추출 방법에 따른 제주산 월동무(Raphanus sativus L.)의 향미 화합물 특성 비교 연구 (A Comparative Research of Taste and Odor Compounds Characteristics in Wintering Radish (Raphanus sativus L.) Produced in Jeju Island by Various Extraction Methods)

  • 본 연구는 전자 센서를 이용하여 다양한 추출방법에 따른 무 추출물들의 향미 특성을 조사하였다. 무의 신맛은 무 흰색 부위의 열수 추출물이 6.9의 센서 값을 나타내어 가장 높았다. 신맛은 8.0의 센서 값을 나타내어 무 초록색 부위의 콜드 브루 추출물이 가장 높았다. 감칠맛은 무 흰색 부위의 열수 추출물이 8.3의 센서 값을 나타내어 가장 높았다. 단맛은 무 초록색 부위의 콜드 브루 추출물이 6.8의 센서 값으로 가장 높은 값을 나타내었고, 쓴맛은 무 초록색 부위의 고온 가압 추출물이 7.5의 센서 값으로 가장 높은 값을 나타내었다. 전자코를 이용하여 무 추출물들에서 총 16개의 휘발성 향기성분을 확인하였다. 무 추출물 속 휘발성 향기성분들 중 methanethiol이 주요한 향기성분으로 확인되었다. 본 연구에서 확인된 결과들은 무 추출물의 향미 특성에 대한 기초 연구 자료로 활용될 수 있을 것으로 판단된다.

Antiviral Effects of Natural Products on the Inhibition of Hepatitis B Virus DNA Replication in 2.2.15 Cell Culture System

  • Evaluation of plant extracts that might inhibit hepatitis B virus (HBV) replication was performed to find potent anti-HBV agents. Eighty-five species of plants from forty-three families were tested for their anti-HBV activities using HBV-producing HepG2-derived 2.2.15 cells. The anti-HBV activity of plant extracts was measured by slot blot hybridization technique and cytotoxicity was determined by crystal violet staining procedure. All plants were extracted with methanol and the extracts were partitioned into n-hexane, ethyl acetate and aqueous layer. The ethyl acetate fractions of Rhus verniciflua $(stem:\;EC_{50},\;8.2{\mu}g/ml;\;CC_{50},\;9.4{\mu}g/ml)$, Gastrodia elata $(root:\;EC_{50},\;17.7{\mu}g/ml;\;CC_{50},\;>20{\mu}g/ml)$, Raphanus sativus $(seeds:\;EC_{50},\;17.3{\mu}g/ml;\;CC_{50},\;>20{\mu}g/ml)$, and Angelica gigas $(root:\;EC_{50},\;8.3{\mu}g/ml;\;CC_{50},\;15.6{\mu}g/ml)$ revealed the anti-HBV activity in 2.2.15 cell culture system and these fractions are under the process of further sequential fractionation by column chromatography to find the active principles against HBV.

부종(浮腫)의 식요방법(食療方法)에 관한 연구 (A study of dietetic therapy on the edema)

  • Objectives : The purpose of this research was to examine dietary therapy on the edema based on the literary findings, for clinical treatment and prevention of the edema. Methods : This Research was examined on histories, compositions, applications, and effects of dietetic treatment about 100 cases of dietary therapy of the edema from Chinese literatures. Results : 1. Various vegetables and animals including herbs, grains, vegetables, fruits, food and drink were used for the dietary therapy. 2. Methods of the preparation for use as therapeutics were decoction, pulvis, gruel, cake, tea, paste, gelatin and etc. 3. Frequently used materials were Cyprinus carpio Linnaeus, Cucumis sativus L. Coix lachrymajobi var. mayuen, Zea mays, Maydis Stigma, Citrullus vulgaris, Zingiber officinale, Cinnamomum cassia Blume, Allium scorodorpasum var. viviparum Regel, Camellia sinensis, Armeniacae amarum Semen, Phyllostachys bambusoides, Luffa cylindrica, Dioscorea batatas Decne, Panax ginseng, Brassica oleracea and Raphanus sativus. Conclusion : Though dietary therapy for the edema is not based on clinical or experimental data, but through experience. It is mostly based on Yin-Yang and five elements, visceral manifestation, channels and their collateral channels and Oriental herbal medicine theories. If we use them properly according to oriental medicine method, it will be effective on treating and preventing the edema.

cDNA Cloning and Overexpression of an Isoperoxidase Gene from Korean-Radish, Raphanus sativus L.

