High-Level Production of Human Papillomavirus (HPV) Type 16 L1 in Escherichia coli

  • Human papillomavirus (HPV), a non-enveloped, double-stranded DNA tumor virus, is a primary etiological agent of cervical cancer development. As a potential tool for prophylactic vaccination, the development of virus-like particles (VLPs) containing the HPV16 L1 capsid protein is highly desired. In this study, we developed a high-level expression system of the HPV16 L1 in Escherichia coli for the purpose of VLP development. The native gene of HPV16 L1 has many rare codons that cause the early termination of translation and result in the production of truncated forms. First, we optimized the codon of the HPV16 L1 gene to the preferable codons of E. coli, and we succeeded in producing the full-size HPV16 L1 protein without early termination. Next, to find the best host for the production of HPV16 L1, we examined a total of eight E. coli strains, and E. coli BL21(DE3) with the highest yield among the strains was selected. With the selected host-vector system, we did a fed-batch cultivation in a lab-scale bioreactor. Two different feeding solutions (complex and defined feeding solutions) were examined and, when the complex feeding solution was used, a 6-fold higher production yield (4.6 g/l) was obtained compared with that with the defined feeding solution.

  • In this paper, the concept of the diversified cultural tourism industry in Korea and China was summarized and the research direction was suggested. Also, the characteristics and status of the cultural tourism industry were analyzed. As a characteristic of the cultural tourism industry in both Korea and China, it exhibited similar cultural tourism forms such as performances, theme parks, festivals, and museums. Korea had a unique cultural tourism form called Hallyu cultural tourism, and China is also a rare cultural arts complex in Korea. And has developed cultural tourism. The strength of the Korean culture and tourism industry is that it is rich in cultural resources and possesses many assets with cultural contents, and the strength of the Chinese culture and tourism industry is that it has abundant tourism resources and huge economic power. As such, Korea and China with many similarities were found to be a country with high interest and potential in the cultural tourism industry, which is growing into a high value-added industry in the future. In addition, in order to continuously and stably develop the cultural tourism industry, continuous efforts and interests and proper research must be conducted so that the two countries can coexist with each other.

Two Cases of Narcolepsy Patient Portraying a Tendency of a Dull Learning Ability and Mistaken as an Idle Student (학습부진 양상을 보이고 나태한 학생으로 오인된 기면증 환자 2 례)

  • Sleepiness is associated with many different conditions and, as a neglected topic, it can be the cause of serious psychological and social disadvantages. In the aspect of learning, additional problems may arise from poor progress in school caused by the effect of sleepiness on concentration, memory, and other cognitive functions. Narcolepsy is by no means the most common cause of excessive sleepiness. Nonetheless, it is not a rarity, especially in young people. The non-specific nature of early features of narcolepsy, combined with very limited awareness that the condition can start in various ways, leads to many misinterpretations. Misinterpretation of narcolepsy symptoms is not confined to the medical profession. Teachers may well be critical of a student with narcolepsy because of their perception of narcolepsy symptoms as laziness, poor motivation, or difficult behavior and dull learning ability. Inappropriate reactions by parents, teachers, and peers, based on misinterpretation of narcolepsy symptoms or the patient's reactions to them, make a difficult situation worse. Especially in Korea, where schooling is focused on college entrance examinations, the problem is very serious and intensified by inappropriate or delayed diagnosis and treatment. Therefore, psychiatrists should be aware that narcolepsy in young adolescents is not rare and that they need to be familiar with its clinical features in both its classic and less obvious forms. Narcolepsy should be suspected if a adolescent's excessive sleepiness can not be explained in other ways. Therefore, we report on two patients who portray the tendency of dull learning ability and are mistaken as idle students. We diagnosed narcolepsy through polysomnography and multiple sleep latency testing. We treated the students with methylphenidate and pemolin. The students showed improvement in learning ability and were able to adapt better to school.

