The location and local arrangement of motor, sensory neurons within brain stem, nodose ganglia, spinal ganglia and sympathetic ganglia projecting to rat's kidney and meridian point BL 23, GB 25 were investigated by HRP immunohistochemical methods following injection of 5% WGA-HRP into left kidney and meridian point BL 23, GB 25. Following injection of WGA-HRP into left kidney, anterogradely labelled sensory neurons were founded within either nodose ganglia and spinal ganglia. The sensory neurons innervating rat's left kidney were observed within spinal ganglia $T_{7}{\sim}L_3$. Sympathetic motor neurons innervating rat's left kidney were labelled within left suprarenal ganglia, either celiac ganglia, superior mesenteric ganglia, and sympathetic chain ganglia $T_{1}{\sim}L_3$. Sympathetic chain ganglia were concentrated in $T_{12}{\sim}L_1$. The sensory neurons innervating rat's meridian point BL 23 were founded within spinal ganglia $T_{2}{\sim}L_2$. They were numerous in spinal in ganglia $T_{10}{\sim}T_{12}$. Sympathetic motor neurons innervating rat's meridian point BL 23 were observed in suprarenal ganglia and greater splanchnic trunk, sympathetic chain ganglia from $T_1$ to $L_3$. They were concentrated in $T_{12}{\sim}L_3$. The sensory neurons innervating rat's meridian point GB 25 were labelled within spinal ganglia $T_{6}{\sim}T_{13}$. They were numerous in from T10 to $T_{12}$. Sympathetic motor neurons innervating rat's meridian point GB 25 were labelled within greater splanchnic trunk and sympathetic chain ganglia $T_{12}{\sim}L_3$. They were concentrated in $T_{13}{\sim}L_1$. This results neuroanatomically imply that the location of rat's motor and sensory neurons innervating meridian point BL 23 and GB 25 were closely related that of innervating kidney.
Heavy metals like Mercury and Cadmium cause various kinds of toxicities in the organs of Liver and Kidney. To observe the results of toxicity in the liver, kidney, and serum when the rats were injected subcutaneously with HgCl$_2$ and CdC1$_2$ and sacrificed after 24 hours and 72 hours from the last injection, we measured variation of lipidperoxide values in rat liver homogenate, variation of aspartate aminotransferase and alanine aminotransferase in rat serum. Variation of lipidperoxide values in rat kidney homogenate and variation of BUN in rat serum. It was found that Mercury and Cadmium administered subcutaneously to the skin in the air could cause the damages of liver and kidney.
Insulin-like growth factors (IGFs) and their binding proteins (IGFBPs) are important regulators on the development of maternal tissues during pregnancy. This study was performed to examine the relationship between maternal IGFs/IGFBPs system (i.e: IGF-I, II, their receptors, and IGFBPs) in pre- and post-partum rats. The liver and kidney are important organs for the synthesis of IGFs and IGFBPs in adults. The levels of materanal IGFs and IGFBPs in serum, liver, and kidney were examined at 14 and 21 days of gestation and at 3, 7, 11, and 14 days after birth. The expression of IGFs and their receptors mRNA was also examined in fetal and maternal rat liver, kidney. IGF-I concentrations in maternal serum and liver were decreased during pregnancy. However, IGF-I concentration in maternal kidney was increased, having maximal effect at 14 days of gestation. IGF-I concentrations were decreased in serum, liver, and kidney of postpartum rat, compared to control (p < 0.05). On the other hand, IGF-II concentrations in serum, liver, and kidney were increased during pregnancy (p<0.05) and gradually decreased to control level in postpartum period. The levels of IGFBP-3 and IGFBP-2 are expressed in serum, liver, and kidney. However, IGFBP-3 is mainly expressed in serum and liver, and IGFBP-2 in kidney. The levels of IGFBP-3 and IGFBP-2 in maternal serum were markedly decreased during pregnancy and gradually recovered to control level during postpartum period by western ligand blotting. However, there was no change of IGFBP-3 and IGFBP-2 levels by western immunoblotting. The levels of IGFBP-3 and IGFBP-2 in maternal liver and kidney also showed the same pattern of serum, although the main IGFBP is different. In normal rat serum, IGF-I 150 kDa and 50 kDa carrier proteins were detected. The level of IGF-I 150 kDa carrier proteins in pregnant rat was decreased compared to normal rat, but that of 50 kDa carrier proteins was increased. IGFBP-3 protease activity was identified in pregnant rat serum and maternal placenta, and it was inhibited by EDTA ($Ca^{2+}$ chelating agent) and aprotinin (serine proteinase inhibitor). Taken together, these results suggest that the changes of IGFs and IGFBPs in maternal rats are regulated by liver and kidney IGFs and their receptors mRNA during the pregnancy.