  • A partial cDNA encoding a Korean radish isoperoxidase was obtained from a cDNA library prepared from 9 day old radish root. In order to obtain Korean radish isoperoxidase cDNA, 5' RACE (rapid amplification cDNA end) PCR was performed and a cDNA (prxK1) encoding a complete structural protein was obtained by RT (reverse transcription)-PCR. Sequence analysis revealed that the length of the cDNA was 945 base pairs, and that of the mRNA transcript was ca. 1.6 kb. The deduced amino acid of the protein were composed of 315 amino acid residues and the protein was 92% homologous to turnip peroxidase, and 46% to 50% homologous to other known peroxidases. The 945 bp cDNA encoding Korean radish isoperoxidase was overexpressed in Escherichia coli up to approximately 9% of total cellular protein. The recombinant fusion protein exhibited 43 kDa on SDS-PAGE analysis and the activity level of the recombinant nonglycosylated protein was two fold higher in IPTG induced cell extracts than that of uninduced ones.

Peroxidase Activity Boosting by Various Nitrogenous Compounds

  • Effects of various nitrogenous compounds on the peroxidative activity of Korean radish (Rophanus sativus L.) isoperoxidase $A_1$ were examined by using anilino substrates, such as dianisidine and phenylenediamine. We also used phenolic substrates such as guaiacol, chlorogenic acid, caffeic acid, ferulic acid and esculetin. The peroxidation of dianisidine was stimulated by adenine and imidazole as much as 5 fold and 11 fold, respectively at pH 8. Moreover, about 4.8 fold and 8 fold stimulation of phenylenediamine peroxidation occurred by adenine and imidazole, respectively at pH 8. The stimulation by adenine and imidazole did not occur at the acidic pH range. The peroxidations of phenolic substrates, such as guaiacol, chlorogenic acid, caffeic acid, ferulic acid and esculetin, were not boosted greatly by any of the nitrogenous compounds tested. Notably, ammonium salt, which has been known for the excellent booster of horseradish peroxidase, did not affect the peroxidation of the Korean radish isoperoxidase $A_1$. The kinetic studies of dianisidine peroxidation with imidazole, as a model of boosting reaction, showed that neither the affinity of imidazole against dianisidine, nor the activation energy of dianisidine peroxidation changed during the activity boosting of isoperoxidase $A_1$.

Allelopathic Effects of Artemisia lavandulaefolia

  • The allelopathic effects of Artemisia lavandulaefolia were studied using several test plants and microbes. Aqueous extracts and volatile compounds of A. lavandulaefolia inhibited seed germination, seedling and root growth of the test species such as Achyranthes japonica. Lactuca sativa, Artemisia princeps var. orientalis. Oenothera odorata, Plantago asiatica. Aster yomena, Elsholtzia ciliata, and Raphanus sativus var. hortensis for. acanthiformis. The root growth of test species was more affected than shoot growth by allelochemicals of A. lavandulaefolia. Essential oil of A. lavandulaefolia had antibacterial and antifungal effects. However, the antimicrobial activity of the essential oil was dependent upon the microbial species and concentrations. Callus growth of Oryza sativa, Brassica campestris subsp. napus var. pekinensis and Achyranthes japonica was sensitive by the essential oil of A. lavandulaefolia. Twenty three chemicals were identified from A. lavandulaefolia essential oil by gas chromatography. Primary allelochemicals among them were 1, 8-cineole, 1-$\alpha$-terpineol, $\alpha$-terpinene. camphor, 2-buten-1-ol and azulene. We concluded that aqueous extract and essential oil of A. lavandulaefolia were responsible for allelopathic effects.

