undertaken to clarify changes in renin response and isoelectric ronin profile to TMB-8 in outer, mid and inner cotices of normotensive and hypertensive rats. The basal rate of renin release was highest in outer cortex of Sprague-Dawley rat (SDR), Wistar rat (WR) and spontaneously hypertensive rat (SHR). The basal renin release from outer and inner cortex of SHR was significantly lower than that from those of SDR. The reponse of renin release to TM8-8 was highest in mid cortex and the increase of renin release in response to TMB-8 from inner cortex of SDR was significantly higher than that in SHR. In dehydrated rats, the basal renin release from renal cortical slices of SDR was increased but that from WR and SHR was not. The response of renin release to TMB-8 from mid and inner cortex of dehydrated WR tended to increase. In dehyrated SHR, increase of renin release from inner cortex was significantly higher than that in euhydrated SHR. No significant differences in the isoelectric renin profile were found both in different cortical areas and strains. In dehydrated rats, the percentage of renin form 2 was decreased and those of renin form 5 and 6 were increased. These results suggest that the heterogeneity of renin release from cortical area of euhydrated and dehydrated rats in response to TMB-8 may be related to the changes of renal blood flow and/or calcium metabolism in cortical area. These data also suggest that the renin forms with different isoelectric points may have an physiological significance.

The Effect of Hypophysectomy and Subsequent Administration of Sex Hormone on Several Endocrine Glands and Plasma Components in Rats (흰쥐의 내분비선 및 혈장성분에 미치는 뇌하수체척출의 영향과 이에 대한 성 Hormone의 효과)

  • The present experiments were carried out to elucidate the effects of hypophysectomy and subsequent administration of sex hormone on thyroid, adrenal gland, gonads and blood plasma components in the rat. The jresults obtained were summarized as follows: The weight of the thyroid gland of both male and female hypophysectiomzed rats decreased markedly from 7 days up to 56 days after the hypophysectomy as compared to the control group. The administration of sex hormone (6 mg of testosterone propionate to male and 6 mg of hexestrol to female) to the hypophysectomized rat gave on effect on the change in the weight of the thyroid gland. The hopophysectomy decreased the uptake of radioactive iodine in the thyroid gland in both male and female rats with time. Subsequent administration of the sex hormone caused no effect. With regard to the histological changes of the thyroid gland, the hypophysectomy caused significant changes in the gland showing a remarkable degeneration. The function of the gland seemed to disa, pp.ar almost completely on 56th day after the hypophysectomy. Upon the administration of sex hormone after the hypophysectomy, however. the epithelia of the follicle which has changed to flat from has partly returned to its functional cubicfrom and nuclei recovered as nearly as normal. These recovery were more remarkable in the female than in the male. The hypophysectomy kept causing a significant decrease in the weight of the adrenal gland in male and female rats during the period of observation (up to 56 days) as in the case of thyroid gland. The administration of sex hormone has on effect in this respect either. The hypophysectomy also caused a marked morphological change in the gland: zona fasciculata and zona reticularis were dicreased in size quichly after the hypophysectomy. The administraton of the sex hormone to the hypophysectomized rat resulted in clear distinction among the three layers of the adrenal cortex which otherwise very diffused. In the male, this phenomenum was more remarkable than in the female and the pattern of the cell arrangements and the thickening of each layer became similar to those of normal rats. The gonads of both sexes have also kept decreasing in the weight and degenerated in morpohology after the hypophysectomy. However, the degenerate follicle became enlarged after the administration of hexestrol in the female. Furthermore, the vacuoles found in interstitial cells of hypophysectomized rat disa, pp.ared after the administration of testosterone in the male and the formation of spermatocytes seemed to be recovered. Hypophysectomy also caused a gradual increase in the contents of total protein, non-protein nitrogen, total lipid, cholesterol and calcium in the blood plasma with time. The concentrations of sodium, potassium and chloride in the blood did not change after the hypophysectomy. Sex hormone caused practically no change in above tendency.

  • This study was performed to evaluate the effect of low-power laser irradiation on the periodontium of the orthodontically moved tooth of rat. The experimental materials were 30 male rats. Orthodontic appliances were placed bilaterally between maxillary first molar and incisor teeth and the force was 1 Oz. Experimental animals were divided into eight groups as follows: no movement, 1st, 2nd, 3rd, 5th, 7th, 14th, and 28th day groups. In all experimental animals except no movement group, low-power laser was irradiated on the unilateral maxillary first molar (experimental side), but on the contralateral side, only orthodontic force was applied (control side). The histologic effects of laser on the periodontium of the orthodontically moved tooth on the consecutive experimental days were as follows: In the experimental side, by the biostimulating effect of laser, 1. Hyalinized tissue was formed later and eliminated earlier than in the control side. 2. Undermining bone resorption was occurred earlier than in the control side. 3. More osteoid tissue was deposited and calcified earlier than in the control side. 4. The most prominent changes were formation of new blood vessels and dilatation of old blood vessels.

