Comparison of Glucosinolate Contents in Leaves and Roots of Radish (Raphanus spp.)

  • Glucosinolates (GSLs), beneficial secondary metabolites for human health are abundantly present in radish vegetable. Radish is a member of Brassicaceae family and its seed, leaf and root contain very important GSLs. The objective of this study was to determine the variation of individual and total GSL contents in leaves and roots of 44 radish (Raphanus spp.) germplasm (26 R. sativus L., 3 R. raphanistrum, and 15 R. sativus L. var. raphanistroides Makino), and compare the GSL contents between leaves and roots among three Raphanus species. Thirteen GSLs were identified, being the glucoraphasatin (GRS) and glucobrassicin (GBS) the most abundant aliphatic and indolyl GSLs in both the leaves and roots. Variation in individual and total GSL contents was found among the germplasm of three Raphanus species. The GRS content was higher in roots than that of leaves in all three Raphanus species but the GBS content was higher in leaves than roots. GRS was represented 87.0%, 92.7% and 94.7% of the total GSL in roots of R. sativus L., R. raphanistrum and R. sativus L. var. raphanistroides (Makino) germplasm, respectively. Germplasm of R. raphanistrum exhibited the highest (average, $79.5{\mu}mol/g\;dw$) total GSL with a ranged from 62.7 to $92.9{\mu}mol/g\;dw$. The germplasm IT119288, Joseonmu and IT119262 from R. sativus L., RA 504 and K046542 from R. raphanistrum, and Gyeongju-2003-32 (G2003-32) and IT302373 from R. sativus L. var. raphanistroides (Makino) had high total GSL contents and these could be good candidates for developing the functional compounds-rich varieties in radish breeding program.

  • Radish추출물을 수용성과 지용성으로 나누어 추출하여 DPPH 저해율, SOD 활성율, Catalase 활성, collagen 함량변화를 측정한 결과, DPPH 저해율과 Catalase 활성은 수용성 추출물(81.0%, 175units/mg)에서 높게 나타났고, SOD 활성율에서는 지용성 추출물(92.3%)이 높게 나타났다. 콜라겐$(21.45{\mu}g)$내의 pyridinoline의 함량은 control에 비해 수용성과 지용성 추출물을 첨가한 반응액에서 적었으며, 두 추출물을 비교해 봤을 때 큰 차이는 보이지 않았다.

Genetic Variation in Korean Populations of Wild Radish, Raphanus sativus var.hortensis f. raphanistroides (Brassicaceae)

  • Raphanus sativus L. var. hortensis f. raphanistroides (wild radish: Brassicaceae), a herbaceous perennial, occurs only on beaches in East Asia. Genetic diversity and population structure of seven Korean populations were investigated using starch gel electrophoresis. Although the Korean populatins are small, isolated with patchy distribution, the population maintain a moderate level of genetic diversity; the mean percentage fo polymorphic loci was 51.4%, mean number of alleles per locus was 1.84, and mean expected heterozygosity was 0.116. A combination of animal-outcrossing breeding system, wide geographical distribution, restricted ecological distribution, and a propensity for high fecundity may in part be explanatory factors contributing the moderate level of genetic diversity within populations. An overall excess of homozygotes relative to Hardy-Weinberg expetations (mean FISa=0.116) indicates that consanguineous mating occur within wild radish populations, leading to a family structure within a circumscribed area. Although population of wild radish experience a limited gene flow, only 5% of the total genetic variation found in Korean wild radish populations examined is due to differences among populations (mean GST=0.052). This value is considerably lower than the mean values of species with similar life history and ecological characteristics. However, significant differences were found in allele frequencies between populations for all polymorphic loci (P<0.01). It is supposed that directional selection toward genetic uniformity (similar gene frequencies) in a relatively homogenous habitat is thought to be operated among Korean wild radish populations.

20일 무(Raphanus sativus L. var. sativus)의 수경재배에 적합한 양액 및 배지의 선발 (Selection of Nutrient Solutions and Substrates for Radish (Raphanus sativus L. var. sativus) Growth)