Paraquat 스트레스를 받는 무 (Raphanus sativus L) 유식물에서 H2O2 분해 효소에 대한 폴리아민의 효과 (Effects of polyamines on hydrogen peroxide-scavenging enzymes in radish seedling plants under paraquat stress)

  • Application of exogenous polyamines (PAs) reduced the paraquat (PQ)-induced cotyledon injuries in radish seedling plants with 1 mM spermidine (Spd) being the most effective protectant. PQ injury symptoms in the cotyledons, e.g., large accumulation of $H_2O_2$, and losses of fresh weight, chlorophyll, and proteins, were significantly alleviated. Likewise, analysis of $H_2O_2$-scavenging enzymes such as catalase (CAT) and guaiacol peroxidase (GPX) showed that pretreatment with Spd among PAs remarkably increased total CAT activity and strongly retarded PQ-induced rapid decline in total GPX activity. In a native gel assay, one CAT isozyme (CAT1) and two GPX isozymes (GPX1 and a newly synthesized GPX isozyme) proved to be more responsible for PQ tolerance, as manifested by the strong increases in their activities by Spd pretreatment. Based on these results, we can suggest that PAs (especially 1 mM Spd) may function as antioxidant protectors by invoking CAT and GPX enzymes which control the endogenous $H_2O_2$ level in radish cotyledons exposed to PQ.

  • Objectives: The objective of this study was to teach patients clinically useful external application of acne. Methods: We investigated the effects and manufacturing methods of the medical herbs published in the woman's monthly magazines according to "Zhong-Yao-Da-Ci-Dian(中藥大辭典)". Results: We investigated that Saururus chinensis Baill.(三白草), Houttuynia cordata Thunb.(魚腥草), Coix lachrymajobi L.(薏苡仁) were discovered 3 times, Brassica oleracea L.(甘藍), Raphanus sativus L.(래복), Daucus carota L. var. Sativa DC.(홍래복), Oryza sativa L.(米皮糠), Spinacia oleracea L.(파래), Cucumis sativus L.(黃爪) 2 times, and Glycyrrhiza uralensis Fisch.(甘草), Gallus gallus domesticus Braisson.(?子白), Aloe vera L.(노회), Phascolus radiatus L.(綠豆), Prunus persica Batsch.(桃葉), Lycopersicon esculentum Mill.(番茄), Taraxacum mongolicum Hand.-Mazz.(蒲公英), Prunus armeniaca L.(杏仁), Musa paradisiaca L. var. Sapientum O. ktze.(香蕉) and Malus pumila Mill.(평과) were discovered once in woman's monthly magazine during last two years. According to the effects, we classificated four groups which were defervescence(淸熱), subsidence of edema(消腫), pyorrhea(排膿) and gloss(滋潤) group. Isolation and Identification of an Autophagy-inducing Compound from Raphani Semen

  • The autophagy-lysosomal pathway is an important protein degradation system, and its dysfunction has been implicated in a number of neurodegenerative diseases, including Parkinson's disease. Raphani Semen, one of the herbs of Yeoldahanso-tang (YH), has neuroprotective effects via the autophagy pathway. The activity-guided method was used to isolate and identify the components of Raphani Semen. In this experiment, the total extract of Raphani Semen was partitioned to n-butanol, methylene chloride, and water fractions. Flow cytometry data showed that only the water fraction showed autophagy-inducing activity in vitro. Compounds 1 and 2 were isolated from this water fraction by preparative HPLC separation. The structures of compounds 1 and 2 were identified as stachyose and raffinose, respectively, by the analysis of various spectral data ($^1H$ NMR, $^{13}C$ NMR, and MS) and comparisons with standard stachyose and raffinose. Of these two compounds, raffinose showed autophagy-inducing activity in PC12 cells through the mTOR pathway.

Allelopathic Effects of Chrysanthemum boreale on Seed Germination and Seedling Growth of the Selected Plants

  • To examine the allelopathic potentials of Chrysanthemum boreale, aqueous extracts and essential oil of the plant were used in these experiments. Seed germination of the receptor species was inhibited by the aqueous extracts and the inhibitory effect was increased in proportion to the concentration of extracts. In contrast, seedling elongation showed varied results. Achyranthes japonica, Bidens bipinnata, Raphanus sativus var. hortensis for. acanthiformis, Plantago asiatica, Pimpinella brachycarpa and Lactuca sativa were inhibited by increasing concentration of the aqueous extract, while Brassica campestris subsp. napus var. pekinensis and Echinochloa crus-galli were stimulated by the extract. Dry weight was also inhibited proportionally by increasing concentration of the aqueous extract, while some species were stimulated by a lower concentration of the extract. The volatile substances of C. boreale did not affect the seed germination of receptor plants, but seedling elongation and dry weight of some species were inhibited dose-dependently. Root hair development of selected plants was inhibited along with the concentration of essential oil. The above mentioned results, therefore, confirmed that the natural substances from C. boreale had allelopathic potentials to other plants.
