Hispidin is a secondary metabolite found in numerous medicinal mushrooms that has attracted significant attention, owing to its distinct biological effects, including antioxidant, anti-inflammatory, antitumor, and cytoprotective properties. Experiments are being carried out to study the interaction of detonation nanodiamonds (DNDs) with synthetic and natural hispidin sourced from extracts of Pholiota sp. fungus. The bioluminescence method is used to determine the adsorption/desorption properties of DNDs toward hispidin. It is found that hispidin forms strong conjugates with DNDs, and the use of various eluents does not result in a significant release of the adsorbed hispidin molecules. DND-bovine serum albumin (BSA) complex, where DNDs serve as a carrier for the protein and the latter acts as a hispidin sorbent, has been developed and applied in hispidin adsorption/desorption tests. The results support the use of the DNDs as a carrier for hispidin in medical applications. They also advocate the application of the DND-BSA complex for isolating the substance from fungal extracts.

In recent years robotics has become an important resource in engineering. Adoption of Robotics and Autonomous Systems (RAS) in activities related to ship inspections has obvious potential advantages, but also arises particular challenges, both from technical and legal viewpoints. The ROBINS project (ROBotics technology for INspection of Ships) is a collaborative project co-funded within the H2020 EU Research and Innovation programme call, aimed at filling the gap between current ship inspections approach and available robotic technology, both from technological and regulatory point of view. Main goal of the present work is to highlight how ship inspections are currently carried out by humans, how they could be improved using RAS, even if not completely autonomous for the time being, at least in selected operational scenarios and how the performances of RAS platforms can be tested to assess their effectiveness in carrying out surveys onboard. In such a framework, a testing facility aimed at assessing RAS' capabilities as well as providing suitable environment for their development has been built and it is still under development along with dedicated testing protocols, able to assess the equivalence between human and RAS inspection of ship and marine structures. The features of a testing facility where RAS can be tested and the testing protocols are presented, showing how technological and regulatory gaps are filled.

Ischemic and reperfusion injuries of the heart underlie the pathogenesis of acute myocardial infarction (AMI) and sudden cardiac death. The mortality rate is still high and is 5-7% in patients with ST-segment elevation myocardial infarction. The review is devoted to pharmacological approaches to limitation of ischemic and reperfusion injuries of the heart. The article analyzes experimental evidence and the clinical data on the effects of P2Y12 receptor antagonists on the heart's tolerance to ischemia/reperfusion in animals with coronary artery occlusion and reperfusion and also in patients with AMI. Chronic administration of ticagrelor prevented adverse remodeling of the heart. There is evidence that sphingosine-1-phosphate is the molecule that mediates the infarct-reducing effect of P2Y12 receptor antagonists. It was discussed a role of adenosine in the cardioprotective effect of ticagrelor.

A significant deficiency of most computer models used for stream floodplain analysis, is that the locations of structures impacted by flood waters, such as roads, buildings, and bridges, cannot be effectively compared to the floodplain location. The purpose of this study is the integration of the HEC River Analysis System(HEC-RAS) with ArcView geographic information system to develop a regional model for floodplain determination and representation. Also this study presents to enable two- and three-dimensional floodplain mapping and analysis in the ArcView. The methodology is applied to a Yeoju of Kyunggi-do, located in South Han River Basin. A digital terrain model is synthesized from HEC-RAS cross-sectional data and a digital elevation model of the study area. The flood plain data developed in ArcView was imported into HEC-RAS where it was combined with the field surveyed channel data in order to construct full floodplain cross sections that reflected accurate channel and overbank data for the HEC-RAS model. The flood plain limits could be expressed more accurately on ArcView by using water level data to be computed in HEC-RAS program. The computed water surface elevations and information of cross-section must be manually plotted in order to delineate floodplains. The resulting of this study provided a good representation of the general landscape and contained additional detail within the stream channel. Overall, the results of the study indicate that GIS combined with HEC-RAS is proven to be very useful and efficient for the automatic generation of flood maps, and an effective environment for floodplain mapping and analysis.

