A Study of Effective Methods for the Formation of Calluses on Trees after Surgical Operation (외구수술 후 수목의 효율적인 Callus 형성 방법에 관한 연구)

  • This study validated the effect of Vaseline and the vegetable hormone-added medicine that can influence the formation of new calluses on the injured part, with the subjects of natural monument trees and large old trees. The medicines can help in the treatment of the pored or injured parts and promote the formation of new callus on natural monument trees and large old trees, which can help vigorous growth activities and prevent any secondary infection by pathogens at the same time. It turned out that the Vaseline that is used for the protection of cambiums after surgical operations on the trees did not have any significant effect on the formation of calluses. However, since it was found that new calluses formed rapidly when applying the medicine using a vegetable hormone, the medicine is highly recommended for use. If further studies should determine proper medicines for the trees according to the kinds of trees and if the medicine is applied in proper concentrations to the injured part for rapid formation of calluses on the trees when the trees undergo surgical operations, it then should be helpful in preventing the sound part(s) of the trees from decay caused by secondary infection and treatment of the injured part.

  • In order to culture the placenta of Capsicum annuum L. in synthetic medium, the experiment was initiated to find out the proper medium for callus formation and growth rate. Linsmaier and Skoog RM 1964 media was more effective than Murashige and Skoog or white media. The addition of 2, $4-D(10^{-5}M)\;and\;kinetin(10^{-6}M)$ into Linsmaier and Skoog RM 1964 media was very effective in callus formation. The growth rate was the most rapid from 2 weeks to 3 weeks after inoculation, and then it showed slow rate.

  • We confirmed conditions for callus formation and plant regeneration of five varieties of orchardgrass (Dactylis glomerma L.). Among five varieties of orchardgrass, "Hapsung 19" expressed the highest rate for both of callus formation and plant regeneration. Otherwise, among SH (Schenk and Hildebrandt), MS (Murashige and Skoog) and N6 medium (Chu), MS and N6 medium were highest degree of efficiencies in callus formation and plant regeneration, respectively. In this study, we determined volume of hormones appended in media; $3\;mg/\;{\ell}$ ofdicamba for callus formation and $1\;mg/\;{\ell}$ of NAA (I-naphtalene acetic acid) and $5\;mg/\;{\ell}$ kinetin (6-furfurylaminopurine) for plant regeneration were appended in their media. We obtained orchardgrass plants from callus about 50~80 days after transferred to regeneration media.ation media.

  • Distraction osteogenesis was pioneered by Ilizarov in the treatment of injured extremities. Its subsequent application to Oral and Maxillofacial Surgery has opened a new chapter in the treatment of facial deformity. Careful monitoring of the period of distraction and consolidation of the callus is important and has been well described. Complications, such as infection, haematoma and premature ossification, are difficult to diagnose and can compromise the outcome after the surgery. Too slow rate of distraction results in premature fusion of cortices and too rapid information of bone 'cyst' within the callus distraction. I experienced 2 patients of congenital unilateral mandibular hypoplasia, so called Hemifacial Microsomia. After distraction surgery, I evaluated callus formation of mandibular distraction with Ultrasound Sonography during distraction and consolidation period. Plain radiography, although it enables accurate measurement of the distraction gap, did not give sufficient detail to allow assessment of early stages of bone formation, But, ultrasound monitoring could enable continuous monitoring of the distraction gap without exposure to ionizing radiation and allow detection of fine detail, which may influence manipulation of the callus.

Regeneration from Storage Root Disk Culture of Purple Sweet Potato

  • Sweet potato has low regeneration capacity, which is a serious obstacle for the fruitful production of transgenic plants. Simple and rapid regeneration method from storage root explants of purple sweet potato (Ipomoea batatas L.) was investigated. The embryogenic callus was observed from 4 cultivars and its highest rate was induced at 1 μM 2,4-D after 5 weeks of culture. Result revealed that a low concentration of 2,4-D and low light intensity was important factors for embryogenic callus formation. After subculture on medium with 5 μM ABA for 4 days, subsequently, occurred the regeneration of shoots within 4 weeks when these embryogenic callus was transferred onto the MS hormone free medium. Regenerated shoots were developed into platelets, and grown normal plants in the greenhouse. We developed a simple and quickly protocol to regenerate plantlets in storage root explants of purple sweet potato. This regeneration system will facilitate tissue culture and gene transfer research of purple sweet potato.

The Effect of Plant Growth Regulators on Callus Induction and Shoot Regeneration from the Dwarf Type Variety, Hibiscus syriacus L.var. Ggoma (왜성 변이 품종 '꼬마' 무궁화의 캘러스 유도 및 지상부 형성에 식물생장조절물질이 미치는 영향)

  • 'Ggoma' is a new Hibiscus dwarf type variety developed by gamma irradiation at the Korea Atomic Energy Research Institute (KAERI). This study was conducted to determine the best optimum cultural callus formation and shoot regeneration condition. Sterilized leaf tissues were cultured on MS (Marashige and skoog's) medium containing 3% sucrose, 0.8% agar with different concentration and combination of TDZ, 2, 4-D, KT, BA, and 2iP for 4 weeks in vitro culture. MS medium containing 2,4-D $0.1mg\;l^{-1}$ and BA $0.5mg\;l^{-1}$ were most effective on callus formation and growth. After 4 weeks, callus was transferred on BA (0.5, 1, $1.5mg\;l^{-1}$) and TDZ (0.1, 0.2, $0.3mg\;l^{-1}$) for shoot formation. The best condition for inducing the shoot from callus was BA $1.5mg\;l^{-1}$ and TDZ $0.3mg\;l^{-1}$. This result will be useful for the rapid multiplication of Hibiscus syriacus L.var. Ggoma.