  • 본 실험은 양액의 종류와 배지의 종류, 그리고 양액의 농도가 20일 무의 생육과 몇가지 품질에 미치는 영향을 알아보고자 실시하였다. 20일 무의 양액재배시 적절한 양액의 구명을 위하여 기존의 Cooper, Hoagland, Yamazaki양액의 비교실험을 실시한 결과, YamaBaki양액이 엽장, 엽수, 지상부와 지하부 생체중 등 전반적으로 Hoagland와 Cooper 양액보다 좋은 결과를 나타내었다. 근형지수는 모든 처리구에서 약 0.66으로 비교적 낮은 수치를 보였다. 14가지 조합의 배지를 이용하여 고형배지경을 실시하였다. 그 결과 모래 배지가 가장 균일하고도 좋은 생육의 20일 무를 배출하였고, 피트모스는 반대의 생육을 보였다. 근형지수의 경우 전반적으로 고형배지경이 순수수경재배보다 높은 수치를 나타내었다. Yamazaki 양액의 적정농도 수준을 구명을 위하여 모래를 배지로 하여 20일 무의 생육과 품질을 비교 조사한 결과, 1.0mS/cm 이상의 농도에서는 전반적으로 20일 무의 생육이 좋았으며, 특히 1.5mS/cm가 가장 좋은 생육을 보였다. 비타민 C의 함량과 티오시아네이트 함량은 1.0mS/cm이상의 처리구에서는 현저한 차이가 없었으나, 0.5mS/cm는 상당히 높은 수치를 보였다. 또한 무기물 함량의 경우는 모든 처리구에서 통계적 유의성이 없었으나, 칼륨이 1.5mS/cm에서 비교적 높은 함량을 나타내었다.

Differential Expression of Three Catalase Genes in the Small Radish (Rhaphanus sativus L. var. sativus)

  • Three catalase cDNA clones were isolated from the small radish (Raphanus sativus L.). Their nucleotide and deduced amino acid sequences showed the greatest homology to those of Arabidopsis. Genomic Southern blot analysis, using RsCat1 cDNA as a probe, showed that catalases are encoded by small multigene family in the small radish. Nondenaturing polyacrylamide gels revealed the presence of several catalase isozymes, the levels of which varied among the organs examined. The isozyme activities were assigned the individual catalase genes by Northern analysis using total RNA from different organs. The three catalase genes were differentially expressed in response to treatments such as white light, xenobiotics, osmoticum, and UV. Their expression in seedlings was controlled by the circadian clock under a light/dark cycle and/or in constant light. Interestingly, RsCat1 transcripts peaked in the morning, while those of RsCat2 and RsCat3 peaked in the early evening. Our results suggest that the RsCat enzymes are involved in defense against the oxidative stress induced by environmental changes.

  • Objectives: The objective of this study was to teach patients clinically useful external application of acne. Methods: We investigated the effects and manufacturing methods of the medical herbs published in the woman's monthly magazines according to "Zhong-Yao-Da-Ci-Dian(中藥大辭典)". Results: We investigated that Saururus chinensis Baill.(三白草), Houttuynia cordata Thunb.(魚腥草), Coix lachrymajobi L.(薏苡仁) were discovered 3 times, Brassica oleracea L.(甘藍), Raphanus sativus L.(래복), Daucus carota L. var. Sativa DC.(홍래복), Oryza sativa L.(米皮糠), Spinacia oleracea L.(파래), Cucumis sativus L.(黃爪) 2 times, and Glycyrrhiza uralensis Fisch.(甘草), Gallus gallus domesticus Braisson.(?子白), Aloe vera L.(노회), Phascolus radiatus L.(綠豆), Prunus persica Batsch.(桃葉), Lycopersicon esculentum Mill.(番茄), Taraxacum mongolicum Hand.-Mazz.(蒲公英), Prunus armeniaca L.(杏仁), Musa paradisiaca L. var. Sapientum O. ktze.(香蕉) and Malus pumila Mill.(평과) were discovered once in woman's monthly magazine during last two years. According to the effects, we classificated four groups which were defervescence(淸熱), subsidence of edema(消腫), pyorrhea(排膿) and gloss(滋潤) group. 부종(浮腫)의 식요방법(食療方法)에 관한 연구 (A study of dietetic therapy on the edema)

  • Objectives : The purpose of this research was to examine dietary therapy on the edema based on the literary findings, for clinical treatment and prevention of the edema. Methods : This Research was examined on histories, compositions, applications, and effects of dietetic treatment about 100 cases of dietary therapy of the edema from Chinese literatures. Results : 1. Various vegetables and animals including herbs, grains, vegetables, fruits, food and drink were used for the dietary therapy. 2. Methods of the preparation for use as therapeutics were decoction, pulvis, gruel, cake, tea, paste, gelatin and etc. 3. Frequently used materials were Cyprinus carpio Linnaeus, Cucumis sativus L. Coix lachrymajobi var. mayuen, Zea mays, Maydis Stigma, Citrullus vulgaris, Zingiber officinale, Cinnamomum cassia Blume, Allium scorodorpasum var. viviparum Regel, Camellia sinensis, Armeniacae amarum Semen, Phyllostachys bambusoides, Luffa cylindrica, Dioscorea batatas Decne, Panax ginseng, Brassica oleracea and Raphanus sativus. Conclusion : Though dietary therapy for the edema is not based on clinical or experimental data, but through experience. It is mostly based on Yin-Yang and five elements, visceral manifestation, channels and their collateral channels and Oriental herbal medicine theories. If we use them properly according to oriental medicine method, it will be effective on treating and preventing the edema.