A Basic Study on the Application of Modular Construction - Focused on the Analysis of Case Study - (모듈러 건축의 현황과 활용에 관한 기초연구 - 사례조사 분석을 중심으로 -)

  • This research was for the investigation and analysis of the illustration of modular construction application which is different use by each school facility since modular construction related examples are rare in domestic situation, and it has a limitation because of its being basic research material for generating the basic form of modular construction. The research results are as followings. First, in case of school facility from illustration investigation results, module measurement of class modulation is as similar as $3m{\times}10m$, but in resident facility the planning of more flexible plane shape can be possible since modules of 6 cases are free and various, and facade form of various types can be appeared by combination of module unit. Second, as a result of the generated characteristics in compared analysis of representative examples, school facilities were highly indicated for movability and duration reduction areas, and the flexibility, economic efficiency, and environment-friendliness was indicated low relatively. Third, the basic planning types of modular construction can be largely divided into layered type, horizontal (straightway) type, and compound type. The layered type has a short traffic line and facility system and is appropriate for the low-rise form unless separate construction method is used since it is susceptible to load. The horizontal type is advantageous for securing an opening since it has wide extent in light but has a long traffic line and facility system. Finally, the compound type can be possible for planning of various forms but needs the combination of various unit modules and traffic line and facility plan for it can be difficult.

The vascular plants of Chujado Island (Jeju-do), Korea (추자도(제주도) 지역의 관속식물)

  • To identify the flora of the vascular plant on Chujado Island in Chuja-myeon, Jeju-si, Jeju Province, Korea, eleven field surveys were conducted between April to September of 2013 and from May to September of 2018. The results of the surveys identified 558 taxa, with 97 families, 311 genera, 481 species, two subspecies, 65 varieties, and ten forms of vascular plants on Chujado Island. Of the 558 taxa identified, seven were endemic species and eight were rare plants according to the Korea Forest Service. Furthermore, 109 of the taxa identified were from floristic environments, while 62 were considered to be naturalized plants, with three of these considered to be invasive and causes of ecosystem disturbances. The highest degree of similarity among the plants from seven sites adjacent to Chujado Island was found on Saengil Island, at 51.7%.

Plant Resources of wetlands in Youngsan River Streams of Downtown in Gwangju Metropolitan City (광주광역시 도심 영산강 수계 습지의 식물자원)

  • The investigated Varcular plants in Youngsan River Streams of Downtown in Gwangju Metropolitan City consist of total 437 taxa: 2 forms, 49 varieties, 386 species, 265 genera, 91 families. Among 53 taxa of hydrophytes, emerged plants were 16taxa, floating-leaved plants were 10taxa, suvmerged plants were 10taxa, and free-floating plants were 3taxa and swamp plants were 14taxa. Based on the list of Rare and Endangered plants, 5taxa were recorded such as Hydrocharis dubia, Euryale Ferox, Penthorum chinense, Prunus yedoensis, Nymphoides coreana. And Korean endemic Plants were appeared as 5taxa: Poa annua, Forsythia koreana, Paulownia coreana, Galium koreanum, Aster koraiensis. From the specific plant species sorted by classes, class I has 10taxa, class II has 2taxa, class III has 2taxa, class IV has 3taxa, class V has 5taxa. Naturalized plant were listed as 62taxa: 15families, 44genera, 59species, 3varieties, and naturalization index was 14.19%. The ecosystem disturbance plants assigned by the Ministry of Environment, 3taxa were recorded: Paspalum distichum, Ambrosia artemisiifolia var. elatior, Solanum carilinense.

Plant Diversity of Qurecus mongolica Forest in Mt. Biryong (비룡산 신갈나무림의 식물 다양성)

  • This study was performed to investigate species diversity of understory in Mt. Biryong. The quadrat size was installed based on the result of analysis of Species-Area Curve. The vascular plants were enlisted 107 taxa: 47 families, 80 genera, 91 species, 11 varieties, 2 subspecies, and 3 forms. The three taxa such as Carex okamotoi, Philadelphus schrenkii var. schrenkii, and Weigela subsessilis were enlisted from the surveyed sites as the Korean endemic plants. Due to the list from the Korea Forestry Administration, Rhododendron micranthum was enlisted as the rare plant. The growth from was categorized followed by forbs, shrubs and trees, respectively.