Multiple Shoot Formation of Gentiana axillariflora Leveille by in Vitro Culture (큰용담의 기내증식에서 multiple shoot의 유기)

  • This study was aimed to proliferate Gentiana axillariflora Leveille which is one of the important medicinal and ornamental plants, by establishment of multiple shoot formation and embryogenesis through tissue culture technique. Callus was formed on MS (Murashige and Skoog) medium supplemented with 2,4-D, CPA, but not formed with BAP. The addition of 2,4-D 2 mg/ l into the medium was effective for callus formation and the rate of callus formation was about 90%. Somatic embryos were obtained on MS medium for two months. When callus was cultured on MS medium with combination treatment of 2,4-D 0.5 mg/ l and BAP 0.5 mg/ l, the number of embryo formed was better than that of other single or combination treatments and the total numbers of embryo a were 18.8 (number of total embryo/number of explants incubated = 753/40) at mean. Callus induction from stem and node explants was increased by addition of TDZ 2 mg/ l in the presence of 2,4-D 2 mg/ l, respectively. The best result about the differentiation of shoots was obtained on MS medium added BAP 2 mg/ l from node culture. Multiple shoots from shoot apex were induced on MS medium containing BAP 1 mg/ I and TDZ 1 mg/ l , BAP 2 mg/ l and TDZ 1 mg/ l. The number of multiple shoots per one explant was above seventy plants. It was the most effective regeneration system for rapid multiplication of Gentiana axillariflora Leveille.

Callus Induction and Somatic Embryogenesis in Suspension Culture of Rehmannia glutinosa (지황의 캘러스 유도와 현탁배양에서 체세포배 발생)

  • This study was carried out to investigate the appropriate medium and constitutionof growth regulators for somatic embryogenesis for development of rapid mass propagation system via somatic embrygenesis in Rehmennia glutinosa. Embryogenic callus formation from leaf explant was more effective when 4mg / l BA with 0.5mg / l NAA than that of treated with only auxins or cytokinins. LS medium was suitable for embryogenic callus formation. LS medium with 4mg / l BA with 0.5mg / l NAA was effective for the maintenance and proliferation of embryogenic callus. In suspension culture, addition of 1mg / l BA to LS medium was proper for somatic embryogenesis. The highest rate of shoot developement form cotyledon stage embryo was obtained in 1/2 LS medium and plantlet survived by 75% after transplanted to the soil. after 4 weeks.

Effect of Plant Growth Regulators on in vitro Micropropagation of Colored Calla Lily(Zantedeschia spp.) (유색칼라 기내 미세번식에 미치는 식물생장조절물질의 영향)

  • To establish rapid micropropagation through organogenesis from apices-derived callus or direct adventitious shoot of three calla lily cultivars(Zantedeschia spp, cv. Sunlight, cv. Chiante, cv. Pink Persuation) were cultured on Murashige and Skoog medium supplemented with different plant growth regulators. The formation rate of callus, organogenesis and in viかo tuber production among the three cultivars were tested. Callus was obtained from cvs. Sunlight, Chiante and Pink Persuasion; the best cultivar was Sunlight. Sunlight induced $53.3\%$ callus and Chiante had the highest rate of $56.7\%$ direct shoot regeneration on medium with 2.0 mg/L BA. Regeneration frequencies ranged from 20 to $70\%$ on medium with 2.0-3.0 mg/L BA. The highest percentage of regeneration and the greatest number of shoots were obtained on medium containing 3.0 mg/L BA in three cultivars. Cytokinins induced multiple shoot formation; 1.0 mg/L of 2ip, 5.0 mg/L of BA, and 1.0 m/L of BA induced 16, 14 and 12 multiple shoots in cvs. Sunlight, Chiante and Pink Persuasion, respectivly. 1.0 mg/L of IAA enhanced root growth in cvs. Sunlight and Chiante while cv. Pink Persuasion exhibited enhanced root growth at 2.0 mg/L of IBA. NAA, however, induced no change in root growth. The addition of 90 g/L sucrose enhanced in vitro tuber formation and following tuber expansion in cv. Sunlight, while 70 g/L of sucrose was effective in cvs. Chiante and Pink Persuasion.

Improvement in Clonal Propagation of Hemidesmus indicus R. Br. through Adenine Sulphate

  • A protocol has been developed for rapid large scale clonal propagation of an aromatic endangered medicinal plant, Hemidesmus indicus R. Br. with the elimination of the problems such as premature leaf fall and callus formation during caulogenesis and rhizogenesis. Multiple shoots were induced from shoot tip and nodal explants on Murashige and Skoog (MS) medium supplemented with 1 mg/L Benzylaminopurine (BAP) and 0.5 mg/L Napthaleneaceticacid (NAA). Addition of 15 mg/L adenine sulphate to the above medium checked leaf abscission completely, reduced the time required for caulogenesis and restored morphogenetic potential after several subcultures. The in vitro grown propagules were rooted in 1/2 MS medium supplemented with 2 mg/L Indolebutyric acid (IBA) +1 mg/L NAA and sucrose 0.7% (w/v). Addition of charcoal at 100 mg/L to the rooting medium quickened root initiation with a complete check on callus formation. The effect of sucrose concentration on both caulogenesis and rhizogenesis was also studied. The resultant plantlets were acclimatized and grown in fields where ninety eight percent of the rooted shoots survived and grew normally. The estimation of the secondary metabolite content in the shoots of the regenerated plant and the mother plant indicated that the concentration of the three secondary metabolites lupeol, vanillin and rutin was similar.