Inhibitory Effects of Yuzu and Its Components on Human Platelet Aggregation

  • Our previous study demonstrated that yuzu has an anti-platelet effect in rat blood. In the present study, we examined whether the anti-platelet effect of yuzu can be extended to human blood by investigating its ability to inhibit aggregations induced by various agonists in human platelet rich plasma (PRP). This study also investigated the underlying mechanism of yuzu focusing on ADP granule secretion, $TXB_2$ formations, and $PLC{\gamma}$/Akt signaling. The results from this study showed that ethanolic yuzu extract (YE), and its components, hesperidin and naringin, inhibited human platelet aggregation in a concentration-dependent manner. YE, hesperidin and naringin also inhibited $TXB_2$ formation and ADP release. The phosphorylation of $PLC{\gamma}$ and Akt was significantly inhibited by YE, heperidin and naringin. Furthermore, we demonstrated that YE, heperidin and naringin has anti-platelet effects in rat ex vivo studies, and lower side effects in mice tail bleeding time studies. The results from this study suggest that YE, hesperidin and naringin can inhibit human platelet aggregation, at least partly through the inhibition of $PLC{\gamma}$ and Akt, leading to a decrease in $TXB_2$ formation and granule secretion.

Effects of Salviae Miltiorrhizae Radix on the Diet-induced Hyperlipidemia in Rats (단삼이 흰쥐의 식이성 고지혈증에 미치는 영향)

  • The purpose of this study was to examine the effects of Salviae Miltiorrhizae Radix on the high fat diet-induced hyperlipidemia in rats. Sixty male Sprague-Dawley rats, around 270g of B.W., were divided into normal, control, sample A and sample B group. Normal and control groups were orally administrated with saline during 4 weeks, sample A and sample B with 252.5mg/kg(rat) and 420.1 mg/kg(rat) of solid extract of Salviae Miltiorrhizae Radix during 4 weeks respectively. Blood samples were collected from each of animals at 4 weeks, and used for the blood chemical analysis. Animals were sacrificed at 4 weeks, and chemical analysis was performed on the collected liver samples. The serum total cholesterol, HDL-cholesterol, LDL-cholesterol levels of sample A group were significantly decreased compared with the control group. On the basis of these of results, it is confirmed that Salviae Miltiorrhizae Radix has the curative effects on the hyperlipidemia in rats induced by high fat diet

Effects of Shiitake Mushroom on Anti-platelet Aggregation and Anti-thrombotic (표고버섯 추출물의 항혈소판 응집 및 항혈전 효과)

  • In in vitro study, the anti-platelet aggregation effect of Shiitake mushroom extract was examined by measuring the collagen induced platelet aggregation and the DPPH radical scavenging. In in vitro study, anti-thrombotic effect of Shiitake mushroom extract was examined using the carotid artery thrombosis rat model. Carotid artery thrombosis rat model was made by 35% $FeCl_3$ treatment. After that, we investigate thrombus weight and blood flow. In platelet aggregation test, the extract significantly inhibited platelet aggregation in a concentration dependent manner(p<.001). Also, extract increased DPPH radical scavenging activity in a concentration dependent manner. Extract significantly inhibited thrombus weight to compare with control group. And blood passage time were shorter in the Shiitake mushroom extract supplemented groups than in control group. These results provide experimental evidence that Shiitake mushroom can be used to prevent platelet aggregation and thrombosis, then could apply the clinical diseases such as cardiovascular disease, and so on.

Hypoglycemic, Hepatoprotective, and Nephroprotective Effects of - 'Saengjinyanghyul-tang($Sh\={e}ngj\={i}ny\v{a}ngxu\`{e}-t\={a}ng$)' - in Streptozotocin-Induced Diabetic SD Rats (생진양혈탕(生律養血湯)이 Streptozotocin으로 유발된 당뇨병 Rat 모델의 고혈당 및 간병증과 신병증에 미치는 영향)

  • Objectives : This study aimed to evaluate the hypoglycemic, hepatoprotective, and nephroprotective effects of 'Saengjinyanghyul-tang($Sh\={e}ngj\={i}ny\v{a}ngxu\`{e}-t\={a}ng$ : SJYHT)' in streptozotocin (STZ)-induced diabetic SD rat model. Methods : SJYHT extracts were once a day dosed for 28 days at a dosage 1000, 500, and 250mg/kg/5ml from 25 days after STZ-dosing, and the changes on blood glucose levels, liver and kidney weight, serum AST, ALT, BUN and creatinine levels were observed with histological changes on the liver and kidney. Results : Increased blood glucose levels, and diabetic hepatopathy & nephropathy including increased serum AST, ALT, BUN and creatinine levels, histological changes after STZ-dosing were dose-dependently reduced by SJYHT extract-dosing. Conclusions : 'SJYHT' water extracts have favorable hypoglycemic, hepatoprotective, and nephroprotective effects on the STZ-induced diabetic SD rats. Therefore, it is expected that 'SJYHT' has potential for use in the management of diabetes and diabetic complications.