Primary Hyperoxaluria in Korean Pediatric Patients

  • Background: Primary hyperoxaluria (PH), a rare inborn error of glyoxylate meta bolism causing overproduction of oxalate, is classified into three genetic subgroups: type 1-3 (PH1-PH3) caused by AGXT, GRHPR, and HOGA1 gene mutations, respectively. We performed a retrospective case series study of Korean pediatric patients with PH. Methods: In total, 11 unrelated pediatric patients were recruited and their phenotypes and genotypes were analyzed by a retrospective review of their medical records. Results: Mutational analyses revealed biallelic AGXT mutations (PH1) in nine patients and a single heterozygous GRHPR and HOGA1 mutation in one patient each. The c.33dupC was the most common AGXT mutation with an allelic frequency of 44%. The median age of onset was 3 months (range, 2 months-3 years), and eight patients with PH1 presented with end stage renal disease (ESRD). Patients with two truncating mutations showed an earlier age of onset and more frequent retinal involvement than patients with one truncating mutation. Among eight PH1 patients presenting with ESRD, five patients were treated with intensive dialysis followed by liver transplantation (n=5) with/without subsequent kidney transplantation (n=3). Conclusion: Most patients presented with severe infantile forms of PH. Patients with two truncating mutations displayed more severe phenotypes than those of patients with one truncating mutation. Sequential liver and kidney transplantation was adopted for PH1 patients presenting with ESRD. A larger nation-wide multicenter study is needed to confirm the genotype-phenotype correlations and outcomes of organ transplantation.

Autosomal Recessive Malignant Infantile Osteopetrosis Associated with a TCIRG1 Mutation: A Case Report of a Neonate Presenting with Hypocalcemia in South Korea

  • Osteopetrosis refers to a group of genetic skeletal disorders characterized by osteosclerosis and fragile bones. Osteopetrosis can be classified into autosomal dominant, autosomal recessive, or X-linked forms, which might differ in clinical characteristics and disease severity. Autosomal recessive osteopetrosis, also known as malignant osteopetrosis, has an earlier onset, more serious clinical symptoms, and is usually fatal. We encountered a 1-day-old girl who was born full-term via vaginal delivery, which was complicated by meconium-stained amniotic fluid, cephalo-pelvic disproportion, and nuchal cord. Routine neonatal care was provided, in addition to blood tests and chest radiography to screen for sepsis, as well as skull radiography to rule out head injuries. Initial blood tests revealed hypocalcemia, which persisted on follow-up tests the next day. Radiographic examinations revealed diffusely increased bone density and a "space alien" appearance of the skull. Based on radiographic and laboratory findings, the infantile form of osteopetrosis was suspected and genetic testing for identification of the responsible gene. Eventually, a heterozygous mutation of the T cell immune regulator 1, ATPase H+ transporting V0 subunit a3 (TCIRG1) gene (c.292C>T) was identified, making this the first reported case of neonatal-onset malignant osteopetrosis with TCIRG1 mutation in South Korea. Early-onset hypocalcemia is common and usually results from prematurity, fetal growth restriction, maternal diabetes, perinatal asphyxia, and physiologic hypoparathyroidism. However, if hypocalcemia persists, we recommend considering 'infantile of osteopetrosis' as a rare cause of neonatal hypocalcemia and performing radiographic examinations to establish the diagnosis.

Characteristics of Vascular Plants in Yongyangbo Wetlands

  • The objective of this study was to provide basic data for the conservation of wetland ecosystems in the Civilian Control Zone and the management of Yongyangbo wetlands in South Korea. Yongyangbo wetlands have been designated as protected areas. A field survey was conducted across five sessions between April 2019 and August of 2019. A total of 248 taxa were identified during the survey, including 72 families, 163 genera, 230 species, 4 subspecies, and 14 varieties. Their life-forms were Th (therophytes) - R5 (non-clonal form) - D4 (clitochores) - e (erect form), with a disturbance index of 33.8%. Three taxa of rare plants were detected: Silene capitata Kom. and Polygonatum stenophyllum Maxim. known to be endangered species, and Aristolochia contorta Bunge, a least-concern species. S. capitata is a legally protected species designated as a Class II endangered species in South Korea. A total of 26 taxa of naturalized plants were observed, with a naturalization index of 10.5%. There was one endemic plant taxon (Salix koriyanagi Kimura ex Goerz). In terms of floristic target species, there was one taxon in class V, one taxon in Class IV, three taxa in Class III, five taxa in Class II, and seven taxa in Class I. Three invasive alien species (Ambrosia trifida L., Ambrosia artemisiifolia L., and Humulus japonicus Siebold & Zucc) were observed. For continuous conservation of Yongyangbo Wetlands, it is necessary to remove invasive alien plants and block the inflow of non-point pollutants.