We prepared two samples of ultrafine ferrihydrite (FH) nanoparticle ensembles of quite a different origin. First is the biosynthesized sample (as a product of the vital activity of bacteria Klebsiella oxytoca (hereinafter marked as FH-bact) with a natural organic coating and negligible magnetic interparticle interactions. And the second one is the chemically synthesized ferrihydrite (hereinafter FH-chem) without any coating and high level of the interparticle interactions. The interparticle magnetic interactions have been tuned by modifying the nanoparticle surface in both samples. The coating of the FH-bact sample has been partially removed by annealing at 150℃ for 24 h (hereinafter FH-annealed). The FH-chem sample, vice versa, has been coated (1.0 g) with biocompatible polysaccharide (arabinogalactan) in an ultrasonic bath for 10 min (hereinafter FH-coated). The changes in the surface properties of nanoparticles have been controlled by XPS. According to the electron microscopy data, the modification of the nanoparticle surface does not drastically change the particle shape and size. A change in the average nanoparticle size in sample FH-annealed to 3.3 nm relative to the value in the other samples (2.6 nm) has only been observed. The estimated particle coating thickness is about 0.2-0.3 nm for samples FH-bact and FH-coated and 0.1 nm for sample FH-annealed. Mössbauer and magnetization measurements are definitely shown that the drastic change in the blocking temperature is caused by the interparticle interactions. The experimental temperature dependences of the hyperfine field hf>(T) for samples FH-bact and FH-coated have not revealed the effect of interparticle interactions. Otherwise, the interparticle interaction energy Eint estimated from the hf>(T) for samples FH-chem and FH-annealed has been found to be 121kB and 259kB, respectively.
To investigate interaction of angiotensin converting enzyme (ACE) inhibitor with local tissue renin- angiotensin system (RAS), changes in gene expression of the RAS components in various tissues in response to chronic administration of an ACE inhibitor, enalapril, were examined in Sprague-Dawley male rats. Enalapril was administered in their drinking water $(3{\sim}4\;mg/day)$ over 8 wk. Plasma and renal ACE activity increased significantly after 4 and 8 wk of enalapril treatment. Renin levels of the plasma and kidney of the enalapril-treated rats markedly increased after 4 wk and decreased thereafter, but still remained significantly higher than those of control rats. Kidney mRNA levels of renin markedly increased after 4 and 8 wk of enalapril treatment, but those of angiotensinogen and ANG II-receptor subtypes, $AT_{1A}$ and $AT_{1B}$, did not change significantly. The liver expressed genes for renin, angiotensinogen and $AT_{1A}$ receptor subtype, but $AT_{1B}$ receptor subtype mRNA was not detectable by RT-PCR. None of mRNA for these RAS components in the liver changed significantly by enalapril treatment. The hypothalamus showed mRNA expressions of renin, angiotensinogen, $AT_{1A}$ and $AT_{1B}$ receptor subtypes. $AT_{1A}$ receptor subtype mRNA was more abundant than $AT_{1B}$ receptor subtype in the hypothalamus as shown in the kidney. However, gene expression of the RAS components remained unchanged during 8-wk treatment of enalapril. In the present study, chronic ACE inhibition increased plasma and renal levels of ACE and renin, but did not affect mRNA levels of other RAS components such as angiotensinogen, ANG II receptor subtypes in the kidney. Gene levels of the RAS components in the liver and hypothalamus were not altered by chronic treatment of enalapril. These results suggest the differential expression of the RAS components in response to enalapril, and localized action and some degree of tissue specificity of enalapril.
Objectives: The aim of the present work is evaluating the special effects of Urtica Dioica and Lamium Album on the serum level of K-Ras and GSK-3 beta in diabetic rats. Methods: In the present experimental study, 32 male Wistar rats randomly divided into 4 groups (Group I: normal control rats; receiving daily PBS, Group 2: diabetic control rats; receiving single dose of streptozotocin (60 mg/kg) and daily PBS, Group 3: Diabetic rats treated with 100 mg/kg of hydroalcoholic extract of the U. dioica, Group 4: Diabetic rats treated with 100 mg/kg of hydroalcoholic extract of L. Album. Diabetes-induced by an intraperitoneal injection of streptozotocin (60 mg/ kg). On the 14 th day of treatment, the weight, fasting blood sugar (FBS) and on 28 th day blood glucose, K-Ras and GSK3 beta was measured. Results: In diabetic group blood GSK- 3 beta increase in comparison to control group (P < 0.05), also blood K-Ras decrease in the diabetic group (P < 0.05). Both extracts reduced GSK-3 beta level, however, this reduction was only statistically significant by U.dioica (P < 0.05). Compared to diabetic group, blood K-Ras level increased by both extract (P < 0.05). Also diabetes induction increase blood glucose levels and both extracts decrease its level significantly (P < 0.05).there is no significant differences among both extract effects on blood glucose, and K-Ras. Conclusion: For the first time shown that both extracts by regulating GSK-3 beta and K-Ras improve blood glucose level. More studies are needed to determine all the effects of these herbs.
Interactions between tumor cells and the extracellular matrix (ECM) strongly influence tumor development, affecting cell survival, proliferation and migration. Fibronectin, a major component of ECM, has been shown to interact with integrins especially the ${\alpha}5{\beta}1$ integrin. Cell invasion and metastasis are often associated with matrix metalloproteinases (MMPs) which are capable of digesting the different components of the ECM and basement membrane. MMP-2 is produced as a latent pro-MMP-2 (72 kDa) to be activated, resulting the 62 kDa active MMP-2. In this study, we investigated the effect of fibronectin on activation of pro-MMP-2 and the cellular invasiveness in H-Ras-transformed MCF10A human breast epithelial cells. Here we show that fibronectin induces activation of pro-MMP-2 and up-regulation of MT1-MMP and TIMP-2 in H-Ras MCF10A cells. These results demonstrate that H-Ras MCF10A cells secrete high levels of active MMP-2 when cultured with fibronectin, suggesting a possible interaction between the ECM network and H-Ras MCF10A cells to generate active MMP-2 which is important for proteolysis and ECM remodeling. Invasive and migratory abilities of H-Ras MCF10A cells were enhanced by fibronectin. Fibronectin up-regulated the expression of ${\beta}1$ integrin which may play a role in cellular responses exerted by fibronectin. Since acquisition of pro-MMP-2 activation can be associated with increased malignant progression, this study provides a mechanism for the cell surface-matrix degrading effect of fibronectin which will be crucial to breast cell invasion and migration.