Alterations of renin-angiotensin system have been suggested as one of the mechanisms increasing arterial blood pressure in experimental and clinical hypertension. But the exact nature of high blood pressure in the early and late phase of renal hypertension is still controversial. To clarify the nature of renin release in both unclipped and clipped kidney of two kidney one clip Goldblatt lypertensive rat, experiments have been done in kidney slices, which were obtained from the rats of 3 and 7 days of operation. Basal rate of renin release was suppressed in unclipped kidney slices compared to clipped kidney Norepinephrine increased renin release from unclipped kidney slices, but not from clipped kidney slices. Suppressions by angiotensin Il and arginine vasopressin of renin release were attenuated in the clipped kidney slices compared to unclipped and sham-operated kidney slices. Increases by verapamil and trifluoperazine of renin release were attenuated in the clipped kidney slices compared to unclipped and sham-operated kidney slices. These results suggest that the negative feedback control mechanism of the renin-angiotensin system by angiotensin Il and arginine vasopressin is attenuated in the clipped kidney of two kidney one clip Goldblatt hypertensive rat, and that one of the altered mechanisms may be caused by certain regulatory changes of intracellular calcium and/or calcium-calmodulin complex in the juxtaglomerular cells.
Sulfamethazine sodium was orally administrated to Sprague Dawley female rats(body weight: 200~250g) with the sonde caude at the dose of 20mg of sulfamethazine sodium per 100g of body weight for 3 days to investigate the depletion rate of the drug from liver, kidney and muscle of rat. The results obtained were summerized as follows; 1. The mean concentrations of sulfamethazine in liver according to the time lapsed after oral administration of the sulfamethazine sodium were decreased from 1.27ppm at day 1 to 0.28ppm at day 4. 2. Sulfamethazine concentrations in kidney according to the time lapsed after oral administration of the sulfamethazine sodium were decreased from 0.77ppm at day 1 to 0. 12ppm at day 4. 3. The mean concentration of sulfamethazine in skeletal muscle according to the time lapsed after oral administration of the sulfamethazine sodium was at or below 0.09ppm within 4 days after withdrawl of medicated solution.
Production of prostaglandins involved in renal salt and water homeostasis is modulated by regulated expression of the inducible form of cyclooxygenase-2 (COX-2) at restricted sites in the rat kidney. COX-2 expression in the kidney is regulated by dietary salt intake, but the mechanism of its action is not fully understood. The effect of ginsenoside (GS) from Panax ginseng on basal and nitro-L-arginine suppressed nitric oxide (NO) production was studied in rat kidney. NO production was determined by conversion to [$^{14C}$]=L-citrulline from [$^{14C}$]-L-arginine both in whole kidney and three renal segments; glomerulus, cortex excluding glomerulus (cortex-) and medulla. Nitro-L-arginine (total dose of 30 mg/kg/3 days, i.p.) significantly reduced NO production in whole kidney, which was prevented by GS pretreatment (30 mg/kg/3 days, i.p.). Relative high dose of GS (120 mg/kg/4 days, i.p..) selectively increased NO production in glomerulus and cortex-. Protein content, on wet weight basis, in cortex- and glomerular DNA content were significantly reduced by GS. Our results confirm the existence of constitutive nitric oxide synthase in kidney and it seems that target nephron segment for volume expansion due to GS'NO-mediated vasodilation and for NO production stimulated by GS is cortex including glomerulus.lus.