Introduction: Epidermal growth factor is a single-chain polypeptide consisting of 53 amino acids with potent mitogenic activity that stimulates the proliferation of a range of normal and neoplastic cells through an interaction with its specific receptor (epidermal growth factor receptor, EGFR). This interaction plays a key role in tumor progression including the induction of tumor cell proliferation. An increased EGFR copy number have been associated with a favorable response to EGFR tyrosine kinase inhibitors therapy. In contrast, K-ras mutations tend to predict a poor response to such therapy. The aim of this study was to determine the correlation between the clinicopathological factors and the up-regulation of EGFR expression and Kras mutations in oral squamous cell carcinoma. Materials and Methods: This study examined the immunohistochemical staining of EGFR, K-ras mutation detection with peptide nucleic acid (PNA)-based real-time polymerase chain reaction (PCR) clamping in 20 specimens from 20 patients with oral squamous cell carcinoma. Results: 1. In the immunohistochemical study of poorly differentiated and invasive oral squamous cell carcinoma, a high level of EGFR staining was observed. The correlation between immunohistochemical EGFR expression and histological differentiation, as well as the tumor size of the specimens was significant (Pearson correlation analysis, significance [r] >0.5, P<0.05). 2. In PNA-based real-time PCR clamping analysis, a K-ras mutation was not detected in all specimens. Conclusion: These findings suggest that the up-regulation of the EGFR may play a role in the progression and invasion of oral squamous cell carcinoma that is, independent of a K-ras mutation.
We have explored the impact of altering the Ras-cAMP pathway on cell survival upon oxidative exposures. A hyperactivated Ras mutant of Saccharomyces cerevisiae, intrinsically more sensitive to heat shock than the wild type, was investigated with regard to oxidative stress. In this paper we report that the response of iral, ira2-deleted mutant (IR2.53) to an oxidant, such as hydrogen peroxide (H2O2) or menadione is more sensitive than that of the wild type. IR2.53 showed a dramatic decrease in survival rate when challenged with 0.1 mM H2O2 for 30 min. The greater sensitivity of IR2.53 was also noticed with treatment of 0.01 mM menadione. Prior to oxidative stresses by these oxidants, both the wild type and the mutant were preconditioned with a mild heat shock (37 ℃, 30 min), resulting in improved survivals against oxidative stresses. Rescue of IR2.53 from menadione stress by heat pretreatment was more clearly demonstrated than that from H2O2 treatment. On the other hand, no significant difference was observed between the wild type and the IR2.53 mutant in their survival rates upon paraquat treatments. These findings imply that the mechanism by which H2O2 and menadione put forth their oxidative effects may be closely associated with the cAMP-Ras pathway whereas that of paraquat is independent of the Ras pathway. Finally, the level of glutathione (GSH) was measured enzymatically as an indicator of antioxidation and compared with the survival rate. Taken all these together, this study provides an insight into a mechanism of the Ras pathway regulated by several oxidants and suggests that the Ras pathway plays a crucial role in protection of cell damage following oxidative stress.