To investigate the ultrastructural changes of kidney and clarify to a cause of its changes in lead intoxicated rats, the 0.5% lead acetate administed orally to the rats and those were sacrifled at 2 day, 1, 2, 4, 6 and 8 week after the treatment of lead acetate. Each extirpated kidney was histopathologically examined under the electron microscopy and histochemical examination was also carried out. Concomitantly, the activity of free radical metabolizing enzyme was determined. The blood levels of lead concentration showed a gradual increase from the first group reaching the plateau at the one or two week group with the slightly decreasing value throughout the whole course of the experiment. And the urinary ALA concentration showed a gradual increase from the first group to the 8 week group. In the kidney tissue of rat sacrified at 6 week, the proximal tubular cells showed dilatation of endoplasmic reticulum, mitochondrial swelling, increased numbers of secondary lysosomes and myelin figure-like residual bodies on electron microscope and oxygen free radicals are identified by histochemistry on light microscope whereas there were no differences in the activity of catalase and glutathione peroxidase between the lead acetate treated group and control group. But the activity of xanthine oxidase was more increased in lead acetate treated rats than control group. Furthermore, the superoxide dismutase activity was significantly increased in the experimental group than the control group. In conclusion, it is assumed the kidney damage in lead intoxicated rat may be induced by free radicals.
Radiological and clinical evidences indicate that hyperthermia combined with irradiation produce a significant improvement in therapeutic effect of cancer The experiences obtained from 90 rats' kidney A single dose of irradiation ranged from 6Gy, 8Gy and 10Gy was delivered on the rat's kidney. The combined therapy group had the same irradiation after hyperthermia at $42\~44^{\circ}C$ for 30 minutes. Microscopic examination and calculation of thermal enhancement ratio were carried out, and the results were as follows: 1. In the group of hyperthermia alone, there were moderate glomerular congestion and mild tubular degeneration on light microscopic examination. 2. In the group of irradiation alone, tubular degeneration was noted in 6Gy irradiation and its severity was increased along with radiation dose. 3. In the group of hyperthermia combined with irradiation, tubular degeneration and necrosis were appeared in 6Gy and 10Gy irradiation, respectively. 4. On electron microscopic examination, proximal convoluted tubular and glomerular changes in irradiation group were similar to that of combined with hyperthermia, and its severity was increased along with observation periods. 5. Thermal enhancement ratio (TER) was 1.0 after evaluation of histipathologic changes in rat's kidney, with combination therapy.

Diabetic nephropathy remains a major cause of morbidity and mortality in the diabetic population and is the leading cause of end-stage renal failure. Despite current therapeutics including intensified glycemic control and blood pressure lowering agents, renal disease continues to progress relentlessly in diabetic patients, albeit at a lower rate. Since synthetic drugs for diabetes are known to have side effects, fungal mushrooms as a natural product come into preventing the development of diabetes. Our previous report showed the hypoglycemic effect of extracellular fungal polysaccharides (EPS) in streptozotocin (STZ)-induced diabetic rats. In this study, we analyzed the differential expression patterns of rat kidney proteins from normal, STZ-induced diabetic, and EPS-treated diabetic rats, to discover diabetes-associated proteins in rat kidney. The results of proteomic analysis revealed that up to 500 protein spots were visualized, of which 291 spots were differentially expressed in the three experimental groups. Eventually, 51 spots were statistically significant and were identified by peptide mass fingerprinting. Among the differentially expressed renal proteins, 10 were increased and 16 were decreased significantly in diabetic rat kidney. The levels of different proteins, altered after diabetes induction, were returned to approximately those of the healthy rats by EPS treatment. A histopathological examination showed that EPS administration restored the impaired kidney to almost normal architecture. The study of protein expression in the normal and diabetic kidney tissues enabled us to find several diabetic nephropathy-specific proteins, such as phospholipids scramblase 3 and tropomyosin 3, which have not been mentioned yet in connection with diabetes.
